1.Clinical observation of fentanyl transdermal system treating bone pain from metastases of lung cancer

Objective To observe the effect and adverse reaction of fentanyl ransdermal system on the cancer pain resulted from lung cancer metastasizing to bone.Methods Thirty -two patients with moderate to severe cancer pain of bone metastasis from lung cancer were treated with fentanyl transdermal system.Analgesic effect was observed.The quality of life and adverse reactions were evaluated.Results The PI evaluation were (7.62 ± 1.35)and (1.93 ±2.41)before /after treated with fentanyl ransdermal system.The differences of bone pain between before and after treated were significant(t =3.983,P <0.01).Among the thirty -two patients with moderate to severe cancer pain used by fentanyl transdermal system,complete relief was in 12 cases(37.5%),apparent relief was in 11 cases(34.3%),moderate relief was in 7 cases(21.8%),mild relief was in 2 cases(6.3%),the total relief rate was 93.7%.The adverse reactions included hypersomnia were in 13 cases,dizziness in 11 cases,nausea and vomiting in 6 cases,constipation in 5 cases,pruritus and erythra in 4 cases,dysuria in 2 cases.All above adverse reactions dis-appeared after stopping fentanyl ransdermal system and /or undergoing symptomatic treatment.Conclusion Fentanyl transdermal system is easy to use,the effect is significant,there is few adverse reactions,which can significantly improve quality of life of cancer patients.
2.INVESTIGATION OF RELATIONSHIP BETWEEN EXPRESSION OF MANNOSE-LIGAND RECEPTOR ON CAPACITATED HUMAN SPERM IN VITRO AND ACROSOME REACTION INDUCED BY SOLUBILIZED MOUSE ZONA PELLUCIDA

Objective To study the correlation between expression of mannose-ligand receptor(MR) on capacitated human sperm in vitro and acrosome reaction induced by zona pellucida(ZP). Methods The swimming-up sperms were incubated in capacitating medium BWW for 6 hours at 37℃, and then treated with solubilized mouse zona pellucida(mZPS).After an hour, the percentages of spermatozoa labeled with FITC-DMA were counted and used to show the expression of MR; the percentages of acrosome-reacted sperm visualized with fluoresceinated Pisum sativum agglutinin(PSA) were recorded. Results There is no correlation between expression of MR on capacitated human sperm in vitro and acrosome reaction induced by mZPS.Conclusion Expression of MR on capacitated human sperm in vitro might not be the essence of capacitation.;
3.Value of Measuring Nuclear DNA Content in Colorectal Cancer

PurposeTo study the relationship between the nuclear DNA contents and colorectal cancer.MethodsThe nuclear DNA contents were measured by automatic image analysis technique in 21 cases with normal colorectal mucosa,and in 32 cases with colorectal cancer. ResultsThe DNA index(DI), proliferation index(PI), S %, > 5C% and detection rate of aneuploidy were significantly higher than those of normal colorectal mucosa group. DNA polidy pattern of colorectal cancer group were type Ⅲ and Ⅳ, but that of normal colorectal mucosa group was only type Ⅰ . ConclusionsThe DNA content was related to the degree of differentiation of tumor, size of tumor, situs of tumor and prognosis, not related to age or sex. Measuring nuclear DNA content in colorectal cancer was useful for diagnosing colorectal cancer, understanding the biological characteristic, assessing patient prognosis.
4.Aberrant methylation in lung cancer

Aberrant promoter methylation in special genes such as tumor suppressor genes is early eventin carcinogenesis of lung cancer. Aberrant methylation, resulting in corresponding mRNA silence or overexpression , closely relates to carcinogenesis, development, treatment response and prognosis of lung cancer and it may become a potential biomarker. Methylation of special genes may provide evidences for individual treatment of lung cancers.
6.Experimental study of apoptosis induced by rhFasL gene transferred into primary cultured synoviocytes from patients with rheumtoid arthritis

