Aim To investigate the effects and possible mechanisms of kaemperol in the rats with chronic cere-bral ischemia. Methods Chronic cerebral hypoperfu-sion model was produced by permanent occlusion of bi-lateral common carotid arteries (2VO)in rats. After KAE treatment, the rats underwent Morris water maze and prehensile traction test.Neuronal morphology was observed using Nissl and HE staining. The activity of SOD and the content of MDA in brain tissue were de-termined.The DJ-1 protein expression was assayed by Western blot. Results Compared with 2VO model group,KAE significantly improved learning and memo-ry and the grasping ability. In addition,KAE signifi-cantly reduced brain tissue pathological injury induced by 2VO. Furthermore, KAE significantly increased SOD activity and enhanced antioxidant protein DJ-1 ex-pression in brain tissue. Conclusions KAE could sig-nificantly attenuate the cognitive impairment, limb bal-ance dysfunction and pathological injury in rats with chronic cerebral ischemia. The mechanism may be re-lated to improving the antioxidant system in vivo.

OBJECTIVE:To explore drug quality dynamic evaluation mechanism based on analysis of drug quality announce-ment data. METHODS:The sorts and types of drugs, places of origin, quantity of production enterprises and frequency distribu-tion of being mentioned were analyzed on the basis of drug quality announcement issued by CFDA and provincial administration in 2013. RESULTS & CONCLUSIONS:Frequency of TCM being mentioned (0.75%) was higher than that of chemical drug (0.30%). Among drugs mentioned more than 10 times, the most chemical drugs were injection (70%) and the most TCM were tablet(66.7%). Among all types, drug types produced by more than 100 enterprises accounted for 62.5%,and those produced by more than 50 enterprises accounted for 93.75%,which indicated the frequency of problems was in relatively direct proportion to the quantity of drug manufacturing enterprises. Top 10 provinces in the list of the number of drug quality announcements showed different characteristics of problem type distribution. In compassion, the quality problems of TCM in Jilin,Guangxi,Mongolia,Si-chuan were prominent, and reference number proportion of involved TCM were 85%,80%,97% and 69%,respectively;the quality problems of chemical drugs in Henan,Shanxi,Jiangsu,Guangdong,Anhui,Shandong should be paid attention, and refer-ence number proportion of involved chemical drugs were 62%,61%,95%,57%,86% and 65%,respectively. It is suggested that the government should carry out the dynamic risk assessment system based on data of drug quality announcement, and strength-en standard management of data collection of local drug quality announcement.

Glutamate decarboxylase (GAD) can catalyze the decarboxylation of glutamate into γ-aminobutyrate (GABA) and is the only enzyme of GABA biosynthesis. Improving GAD activity and thermostability will be helpful for the highly efficient biosynthesis of GABA. According to the Ramachandran plot information of GAD 1407 three-dimensional structure from Lactobacillus brevis CGMCC No. 1306, we identified the unstable site K413 as the mutation target, constructed the mutant GAD by site-directed mutagenesis and measured the thermostability and activity of the wide type and mutant GAD. Mutant K413A led to a remarkably slower inactivation rate, and its half-life at 50 °C reached 105 min which was 2.1-fold higher than the wild type GAD1407. Moreover, mutant K413I exhibited 1.6-fold higher activity in comparison with the wide type GAD1407, although it had little improvement in thermostability of GAD. Ramachandran plot can be considered as a potential approach to increase GAD thermostability and activity.

