1.Clinical Observation of Heat-sensitive Moxibustion for Bladder Dysfunction After Poisonous Snakebite

Objective To discuss the safety and effectiveness of heat-sensitive moxibustion for bladder dysfunction after poisonous snakebite. Method Twenty-six patients with bladder dysfunction due to poisonous snakebite were intervened by heat-sensitive moxibustion, and the changes of residual urine volume and uroflowmetry parameters were analyzed by using type-B ultrasonic scanning and uroflowmetry. Result According to the ultrasonic scanning and uroflowmetry, the residual urine volume was significantly changed after treatment in the 26 subjects (P<0.05), and the maximum urine flow rate (Qmax) was also significantly different from that before treatment (P<0.05). Conclusion Heat-sensitive moxibustion can improve the residual urine volume and Qmax in bladder dysfunction after poisonous snakebite, and it’s a safe and reliable method.
2.Significance of platelet rich plasma in promoting bFGF and VEGF expression in would healing of rabbit buns

Objective To discuss the significance of platelet rich plasma in promoting bFGF and VEGF expression in would healing of rabbit buns .Methods The 24 rabbits were randomly divided into control group and experimental group .The sulfadiazine silver paint to the wound for control group and the platelet rich plasma gel evenly spread to the wound for experimental group .At 7th ,10th ,14th day ,4 rabbits of each group were randomly selected to sacrificed after anesthesiaed ,wound healing rate was compare in 2 groups ,HE staining and testimmunohistochemistry were conducted in each group .Results At 7th ,10th ,14th day ,the wound healing rate of experimental group were higher than those of control group .At 7th d ,the focal granulation ,cells and vessels inten-sive were more significant in experimental group than in control group ,at 10th day ,the wound fibroblast cells and capillary number are more significant in experimental group ,and at 14th day ,the most of fibroblasts translate into fiber cell and the capillary number decreased ,the fibroblasts proliferation was still active and fiber cell was less in the control group .At 7th ,10th day ,the expression of bFGF and VEGF were both higher ,but experimental group is obvious higher than control group(P<0 .05) ,but at 14th day ,the ex-pression of bFGF and VEGF were gradually declined and was not obvious statistics differences in the two group (P>0 .05) .Conclu-sion The platelet rich plasma could promote the big white rabbit scald wound healing ,while the main mechanism maybe the ex-pression of bFGF and VEGF increased in the early .
3.Impact of heme oxygenase 1 on monocyte chemoattractant protein 1 expression of human umbilical vascular endothelial cell in uremic milieu

Objective To investigate the influence of uremic serum on the monocyte chemoattractant protein 1 (MCP-1) expression of human umbilical vascular endothelial cells (HUVECs) in vitro and the effect of up-regulation of berne oxygenase 1 (HO-1) on the synthesis of MCP-1 of HUVECs in uremic milieu. Methods HUVECs were incubated to confluence and then preineubated with heroin and/or protoporphyrin zinc IX (ZnPP)for 6 hours.The cultures were subsequently incubated with M199 cell medium containing 10% serum of healthy people or with medium containing 10% serum of maintenance hemodialysis (MHD) patients.HO-1 protein and mRNA expression was detected by immunohistochemistry and semi-quantitative RT-PCR.MCP-1 mRNA expression was measured by semi-quantitative RT-PCR,and MCP-1 protein was quantified by ELISA. Results Up-regulated expression of MCP-1 mRNA and protein was detected in HUVECs incubated with medium containing 10% serum of MHD patients.The protein synthesis was 2.95 folds of the control.Heroin induced expression of HO-1 mRNA and protein,and concurrently inhibited the up-regulated MCP-1 expression induced by uremic serum.Such effects of heroin could be blocked by ZnPP. Conclusions Uremic serum induces the expression of MCP-1 in HUVECs.Up-regulated expresson of endothelial HO-1 induced by heroin inhibits the enhancement of MCP-1 synthesis.HO-1 may be beneficial to the alleviation of endothelial cell injury in uremic milieu.
4.Development and evaluation of whole breast irradiation with volumetric modulated arc therapy-based hybrid intensity-modulated radiotherapy after breast conserving surgery for breast cancer