Objective To observe apoptosis of primary cultured synoviocytes from patients with rheumtoid arthritis (RA) which induced by transfected recombinant plasmid carring Fas ligand (FasL) gene, aimed to detect the aim targets and develope gene therapy for RA by intra-articular injection. Methods FasL cDNA was introducted into the plasmid pcDNA3.1-neo by reverse transcriptase-polymerse chain reaction (RT-PCR) to set up the recombinated human FasL gene expression carrier, Then the pcDNA3.1-FasL was transducted into the primary cultured synovial cells by catholyte liposomes. After screened by G418, transgenic synovial cells were choosen. Finally through the microscope, TUNEL, Annexin Ⅴ-FITC and PI staining with flow cytometry, we observed the apoptosis state of each synovial cell. Results Prokaryotic and eukaryon carrier with FasL gene were established successfilly. The gene sequencing had completely coincident with the lierature. When eukaryon carrier with FasL (pcDNA3.1-FasL) was transducted into the index number livinging RA synovial cells after 15 h, most synovial cells presented distortion transformation, and the minority sheded from the bottom wall. The transgenic RA synovial cells after screened by G418, only a little amount survived about 1~2 per sight, and the cells grew slowly, the normal matched cells were cloned after 2~4 weeks. The FasL gene transgenic RA synovial cells presented positive findings by TUNEL staining. Early apoptosis cells by Annexin Ⅴ/PI staining, but the difference wasn′t significant. Conclusion The eukaryon carrier with FasL (pcDNA3.1-FasL) can be transferred into RA synovial cells. It can induce synovial cell apoptosis.
7.EFFECTS ON SPERM OOCYTE PENETRATING BY ANTI SERUM TO HUMAN SPERM MANNOSE RECEPTOR

Objective To study the effect of mannose receptor(MR) from human spermatozoa on sperm-egg fusion. Methods The Balb/c mouse was immunized with the purified mannose receptor(pMR) which was isolated from motile human sperm by mannose-agarose gel affinity chromatography and the anti-MR serum was harvested. Human sperm after induction of acrosome reaction was pretreated with anti-MR serum and subsequently processed in sperm penetration assay (SPA). Results The anti-MR serum can inhibit human spermatozoa from binding and penetrating zona-free golden hamster eggs with a dose-dependent reduction of Fertilization rate(FR) and Penetration index(PI).Conclusion The results indicate that MR of human sperm plays an important role in sperm-oocyte fusion.

TGF ? plays a crucial role in the renal fibrosis and Smads mediate the intracellular signal transduction of TGF ?.Overexpression of Smad7,which can mediate TGF ? by the feedback action,is able to change the physiopathology equilibrium between the R Smads response and the I Smads inhibition to the fibrosis induced by TGF ?.It may help to understand the mechanism of renal fibrosis and Smads TGF ? signaling transduction, and it may also provide a new approach to the renal fibrosis.
9.Treatment of advanced gastric cancer with the regimen of etoposide,folinic acid and fluorouracil

Purpose:To observe the response and tolerance to the regimen of etoposide,folinic acid and fluorouracil (ELF) in advanced gastric cancer. Methods:Thirty two patients with advanced gastric cancer received chemotherapy of ELF regimen(VP 16 100 mg iv d 1—3 ,CF 100—300 mg iv d 1—3,5 FU 750 1 000 mg iv d 1—3 Results:The overall response rate was 37.5% (12/32) and complete response rate was 3.1% (1/31), the partial response rate was 33.4% (11/32). The main toxicity was myelosuppression and alopecia. Leukopenia was observed in 81.6% of the patients, but grade 3 and 4 in only 12.5%. Anemia and throbopenia was observed in 21.8% and 35.1% of the patients respectively, alopecia in 100%. Other side effects were uncommon. Conclusions:This study shows that the regimen of ELF is effective and tolerable for treatment of advanced gastric cancer. [
10.Transcription activator protein 1 (AP-1)-related study in basophils from patients with chronic idiopathic urticaria

Objective To investigate the role of AP-1 in the pathogenesis of chronic idiopathic urticaria (CIU). Methods By using immunomagnetic separation technology, peripheral blood basophils were isolated from 10 CIU patients and 10 normal human controls followed by the extraction of nuclear protein from the basophils. TransAMTM AP-1 family kit was used to detect the DNA binding activity changes of AP-1 family transcription factors in basophils, and Western blotting to detect the expression of P-c-jun protein. Results There were some differences in the DNA binding activity of AP-1 family transcription factors in basophils between CIU patients and normal controls. The DNA binding activity of Phospho-c-jun, c-fos, Fos-B, Jun-B and Jun-D factors was increased in CIU patients compared with the controls, and the increase in that of P-c-jun and Jun-D was statistically significant (both P < 0.05). There was an insignificant decrease in the DNA binding activity of Fra-1 factor in the CIU patients compared with the controls (P > 0.05). The P-c-jun (Ser73) protein expression was higher in CIU patients than that in the controls (0.527 ± 0.312 vs. 0.435 ± 0.042, P < 0.05),whereas there was no significant difference in the P-c-jun (Ser63) protein expression level. Conclusion Some changes in DNA binding activity of AP-1 and overexpression of P-c-jun (Ser73) protein in basophils may be involved in the pathogenesis of CIU.
