1.Expression and distribution of aquaporin 3 during early embryonic development of mouse

Objective To investigate the expression and distribution of aquaporin 3 (AQP3) in mouse early embryos at different stages.Methods Controlled ovarian hyperstimulation model of Kunming mouse was used to collect four-cell embryos,eight-cell embryos,morula stage,and early blastocysts.Immunofluorescence microscopy and laser confocal microscopy were used to detect expression and distribution of AQP3 channels in these stages.Results Fluorescence signal of AQP3 was found in four embryonic stages of mice.Distribution within embryo was different at different embryonic stages.AQP3 was mainly expressed on the karyotheca of blastomeres at four-cell and eight-cell stage.In morula stage,AQP3 was mainly expressed on cell membrane of each blastomere.In early blastocysts,AQP3 was predominantly expressed on the cell membrane and cytoplasm of trophoblastic cell.Conclusions AQP3 trans-membrane channel might have potential regulation function on mouse embryonic development.
2.Effect of combining loop electrosurgical excision procedure and recombinant human interferon α2b suppository in treatment of cervical intraepithelial neoplasia

Objective To study the effect of combining loop electrosurgical excision procedure (LEEP) and recombinant human interferon α2b (rhIFNα-2b) suppository in treatment of cervical intraepithelial neoplasia (CIN).Methods Prospective,random and control study was conducted in 82 patients with CINⅠ-CINⅢ.Before canting out LEEP to these patients,all women were performed HPV DNA detection by the method of Hybri Max.Among these patients,41 patients were assigned to the studying group,in which the patients were given rhIFNα-2b suppository for three courses of treatment after LEEP.The other 41 patients who carried out LEEP simply were assigned to the control group.Liquid-based ThinPrep cytology test (TCT) and HPV DNA were examined in the sixth and twelfth month after treatment.Results In the studying group,the cure rate was 90.2% when LEEP ended six months,and the cure rate was 100.0% when LEEP ended twelve months.In the control group,the cure rate was 43.9% when LEEP ended six months,and the cure rate was 61.0% when LEEP ended twelve months.In the sixth and twelfth month after LEEP,the difference was significant when we compared the cure rate between the two groups (x2 =19.93,19.89,all P < 0.05).Conclusion The clinical effect of combining LEEP and rhIFNα-2b suppository is better than LEEP in treatment of CIN.The method can remove or destroy the cervical lesions effectively and inhibit HPV replication and spread of HPV infection.
3.The application of DTI and DTT in benign lesion of spinal cord

Objective To explore the application value of diffusion tensor imaging and diffusion tensor tractography in the benign lesion of spinal cord.Methods 30 cases with the benign lesions of spinal cord and 10 healthy volunteers underwent MRI and DTI scanning by 1.5T MR.Results Compared with normal control group,ADC values [(1.29±0.24)×10 -3 mm2/s]of extramedullary group-Ⅰwere increased slightly,and FA values (0.46±0.06)were decreased slightly(P <0.05).In extramedullary group-Ⅱ,ADC values [(2.03±0.19)×10-3 mm2/s]were significantly increased and FA values (0.37±0.03)decreased in the lesion level (P <0.05).The ADC values [(1.71±0.24)×10 -3 mm2/s]of intramedullary group was significantly elevated,and the FA values (0.30±0.06)was obviously reduced (P <0.05 ).The fiber tracts showed the abnormality of the white matter,including compression,sparseness, grow downwards,interruption and other changes.Conclusion The changes of the FA and ADC values,diffusion tensor imaging of the bundle can quantitatively evaluate the severity of spinal cord damage and stereo display fiber bundle damage.
4.Relationship between blood pressure variability and arterial elasticity in elderly patients with stroke

Objective To explore the correlation between blood pressure variability (BPV) and arterial elasticity in elderly patients with stroke.Methods One hundred and eighty-three stroke patients (121 with cerebral ischemic stroke and 62 with hemorrhagic stroke),admitted to our hospital from May 2014 to December 2016,and 61 non-stroke individuals were enrolled according to the inclusion and exclusion criterion.Carotid-radial pulse wave velocity (CrPWV) was calculated and carotid artery ultrasound was used to measure intima-media thickness (IMT);24 h ambulatory blood pressure monitoring was performed to calculate the BPV parameters.The arterial stiffness index β was calculated.Results As compared with those in the non-stroke subjects,morning blood pressure surge (MBPS),standard deviation of systolic blood pressure (SBP),variable coefficient of SBP,systolic blood pressure fall (SBPF),CrPWV,IMT and stiffness index β in stroke patients were significant increased (P<0.05).Correlation analysis indicated that MBPS,standard deviation of SBP,weighted standard deviation of SBP,variance of SBP,variable coefficient of SBP,SBPF,and diastolic blood pressure fall were signficantly correlated with CrPWV and stiffness index β (P<0.05);IMT was closely related to MBPS,standard deviation of SBP,variable coefficient of SBP,and SBPF (P<0.05).Multivariate linear regression analysis showed that MBPS,standard deviation of SBP and SBPF were the independent influencing factors of IMT,CrPWV and stiffness index β5.Conclusion BPV showed significant relationship with vascular elasticity in stroke patients,and reduction of BPV helps to delay the process of atherosclerosis.
5.Advances in the application of 5×FAD transgenic mice in Alzheimer's disease research