Objective To develop whole breast irradiation with volumetric-modulated arc therapy (VMAT)-based hybrid intensity-modulated radiotherapy (IMRT) after breast conserving surgery for breast cancer,and to evaluate its value in clinical application.Methods Ten patients with breast cancer undergoing breast conserving surgery were enrolled.Two radiotherapy plans were designed based on hybrid fixed-beam IMRT/IMRT technique and hybrid VMAT/IMRT technique,respectively.One group received whole breast irradiation with a prescribed dose of 50 Gy in 25 fractions.The other group received whole breast irradiation with a prescribed dose of 50 Gy in 25 fractions,as well as simultaneous integrated boost to the tumor bed with 60 Gy in 25 fractions.The dosimetric parameters and delivery efficiency were compared between the two plans using paired t test.Results For patients treated with whole breast irradiation alone,there were no significant differences in conformity index and homogeneity index of target volume between the two plans (P=0.866,0.056);however,compared with the hybrid fixed-beam IMRT/IMRT technique,the hybrid VMAT/IMRT technique significantly increased the doses to organs at risk (OARs) and the number of monitor units (MUs) in the intensity-modulated field (P=0.000-0.050,P=0.002).For patients treated with whole breast irradiation with a simultaneous integrated boost to the tumor bed;however,the hybrid VMAT/IMRT technique significantly reduced the doses to the lung and spinal cord,number of MUs in intensity-modulated field,and delivery time compared with the hybrid fixed-beam IMRT/IMRT technique (P=0.004,0.001,0.000,0.000).Conclusions For patients treated with whole breast irradiation with a simultaneous integrated boost to the tumor bed,the hybrid VMAT/IMRT technique is highly promising for clinical application due to better OAR sparing and higher delivery efficiency.
5.Effects of resveratrol on proliferation of ARPE-19 cells

AIM:To investigate the effects of resveratrol ( Res) on the proliferation of ARPE-19 cells and to ex-plore the possible mechanisms.METHODS:After ARPE-19 cells were treated with Res at concentrations of 0, 50, 100, 150, 200 and 300 μmol/L for 24 h, 48 h and 72 h, the effects of Res on the proliferation of the cells were tested by CCK-8 assay.The ARPE-19 cells were treated with Res at concentrations of 0, 100, 150 and 200 μmol/L for 48 h.The effects of Res on the cell cycle and apoptosis were detected by flow cytometry with Annexin V-FITC/PI staining.The protein expression of proliferating cell nuclear antigen (PCNA) was detected by immunofluorescent assay.The mRNA expression of PCNA, P21 and P27 was determined by real-time PCR.RESULTS:The results of CCK-8 assay showed that Res inhibited the prolifera-tion of ARPE-19 cells in a time-and dose-dependent manner.The treatment with Res for 48 h resulted in an arrest of cell cycle at S phase without increasing cell apoptosis.Res inhibited the protein expression of PCNA in ARPE-19 cells.The re-sults of real-time PCR showed that Res increased the mRNA expression of P21 and P27, and decreased the mRNA expres-sion of PCNA.CONCLUSION: Res inhibits the proliferation of ARPE-19 cells and induces the cell cycle arrest at S phase.The mechanism may be related to up-regulation of P21 and P27, and down-regulation of PCNA.
6.Cervical Vestibular Evoked Myogenic Potential in Diabetes Rats

