1.Correlation analysis of the treatment of the time of maternal pulmonary tuberculosis and the prognosis of their children

Objective To evaluate the correlation of the diagnostic chance of maternal pulmonary tuberculosis and the prognosis of their children. Methods One hundred and fifty-six patients with maternal pulmonary tuberculosis were analyzed,which were assigned to two groups based on the diagnostic chance: pregnant group ( 60 cases) and postpartum group ( 96 cases). Living quality of their children was counted, and viewed in parallel with children (98 cases,control group) of healthy mother at the same time. Results The morbidity in postpartum group was 69.79% (67/96),significantly higher than that in pregnant group [l3.33%(8/60)](P<0.05) and control group [4.08%(4/98)](P<0.05). The mortality in postpartum group was 8.33% (8/96), significantly higher than that in pregnant group (0)(P< 0.05) and control group (0)(P< 0.05).The morbidity and mortality in pregnant group were not significant difference compared with control group(P > 0.05 ). Conclusion Diagnosis and treatment before the parturition of maternal pulmonary tuberculosis can reduce the morbidity and mortality of the children and improve their life quality.
2.ARTIFICIAL IMMUNE ALGORITHM OF MULTICELLULAR GROUP AND ITS CONVERGENCE

Objective To find out more extrema simultaneously including global optimum and multiple local optima existed in multi-modal functions. Methods Germinal center is the generator and selector of high-affinity B cells, a multicellular group's artificial immune algorithm was proposed based on the germinal center reaction mechanism of natural immune systems. Main steps of the algorithm were given, including hyper-mutation, selection, memory, similarity suppression and recruitment of B cells and the convergence of it was proved. Results The algorithm has been tested to optimize various multi-modal functions, and the simulation results show that the artificial immune algorithm proposed here can find multiple extremum of these functions with lower computational cost. Conclusion The algorithm is valid and can converge on the satisfactory solution set D with probability 1 and approach to global solution and many local optimal solutions existed.
3.Effect of curcumin on transforming growth factor-β1 level in lung tissue of mice with acute lung injury

ObjectiveTo observe the level of transforming growth factor-β1 ( TGF-31 ) in lung tissue of mice with acute lung injury induced by bleomycin. MethodsSixty Kunming male mice were randomly (random number) divided into sham operation group, model group, curcumin high dose group,medium dose group and low dose group (n =12 in each group). The models of acute lung injury were made by intratracheal instillation of bleomycin for once in the mice of model group and curcumin treated groups,and equal volume of saline was used in mice of sham operation group instead of bleomycin. Curcumin was given in doses of 200 mg· kg-1 · d-1 , 100 mg· kg-1 · d-1and 50 mg · kg-1 · d-1 to the mice of high dose group, medium dose group and low dose group, respectively 24 hours after modeling until the day before sacrifice, and the mice of sham operation group and model group were fed with equal volume of saline instead. Six mice of each group were sacrificed on the 3rd and 7th day separately for detecting lung wet to dry ratio (W/D), pathological changes of lung tissue stained with HE, and level of the transforming growth factor-β1 in the lung tissues stained with immunohistochemistry in mice of each group. Data were analyzed to express as mean ± standard deviation. -PSS11.5 statistical package was used for analysis. The comparison of the mean diversity carried out by using single factor analysis of variance. Results Morphological changes in lung tissue observed under light microscope showed that obvious morphological abnormalities,alveolar edema fluid and inflammatory cells were not found in mice of sham operation group, and lung capillaries dilated, widened alveolar septum, edema fluid and inflammatory cells infiltration in the alveolar in lung tissue were seen in mice of model group, and different degrees of pathological changes were found in mice of three curcumin treated groups, but those changes were less severe than those in mice of model group. Lung wet to dry ratio (W/D) was significantly higher in mice of model group than that in mice of sham group ( P < 0. 01 ), but that in mice of three curcumin treated groups was significantly lower than that in mice of model group ( P < 0. 01 ). Immunohistochemistry showed low level of TGF-β1 in lung tissue of mice in sham operation group, and the level of TGF-β1 in lung tissue of mice in model group was significantly higher than that in sham operation group ( P < 0. 01 ). The level of TGF-β1 in lung tissue of mice in three curcumin treated groups was significantly lower than that in mice of model group ( P < 0. 01 ),but that was still higher than that in sham group ( P < 0. 01 ). ConclusionsCurcumin can inhibit the TGF-β1 in lung tissue and play a protective effect on acute lung injury induced by bleomycin, reducing the degree of lung injury. The protective mechanism may be associated with the inhibition of TGF-β1 in lung tissue.
4.The study of dendritic cell phenotypic antigens in human ovarian epithelial carcinoma

