1.Evaluation of the treatment effect of psychotherapy on vitiligo

Objective To investigate the role of psychotherapy plays in the treatment of vitiligo.Methods Psychological health questionnaire(SCL-90)was used in the study to evaluate the psychological well being of 120 vitiligo patients.66 subjects suffering from psychological problems were divided randomly into groups Ⅰand Ⅱ while 54 subjects free of psychological problems randomly into groups Ⅲ and Ⅳ.Psychotherapy was applied together with drug and phototherapy to those subjects from groups Ⅰ and Ⅲ while groups Ⅱ and Ⅳ having no psychotherapy accompanied.After 3 months of treatment,evaluation was done on the treatment effect with psychological testing once again.The correlation between the improvement of psychological conditions of the subjects and the lesion development was then evaluated by"P"value of the four groups.Results Significant difference was achieved between group Ⅰ and group I1I(P<0.05).There was no significant difference between groupsⅢand IV(P>0.05).There was significant difference between groups Ⅰ and Ⅱ.There was no significant difference between groups Ⅱ and Ⅳ.Conclusions The psychotherapy is helpful in improving the early treatment effect and quality of life in vitiligo patients.
2.Anoikis-suppression and invasion induced by tyrosine kinase receptor B in OVCAR3 ovarian cancer cells

Objective To study the relationship between tyrosine kinase receptor B (TrkB)expression and anoikis-suppression and invasion in OVCAR3 ovarian cancer ceils. Methods The expression of TrkB mRNA in OVCAR3 ovarian cancer cells under two culture conditions :adhesive cells and ceil-spheroids were evaluated by RT-PCR and real-time PCR.The relationship between TrkB expression and anoikis-suppression of OVCAR3 ovarian cancer ceils was examined by RNA interference (RNAi) technic,anchorage independent culture and fluorescence-activated ceil sorting analysis.The difference in invasion and metastatic ability of OVCAR3 cells under two culture conditions and with or without TrkB silenced by small interfering RNA (siRNA) was investigated by matrigel invasion assay and in vivo studies.Results The expression of TrkB mRNA was highest in OVCAR3 ovarian cancer ceils,0.0240 ~ 0.0017,compared with the other three cell lines,0.0030±0.0006,0.0027±0.0009 and 0.0087±0.0003 respectively,andthe expression in OVCAR3 multicellular spheroids was significantly higher than that in ceils under monolayer adhesive culture,0.0437±0.0021 versus 0.0240±0.0017 (P<0.01) . TrkB mediated anoikissuppression in OVCAR3 ovarian cancer ceils.OVCAR3 multiceilular spheroids had a higher invasion ability than OVCAR3 cells under monolayer adhesive culture,and the penetrating cells of the two groups were 71.8± 0.8 and 47.7±0.8 respectively (P<0.01 ).The metastatic ability of OVCAR3 cells was attenuated when TrkB was silenced,and the volume of the tumors developed by OVCAR3 adhesive cells and OVCAR3 adhesive cells with TrkB silenced were (16.3±4.7) mm3 and(6.0±1.4) mm3 respectively (P<0.01).Conclusion As an anoikis-suppressur,TrkB may increase the invasion and metastasis of OVCAR3 ovarian cancer cells.
3.Effects of Pediatric Early Warning Score nursing record template on pediatric pneumonia

Objective? To explore the effects of Pediatric Early Warning Score (PEWS) nursing record template on pediatric pneumonia. Methods? The medical records of totally 40 pediatric pneumonia patients admitted between June and December, 2017 in the Department of Respiratory Medicine of a Class Ⅲ Grade A children's hospital were selected into the observation group, while the medical records of another 40 pediatric pneumonia patients admitted between June and December 2016 were selected into the control group by random sampling. The medical records of the control group were written in accordance with the Medical Record Writing Standards of Anhui Province (Version 2013), whereas the medical records of the observation group were written using the PEWS nursing record template. The nursing document quality scale of the two groups was viewed, and objectivity, expression of the specialty and misuse of medical terms were compared between the nursing records of the two groups; the time used to write the nursing records by nurses with different length of service was compared. Results? Totally 35 errors in objectivity, expression of the specialty and use of medical terms were found in the nursing records of the observation group, while those of the control group was 211, and there was statistically significant difference between the two groups (χ2=12.81, P<0.05). Conclusions? The use of PEWS nursing record template can reduce the errors in the nursing records of pediatric inpatients, improve the quality of nursing documents, and save the time of writing nursing records for nurses, which is worth promoting in clinical practice.
4.Interaction of olfactory ensheathing cells with nerve repairing scaffolds.