Objective To verify whether cervical vestibular evoked myogenic potential (cVEMP) can be used as an effective method to assess vestibular functions in diabetic rats ,we examined and analyzed cervical vestibular e-voked myogenic potential (cVEMP) in different duration diabetic rats and normal rats .Methods A total of 220 male SD rats were randomly divided into control group (n=20) and diabetes group (n= 200) .The diabetic rats were further divided into 4 ,6 ,8 ,10 ,and 12 week groups (40 rats each group) according to the diabetes duration . Diabetes was induced by STZ .The general situation ,ABR and cVEMP were conducted in all the groups .Results All diabetic groups appeared typical diabetic symptoms including polydipsia ,polyuria ,more food ,weight loss (P<0 .01) and blood glucose elevation (P<0 .01) .The threshold of ABR was significantly elevated (P<0 .05) in the 6 , 8 ,10 ,and 12 week diabetic group .Compared to the normal group ,the threshold of cVEMP was significantly elevat-ed in 8 week(53 .87 ± 11 .16 dB nHL) ,10 week(67 .00 ± 12 .74 dB nHL) ,12 week(67 .00 ± 9 .23 dB nHL) diabetes groups(P<0 .01) and the mean latency of P1 peak was significantly prolonged (P<0 .01) in 8 week(5 .01 ± 0 .33 ms) ,10 week(5 .37 ± 0 .45 ms) ,and 12 week(5 .39 ± 0 .24 ms) .The mean latency of N1 peak was significantly pro-longed (P<0 .01) in 10 week (8 .98 ± 0 .86 ms) ,and 12 week (9 .08 ± 0 .45 ms) compared to the control group .As to P1 - N1 amplitudes ,there was no significantly difference among all groups (P> 0 .05) .Conclusion In the cVEM P test ,the threshold increased ,the PI peak latency and N1 peak latency prolonged from the 8 th week of diabe-tes rats models .Vestibular functions were descent gradually due to different degrees of vestibular end -organ impairment . All data indicated that VEM P could be used as an early diagnostic method .
7.Correlation between risk factors of hearing lose and results of initial hearing screening in 1021 neonates.

OBJECTIVE:
To explore the risk factors of the newborns who failed initial hearing screening by analysing the distortion production otoacoustic emission (DPOAE) results of 1021 newborns with potential risk factors of hearing loss.
METHOD:
All newborns, who were born in obstetrical department and admitted in the neonatal department of the Nanfang Hospital during June 2009 to January 2012 and underwent initial hearing screening, were included in this study. Their clinical data and DPOAE results were analyzed retrospectively in order to identify the risk factors for failure of initial hearing screening in infants; cases who failed the DPOAE test were followed up by telephone interviews.
RESULT:
(1) One hundred and thirty-seven cases (13.42%) of the 1021 newborns did not pass the hearing screening. 51 cases (5.00%) did not pass the test in both ears. Meanwhile, left ear in 47 cases (4.60%) and right ear in another 39 cases (3.82%) failed the test respectively. (2) Univariate analysis showed that 14 factors had significant influence on the hearing screening results, such as birth weight, small for gestational age, multiple pregnancy, gestational age, delivery mode, oligohydramnion, oxytocin, blood sugar level of newborn, Apgar scores at 1 min, exposed prenatally to glucocorticoid, maxillofacial deformity, hypoxic-ischemic encephalopathy, neonatal respiratory distress syndrome and neonatal asphyxia (P < 0.01). (3) Multivariate Logistic regression analysis suggested that birthweight less than 1500 g, multiple pregnancy, Apgar scores of 0-4 at 1 min, exposed prenatally to glucocorticoid and maxillofacial deformity were risk factors for failure of initial hearing screening (OR were 3.132, 1.808, 2.615, 1.827 and 12.174 respectively; 95% CI were 1.466-6.691, 1.120-2.917, 1.317-5.336, 1.130-2.953 and 1.986-74.632 respectively). (4) Results of telephone interviews revealed that Apgar scores of 0-4 at 1 min would be a risk factor of language development.
CONCLUSION
Birthweight less than 1500 g, multiple pregnancy, Apgar scores of 0-4 at 1 min, exposed prenatally to glucocorticoid and maxillofacial deformity are risk factors of failure of initial hearing screening among newborns with potential hearing loss. Monitoring of the hearing condition of the infants at risk should be strengthened.