Objective To investigate the mechanisms of quercetin in retinal angiogenesis via in vitro and in vivo studies. Methods Human umbilical vein endothelial cells (HUVECs) was used in in vitro study, and oxygen-induced retinopathy (OIR) and laser-induced choroidal neovascularization (CNV) animal models were used in in vivo study. In the in vitro cell study, normal control group,VEGF-165 treatment group and VEGF-165 combined with quercetin treatment group were used.Cell counting kit-8 (CCK8) and Transwell were used to measure HUVECs cell viability, including proliferation and migration.OIR and CNV animal models were administrated through intraperitoneal injection of quercetin, and the non-perfusion area and CNV aera were evaluated.Western blot assay was used to detect the expression of integrin α5 and integrin β3 expression. In the animal study,18 breastfeeding mice and 18 infant mice were randomly divided into normal control group, model control group and quercetin treatment group,6 for each group. The animal feeding and use was in accordance with the standards set by the ARVO,and the experiment was approved by the Ethic Committee for Experimental Animal of Fu Xing Hospital,Capital Medical University (2016-KY-0036).Results In the in vitro study,VEGF-165 (20 ng/ml) promoted the proliferation and migration of HUVECs,while quercetin (50 μmol/L) inhibited HUVECs proliferation and migration significantly, comparing to the VEGF-165 treatment group (proliferation:Fgroups =18.51,P =0.00;migration:F =85.74,P =0.00).In the in vivo studies, quercetin 20 mg/(kg·day) decreased the non-perfusion area and CNV area comparing to the untreated groups, with significant differences between them (t =6.02,P =0.00;t =5.79,P =0.00).Besides,quercetin down-regulated the expression of integrin α5 and integrin β3 significantly both in the in vitro and in vivo studies.Western blot test showed that integrin α5 and integrin β3 in VEGF-165+quercetin processing 24 hours group were significantly decreased than those in the VEGF-165 group, with significant differences between them (t =4.46,P<0.05;t =5.18,P<0.01).Compared with the VEGF-165 +quercetin 24 hours group, the integrin α5 and integrin β3 in VEGF-165 +quercetin processing 48 hours group were increased, but they were still significantly increased than those in the VEGF-165 group, with significant differences between them (t =6.54,P < 0.05;t =7.17,P < 0.01).For the animal studies, quercetin also inhibited the levels of integrin α5 and integrin β3 in OIR and CNV models (t =5.44,13.52;both at P=0.00).Conclusions Quercetin can inhibit the retinal and choroidal neovascularization through integrin pathway, which provides a new treatment strategy for clinical therapy.

Journal of Biomedical Engineering 2005;22(4):677-680
We analyzed the multifractal singularity spectrum of synchronous 12-lead ECG (electrocardiogram) signals from heart and brain disease patients, and found that multifractal curves of different leads do not overlap each other. After calculating the scope of the singularity strength, we noticed that the averages of scope are not the same in different subjects, and the dispersing degree is also different in someone's different leads. Both the deltaalpha, the average of the deltaalpha, and the dispersing degree deltaalpha (be defined as standard deviation) of the deltaalpha of every one's 12-lead ECG were computed. Then, a comparison was made between the spectrum of the healthy subjects and the heart disease patients. The results showed that their deltaalpha are close, but their deltaalpha are markedly different. Also the healthy subjects and brain disease patients were compared, we found that their deltaalpha are close, but their deltaalpha are markedly different. These indicate that the character of multifractal spectrum is controlled by both the neurosystem of body and the self-syntonic property of cardiac structures. Furthermore, the deltaalpha is related with the neuroautonomic control of people's body on the ECG, and the deltaalpha is related with the anisotropy of the heterogeneous tissue and the electric signal propagation in heart.

Journal of Biomedical Engineering 2007;24(6):1211-1214
By analysing the f(a) singularity spectra of the ST segments of the synchronous 12-lead ECG, we have found that the singularity spectrum is close to monofractality and its area is only half the area of the synchronous 12-lead ECG f(alpha) singularity spectrum. The ST segments of the synchronous 12-lead ECG signal also has f(alpha) singularity spectra distribution and it also has a reasonable varying scope. We have also found that the lead number of the ST segment f (alpha) singularity spectra for adults having coronary heart disease overstep the reasonable scope tends to increase over that of the ECG f(alpha) singularity spectra. These findings show that using the ST segments f(alpha) singularity spectra distribution of the synchronous 12-lead ECG is more effective than using the synchronous 12-lead ECG on the clinical analysis.
