1.Advances in basic immunology and implications for ocular immunology

Immune homeostasis is among the mechanisms of keeping organisms healthy and capable of responding to unfavorable insults either from exogenous or endogenous environments.In recent years,major progress has been made in basic immunology as spotlighted by innate immunity,characterization of novel subclasses of dendritic cells,T or B lymphocytes,and modulation by microRNA (miRNA),long non-coding RNA (LncRNA) and epigenetics.Meanwhile,the eye poses an organ with complex structure and function with specific requirement for immune processes.Pathogenesis or treatment of major ocular disorders involves transplantation immunology,infection immunology,autoimmunity,hypersensitivity,etc.Hence progression in basic immunology lends valuable insights to scientists or ophthalmologists,helping them to understand ocular immunology concerning eye diseases.This review briefs recent updates and future directions in the basic immunology,aiming to provide more reference to the society of ocular immunology.

Gene Expression Omnibus (GEO)can act as an international public resource platform storing and sharing experimental data generated from high-throughout research projects that utilize genome-wide expression strategies like microarray screening and analysis.Numerous data involving inflammatory carcinoma models is helpful for dissecting the regulatory network and molecular mechanism of oncogenesis,thus offering favorable clues for new thoughts in prevention and treatment of cancers.GEO originated from various disease models or processed experiments also provides researchers with powerful evidence for tracing functions of novel predictive genes.
3.Effect of triamcinolone acetonide on the activation and barrier function of human retinal pigment epithelium

Objective To observe the effect of triamcinolone acetonide(TA) on activation and barrier function of human retinal pigment epithelium (RPE). Methods ARPE-19 cells were cultured in 96-well tissue culture plate. Four weeks later, TA with different concentration (0.02 and 0.05 mg/ml)was added to the cells and culture for 3 or 7 days. The activation of ARPE-19 cells was assessed by methyl thiazolyl tetrazolium (MTT). ARPE-19 cells were cultured on polyester microporous filters for 4 weeks, and the transepithelial resistance (TER) was recorded. TA (0.02 and 0.05 mg/ml) was added to the culture fluid respectively, and after cultured for 1 week TER was measured again. The RPE permeability was detected by enzyme-linked immunosorbent assay (ELISA) with horse radish peroxidase as the tracer. [WTHZ]Results In the culture fluid with 0.02 mg/ml TA cultured for 3 or 7 days, the average survival rate of ARPE-19 cells was 93.70% and 90.63% respectively, without statistic difference compared with the control (P=0.147, 0.091). While in the 0.05 mg/ml TA group after cultured for the same duration, the activation of ARPE-19 cells decreased significantly compared with the control (with the average survival rate of 87.75% and 88.98%; P=0.025, 0.043). 4.Transcatheter hepatic arterial chemoembolization for spontaneously ruptured hepatocellular carcinoma

Objective To investigate the practical value of transcatheter hepatic arterial chemoembolization (TACE) in the treatment of spontaneously ruptured hepatocellular carcinoma. Methods Using Seldinger technique,we treated 14 patients with ruptured hepatocellular carcinoma by TACE, in which a suspension of gelatin sponge, iodide oil and chemotherapeutic agents was used as embolus. Results Of 12 patients with active bleeding, successful hemostasis was achieved in 10 patients, with an effective rate of 83 3% (10/12), and 2 patients died from hemorrhagic shock within 48 hours, with an in-hospital mortality of 14 3% (2/14). One patient succumbed to hepatic failure two months after the TACE, and 1 patient received a radical operation two months after the TACE. Follow-up in 10 patients revealed that 8 patients underwent TACE for 1 ~ 4 times (mean survival time, 7 8 months) and 5 patients existed for more than 1 year (35 7%, 5/14). Conclusions TACE is effective in the management of spontaneously ruptured hepatocellular carcinoma.
5.Differentiate Indicator of Random Test of MMPI-2

Objective:To find valid indicator to differentiate random test of MMPI-2.Methods:According to four coincidence indicators of MMPI-2 of 1278 psychiatric cases (male, 623; female,555), including VRIN,│F-Fb│,VRIN+│F-Fb│,F+Fb+│F-Fb│,differentiate random test of MMPI-2 were studied.Results:VRIN+-F-Fb-,-F-Fb-were the best indexes. VRIN was a fine index. F+Fb+ and│F-Fb│were poor.Conclusions:It is not good to use one coincidence indicator, but it can eliminate 91.50% of random test if the four coincidence indicators are used together.
6.Cloning and expression of Escherichia coli O157∶H7 Stx2B subunit

Objective: To clone,identify and express the Shigalike toxin 2B(Stx2B) subunit gene of EHEC O157∶H7.Methods: A pair of primers were designed based on the Stx2B subunit gene sequence of EHEC O157∶H7.The Stx2B gene was amplified from the EHEC O157∶H7 chromosome by PCR and cloned into the pMD18-T vector.Thereafter,the gene was cut from the pMD18-T vector and cloned into the prokaryotic expression plasmid pET-28a vector.Then the recombinant plasmids were transformed into E. coli BL21(DE3) and the transformed host strain induced by IPTG.The expression protein was detected by SDS-PAGE and Western-blot analyses.Results: The Stx2B gene was successfully cloned into pMD18-T and pET-28a vectors,and the expressed protein identified by SDS-PAGE and Western blot.The molecular mass(Mr) of the expressed product was about 7 500,and the expression rate about 40%. Conclusion: The Stx2B gene was successfully cloned and effectively expressed in the prokaryotic expression system.
7.Analysis of Therapeutic Efficacy of Shensong Yangxin Capsule Combined with Low-dose Amiodarone in the Treatment of Ventricular Tachyarrhythmia