Transgenic 5 × FAD mice are APP/PS1 transgenic mice carrying five familial Alzheimer's disease(AD)gene mutations.Beta-amyloid precursor protein(amyloid precursor protein,APP)expression is related to the K670N/M671L(Swedish),1716V(Florida),and V7171(London)mutations,and presenilin 1(PSI)is affected by the M146L and L286V mutations.5 × FAD mice express high levels of β-amyloid in the brain at 1.5 months old,and neuritic plaques began to appear at 2 months old.The pathological phenotypes of 5 × FAD mice include amyloid plaque aggregation,neuronal loss,gliosis,and memory dysfunction,while their biological characteristics include changes in the formation of brain β-amyloid plaques,hyperphosphorylation of Tau protein,synaptic dysfunction,neuroinflammatory response,mitochondrial dysfunction,blood-brain barrier injury,neuronal injury,endoplasmic reticulum stress,and eye lesions.As a classic animal model of AD,5 × FAD transgenic mice can simulate the neuropathological process and behavioral manifestations of late-stage AD in humans,and these mice are thus widely used in research into the pathogenesis of AD and the development of new drugs.In this review,we summarize the model construction,biological background,and biological characteristics of 5 x FAD transgenic mice,and the development and application of drugs for the prevention and treatment of AD,to provide references for the application of 5 x FAD transgenic transgenic mice in AD research.
6.Physiological regulation of salicylic acid on Helianthus tubeuosus upon copper stress and root FTIR analysis.

Phytoremediation plays an important role in the treatment of heavy metal pollution in soil. In order to elucidate the mechanism of salicylic acid (SA) on copper absorption, seedlings from Xuzhou (with strong Cu-tolerance) and Weifang Helianthus tuberosus cultivars (with weak Cu-tolerance) were selected for pot culture experiments. 1 mmol/L SA was sprayed upon 300 mg/kg soil copper stress, and the photosynthesis, leaf antioxidant system, several essential mineral nutrients and the changes of root upon copper stress were analyzed to explore the mechanism of copper resistance. The results showed that Pn, Tr, Gs and Ci upon copper stress decreased significantly compared to the control group. Meanwhile, chlorophyll a, chlorophyll b and carotenoid decreased with significant increase in initial fluorescence (F0), maximum photochemical quantum yield of PSⅡ (Fv/Fm), electron transfer rate (ETR) and photochemical quenching coefficient (qP) content all decreased. The ascorbic acid (AsA) content was decreased, the glutathione (GSH) value was increased, the superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX) activity in the leaves were decreased, and the peroxidase (POD) activity was significantly increased. SA increased the Cu content in the ground and root system, and weakened the nutrient uptake capacity of K, Ca, Mg, and Zn in the root stem and leaves. Spray of exogenous SA can maintain the opening of leaf stomata, improve the adverse effect of copper on photosynthetic pigment and PSⅡ reaction center. Mediating the SOD and APX activity started the AsA-GSH cycle process, effectively regulated the antioxidant enzyme system in chrysanthemum taro, significantly reduced the copper content of all parts of the plant, and improved the ion exchange capacity in the body. External SA increased the content of the negative electric group on the root by changing the proportion of components in the root, promoted the absorption of mineral nutrient elements and the accumulation of osmoregulatory substances, strengthened the fixation effect of the root on metal copper, and avoided its massive accumulation in the H. tuberosus body, so as to alleviate the inhibitory effect of copper on plant growth. The study revealed the physiological regulation of SA upon copper stress, and provided a theoretical basis for planting H. tuberosus to repair soil copper pollution.

7.Research status of premyopia

The visual impairment and blindness caused by myopia have become a global burden, and the World Health Organization has included the prevention and control of myopia in the global program for preventing blindness. In China, the development of myopia is showing a trend with higher incidence, younger age, and higher refractive errors. Moving forward the port of prevention and control myopia has become an important strategy to address the current predicament. Premyopia refers to the stage in children where the refractive power is ≤+0.75 D and >-0.50 D, and there are multiple risk factors during this stage that can potentially lead to myopia. Currently, the incidence of premyopia and its transformation into myopia is high, and the key prevention and control measures include building a predictive model for the transformation of premyopia into myopia, emphasizing the reduction of exposure to risk factors, using low-concentration atropine eye drops, red light therapy, and optical defocus intervention. This article provides a comprehensive review of the current situation regarding the incidence of premyopia and its transformation into myopia, as well as the research progress on existing prevention and control measures, with the aim of providing relevant references for the prevention and control of myopia during the premyopia stage.
