1.A Preliminary Research on Characteristics of Brain Laterality in Patients with Childhood Onset Schizophrenia

Objective: To investigate the characteristics of brain laterality in patients with childhood onset schizophrenia. Methods: The study employed a dichotic Chinese word listening test(DL) as a chief measure and WISC-R as an assistant measure to examine 16 patients and 20 normal children as controls. Results: DL indicated that the patients differed from the controls in ear advantages and their performances were significantly poorer than the controls in left ear. The performances of the patients on WISC-R showed that their PIQ was significantly lower than VIQ. Conclusion:The patients with childhood onset schizophrenia differed from the controls in brain laterality. They might be involved with right hemisphere impairment.
2.The relationship between the expression of EGFR NF-κB and radiosensitivity in human nasopharyngeal carcinoma

Objective:To detect the expression of EGFR and NF-ΚB and the relationship between the two fac-tors and radiosensitivity in human nasopharyngeal carcinoma. Methods The expression of EGFR and NF-ΚB were detected with immunohistochemical staining SP methods in forty-one patients. The radiotherapy effect was assessed by follow up visit and clinical judgement. All of the forty-one patients were divided into three groups: the complete response group,the partial response(or the stable disease)group and the progressive disease group. Result:The ex-pression of EGFR and NF-ΚB were detected in human nasophaTyngeal carcinoma tissues, which were not in normal ones. EGFR and NF-ΚB were significant difference of the radiosensitivity in nasopharyngeal carcinoma tissues (P< 0. 05). The expression of EGFR and NF-ΚB in the complete response-group and the partial response(or the stable disease)-group were lower than the progressive disease-group(P<0. 05),and there was no significant difference between the complete response-group and the partial response(or the stable disease)-group(P>0. 05). The two factors'expression had negative correlation with radiosensitivity. Conclusion: EGFR and NF-ΚB, as important tar-gets, could predict the radiosensitivity of NPC, which could be helpful for targeting treatment and promote the effect of radiotherapy of NPC.
3.Treatment and outcome of polypropylene mesh or tape related pain after reconstructive pelvic surgery

Objective To investigate clinical treatment and outcome of mesh-related pain after pelvic floor reconstruction. Methods Twelve patients were referred to Peking University People′s Hospital for post-operation pain related to polypropylene mesh or tape used in pelvic floor reconstruction and received reoperation from January 2007 to December 2014. The demographic characteristics, clinical manifestation, operation method and follow-up outcome were retrospectively analyzed, and current literature was also reviewed. Results (1)General information:the median age was 54 years old(range 43-74 years old). The median vaginal delivery times was 2 (range 1-5). Initial pelvic floor operation procedures included 4 cases vaginal mesh, 5 cases vaginal mesh plus tension-free vaginal tape (TVT), 1 case rectal surface mesh implantation, and 2 cases sacrocolpopexy. (2) Related complications:all 12 patients had pain-related post-operation symptoms including dyspareunia, perineal pain, urination and defecation difficulty. Seven patients also complicated with other symptoms including discharge and bleeding. Mean time from first operation to medical consultation was 30 months (range 1-72 months). (3) Reoperation results:all 12 patients received reoperation on mesh revision or tape, 10 cases of them reoperated by vaginal approach, while the other 2 cases by laparoscopy. The range of the mesh or tape depended on tender or firm tissue and scar by palpation. The visual analogue scale (VAS) was significantly improved, which was 7.1 ± 2.2 pre-operation, and 0.9 ± 1.9 post-operation (95%CI: 4.48-7.86, P<0.01). Conclusions Pain is one of the reason why patients was removal of the mesh or tape after pelvic floor reconstruction operation. The surgery procedure may be considered as partial or entire mesh or tape excision by vaginal or abdominal way. Mesh or tape removal surgery could relieve related pain and other complications, and the VAS score was significantly improved after surgical management.
4.Impedance characteristics of ear acupoints in identifying excess or deficiency syndrome of stroke.

Objective: To explore the impedance characteristics of ear acupoints in stroke patients with excess or deficiency syndrome, and to provide basis data for objective study of the syndromes of stroke. Methods: The data of electrical characteristics of ear acupoints in stroke patients and healthy people were collected, and excess syndrome and deficiency syndrome of stroke were identified by quantifying the syndromes of stroke using scales. The differences in impedance characteristics of ear acupoints between stroke patients and healthy people were analyzed, and the differences in impedance characteristics of ear acupoints between stroke patients with excess syndrome and stroke patients with deficiency syndrome were analyzed too. The correlation among impedance characteristics of ear acupoints, stroke and the syndromes was also analyzed. Results: There were significant differences in impedance characteristics of ear acupoints between stroke patients and healthy people (P<0.05,P<0.01). The ear acupoints CO12 (Gan) and CO13 (Pi) had a significant role in diagnosing stroke as compared with CO18 (Neifenmi), AT3.4.AH12i (Naogan), CO10 (Shen), TG2p (Shenshangxian), AH6a (Jiaogan), AT4 (Pizhixia), and CO15 (Xin). There were significant differences in impedance characteristics of ear acupoints between stroke patients with excess syndrome and stroke patients with deficiency syndrome (P<0.05, P<0.01). The ear acupoints AH6a (Jiaogan) and CO10 (Shen) played an important role in differentiation diagnosis of excess syndrome and deficiency syndrome of stroke, followed by CO18 (Neifenmi), TF4 (Shenmen) and TG2p (Shenshangxian). Conclusion: Some ear acupoints with diagnostic value for stroke may provide basis of objective research for stroke diagnosis as well as identifying excess syndrome and deficiency syndrome of stroke.
5.The Effects of Differrent Disinfection Methods for Ventilator Circuits

