Objective:To investigate a valuable method of gene therapy to treat chronic arterial ischemic diseases in lower extremity.Methods:VEGF was selected as the target therapeutic gene.Recombinant plasmid phVEGF 165 constructed in vitro as the eukaryotic vector of human VEGF cDNA,was directly injected into the ischemic hindlimb muscles in rabbit models.In situ hybridization and RT PCR were carried out to detect the expression of hVEGF 165 in the muscle.Immunohistochemistry assay and Western blotting were carried out to detect the expression of hVEGF 165 protein.Results:The expression of external hVEGF 165 peaked at l week and sustained 2～3 weeks.The expression of protein peaked at 2 weeks and lasted 4 weeks after injection.Conclusion:Skeletal muscle can extract and express external hVEGF 165 ,and further translate the external gene into protein.The effect of direct intramuscular injection of phVEGF 165 is limited in local muscles,and no longer than 1 month.So this method of gene therapy is quite safer.

ObjectiveTo compare the influence of laparoscopic and abdominal hysterectomy on cellular immune function. Methods There were 28 patientsreceived laparoscopic hysterectomy (laparoscopic group) and 28 patients treated with abdominal hysterectomy (abdominal group).The surgical effects and the changes of preoperative and postoperative T lymphocyte subsets of two groups were compared.ResultsThe intraoperative blood loss,anal exhaust time,hospital stay and postoperative morbidity of the laparoscopic group were significantly better than those of the laparotomy group[ (84.7 ± 21.7) ml vs.( 108.0 ± 23.8) ml,(19.3 ±4.1) h vs.(23.8 ±3.8) h,(5.12 ± 1.14) d vs.(7.81 ±2.27) d,7.1％ (2/28) vs.17.9％(5/28),P ＜ 0.05 ].CD4 and CD8 in the 1st day after operation of two groups were significantly lower than that before surgery respectively [ laparoscopic group:(38.41 ± 5.52)％ vs.( 40.72 ± 6.46)％,(24.41 ± 3.78 )％ vs.(26.33 ± 4.17)％ ;abdominal group:(38.41 ± 4.97)％ vs.(40.13 ± 6.12)％,(24.41 ±6.32)％ vs.(26.25 ±4.56)％,P ＜ 0.05 ]; but CD4 and CD8 in the 3rd day after operation in the laparoscopic group[ (40.15 ±6.29)％,(27.23 ± 5.12)％] almost returned to normal level before surgery,while CD4 and CD8 in the 3rd day after operation in the abdominal group [ (36.15 ± 5.12)％,(23.15 ± 4.87 )％] still had significant differences compared with that before surgery(P ＜ 0.05).ConclusionsLaparoscopic hysterectomy with less blood loss,rapid postoperative recovery,fewer complications and less impact on cellular immune function,is superior to abdominal hysterectomy.Therefore,it deserves promotion and wide application.

Objective To research the value of dynamic CT enhancement in diagnosing lung cancer.Methods 35 patients with benign disease and 25 patients with lung cancer underwent dynamic CT scan,the results of CT were analysed in comparison with that of pathological specimens,the relationship between the dynamic CT enhancement phenomena and histological results.Results CT enhanced peak,the enhanced ratio of focus and aorta,the form of enhancement and time-dense curve were different between two groups.CT enhanced peak was related to the histological types and clinical stages of pulmonary cacinoma,and metastasis of lymph nodes.Conclusion Dynamic CT enhanced scan can reflect the characteristic of blood in lung cancer.The invasion and metastasis in lung carcinoma can be predicted according to the CT enhanced peak.

Objective To research the relationship between the expression of cyclooxygenase-2(COX-2),vascular endothelial growth factor(VEGF),microvessel density(MVD)and the CT appearances of lung cancer.Methods CT scans were performed respectively in 35 patients with benign disease and 25 patients with lung cancer confirmed pathology.The immunohistochemistry of all pathologic specimen was analysed using P-V method.The relationship between the expression of COX-2,VEGF,MVD and the CT appearances,histological types,clinic stages,lymph nodes metastasis of lung cancer was evaluated.Results The expression level of COX-2,VEGF,and MVD in lung carcinoma was(64.8?13.8)%,(76.4?14.1)% and(62.5?14.6)/HP respectively,it was higher than that in the lung benign disease group.The level of expression of COX-2,VEGF and MVD was closely related to the CT appearances,histological types,clinic stages and lymph nodes metastasis of lung cancer,but there was no relationship with differentiation of carcinoma.Conclusion COX-2,VEGF and MVD may be important molecular biological predictors in diagnosing lung cancer early and can evaluate prognosis of patients with lung cancer.CT appearances can reflect the vascular characteristics of lung cancer,and can predict the involvement,metastasis and prognosis of lung carcinoma according to the appearances of CT.

