1.Effect of prone-position ventilation on oxygenation in patients with acute respiratory distress syndrome

Objective To determine the effect of prone position (PP) ventilation on oxygenation in patients with acute respiratory distress syndrome (ARDS) .Methods Twenty-three patients of both sexes (14 males, 9 females) with ARDS (within 72 h after onset of ARDS) were included in this study. The patients ranged in age from 43-78 years. Ten minutes before the patients were placed in prone position, midazolam 2-5 mg was given i. v. followed by continuous infusion at 2-3 mg?h-1 for sedation and a bolus of vecuronium 4 mg per hour for muscle relaxation. The patients were mechanically ventilated (VT 6-8 ml?kg-1, inspiration-time 1.0-1.2 s, RR 12-20 bpm, FiO2 0.4-1.0, PEEP 6-18 cm H2O) . The patients were placed in prone position for 2 h. Radial artery was cannulated and S-G catheter placed. MAP, CVP, MPAP, PAWP, cardiac output, PaO2 , PaCO2 peak inspiratory pressure (PIP) and airway resistance (Raw) were measured and recorded immediately before (T0) and 0.5, 2 h after the patients were placed in prone position (T1 , T2) and 2 h after supine position was resumed(T3) . Objective:To explore the effects of total saponins of Cornus offlcinalis Sieb.(TSCO) on the contractility and relaxation of mesenteric artery in streptozotocin (STZ)-induced diabetic rats.Methods:Sprague Dawley rats were administrated STZ intra-peritoneally at a dosage of 60 mg/kg to induce diabetes.TSCO was administrated at a dosage of 60 mg/kg or 120 mg/kg (per os) to the diabetic rats for 28 days.Glucose and insulin in the serum as well as the responsiveness ofmesenteric artery rings were determined.Results:TSCO decreased the contractile responsiveness to phenylephrine of mesenteric artery rings from diabetic rats,but increased the reactivity to acethylcholine.TSCO (120 mg/kg) ameliorated the baseline release of nitric oxide of mesenteric artery,but had little effect on the induced release of nitric oxide.Rosiglitazone had less effect on the mesenteric artery function than that of TSCO,though it was more effective on lowering blood glucose.Conclusion:The effects of TSCO on mesenteric artery of diabetic rats are dose dependent,and are possibly exerted by lowering blood glucose and ameliorating the release of nitric oxide in the endothelium.

Parkinson's disease(PD)is a neurodegenerative disease that predominantly affects the elderly.Dopamine mimicking drug is the mainstay in the treatment of Parkinson's disease.However,there is a large of interindividual variability in response to anti-Parkinson's disease drugs.It is thought that genetic variability in dopamine system genes is one of the important factors in determining interindividual variability in drug response.There are a lot of studies focused on the relationship between the risk of Parkinson's disease and genetic variations in domestic,while it is a lack of the pharmacogenetics study on Parkinson's disease.This review summarizes the relationship between the polymorphism of genes encoding dopamine transporter,dopamine-metabolizing enzymes and dopamine receptors and Parkinson's disease treatment.
4.Expression of programmed cell death 5 in endometrial adenocarcinoma and its clinical significance

Objective This study was designed to investigate the relationship between programmed cell death 5 (PDCD5) and endometrial carcinoma .Methods 60 tissue samples of endometrial carcinomas ,40 normal proliferative endometrium tissues and 28 endometrial atypical hyperplasia tissues were obtained from patients who underwent operation.The expression of PDCD5 was detected by Real-time PCR and immunohistochemical analysis . Results In normal proliferative endometrium , atypical hyperplasia tissues and endometrial carcinoma , the PDCD5 mRNA was gradually decreased[(1.21 ±0.16),(0.83 ±0.07),(0.42 ±0.02),F=5.637,P<0.05];The posi-tive rate of protein was gradually decreased (82.5%,64.3%,45.0%,χ2 =27.663,P<0.05);The expression of PDCD5 was correlated with TNM stage , histological grade , depth of myometrial invasion and lymphatic metastasis (χ2 =20.875,11.915,9.409,11.148,all P<0.05),but was not correlated with ages (χ2 =0.287,P>0.05). Conclusion PDCD5 might become a potential molecular marker of early diagnosis of endometrial carcinoma .

6.Effect of δ-aminolevulinic acid concentration on biological characteristics of skin fibroblasts

Objective To investigate the effect of δ-aminolevulinic acid(ALA)-induced photodynamic reaction on biological characteristics of skin fibroblasts.Methods Human skin fihroblasts were isolated from the foreskin of children and cultured in the presence of 0.5 to 5.0 mmol/L ALA;three hours later,they were irradiated with 632.8 am laser followed by additional 12-hour culture.Then,MTI"assay and flow cytometry were used to measure the proliferation and apoptosis of cells.The levels of matrix metallopmteinase(MMP)1,-2 and-3 and hydroxyproline in the culture supematant of irradiated cells were determined by avidinbiotin complex-based ELISA and alkaline hydrolysis-based method,respectively.Results With the rise of ALA concentration from 0.5 to 5.0 mmol/L.the proliferation level of fibroblasts expressed as the absor-bance at 570 nm dropped from 0.45±0.05 to 0.32±0.04.and cell death rate increased from 6.4%±2.0% to 29.6%±2.2%.Meanwhile,the contents of MMP-1,-2 and -3 increased at the early stage,but decreased at the late stage,whereas the hydroxyproline level showed an inverse tendency during the increase of ALA concentration from 0.5 to 5.0 mmol/L.Conclusions Proper intensity of photodynamic reaction induced by ALA could enhance the secretion of MMP and inhibit the synthesis of collagen by skin fibroblasts,however,high concentration of ALA may exert an inverse effect.
7.Effects of simvastatin on neurons autophagy in hippocampus following intrac-erebral hemorrhage in rats