8.Porcine parvovirus nonstructural protein NS1 activates NF-κB and it involves TLR2 signaling pathway

Background:
Porcine parvovirus (PPV) is a single-stranded DNA virus that causes porcine reproductive failure. It is of critical importance to study PPV pathogenesis for the prevention and control of the disease. NS1, a PPV non-structural protein, is participated in viral DNA replication, transcriptional regulation, and cytotoxicity. Our previous research showed that PPV can activate nuclear factor kappa B (NF-κB) signaling pathway and then up-regulate the expression of interleukin (IL)-6.
Objectives:
Herein, the purpose of this study is to determine whether the non-structural protein NS1 of PPV also has the same function.
Methods:
Real-time quantitative reverse transcription polymerase chain reaction (RT-qPCR), enzyme-linked immunosorbent assay, western blot, immunofluorescence assay and small interfering RNA (siRNA) were used.
Results:
Our findings demonstrated that PPV NS1 protein can up-regulate the expression levels of IL-6 and tumor necrosis factor-alpha in a dose-dependent manner. Moreover, PPV NS1 protein was found to induce the phosphorylation of IκBα, then leading to the phosphorylation and nuclear translocation of NF-κB. In addition, the NS1 protein activated the upstream pathways of NF-κB. Meanwhile, TLR2-siRNA assay showed TLR2 plays an important role in the activation of NF-κB signaling pathway induced by PPV-NS1.
Conclusions
These findings indicated that PPV NS1 protein induced the up-regulated of IL-6 expression through activating the TLR2 and NF-κB signaling pathways. In conclusion, these findings provide a new avenue to study the innate immune mechanism of PPV infection.
9.Observation of pelvic floor muscle contractility in the second natural parturition women by intelligent ultrasound

Objective:To observe pelvic floor muscle′s contraction in women with the second and full term natural parturition by intelligent ultrasonic examination.Methods:Ninety-five postpartum women who underwent natural parturition and post-partum check (6-9 weeks after delivery) in Henan Provincal People′s Hospital were recruited in this study from Jul to Dec 2018. Fifty-four puerpera were first parturition and 41 were second parturition. The morphology of urogenital hiatus was observed during both rest and contract maneuver by real-time 3D ultrasound. Several parameters were measured by intelligent ultrasound, such as hiatal area, anteroposterior diameter, transverse diameter, and the thickness of puborectalis muscles, and the D-values of all parameters were calculated in different maneuver. Meanwhile, the strength of pelvic muscle in two groups were measured and the ultrasonic diameters and muscle strength were compared.Results:The hiatal area, anteroposterior diameter, transverse diameter, and the thickness of bilateral puborectalis muscles between first natural parturition and second natural parturition during rest maneuver were (15.92±2.76)cm 2 vs (16.65±2.63)cm 2, (40.93±5.63)mm vs (40.27±6.21)mm, (55.54±6.05)mm vs (57.92±5.27)mm, (7.03±1.51)mm vs (7.44±1.23)mm, (7.49±1.44)mm vs (7.44±1.44)mm, there was no statistical difference( P>0.05). During contract maneuver, these data were (11.76±2.62)cm 2 vs (12.09±2.78)cm 2, (37.57±5.46)mm vs (37.18±4.71)mm, (42.21±5.69)mm vs (43.03±7.13)mm, (8.92±1.54)mm vs (8.87±1.23)mm, (8.90±1.60)mm vs (9.30±2.71)mm, there was no statistical difference( P>0.05). The D-values of all diameters were (4.16±2.38)cm 2 vs (4.43±2.70)cm 2, 3(1.00, 5.00)mm vs 1(1.00, 5.25)mm, (13.33±6.07)mm vs (14.41±6.54)mm, (1.90±1.78)mm vs (1.68±1.31)mm, 1(0.48, 2.40)mm vs 1.25(0.20, 2.13)mm, there was no statistically significant difference between two groups ( P>0.05). The non-eligibility rate of typeⅠmuscle fiber strength in two groups were 65% vs 68%(35 vs 28) and typeⅡmuscle fiber strength were 74% vs 79%(40 vs 32), there was no statistically difference between two groups ( P>0.05). Conclusions:In comparison to the primary and full term natural parturition women, there is no significant reduction of the pelvic floor muscle′s contraction by intelligent ultrasund in second and full term natural parturition women without abnormal pregancy history and complications. This could provide theoretical basis for the choice of the mode of second parturition and the rahabilitation of postpartum.