OBJECTIVE:
To investigate a new way to yield plenty of high purity olfactory ensheathing cells (OECs) and its biocompatibility with appropriate scaffolds.
METHODS:
OECs were prepared from neonatal Wister rats and co-cultured with poly [LA-co-(Glc-alt-Lys)] (PLGL). Its contact angle, adherent rate, and activity rate were tested.
RESULTS:
The contact angle of poly (D, L-lactic acid) (PDLLA) (84.5 degree+/-1.5 degree) was significantly higher than that of PLGL (52.6 degree+/-0.8 degree), the adherent rate of PLGL (80%) was significantly higher than that of the PDLLA (57%), and the activity rate of PLGL (88%) was much higher than that of the PDLLA (76%).
CONCLUSION
PLGL possesses better hydrophilicity and biocompatibility than PDLLA, and it can provide a better cell growth circumstance which is helpful for the effective treatment of nerve injury.

5.Analysis of prognostic factors of the first relapsed/refractory primary central nervous system lymphoma

Objective: To analyze the clinical characteristics of patients with relapsed/refractory primary central nervous system lym-phoma (PCNSL) and to explore the factors that influence the prognosis, in order to provide evidence for the clinical diagnosis and treat-ment. Methods: Sixty-four patients with relapsed/refractory PCNSL diagnosed from October 2006 to August 2015 were selected. The clinical features, treatment plans, and laboratory examination data were retrospectively analyzed. Cox regression was used for multi-variate analysis. Results: Univariate and multivariate analyses showed that progression-free survival of first time (PFS1)≤1 year and Kar-nofsky performance status (KPS) score<70 points were independent prognostic factors in patients with first relapsed/refractory PCNSL. The median PFS2 and overall survival of second time (OS2) were 19 and 21 months, respectively, in patients with PFS1≥1 year, where-as the median progression free survival of second time (mPFS2) and OS2 were 10 and 14 months, respectively, in patients with PFS1<1 year. The median PFS2 (mPFS2) in patients with first relapse/refractory KPS score≥70 points and those with KPS score<70 points were 40 and 10 months, respectively, and the median OS2 were 43 and 12 months, respectively. The median PFS for the methotrexate (MTX) and non-MTX groups was 18 and 10 months, respectively. Multivariate analysis showed that the salvage therapy was a relevant factor influencing the patient's PFS. However, univariate analysis showed that the median OS2 in the MTX and non-MTX groups was 23 and 12 months, respectively, with significant difference but without any correlation with prognosis. Conclusions: progression-free sur-vival (PFS)≤1 year and KPS score<70 were independent prognostic factors in patients with first relapsed/refractory PCNSL. Patients with relapsed/refractory PCNSL who continuously received high-dose MTX-based treatment may have improved long-term treatment outcomes.
6.Preparation and In Vitro Characterization of Gelatin Methacrylate for Corneal Tissue Engineering

BACKGROUND:
Corneal disease is second only to cataract considered as the leading cause of blindness in the world, with high morbidity. Construction of corneal substitutes In Vitro by tissue engineering technology to achieve corneal regeneration has become a research hotspot in recent years. We conducted in-depth research on the biocompatibility, physicochemical and mechanical properties of rat bone marrow mesenchymal stem cells (rBM-MSCs)-seeded gelatin methacrylate (GelMA) as a bioengineered cornea.
METHODS:
Four kinds of GelMA with different concentrations (7, 10, 15 and 30%) were prepared, and their physicchemical, optical properties, and biocompatibility with rBM-MSCs were characterized. MTT, live/dead staining, cell morphology, immunofluorescence staining and gene expression of keratocyte markers were performed.
RESULTS:
7%GelMA hydrogel had higher equilibrium water content and porosity, better optical properties and hydrophilicity. In addition, it is more beneficial to the growth and proliferation of rBM-MSCs. However, the 30%GelMA hydrogel had the best mechanical properties, and could be more conducive to promote the differentiation of rBM-MSCs into keratocyte-like cells.
CONCLUSION
As a natural biological scaffold, GelMA hydrogel has good biocompatibility. And it has the ability to promote the differentiation of rBM-MSCs into keratocyte-like cells, which laid a theoretical and experimental foundation for further tissue-engineered corneal stromal transplantation, and provided a new idea for the source of seeded cells in corneal tissue engineering.
7.Influencing factors of cancer related fatigue in patients with cancer

8.Influencing factors of nurses' work engagement in cancer hospitals: the mediating effect of work acceptance

