1.Evaluation of the body fluid mode on Sysmex XN-2000 for detecting of tumor cells in ascites and pleural effusion

Objective To evaluate the capability for detecting of tumor cells in ascites and pleural effusion by Sysmex XN‐2000 hematology analyzer .Methods Determination of 84 samples of ascites and pleural effusion specimens with Sysmex XN‐2000 hema‐tology analyzer humoral model to analyze the study parameters highly fluorescent cells absolute value (HF‐BF # ) and percentage (HF‐BF% ) of ascites and pleural effusions screening of tumor cells ,and related data for statistical analysis .Results In 84 cases of pleural effusion and ascites samples ,cytology found 14 cases of malignant cells ,cancer cells are not found in 70 cases .14 cases of malignant cases ,HF‐BF # average of 169 /μL(0-2 001) ,HF‐BF% with an average of 29 .7% (0% -261% );70 cases of benign cases ,HF‐BF # an average of 39 /μL(0-524) ,HF‐BF% with an average of 4 .2% (0% -27 .8% ) ,both groups were statistical differences in the mean significance(P<0 .05) .ROC curve analysis with high absolute value and percentage of fluorescent cells to tumor cells detection function ,HF‐BF # area under the curve(AUC) was 0 .493 ,the area(AUC)% HF‐BF under the curve was 0 .222 .Conclusion Sysmex XN‐2000 Pattern of humoral parameters of HF‐BF # and HF‐BF% ,although the tumor cells may pro‐vide some screening information ,but ineffective ,so we believe that in our daily work can′t be over‐reliance on the instrument HF‐BF# ,HF‐BF% and other parameters should be performed on every sample smear staining ,thus improving the detection rate of ma‐lignant cells .
2.RECONSTRUCTION AND FUSION WITH MRI OF CAUDATE NUCLEUS, LENTIFORM NUCLEUS AND THALAMUS

Objective To reconstruct caudate nucleus,lentiform nucleus and thalamus three-dimensionally,fuse these neural structures with (patient's) radiological images and provide anatomical data for diagnosis and treatment of neurosurgical diseases. Methods Chinese Virtual Human dataset was segmented using image-transparency method at first and each image was smoothed by eroding and dilating, then fused neural structures with MRI,the neural structures and brain in MRI were reconstructed and displayed using surface rendering and volume rendering lastly. Results The reconstructed neural structures were smooth,natural and realistic.Their shapes and positions was clearly displayed and could be rotated, observed in any direction.On the fused images,the neural structures and tumor could be seen at the same time.Conclusion The reconstructed structures and fused images have great reference value to teaching and learning and diagnosis and treatment of neurosurgical diseases.
3.Effect of early enteral nutrition on postoperative recovery in patients with colon cancer

Objective To investigate the clinical application of early postoperative enteral nutrition in patients undergoing colon cancer operation.Methods 90 patients suffering from colon cancer were randomly divided into the enteral nutrition group,indwelling stomach tube group and the control group.The control group was given conventional treatment and the indwelling gastric tube group was received postoperative intermittent clamping of stomach tube and enteral nutrition.Moreover,patients in the enteral nutrition group were pulled out the tube on the first postoperative day while giving enteral nutrition.All patients were observed for exhaust defecation time,length of hospital stay postoperative and nutrition indicators.Results Exhaust defecation time,length of hospital stay postoperative had significant difference among three groups.Indwelling stomach tube group set minimum length of stay and first passage of flatus and defecation in the three groups [the enteral nutrition group:(50.07±11.59) h,(76.75±27.37) h,(10.1 1±1.57) d,the control group:(62.03±12.31) h,(90.67±25.64) h,(11.80±1.83) d,indwelling stomach tube group:(43.53±11.94) h,(61.17± 22.67) h,(8.70±1.53) d (P < 0.05)].The levels of hemoglobin and plasma albumin one week after operation were statistically lower than pre-operation,while that the level of these nutrition indicators were higher in indwelling stomach tube group compare to control group (all P < 0.05).The same result was observed between enteral nutrition group and control group.Whereas,the change of nutrition indicators between the enteral nutrition group and the control group was not statistical significant (P > 0.05).Conclusion Indwelling stomach tube combined with enteral nutrition is a reasonable choice for patients after colonic cancer surgery.
4.Observation of Therapeutic Effects of the Conventional Therapy plus Melitracen and Flupentixol in the Treatment of Functional Dyspepsia

OBJECTIVE: To observe the therapeutic effects of the conventional therapy plus Melitracen and Flupentixol in the treatment of functional dyspepsia(FD).METHODS: 320 FD cases were divided into gastrointestinal dyskinesis type(Type A,n=120 cases),ulceration type(Type B,n=100 cases),and nonspecific type(Type C,n=100 cases) based on their clinical symptoms.Patients of different clinical types were randomly assigned to receive conventional therapy alone or in combination with Melitracen and Flupentixol for 4 weeks.The curative effects were observed using gastrointestinal symptom rating scale(GSRS) and the total effective rate were computed.RESULTS: After treatment,the GSRS score of 3 different clinical types of FD in the combination therapy group versus the conventional therapy alone were 1.53?1.68 vs.3.10?1.78,2.02?1.63 vs.3.48?1.80 and 3.46?2.95 vs.5.18?3.21 respectively(P
5.Levels and clinical significance of serum SP-D and MBL in infants with cytomegalovirus pneumonia