Objective:To investigate the numbers and distributions of dendritic cells and the expressive intensity of CD1c,S 100 in different human ovarian carcinoma tissues,and to discuss the relationship between them and the mechanism of development.The study can provide the experimental date of biological therapy of ovarian carcinoma.Methods:The samples of 31 patients with ovarian carcinoma were analyzed by ABC immunocytochemistry and computer imaging techniques.Results:The results showed that the number of positive dendritic cells in ovarian carcinoma reduced greatly with significant and the expressive intensity of CD1c,S 100 was weaker in ovarian carcinoma than in normal ovarian tissues.Conclusion:The total expression of dendritic cells phenotypic antigens was reduced in ovarian epithelial carcinoma,which may play an important role in initiating immunity against ovarian carcinoma.
5.Percutaneous transhepatic biliary preset implantation of after-loading radiotherapy catheter for the treatment of bile duct carcinoma of ampulla: preliminary results of 10 cases

Objective To discuss the safety and feasibility of percutaneous transhepatic biliary preset implantation of after-loading radiotherapy catheter in treating bile duct carcinoma of ampulla.Methods A total of 10 patients with carcinoma of ampulla were collected.After percutaneous transhepatic biliary stent implantation was accomplished,the after-loading radiotherapy catheter was inserted via the guide wire.Based on the lesion's location,the positioning of both the catheter tip and the simulation radioactive source implanted through catheter was conducted.Each time before irradiation,the positions of the catheter and the radioactive source were reset under CT or fluoroscopic guidance in order to ensure that the lesion could get adequate internal irradiation dose.Results Percutaneous transhepatic biliary implantation of after-loading radiotherapy catheter and the positioning of simulation radioactive source were successfully accomplished in all 10 patients,and the total internal irradiation dose was completed within 5-7 days after catheter implantation.No severe complications occurred during the whole therapeutic process.Conclusion The technology of percutaneous transhepatic biliary preset implantation of after-loading radiotherapy catheter is safe and feasible,it carries high success rate with less complications.This therapy can improve the patency rate of biliary stent,and,as a palliative treatment,it is suitable for patients with carcinoma of ampulla.This treatment is worthy of application and promotion.
6.A new aurone glycoside from Veratrum dahuricum (Turcz.) Loes. f.

To study the chemical constituents of Veratrum dahuricum (Turcz.) Loes. f., a new aurone glycoside named as (Z)-7, 4'-dimethoxy-6-hydroxyl-aurone-4-O-β-glucopyranoside was isolated from the 95% ethanol extracts of the rhizomes and roots of Veratrum dahuricum (Turcz.) Loes. f. by repeated column chromatography on silica gel and recrystallization. Its structure was established by extensive spectroscopic analyses, and its cytotoxicities against HepG-2, MCF7 and A549 cell lines were measured in vitro.
7.Interventional therapy of acute mesenteric venous thrombosis

Objective To assess the efficiency and methods of the interventional management in acute mesenteric venous thrombosis (AMVT). Methods Fifteen patients with AMVT who diagnosed by imageology were treated by interventional procedures. Eight patients were treated by transcatheter superior mesenteric artery thrombolysis with urokinase, 5 cases by percutaneous transhepatic treatment, 2 cases by transjugular intrahepatic portosystemic shunt approach. Results The technical success was achieved in all the 15 cases without complications. The majority of the thrombus was cleared by interventional procedures and flow restorated on the angiograms. All the patients with follow-up from 10 to 22 months showed no recurrence. Conclusion The minimally invasive interventional techniques are safe and effective in the treatment of mesenteric venous thrombosis without necrosis.
8.Oncogenic osteomalacia in renal cell carcinoma: a case report and literature review