OBJECTIVE To explore disinfection effect of ventilator circuits and monitor the best one from two methods for sampling bacterial culture of pre-and post-mechanical physical disinfection ventilator circuit as well as different culture media.METHODS Pre-and post-disinfection ventilator circuits were sampled and cultured by the direct smudge method and the dilution method respectively which simultaneously the ordinary nutrient agar and the blood cultures used.RESULTS Bacterical positive rate of the direct smudge method was significantly higher than that of the dilution method in pre-and post-disinfection ventilator circuits(P0.05).CONCLUSIONS The direct smudge method is superior to the dilution method in monitoring the disinfection effect of ventilator circuit,it can effectively reduce the false negative rate and make the disinfection effect examination to be more accurate.
6.Study on peripheral nerve deficit bridged by amnion allograft combined with nerve growth factor

Aim To investigate the feasibility of using arnnion allograftcombined with nerve growth factor to bridge peripheral nerve deficit.Methods 48 SD rats as models of sciatic nerve deficit were randomlyassigned into one of the following groups: autograft (Group A), allograftwith Cyclosporine A (CSA) 5 mg/(kg @ d) for 5 weeks (Group B), deficitbridged by amnion allograft combined with nerve growth factor (Group C) ,and allograft with no immune treatment (Group D) . 6 Wistar rats weresacrificed as the donors of the sciatic nerve allograft. Observation wascarried under light microscope and electron microscope examination at 12weeks post-operatively for morphological studying; The latency, ampli-tude, conduct velocity and negative area under the curve were recordedas electrophysiologic index; Maximum contractility of gastrocnemiusmuscle and thickness of myelin of the regenenative nerve were measured;Axons counting were also performed to evaluate the quality of regenera-tion. Results At 12 weeks, group A, B and C recovered better (Indexessuch as latency, amplitude, conduct velocity , negative area under thecurve, maximum contractility of gastrocnemius muscle, thickness of myelinand axons counting were compared by ANOVA test, F= 12.87, P <0.05; F=19.54, P <0.05; F=35.21, P <0.01; F=56.33, P <0.01. F=75.26, P <0.05; F=14.83, P <0.05; F=96.11, P <0.01), and no significant difference was found among these groups ( P >0.05). Conclusion For long distance peripheral nerve deficit, usingamnion allograft combined with nerve growth factor to bridge the deficit canfacilitate the regeneration of nerve and the recovery of function.

8.Transfection and expression of Smad7 inhibits transforming growth factor-?_1 effects on renal tubular cells

Objective To explore the effect of Smad7 gene transfer on tubular cell cycle arrest, tubular apoptosis and fibronectin (FN) synthesis by transforming growth factor ? 1(TGF ? 1) induced. Methods Mouse Smad7 gene was transfected into renal tubular cells in primary cell culture by using Tfx 50 cationic liposome. Tubular cell proliferation was measured by MTT and cell cycle was observed by flow cytometry. The level of FN secretion in the supernatant was determined by ELISA. Results At 48 h after administration of 10 ng/ml TGF ? 1 to renal tubular cells, cell proliferation declined, G 0/G 1 arrested in cell cycle, and cell FN secretion increased significantly. These abnormalities were attenuated by liposome mediated Smad7 gene transfection. Conclusion Transfection and expression of Smad7 can markedly inhibit TGF ? 1 responses in renal tubular cells, which is helpful for further studies of Smad7 gene therapy in vivo .
9.Application of CNVplex combined with STR assay for genetic etiology exploration in chorionic villus of recurrent spontaneous abortion

Objective The study is to present a novel assay for rapid detection of fetal aneuploidies in chorionic villus for spontaneous abortion.Methods Fetal chorionic villus samples were collected from 60 cases of women diagnosed with recurrent spontaneous abortion (RSA) before 13 weeks gestation.All samples were analyzed using CNVplex (copy numbcr variations multiplex) assay and fluorescence in situ hybridization (FISH) in addition to chromosome analysis.All villi specimens were cell cultured and karyotyped to confirm the fetal chromosomal status.Results Among 48 successfully cultured and karyotyped samples,the chromosomal abnormality rate was 60.42 %.The results of karyotyping and the CNVplex assay were identical,both yielding 20 cases of euploidies,23 autosomal aneuploidies,3 triplodies and 2 × monosomies(Tumer Syndrome).However,FISH obtained only 38 results identical to karyotyping.Two cases of deletion and duplication of chromosome were also identified by CNVplex but not always by karyotyping.As for non-mosaic and non structural abnormity samples,the concordance between cytogenetics and genoty ping was 100% in CNVplex and 79.17% in FISH.Conclusion With CNVplex combined with STR(short tandem repeat) assay,we can detect the aneuploidy abnormalities as effectively as routine karyotyping without the need for cell culture,while also analyzing deletions and duplications(larger than 5 Mbp) that are not always detected by karyotype analysis.Our study demonstrates that CNVplex assay is an efficient,convenient,and accurate method to explore the etiology of miscarriage.