Objective To investigate the levels of serum surfactant protein D (SP-D) and mannosebinding lectin (MBL) in infants with cytomegalovirus (CMV) pneumonia with the severity of disease.Methods A total of 101 hospitalized infants with CMV pneumonia were enrolled from January 2011 to December 2012.These patients were divided as the severe pneumonia group (n =48) and the mild pneumonia group (n =53) according to physical sign of lung and complication.Another 55 infants who were hospitalized in the same period with non-infectious diseases were used as the control group.Serum levels of SP-D and MBL were detected by enzyme-linked immunosorbent assay.Blood gas analyzer was used to measure arterial partial pressure of oxygen (PaO2) of the blood in severe patients.Results The mean serum SP-D levels in the severe pneumonia group [(150.08 ±52.59)ng/ml] and the mild pneumonia group [(109.67 ±31.39)ng/ml] were significantly higher than those in control group [(41.33 ± 16.42) ng/ml] (P < 0.01), and higher in the severe pneumonia group than in the mild pneumonia group (P < 0.01).However, there was no significant difference in serum MBL between all groups (P > 0.05).In severe patients, serum SP-D levels were negatively correlated with PaO2 (r =-0.565, P < 0.01).Conclusion Serum SP-D is associated with the severity of CMV pneumonia, but MBL shows no relation.The serum SP-D levels has an important clinical significance in judgment the sererity of infants with CMV pneumonia.

Objective To observe the effects of glucagon like peptide-1 (GLP-1) analogues liraglutide on expressions of nitric oxide synthase (NOS) and cyclo-oxygen-ase (COX)2 in renal medulla of type 2 diabetes rats, and the mechanism of its lowering blood pressure and promoting excretion of water and salt in kidney. Methods Type 2 diabetes model rats were generated by high-fat and high-sugar feeding for 8 weeks followed by intraperitoneal injection of streptozotocin (STZ). Subse?quently, eighteen type 2 diabetes rats were divided into two groups: liraglutide treatment group (DMT) and diabetes group (DM). Twelve normal rats were divided into two groups: liraglutide treatment wild type group (WTT) and wild type group (WT). DMT and WTT groups were given liraglutide (200μg/kg) by subcutaneous injection, DM and WT groups were given equivalent normal saline by the same way. The levels of blood glucose and blood pressure were detected at 0, 2, 4 and 6 weeks after treatment in groups of rats. Samples of urine were collected for detecting ion concentrations (K+, Na+and Cl-) af?ter treatment for six weeks. Rats were sacrificed and blood samples were collected for detecting ion concentrations (K+, Na+and Cl-). The expression levels of NOS and COX2 mRNA and protein in renal medulla were detected by real-time PCR and Western blot assay. Results After treating with liraglutide, the values of blood glucose (F=5.933, P<0.05) and blood pres?
sure (F=22.070, P<0.05) were gradually decreased in DMT group. After treatment with liraglutide for 6 weeks, the values of blood glucose (mmol/L:12.78 ± 3.82 vs. 18.75 ± 1.68) and blood pressure (mmHg:119.98 ± 4.43 vs. 136.42 ± 4.48) were signifi?cantly decreased (P<0.05) in DMT group than those of DM group (P<0.05). There were no significant differences in the concentrations of K+, Na+and Cl-between the two groups. There were higher levels of K+(mmol/L:46.55 ± 6.43 vs. 33.13 ± 9.71), Na+(mmol/L:56.33±8.83 vs. 41.20±7.25) and Cl-(mmol/L:159.81±25.06 vs. 71.44±12.99) in urine in DMT group than those of DM group (P<0.05). The mRNA levels and protein expressions of NOS and COX2 in renal medulla were significant?ly increased in DMT group than those of DM group (P<0.05). Conclusion GLP-1 analogues liraglutide may enhance the expression of COX2 by increasing the expression of NOS to excrete water and salt, and decrease blood pressure.
9.Analysis of genomic copy number variations in two sisters with primary amenorrhea and hyperandrogenism.

OBJECTIVETo analyze genomic copy number variations (CNVs) in two sisters with primary amenorrhea and hyperandrogenism.
METHODSG-banding was performed for karyotype analysis. The whole genome of the two sisters were scanned and analyzed by array-based comparative genomic hybridization (array-CGH). The results were confirmed with real-time quantitative PCR (RT-qPCR).
RESULTSNo abnormality was found by conventional G-banded chromosome analysis. Array-CGH has identified 11 identical CNVs from the sisters which, however, overlapped with CNVs reported by the Database of Genomic Variants (http://projects.tcag.ca/variation/). Therefore, they are likely to be benign. In addition, a -8.44 Mb 9p11.1-p13.1 duplication (38,561,587-47,002,387 bp, hg18) and a -80.9 kb 4q13.2 deletion (70,183,990-70,264,889 bp, hg18) were also detected in the elder and younger sister, respectively. The relationship between such CNVs and primary amenorrhea and hyperandrogenism was however uncertain. RT-qPCR results were in accordance with array-CGH.
CONCLUSIONTwo CNVs were detected in two sisters by array-CGH, for which further studies are needed to clarify their correlation with primary amenorrhea and hyperandrogenism.

10. Research progress on experimental pharmacology models of sarcopenia from the view of senescence

Sarcopenia, characterized as the progressive decrease in skeletal muscle mass, strength, and function, has been becoming one of chronic musculoskeletal diseases in aging people. In basic research studies, a reliable experimental model would be vital significance for deeply understanding pathophysiological mechanism of sarcopenia and developing novel drugs. This review provided a preliminary summary on the potential mechanisms involved in senescence-induced sarcopenia, followed by a discussion on research progress on pharmacology models based on molecular mechanism of senescence, especially from in vitro cell models and in vivo animal models.