Objective To investigate the clinical manifestations, pathologic features, treatment and prognosis of oncogenic osteomalacia (OO). Methods A 17-year-old male patient was admitted.The patient presented with a 4-year history of progressive lower limb weakness, muscle atrophy and joint pain. Lab tests demonstrated hypophosphatemia. Lumbar and pelvic X-ray showed wedging of the lumbar spine and decreased pelvic bone density. Ultrasonography revealed a low-echo and bloodrich mass measuring 15.1 cm× 14.5 cm× 13.2 cm located at the lower pole of left kidney. Computed tomography showed a voluminous enhanced tumor, supplied by the left renal artery, vice renal artery and a branch of the inferior mesenteric artery. A tortuous vein at the lower part of the tumor drained into the left renal vein. A radioactive octreotide scan was used to study an increased radionuclide-uptake lesion which was initially suspected on the CT scan. The patient was preoperatively diagnosed with left malignant kidney tumor and oncogenic osteomalacia. During a left radical nephrectomy performed through a transperitoneal anterior subcostal incision, a round diameter 15 cm tumor covered by twisted veins was found. The patient recovered well postoperatively. Results The histological diagnosis was renal clear cell carcinoma. Immunohistochemical staining detected Vimentin and RCC were positive and Ki-67antigen index was about 2 %. After lesion removal, the paraneoplastic syndrome resolved. At the 11-month follow-up, no recurrence of the disease was observed. Conclusions Oncogenic osteomalacia is rare and OO with renal cell carcinoma has not been reported so far. OO or rickets should be suspected in those patients who presented with metabolic bone disease associated with hypophosphatemia and inappropriate phosphaturia. If OO syndrome was suspected, in addition to past medical history and imaging, a standard meticulous examination should be initiated promptly. Surgical treatment is still necessary once confirmed. And all of them achieved a good prognosis.
9.Association between ACTN3 gene R577X polymorphism and blood lipid levels in an elderly Chinese Han population in Rugao,China

Objective To explore the association between ACTN3 gene R577X polymorphism and serum lipid levels in an elderly Chinese Han population.Methods This study was based on the ageing arm of The Rugao Longevity and Ageing Study (RtLAS).Genotyping was performed by Taqman MGB method.Lipids includedtotal cholesterol (TC),triglyceride (TG),high-density lipoprotein cholesterol (HDL-C) and low-density lipoprotein cholesterol (LDL-C).Cut points of dyslipidemia were based on that reported in the NCEP ATPIII.We explored the associations between R577X polymorphism loci and dyslipidemia by using analysis of covariance and logistic regression analysis.Results Overall 1 618 samples was included (741 males and 877 females) in this study.Covariance analysis found that TC level of RR,XX and RX genotype in the general population were 1.34,1.37,1.43 mmol/L with an increasing trend (P =0.024);TC and LDL-C levels of RR,XX and RX genotype (TC:5.13,5.29,5.43 mmol/L,P =0.004;LDL-C:2.76,2.88,3.00 mmol/L,P =0.004) were significantly different in females.Logistic regression analysis showed that the increased copies of X allele were significantly associated with increased TC and LDL-C levels in the general population and females.For the general population,TC:OR=1.184,95%CI:1.030-1.361,P =0.018;LDL-C:OR =1.334,95%CI:1.101-1.588,P=0.003;For females,TC:OR=1.332,95% CI:1.102-1.616,P =0.003;LDL-C:OR =1.549,95 %CI:1.208-1.986,P =0.001.After adjusting for other covariates,the above associations remained significant.Conclusions ACTN3 gcnc R577X polymorphism is associated with plasma TC and LDL-C levels in elderly woman of Han populations in Rugao,China.
10.Influences of extracellular matrices secreted by ultraviolet B-induced senescent fibroblasts on ERK signaling in HaCaT cells

Objective To explore the influences of extracellular matrices (ECM) secreted by ultraviolet B (UVB)-induced senescent fibroblasts on the proliferation of and extracellular signal-regulated kinase (ERK) signaling in HaCaT cells. Methods Fibroblasts were irradiated with UVB of 15 mJ/cm2 once daily for 5 days to induce premature senescence, which was identified by SA-β-gal staining 72 hours after the last irradiation.HaCaT cells were divided into 3 groups and inoculated into plates coated with extracellular matrices secreted by non-senescent (PRE-ECM) or senescent fibroblasts (SIPS-ECM) or into uncoated plates (NON-ECM), fol-lowed by additional culture. U0126, an inhibitor of ERK1/2, was used to treat the HaCaT cells 1 hour before inoculation. Then, MTT assay was carried out to detect the proliferation of HaCaT cells after a 3-day culture,Western blot to assess the phosphorylation of ERK at 0.5, 1, 2 and 4 hours after the inoculation, flow cytometry to analyse cell cycle and apoptosis after 24 hours of culture. Results The most rapid and intense phosphory-lation of ERK was observed in SIPS-ECM group. Inhibiting the activation of ERK pathway with U0126 could completely suppress the promoting effect of ECM from senescent fibroblasts on the proliferation of HaCaT cells.After the blocking of ERK activation, the proportion of HaCaT cells in S and G2/M phase decreased from 37.40%, 41.34% and 43.31% to 29.41%, 36.48% and 39.96%, respectively, in NON-ECM, PRE-ECM and SCIP-ECM group. Conclusion The ECM produced by UVB-induced senescent fibroblasts promote the prolifera-tion of HaCaT cells via inducing the phosphorylation of ERK.
