BACKGROUND: Experiments have confirmed that there are integrin receptors on bone marrow mesenchymal stem cells (BMSCs), but it is still uncertain whether angiogenin-1(Ang-1) can promote survival of BMSCs and protest against its apoptosis via integrins. OBJECTIVE: To investigate the effect of human recombinant angiopoietin-1 on adhesion, vigour, anti-apoptosis of rat BMSCs and its mechanism of signal transduction. DESIGN, TIME AND SETTING: The cytological in vitro controlled study was performed at the Opening Laboratory, Zhongshan Hospital Affiliated to Xiamen University from January to October 2008. MATERIALS: A total of 40 clean Sprague Dawley rats were supplied by Shanghai Experimental Animal Center, Chinese Academy of Sciences. Human Ang-1 (R&D) was used in this study. METHODS: Combining density gradient centrifugation and adherence separation was used to separate, cultivate and identify rat BMSCs. At the third to sixth passages, BMSCs were used for this study. Various matrix was used to coat 24-well culture plates for 1 hour. Following washing, monoplast suspension was added. Seven groups were set up. Group 1 served as control, BMSCs were coated with phosphate buffered saline containing 0.1% bovine serum albumin. BMSCs in the groups 2, 3 and 4 were incubated with vitronectin, fibronectin and Ang-1. BMSCs in the groups 5, 6 and 7 were firstly incubated in ethylenediaminetetraacetic acid, ?1 antibody and RGD antibody for 10 minutes, and then placed in a 24-well plate, and coated with Ang-1. MAIN OUTCOME MEASURES: Adhesion, trypan blue staining method, MTT test, Hoechst staining and Annexin V/PI were used to measure biological effects of Ang-1 on adherence, activity and anti-apoptosis of BMSCs. Combined with integrin antibody and signal transduction blocker, the expression of phosphoserine/threonine protein kinase B (pAkt), total Akt, Bcl-2 and ?-actin was examined by western blotting. RESULTS: Rat BMSCs were successfully isolated. The purity of BMSCs was 98.3% using flow cytometry. Cell adhesion assay shows Ang-1 promoted the adhesion of BMSCs at 1, 2 and 3 days compared with control group (P

Objective To explore the difference between the stapes prosthesis surgery with microscope and with otoscope. Methods Fifty-four patients with otoselerosis accepted stapes prosthesis surgery with the same kind of stapes prosthesis, done in the same operative method by the identical surgeon. All the operations were done with microscopes or with otoscopes. The operative difference and therapeutic effects between microscope surgery and otoscope surgery were analysed. Results There was no difference in the postoperative heating improvement between microscope surgery and otoscope surgery; the operative time of microscope surgery [(30±7)] min was much shorter than that of otoscope surgery [(60±5)] min, with significant statistical difference. Conclusions Stapes prosthesis surgery with microscope, which has advantages of bimanualness, easiness, and short operative time, is much better than that with otoscope. Thus, microscope is still the preferred magnifying equipment in stapes prosthesis surgery.

Objective:To evaluate the vestibular function of motion sickness. Method:VNG, which tests the vestibular function of horizontal semicircular canal, and CPT, which tests vestibulospinal reflex and judge proprio-ceptive,visual and vestibular status, were performed in 30 motion sickness pations and 20 healthy volunteers(control group). Graybiel score was recorded at the same time. Result:Two groups' Graybiel score (12. 67±11. 78 vs 2. 10±6. 23;rank test P<0. 05),caloric test labyrinth value [(19. 02±8. 59)°/s vs (13. 58±5. 25)°/s ;t test P< 0. 05],caloric test labyrinth value of three patients in motion sickness group exceeded 75°/s. In computerized pos-tourgrphy testing(CPT) , motion sickness patients were central type(66. 7%) and disperse type(23.3%);all of contral group were central type. There was statistical significance in two groups' CTP area, and motion sickness group was obviously higher than control group. While stimulating vestibulum in CPT, there was abnormality (35%-50%)in motion sickness group and none in contral group. Generally evaluating CPT,there was only 2 pro-prioceptive hypofunction,3 visual hypofunction, and no vestibular hypofunction, but none hypofunction in contral group. Conclusions Motion sickness patients have high vestibular susceptible, some with vestibular hyperfunction. In posturography,a large number of motion sickness patients are central type but no vestibular hypofunction, but it is hard to keep balance when stimulating vestibulum.

Objective To understand the current state of staff for Plasmodium microscopy examinations in Changzhou City in the early stage of eliminating malaria action,so as to provide the evidence for achieving the target of malaria elimination. Methods The staff for Plasmodium microscopy examinations from medical institutions and centers for disease control and prevention at differ-ent levels were investigated by questionnaires and tests of the malaria related theory and microscopic operation. Results Totally 95 people were investigated, and among them,40.0%had college degree and 45.3%were university educated. There were 18.9%of them working on Plasmodium microscopy examinations for less than 1 year,40.0%for 2-5 years,18.9%for 6-10 years and 22.1%for more than 10 years. The numbers of person-time of provincial and municipal level training,district-level training, and unit-level training in the last year were 0.57,0.59,and 0.14,respectively. Totally 18.9%of them had the experience of finding Plasmodium at work, and 97.9% of them considered it was necessary or very necessary to do Plasmodium microscopy examina-tions. However,57.9%and 8.4%of them considered Plasmodium microscopy examinations increased their work load and work dif-ficulty,respectively. The average correct rates of knowledge tests on malaria before and after training were 72.5%and 91.6%re-spectively(P<0.05). The average mark of Plasmodium microscopic tests was 25.3 points(full mark being 50 points),and the passing rate(≥30 points)was 58.9%. Conclusion The working experience of staff for Plasmodium microscopy examinations in Changzhou City is relatively poor, and the related training should be enhanced.

Objective To analyze left ventricular (LV) global systolic function in patients with coronary artery disease(CAD) and their changes after percutaneous coronary intervention(PCI) using velocity vector imaging(VVI). Methods Eighteen healthy adults and twenty two patients with CAD were enrolled in this study. Two-dimensional dynamic images of standard apical four-chambers, two-chambers and parasternal short-axis views at the level of mitral valve and apex were obtained in VVI condition. All patients were examined 1 day before PCI, 1 week, 1 month and 3 months after PCI respectively. LV rotation degree, rotation velocity at baseline and apex were measured using the off-line syngo US workplace software and LV twist, torsion were calculated as global motion condition. Results LV ejection fraction(LVEF) and peak torsion at myocardial infarction group were lower than those at normal control and myocardial ischemia group. After PCI, LVEF were gradually improved in both myocardial infarction group and myocardial ischemia group, while changes of LV apex rotation degree and velocity, peak twist and peak torsion between two groups were opposite: those parameters of the former increased 1 week or 1 month after PCI but decreased 3 months after PCI,while those of the latter decreased 1 week after PCI but increased gradually 1 month and 3 months after PCI. Conclusions LV twist and torsion can be evaluated exactly by VVI.Compared with LVEF, LV twist and torsion can preferably reflect the different characteristic of LV global systolic function after coronary artery recanalization in myocardial infarction group and myocardial ischemia group.
