1.Research on the application rules of Chinese herbs of National TCM Master Li Jiren on treating atrophy diseases with deficiency syndrome based on data mining

Objective:To study the application of herbs of National TCM Master Li Jiren on treating atrophy with deficiency syndrome. Methods:By collecting 195 cases of atrophy-disease treated by Professor Li in Out-patient Medical Record System dated from January 2013 to December 2018, and FP-Growth computing paradigm was used to analyze frequency, FP-Growth enhanced computing paradigm was used to analyze the frequencyand association of symptoms and herbal pairs. Professor Li’s experience was accordingly summurized with clinic situation and Traditional Chinese Medicine theory. Results:There are 121 main symptoms among 195 selected cases, and amyotrophy (79 times), decreased muscle strength (69 times), difficulty in walking (52 times), weakness of upper extremities (43 times) and poor sleeping quality (33 times) rank the top 5 symptom. In terms of tongue manifestations, the color pink, pale, red and purple are the most; while in terms of pulses, weak, thread, deep and stringy were frequent. A total of 159 sorts of herbs were collected and Astragali Radix (140 times), Angelicae Sinensis Radix (103 times), Cistanches Herba (93 times), Psoraleae Fructus (87 times), Morindae Officinalis Radix (81 times) were the top 5. The herbs used are mainly with the function of tonifying deficiency, dispelling wind and dampness promoting blood circulation and removing blood stasisand. most of the herbs belong to liver, kidney and spleen meridians with warm character, sweet and bitter in taste. Association analysis between high-frequency herbs indicated that Astragali Radix is the core of most formulas, which conformed to Li’ s clinical experience. Three prescriptions were also obtained through cluster analysis. Conclusion:Professor Li treated atrophy diseases with deficiency syndrome by the unique idea of strengthening primordial qi and consolidating body resistance of Xin’an Medicine school. On the ground of using herbs to tonify, synergy of herbs, which were able todispel pathogenic wind, dredge channels and promote blood circulation for removing blood stasis, was achieved based on differentiation.

Stem cells under the controlled condition may directionally differentiate into function cells in organism,formed various types of the organization and the organ.This article narrated the concept and classification of stem cells and regeneration medicine,discussed the application of stem cells to diabetes,myocardial cell damage disease,and neurotrosis disease,analyzed and summarized the problems of stem cells and regeneration medicine.The analysis processing material demonstrated that stem cells have been widely applied to clinical trials addressing cardiovascular system disease,nervous system disease,skeleton muscle related disease,diabetes and so on.This has obtained preliminary curative effect and has demonstrated the widespread clinical practice prospect.
3.Effects of living donor liver transplantation on postoperative delirium, cognitive impairment and prognosis in children with biliary atresia

Objective:To explore the changes of neuron-specific enolase (NSE) and S-100β protein (S-100β) during perioperative period in infants undergoing living liver transplantation and examine the effect of brain injury.Methods:From January 2015 to January 2016 in Department of Anesthesiology First Central Clinical College Tianjin Medical University, study group was composed of forty infants of congenital biliary atresia with an age range of (4-12) months, a body weight of (4-10) kg and American Society of Anesthesiologists (ASA) class Ⅲ/Ⅳ. Another 40 infants undergoing general surgery were selected as control group. In study group, blood samples were harvested from central vein pre-operation (T0), before skin incision (T1), 30 min after anhepatic phase (T2), 1 h of neohepatic phase (T3) and 24h after hepato-reperfusion (T4). In control group, blood samples were collected at pre-operation (T0) and 24 h post-operation (T4). Serum levels of S-100β, NSE, heart rate (HR), mean arterial blood pressure (MAP), central venous pressure (CVP) and bispectral index (BIS) were monitored at T1-4 and end of surgery. All children were assessed by Bayley Scale of Infant Development (BSID) at Day 1 pre-operation and 2/4 weeks post-operation for observing mental and motor development status. The results were described with mental development index (MDI) and psychomotor development index (PDI). Pediatric anesthesia emergence delirium (PAED) was employed for evaluating the severity of delirium during the recovery stage at 30 min and 2/4h post-extubation.Results:In study group, serum levels of S-100β and NSE changed significantly during non-hepatic and neohepatic reperfusion phases. After inferior vena cava occlusion, serum concentrations of S-100β and NSE spiked ( P<0.05) and gradually recovered during neohepatic reperfusion period ( P<0.05). No significant inter-group difference existed in serum S-100β or NSE at T4 ( P>0.05). In study group, as compared with Day 1 pre-operation, MDI/PDI decreased at Week 2 post-operation ( P<0.05) and increased from Month 1 post-operation ( P<0.05). Both MDI and PDI were lower than control group before and at Week 2 post-operation ( P<0.05). MDI/PDI of study group basically reached the preoperative level at Month 1 post-operation ( P<0.05). In control group, no significant difference existed in MDI/PDI at Day 1 pre-operation and Week 2/4 post-operation ( P>0.05). In study group, the delirium rate was up to 30% post-extubation and decreased at 2/4h post-extubation. In control group, the incidence of delirium was low at 30 min and 2/4h post-extubation ( P<0.05). Conclusions:Perioperative evaluations of serum levels of NSE and S-100β are significant for predicting the postoperative onsets of delirium and cognitive impairment in children with living donor liver transplantation.
4.Content Determination of Loxoprofen Silver in Loxoprofen Silver Cream by HPLC

