1.The mechanism of the role of ABC transporters in the multi-drug resistance of SP cells in SPC-A1 cell lines

Objective: Recent researches suggest that tumors are organized in a hierarchy of heterogeneous cell populations and that the capability of maintaining tumor formation/growth specifically resides in a small population of cells called cancer stem cells(CSCs).CSCs are resistant to traditional cancer chemotherapy and radiation therapy because they highly express ATP-binding cassette(ABC) drug transporters and are relatively quiescent,with higher DNA repair and anti-apoptosis abilities.The purpose of this study is to investigate the relationship of ABC transporters with the multi-drug resistance and cancer stem cells of SPC-A1 cell lines.Methods: On the basis of Docetaxel-resistant cell line-SPC-A1/ Docetaxel,we compared the content and biological characteristics of SP cells,the expression of ABC transporters and its effect on the multi-resistance to Docetaxel between SPC-A1 and SPC-A1/ Docetaxel cell lines.Results: SP cells existed in both the SPC-A1 and the SPC-A1/ Docetaxel cell line,with a higher content in the latter.P-gp and BCRP obviously expressed in the SPC-A1/ Docetaxel and SPC-A1/ Docetaxel-SP cells,but only the expression of BCRP was increased in SPC-A1-SP cells.SPC-A1-SP cells also showed obviously higher abilities of proliferation,cloning and tumor formation than SPC-A1/ Docetaxel-SP cells.The multi-drug resistance of SPC-A1/ Docetaxel cells could be reversed with verapamil,but their resistance to Docetaxel could not be reversed completely with the same concentration of the drug.Conclusion: BCRP plays a major role in separating SP cells with tumor stem cell traits and its expression counts for the multi-drug resistance of SPC-A1-SP cells.
2.Explore the Mechanism of Huangqi Sijunzi Decoction in the Treatment of Hepatocellular Carcinoma Based on Network Pharma-cology and Bioinformatics

[Objective]To explore the potential mechanism of action of Huangqi Sijunzi Decoction in the treatment of hepatocellular carcinoma(HCC)by network pharmacology combined with bioinformatics methods.[Methods]The bioactive compounds of drugs and their targets were identified using the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)database.Differential gene and module gene of HCC were identified by differential analysis and weighted gene co-expression network analysis(WGCNA).On this basis,the network of"Components-targets of Chinese drug"was constructed.Subsequently,the biological function of the target was investigated and protein-protein interaction(PPI)network was constructed to identify key targets for drug therapy of HCC.Finally,the interaction between the target and the compound was explored by molecular docking.[Results]A total of 156 compounds and 227 targets of 5 Chinese drugs were identified,as well as 2 477 and 685 differential genes in The Cancer Genome Atlas(TCGA)and Gene Expression Omnibus(GEO)datasets,and 2 104 and 2 165 WGCNA association module genes.Then,the Chinese drug component-target network containing 4 drugs,85 compounds and 9 targets was constructed.Biological function analysis showed that the 9 targets were mainly related to cell cycle,p53 signaling pathway and cell senescence.Meanwhile,two core components of kaempferol and quercetin and five core targets of cyclin A2(CCNA2),estrogen receptor alpha(ESR1),cyclin B1(CCNB1),cyclin-dependent kinase 1(CDK1)and topoisomerase Ⅱ alpha(TOP2A)were screened.Molecular docking results showed that the kaempferol and quercetin had stable binding ability to the corresponding core targets.[Conclusion]Huangqi Sijunzi Decoction may play a therapeutic role in HCC by regulating pathways and proteins related with cell cycle.This study provides research ideas and theoretical support for the treatment of HCC with Huangqi Sijunzi Decoction.
3.Analysis of iodine nutritional status among pregnant women after implementation of a new standard of iodized salt in Henan Province in 2018