The result regarding the application of McAbs against PwMJ-SAg in the detection of the circulating antigens(CAg) in sera from the patients (early stage and nonpulmonary type) with paragonimiasis westermani was reported in this paper. CAg in the sera from 35 patients in the early stage and 15 nonpulmonary type of patients was 100% positive by using McAb IB1 and JB4 out of 8 McAbs against PwMJ-SAg. Furthermore, these two McAbs didnot react crossly with the sera from 25 normal subjects, 15 patients with clonorchiasis, 15 patients with fasciolopsiasis, 15 patients with schistosomiasis japonica, 16 patients with Brugian filariasis or 10 patients with pulmonary tuberculosis. However, they crossly reacted with 8 samples of sera from 15 patients (53.3%) with pagumogonimiasis skrjabini. The McAb IB1 and IB4, threfore, are appropriate as the reagents to diagnose paragonimiasis westermani (early stage or nonpulmonary type). Objective To systematically evaluate the curative effect of sequential treatment and standard triple therapy in eradicating Helicobacter pylori and the risk of adverse events between the two therapies.Methods The related studies were searched with electronic databases to evaluate the efficacy of sequential therapy and standard triple therapy in eradicating H.pylori.Information was retrieved including the clinical features of patients,quality of research,eradication rate of H.pylori and incidence of adverse events and so on.The pooled relative risk(RR)and 95%confidence intervals(95%CI)were calculated,and the publication bias was assessed with funnel plot. Results Twelve randomized trials were performed,and 3183patients met the internalization criterion.Meta analysis demonstrated a clear superiority of the sequential therapy over 7-day triple therapy with an RRof 1.23(95%CI:1.19-1.27),over 10-day triple therapy with aRRof 1.17(95%CI:1.10-1.24),and over 14-day triple therapy with a RRof 1.25(95%CI:1.06-1.48).The incidence of adverse event was similar with sequential therapy versus that of triple therapy(RR=0.96,95% CI:0.75-1.23).Conclusions Sequential therapy is associated with a higher eradication rate of H.pylori compared with 7-day,10-day and 14-day triple therapy.The adverse event doesn't increase significantly.

Objective To summarize the experience of endoscopic thyroidectomy for hyperthyroidism.Methods Endoscopic total or subtotal thyroidectomy was performed through anterior chest wall approach in 7 patients with primary or secondary hyperthyroidism.Results The operation was successfully performed in all the 7 patients.The operation time was 130~260 min(mean,168 min),and the intraoperative blood loss was 10~200 ml(mean,70 ml).No recurrent laryngeal nerve or superior laryngeal nerve injuries,or postoperative hemorrhage,or conversions to open surgery were encountered.The postoperative recovery was uneventful.Short-term follow-up observations demonstrated satisfactory cosmetic results and no recurrence.Hypothyroidism occurred in 2 patients and thyroid functions restored to normal levels in 1 of them 2 months after operation.Conclusions Endoscopic thyroidectomy is a safe and effective procedure for hyperthyroidism.Apart from conventional pre-operative preparation,CT examination is also necessary for identifying the measurements of thyroid glands and determining the proportion and location of residual glands.

Objective To analyze the expressions of tumor stem cell markers Lgr5 and CD44 in esophageal carcinoma and its clinical pathological significance.Methods A total of 108 cases of patients with esophageal cancer, 35 cases of stage Ⅱ, 40 cases of stage Ⅲ, 4 cases of stage Ⅳ. After tumor specimens were stained, the expressions of Lgr5 and CD44 in esophageal cancer tissue were detected and its relationship with the clinical features were analyzed.Results There were relations between expressions of Lgr5 and CD44 in esophageal cancer tissue and clinical staging, the later the stageing, the higher expression rate of Lgr5 and CD44, but there was no significant differece with Chi-square test. The Spearman correlation analysis showed there was no correlation between Lgr5 or CD44 and esophageal cancer condition. Conclusion CD44 and Lgr5 are highly expressed in esophageal cancer tissue, but the correlation between CD44 and Lgr5 expression and esophageal cancer condition is uncertain.

The soybean high branched-chain amino acid mixture was composed of soybean protein hydrolyzed by special method with a few types additional amino acids in small amount. The mixture contained 68.9% of essential ami-no acids and 42.9% of branched-chain amino acids.The animal experiment showed that the mixture could promote the growth of young rats and maintain the normal levels of Hb, serum total protein and albumin. Its protein efficiency ratio (PER), true protein digestibility (TD), biological value (BV) and net protein utilization (NPU) were 2.78, 94.0%, 85.0 and 80.0% respectively. These values were significantly higher than that of casein. The results indicated that the soybean high branched-chain amio acid mixture did not produce amino acid antagonism caused by the high amount of branched-chain amino acids and had higher nutritional quality.

Objective To evaluate the serum changes in glioma patients, and in relations with clinicopathalogy parameters response to treatment and survival in patients with glioma via the examination of the significance of serum VEGF, Flt-1 and KDR levels. Methods Serum VEGF,Flt-1 and KDR levels were analyzed in patients of glioma before and after treatment, and patients with brain metastasis, as well as in healthy controls. Serum VEGF, Flt-1 and KDR levels were assessed by Enzyme-Linked Immunosorbent Assay(ELISA) and the data were processed by SPSS 11.5 for t-test and relevant analyses. Results Serum VEGF, Flt-1 and KDR levels were higher in patients with glioma than in healthy control (P =0.001, P =0.043 and P =0.045 respectively). Significant difference on levels of VEGF was found (P =0.032) comparing glioma group with brain metastasis group. The pretherapeutic serum levels of VEGF and Flt-1 were significantly different with disease recurrence or persistence after treatment (P =0.026 and P =0.038).