OBJECTIVE:To establish a method for the content determination of loxoprofen silver in Loxoprofen silver cream, and provide reference for the quality control of the preparation. METHODS:HPLC was performed on the column of Diamonsil C18 with mobile phase of methanol-water-glacial acetic acid-triethylamine(55:45:0.1:0.1,V/V/V/V)at a flow rate of 1.0 mL/min,de-tection wavelength was 223 nm,column temperature was 30 ℃,and the injection volume was 20 μL. RESULTS:The linear range of loxoprofen silver was 6.53-130.7μg/mL(r=0.9999);the limit of quantitation was 0.253μg/mL,limit of detection was 0.076μg/mL;RSDs of precision,stability and reproducibility tests were lower than 2%;recovery was 96.79%-103.68%(RSD=2.23%, n=9). CONCLUSIONS:The method is simple,accurate and reproducible,and can be used for the content determination of Loxo-profen silver cream.
5.Study on quality control of ELISA method for screening TORCH infection

Objective Onto investigate the indoor quality control method for qualitatively detecting the laboratory indicators of TORCH infection (rubella virus IgG ,cytomegalovirus IgG and IgM ,toxoplasma IgG and IgM ) .Methods The statistical method , normal distribution data ,ratio and standard deviation of positive rate detected by the ELISA method were adopted ,1+2s was set as the out of control rules ,the semi Lerey‐Jennings quality control chart was drawn;the direct probability calculation method was a‐dopted for the non‐normal distribution data and small probability event .The testing data of 57 batches were retrospectively ana‐lyzed .Results The positive rate of rubella virus IgG was 86 .66% ,cytomegalovirus IgG/IgM positive rates were 98 .87% and 0 .13% ,toxoplasma gondii IgG/IgM positive rates were 2 .43% and 1 .71% ,the data of 151 ,3 ,5 ,176 ,27 samples had the critical value range of five indicators .The number of out of control was once for cytomegalovirus IgG ,once and 4 times for Toxoplasma gondii IgG/IgM .Conclusion The indoor quality control for the ELISA qualitative detection of TORCH infection can adopt the data of daily detection positive rate or negative rate for monitoring the false positive .The critical value range of specimens should be fur‐ther conducted the recheck or confirmation experiment .
6.Current progress of mesenchymal stem cells in tissue engineering trachea

Tissue engineering can regenerate damaged tissues and restore the biological functions by cell or tissue reconstruction,and is becoming a promising method for trachea replacement.Seed cells,cell growth factors and tracheal scaffolds are the three major elements of tissue engineering trachea,as a result researchers have paid a lot of attention to find ideal seed cells.Mesenchymal stem cells (MSCs) are a kind of stem cells with high self-renewal ability and muhi-directional differentiation potential.MSCs are widely distributed in bone marrow,umbilical cord,adipose tissue,myocardial tissue,brain,muscle and skin,and can differentiate into a variety of cells,including osteocytes,chondrocytes,adipocytes and neurocytes.MSCs have the characteristics of high proliferation ability,wide differentiation range and immunomodulatory function,which can be used to repair damaged tissue.These advantages make the MSCs an ideal candidate of seed cells for tissue engineering trachea.This review mainly summarized the application of MSCs in tissue engineering trachea.
7.Cellular biocompatibility and biomechanical properties of 3D printed tracheal graft

Objective To prepare 3D printed tracheal graft and investigate its cellular biocompatibility and biomechanical properties.Methods Bone marrow was isolated from tibial plateau of young New Zealand white rabbit,and bone mesenchymal stem cells (BMSCs) were obtained by whole bone marrow culture method and adherent purification method.Biomechanical test was performed for 3D printed trachea graft.After co-cultured of 3D printed trachea graft and BMSCs,cell morphology was observed and the proliferation index of the cells on 3D printed trachea graft was quantified using sulforhodamine B (SRB) assay.Results 3D printed trachea graft showed excellent biomechanical properties.Cell morphology was normal and cells grew well after co-culture with 3D printed trachea graft.The SRB assay indicated good proliferation of BMSCs on 3D printed trachea graft.Conclusions 3D printed trachea graft shows favorable cellular biocompatibility and biomechanical properties,and therefore can be used as a scaffold material for tissue-engineered trachea.
8.Blood glucose recovery following allogenetic hematopoietic stem cell transplantation in patients with hematological diseases complicated with diabetes mellitus