OBJECTIVE:To observe therapeutic efficacy and safety of Shensong yangxin capsule combined with low-dose ami-odarone in the treatment of ventricular tachyarrhythmia. METHODS:In retrospective analysis,106 patients with ventricular tachyar-rhythmia were divided into control group and observation with 53 cases in each group according to drug use. Based on conventional treatment,control group was additionally given Amiodarone tablet (first week:0.2 g each time,3 times a day;second week:0.2 g each time,2 times a day;third week to the end of the treatment:0.2 g each time,1 times a day),orally. Observation group was given Amiodarone tablet (first week:0.2 g each time,twice a day;second week to the end of the treatment:0.2 g each time,1 times daily),orally +Shensong yangxin capsule 1.6 g,3 times a day,orally. Both groups were treated for 8 weeks. Clinical effica-cy,blood pressure,heart rate and electrocardiogram(PR period and QT period)and the cardiac cardioversion time were compared between 2 groups,and the occurrence of ADR was also recorded during treatment. RESULTS:After treatment,total response rate of observation group(94.34%)was higher than that of control group(81.13%),the incidence of ADR was lower than that of con-trol group (3.77% vs. 13.21%) and the cardiac cardioversion time was shorter than that of control group,with statistical signifi-cance(P<0.05). Before treatment,there was no statistical significance in heart rate between 2 groups;after treatment,heart rate of 2 groups were significantly lower than before,and observation group was significantly lower than control group,with statistical significance(P<0.05). There were no statistical significance in blood pressure、PR period and QT period between 2 groups before and after treatment (P>0.05). CONCLUSIONS:Based on conventional treatment, Shensong yangxin capsule combined with low-dose amiodarone have good therapeutic efficacy with rapid response for ventricular arrhythmia,it can significantly improve the heart rate of the patients,and the effect is faster with good safety .
8.Review on pathogenicity of Staphylococcus aureus in food

Staphylococcus aureus,an important foodborne pathogen,can contaminate foods through variety of ways and produce enterotoxin that may cause Staphylococcal food poisoning.In addition to food safety problems,Staphylococcus aureus could also cause clinical infection.The study of its pathogenicity is not only beneficial to prevent and control foodborne diseases,but also provide a new point for clinical treatment.This paper analyzes the types and characteristics of common strains of Staphylococcus aureus in food,and summaries the effect of food processing on pathogenicity and the methods for pathogenicity research in order to provide reference for the related study on pathogenicity of Staphylococcus aureus.
9.Expression and characterization of recombinant flagellin FlgE of Pseudomonas aeruginosa

Objective To construct the prokaryotic expression and purification protocols for Pseudomonas aeruginosa type E flagellin (FlgE) and to study its bioactivity.Methods With analysis of Pseudomonas aeruginosa flagellin FlgE sequences,the whole-length FlgE gene was amplified from Pseudomonas aeruginosa genomic DNA by using PCR and primers with proper restriction enzyme sites.The amplified FlgE fragment and prokaryotic expression plasmid pET24a were digested with Nde Ⅰ and Hind ⅢⅣ respectively.The target fragment and vector were recovered and ligated to obtain the recombinant plasmid pET24a-FlgE.DNA sequencing of positive clone confirmed that the target gene and the junctions with vectors were all correct.The plasmid pET24a-FlgE was transformed into BL21 bacteria.The culture conditions like temperature,rotation speed,inducer concentrations,time length were optimized to achieve maximal expression of the target recombinant FlgE with 6×His tag at C terminal.FlgE-His proteins were purified using His-Trap affinity chromatography columns and identified by SDS-PAGE.The purified proteins were further subjected to endotoxin elimination with proper kits.The purified recombinant FlgE was added to cultured corneal epithelial cells for 4 h and the expression of several inflammation-related molecules was examined by using real-time quantitative PCR.Results The recombinant plasmid pET24a-FlgE was successfully constructed and high level FlgE expression was achieved in BL21 with rotation at 16℃ and 1 mmol/L isopropyl-β-D-glucopyranosyl-galactosidase induction for 20 h.Purified recombinant FlgE-His was obtained and used for primary bioactivity assay.After treatment of corneal epithelial cells with 20 μg/ml FlgE for 4 h,the expression of inflammatory cytokines IL-6,IL-8 were significantly increased.Inactivation of the FlgE with ethanol abolished its stimulatory activity.Conclusion The prokaryotic expression and purification system for recombinant Pseudomonas aeruginosa flagellin FlgE was set up,and the recombinant FlgE stimulated the expression of inflammatory factors in corneal epithelial cells.
10.Clinical analysis of 163 cases of pulmonary thromboembolism

Objective To explore the role and significance of standardized scheme for diagnosis and treatment of pulmonary thromboembolism(PTE).Methods The clinical data of 163 consecutive PTE patients who were treated in our hospital from Jan.1972 to Dec.2006 were retrospectively reviewed.The patients were divided into group A and group B based on the time of application of standardized treatment and diagnosis for PTE.The clinical data of the two groups were analyzed and compared.Results The main risk factors included deep vein thrombus,operation,injury,fracture and tumors,etc.Dyspnea was the most important clinical symptoms.The incidences of dyspnea in group A and B were 92.5% and 84.6%,respectively.The diagnostic case and the ratio of final diagnosis in group B was increased compared to those in group A on an annual basis.The median time for diagnosis was shortened(P