Objective:To understand the current status of nurses' work engagement in cancer hospitals, explore its influencing factors and the mediating effect of work acceptance between hospital magnetic level and work engagement.Methods:In September 2019, 420 nurses from a ClassⅢ Grade A cancer hospital were selected as the research object by convenient sampling. The General Information Questionnaire, the Chinese version of the Essentials of Magnetism ScaleⅡ (EOMⅡ) , Work-related Acceptance and Action Questionnaire (WAAQ) and the Utrecht Work Engagement Scale (UWES) were used to conduct a cross-sectional survey of nurses. Single factor analysis, correlation analysis and structural equation model were used to analyze the relationship among hospital magnetic level, work acceptance and work engagement of nurse. A total of 420 questionnaires were distributed and 408 valid questionnaires were collected.Results:Among 408 nurses, the scores of EOM Ⅱ, WAAQ and UWES were (141.53±22.73) , (33.79±8.38) and (45.11±17.27) respectively. Work engagement was positively correlated with hospital magnetic level and work acceptance with statistical differences ( r=0.549, 0.564; P<0.01) . The hospital magnetic level had a positive predictive effect on work acceptance (β=0.39) and work engagement (β=0.40) , and work acceptance had a positive predictive effect on work engagement (β=0.47) . Work acceptance had a partial mediating effect between hospital magnetic level and work engagement, and the mediating effect accounted for 31.49% of the total effect. Conclusions:Work acceptance plays a part of the mediating role between hospital magnetic level and work engagement. Nursing managers can create a magnetic nursing work environment to promote nurses' work acceptance and increase their work engagement.
9.Effect of hepatitis B virus X in inhibiting the apoptosis of trophoblastic cells and its potential mechanism

【Objective】 To investigate the relationship between hepatitis B virus X (HBx) protein and EGFR promoter, and the role of HBx protein in activating EGFR/PI3K/p-Akt signaling pathway and inhibiting apoptosis. 【Methods】 EGFR promoter plasmids were constructed and the relationship between HBx and EGFR promoters was characterized using a luciferase reporter assay. EGFR-overexpressing trophoblast cells were constructed, and EGFR expression in the overexpressing cells was knocked down using EGFR shRNA. The expression and localization of EGFR/PI3K/p-Akt were detected by Western blotting and confocal laser microscopy. Cell apoptosis was analyzed using flow cytometry. HBV plasmids carrying either full-length HBx or HBx with a deletion mutation (ΔHBx) and HBx plasmids were transfected into two types of trophoblast cells; HBx and PI3K/p-Akt protein expressions were detected by Western blotting. Cell apoptosis was analyzed using flow cytometry. 【Results】 Co-transfection of HBx and EGFR promoter plasmids in JEG-3 and HTR-8/Svneo cells significantly elevated the expression of EGFR promoter driven luciferase compared with the control group (P<0.01). In EGFR-overexpressing cells, the expression of PI3K/p-Akt was significantly increased (P<0.01), whereas the apoptosis rate was significantly decreased for JEG-3 cells and HTR-8/Svneo cells (both P<0.01). These results were reversed in the EGFR-knock down group. When the intracellular HBx protein was expressed in JEG-3 and HTR-8 cells, PI3K/p-Akt protein expression was significantly increased (both P<0.05), and the proportion of apoptosis was significantly decreased (both P<0.05). 【Conclusion】 In placental trophoblast cells, HBx protein activates the expression of EGFR by acting on the EGFR promoter, and inhibits the apoptosis of trophoblast cells via the downstream EGFR/PI3K/p-Akt signaling pathway.
10.Nanoparticles (NPs)-mediated Siglec15 silencing and macrophage repolarization for enhanced cancer immunotherapy.

T cell infiltration and proliferation in tumor tissues are the main factors that significantly affect the therapeutic outcomes of cancer immunotherapy. Emerging evidence has shown that interferon-gamma (IFNγ) could enhance CXCL9 secretion from macrophages to recruit T cells, but Siglec15 expressed on TAMs can attenuate T cell proliferation. Therefore, targeted regulation of macrophage function could be a promising strategy to enhance cancer immunotherapy via concurrently promoting the infiltration and proliferation of T cells in tumor tissues. We herein developed reduction-responsive nanoparticles (NPs) made with poly (disulfide amide) (PDSA) and lipid-poly (ethylene glycol) (lipid-PEG) for systemic delivery of Siglec15 siRNA (siSiglec15) and IFNγ for enhanced cancer immunotherapy. After intravenous administration, these cargo-loaded could highly accumulate in the tumor tissues and be efficiently internalized by tumor-associated macrophages (TAMs). With the highly concentrated glutathione (GSH) in the cytoplasm to destroy the nanostructure, the loaded IFNγ and siSiglec15 could be rapidly released, which could respectively repolarize macrophage phenotype to enhance CXCL9 secretion for T cell infiltration and silence Siglec15 expression to promote T cell proliferation, leading to significant inhibition of hepatocellular carcinoma (HCC) growth when combining with the immune checkpoint inhibitor. The strategy developed herein could be used as an effective tool to enhance cancer immunotherapy.