BACKGROUND:Recent studies have shown that the large-dose regular insulin therapy used to control blood glucose levels can cause 50%of patients suffering from vascular, optic nerve and kidney complications. Previous results from authors exhibit that when al ogeneic hematopoietic stem celltransplantation is applied for treatment of leukemia, diabetic symptoms in patients disappear. Dose it prompt that al ogeneic hematopoietic stem celltransplantation is an effective therapy for treatment of diabetes mel itus? OBJECTIVE:To explore the feasibility of hematopoietic stem celltransplantation for treatment of diabetes mel itus. METHODS:A retrospective analysis was done regarding the data of patients with hematological diseases complicated with diabetes mel itus who underwent al ogenetic hematopoietic stem celltransplantation. Four patients with acute lymphocyte leukemia, chronic myelogenous leukemia, aplastic anemia, and myolodysplastic syndromes, respectively, were complicated with diabetes mel itus. Conditioning regimen was cyclophosphamide+total body irradiation protocol. Cyclosporin A and short-term methotrexate were used for graft-versus-host disease prophylaxis. Blood glucose was control ed by oral hypoglycemic drugs or insulin injections before transplantation. RESULTS AND CONCLUSION:Al the four patients were successful y engrafted. Fasting glucose level of the four patients recovered at 4-6 months after hematopoietic stem celltransplantation (without hypoglycemic drugs). One patient died of leukemia relapse after 12 months of hematopoietic stem celltransplantation. The other three patients had disease-free survival until the time of fol ow-up.
9.Effects of ulinastatin on myocardial injury in patients undergoing live donor liver transplantation

Objective To investigate the effects of ulinastatin on the myocardial injury in patients undergoing live donor liver transplantation.Methods Forty patients (AHA classification grade A or B),aged 40-64 yr,with a body mass index of 18-25 kg/m2,scheduled for live donor liver transplantation,were randomly divided into 2 groups ( n =20 each):control group (group C) and ulinastatin group (group U).Anesthesia was induced with midazolam,sufentanil,and cisatracurium besilate.The patients were tracheal intubated and mechanically ventilated.Ulinastatin 300 000 IU in 100 ml of normal saline was infused intravenously over 30 min after anesthesia induction and then the infusion was repeated at 4 h interval until the end of operation in group U,while the equal volume of normal saline was given in group C.Blood samples were taken from the central vein immediately before skin incision (T0,baseline),at 30 min of anhepatic phase (T1),at 30 min of neohepatic phase (T2),and at 0,4 and 24 h after operation (T3-5) for determination of the concentrations of serum cardiac troponin Ⅰ (cTnI),creatine kinase-MB (CK-MB) and N-terminal pro-brain natriuretic peptide (NT-proBNP).The changing rates of cTnI and CK-MB at T1-5 were calculated.The use of cardiovascular drugs and cardiovsscular accidents were recorded during operation.Results The serum cTnI,CK-MB and NT-proBNP concentrations were significantly higher at T2-5 than at T0 in the two groups ( P < 0.05).Compared with group C,the serum cTnI,CK- MB and NT-proBNP concentrations at T2-5 were significantly deceased in group U ( P < 0.05).The maximal changing rates of cTnI,CK-MB and NT-proBNP concentrations were 4.71 ± 1.62,6.85 ± 1.53 and 4.96 ± 1.23 respectively in group C,decreased to 3.26 ± 1.51,4.56 ± 1.62 and 3.67 ± 1.02 respectively in group U.There was no significant difference in the incidence of cardiovascular accidents and the use of dopamine between the two groups.Conclusion Intravenous infusion of ulinastatin can attenuate the myocardial injury to some extent in patients undergoing live donor liver transplantation.
10.Effect of propofol on brain injury during hepatic ischemia-reperfusion in preadolescent mice

Objective To evaluate the effect of propofol on brain injury during hepatic ischemiareperfusion (I/R) in preadolescent mice.Methods Forty-eight healthy male C57BL/6 mice.aged 2 weeks,weighing 6-9 g,were allocated to one of 3 groups (n=16 each) using a random nunber table:sham operation group (group S),hepatic I/R group (group I/R) and propofol group (group P).Hepatic I/R was produced by occlusion of the blood supply to the median and left lobes of the liver for 60 min followed by reperfusion in anesthetized mice.In group P,1% propofol 30 mg/kg was intraperitoneally injected at 30 min before skin incision,while the equal volume of normal saline was given instead in S and I/R groups.Blood samples were collected and brains were removed at 24 h of reperfusion,and hippocampi were then isolated.The levels of tumor necrosis factor-alpha (TNF-α) and interleukin-1beta (IL-1β) in hippocampi and levels of TNF-α,IL-1β,S-100β protein and neuron-specific enolase (NSE) in serum were determined by enzyme-linked immunosorbent assay.The pathological changes of hippocampi were examined with a light microscope,the apoptosis in hippocampal cells was measured using TUNEL,and the apoptotic index was calculated.Results Compared with group S,the TNF-α and IL-1β levels in serum and hippocampi,levels of S-100β protein and NSE in serum and apoptotic index were significantly increased (P<0.01),and the pathological changes of hippocampi were aggravated in I/R and P groups.Compared with group I/R,the TNF-α and IL-1β levels in serum and hippocampi,levels of S-100β protein and NSE in serum and apoptotic index were significantly decreased (P<0.05),and the pathological changes of hippocampi were attenuated in group P.Conclusion Propofol reduces brain injury induced by hepatic I/R,and the mechanism may be related to inhibition of systemic and central inflammatory responses of preadolescent mice.