Objective:To investigate the iodine nutritional status of pregnant women in Henan Province after implementation of new standard of iodized salt, and to provide evidence for scientific adjustment of control strategy.Methods:In 2018, according to "Henan Surveillance Program on Iodine Deficiency Disorders", 5 townships were randomly picked out based on their sub area positions of east, west, south, north and middle in each county in the non-high iodine areas of 156 non-high iodine and high iodine counties (cities, districts). Then 20 pregnant women were sampled in each chosen township to collect and determine their salt and urinary iodine contents. The iodine level in salt was determined by direction titration; the salt samples from Sichuan and other enhanced salt samples were detected by arbitration; iodine content in urine was tested by arsenic cerium catalytic spectrophometry.Results:In total, 15 430 household salt samples of pregnant women were collected and determined; the coverage rate of iodized salt was 95.4% (14 721/15 430) and the consumption rate of qualified iodized salt was 87.0% (13 426/15 430); the median of salt iodine was 25.8 mg/kg. Totally 15 378 urine samples were collected and the median urinary iodine was 188.0 μg/L. The medians of urinary iodine of early, middle and late pregnant were 190.2, 188.9 and 186.0 μg/L, respectively.Conclusions:After the implementation of new standard of iodized salt, the iodine nutritional status of pregnant women in Henan Provence is totally appropriate. The surveillance of iodine status and the universal salt prevention and control strategies among pregnant women should be continuously presented to ensure the moderate amount of iodine intake.
4.Study on the relationship between habitat factors and the distribution

Objective To explore the relationship between the distribution characteristics and the habitat factors of the invasive B. straminea in South China. Methods From October 2016 to August 2017, the breeding condition and habitat factors of B. straminea were investigated in the rivers of Shenzhen and its adjacent areas in the dry season, normal season and wet reason. The generalized additive model (GAM) was used to study the main habitat factors affecting the distribution density of B. straminea. Results The distribution characteristics of B. straminea showed obvious aggregation and unevenness in space. In terms of time, the density of snails was the highest in the dry season, followed by the normal water season and the least in the wet season. The GAM model analysis showed that the main habitat factors affecting the distribution density of B. straminea were water depth, water temperature, flow velocity, dissolved oxygen, and total phosphorus. When the flow velocity and water temperature were 0.25 m / s and 26 °C, respectively, the largest distribution density of snails might appear. The distribution density of B. straminea was positively correlated with dissolved oxygen and total phosphorus. Conclusion B. straminea is suitable to live in the water environment with poor water quality. In the future, the monitoring should be strengthened to provide reference for the prevention and control of the spread of the snails.
5.Pharmacoeconomic evaluation of clopidogrel versus aspirin for secondary prevention of ischemic stroke

OBJECTIVE To evaluate the cost-effectiveness of clopidogrel versus aspirin monotherapy regimens for secondary prevention of ischemic stroke and to provide economic evidence and reference for clinical medication and decision-making. METHODS Based on the CAPRIE trial, a Markov model was constructed; the probabilities of risk events, health utility values, and costs of risk event management were obtained from relevant literature. The cycle length was 6 months, and the time horizon was 10 years. A discount rate of 5% per year was applied. The primary outcomes were total costs, quality-adjusted life-years (QALYs), and incremental cost-effectiveness ratio (ICER). Cost-utility analysis was performed for above 2 regimens by using TreeAge Pro software. The one-way sensitivity analysis, probabilistic sensitivity analysis and scenario analysis were conducted to validate the robustness of the analyses. RESULTS Compared with the aspirin regimen (325 mg/d of CAPRIE trial dose), the ICER values of clopidogrel regimen for secondary stroke prevention for 10 years, 20 years and 30 years were 4 284.06, 4 201.20 and 3 986.78 yuan/QALY, respectively, which were E-mail:liuxiaoyanrj@sjtu.edu.cn all less than the willing-to-pay (WTP) threshold of one time 。 China’s per capita gross domestic product (GDP) in 2021. E-mail:scilwsjtu-wb@yahoo.com Compared with the aspirin regimen (clinically recommended dose in China, 100 mg/d), the ICER values of clopidogrel regimen for stroke secondary prevention for 10 years, 20 years and 30 years were 58 238.27, 42 164.72 and 36 164.77 yuan/QALY, respectively, which were all less than WTP threshold. When comparing with aspirin regimen of 325 mg/d, results of one-way sensitivity analysis showed that the cost of clopidogrel and aspirin, probability of the first recurrence of ischemic stroke were sensitive factors of model. Results of probabilistic sensitivity analysis showed that when WTP was set at one time GDP per capita in China in 2021, clopidogrel had a probability of being cost- effective of about 66.5%. Results of scenario analysis showed that neither changing the time horizon to 10, 20 or 30 years nor using different doses of aspirin (50, 100, 150, 200 or 250 mg/d) would not alter any conclusions. CONCLUSIONS Compared with aspirin monotherapy, clopidogrel monotherapy is more cost-effective for secondary prevention of ischemic stroke.