Objective:To evaluate effect of simvastatin on neurons autophagy in the hippocampus following intracerebral hemorrhage( ICH) in rats.Methods:All rats were divided randomly into sham group;ICH group and SIM group.The rat model of ICH was made by injection of Ⅶ type collagenase.The behavioral changes were evaluated with Garcia′s neurologic deficit score.Wet-dry weight method was used to evaluate brain edema.Additionally,localization of LC3 protein was determined by immunohistochemistry, and the expression of Beclin-1 in hippocampus was tested by Western blot methods.Results: Compared with ICH group,simvastatin treatment after ICH significantly improved neurologic deficit score ( P<0.05 ) , reduced the water content of brain tissue ( P<0.05 ) , inhibited the expression of LC3 and Beclin-1 protein(P<0.05).Conclusion:Our findings indicated that administration of simvastatin had neuroprotective effects in ICH rat model,which could be involved in inhibiting activation of autophagy in the hippocampus,finally reducing brain edema and neurobehavioral deficit.
8.Clinicopathological analysis of 306 cases with endometrioid cancer in northern Shandong area

Objective To explore the clinicopathology characteristics of endometrial adenocarcinoma and non-endometrial adenocarcinoma.Methods The clinical data of 306 patients with endometrioid cancer were analyzed retrospectively.Results The patients with endometrial adenocarcinoma accounted for 90% of all patients,and the patients with non-endometrial adenocarcinoma accounted for 10%.Compared with the patients with endometrial adenocarcinoma,the average age of the patients with non-endometrial adenocarcinoma was older[(59.32 ± 7.77)years],the constitution ratio of postmenopausal patients was higher(90%),the clinicopathologic stage was later(P =0.000),the grade of histology was higher(P =0.000),the myometrial invasion was deeper(P =0.014).The patients with nonendometrial adenocarcinoma were prone to occur lymph metastasis(P =0.006),and the recurrence rate was higher(P =0.000).Conclusion There is significant defference between endometrial adenocarcinoma and non-endometrial adenocarcinoma,the patients with non-endometrial adenocarcinoma has poor pronosis,and we should pay more attention to it.
9.Comparison of Components in Solutions of Fuzhuang Capsules Extracted by Four Methods

Objective: The extract technologies for the components of Fuzhuang Capsules were optimized. Methods: Four methods: the semi bionic extraction (SBE), the water extraction (WE), the semi bionic extraction by precipitation with alcohol (SBAE) and the water extraction by precipitation with alcohol (WAE) were used to extract Fuzhuang Capsules, with panaxadiol, panaxatriol, ? sitosterol, icariine, betaine, total sugar and dried extractive taken as the marker, under the same conditions of drug granularity, solvent amount, decocting temperature, filtration, concentration, etc., and then four methods were compared and studied. Results: Through the comprehensive evaluation of seven markers, their comprehensive values Y decreased in the order of SBE, WE, SBAE and WAE. Conclusion: SBE is better than the other three methods in the extraction of the components of Fuzhuang Capsules, which is showed by the fact that the pH values of the water for the 1st, 2nd and 3rd decoctions are 3.4, 7.5 and 8.4, and the extraction times are 2.0h, 1.5h, and 1.5h respectively.
10.Effect of IL-18 on expression of molecule on surface of 9 L in vitro and in vivo

Objective:To explore the influence of IL-18 on the expression of MHCⅠ,MHCⅡ,CD80,CD86 on the surface of 9L cells,to identify the effect of IL-18 on the immunogenicity and the efficiency of tumor antigen presentation of 9L.Methods:Retroviruses were used to transduce the mIL-18 gene into rat glioma cells and the cell clones (9L/IL-18) which steadily expressing mIL-18 gene were obtained ,and got the control cells of 9L/LXSN by the same method.The expression of MHCⅠ,MHCⅡ,CD80,CD86 on the cell surface were assessed by flow cytometry.The cell suspension of 9L/IL-18,9L/LXSN and 9L cells were inoculated into the brain of F344 rat to establish the animal models by the stereotactic technique ,got the tumor tissues to analyze the expression of MHCⅠ, MHCⅡ,CD80 and CD86 on the surface of tumor cells after 14 days.Results:The expression of MHCⅠ,MHCⅡ,CD80 and CD86 on the surface of 9L/IL-18 were (10.9±1.44)%,(0.61±0.14)%,(1.01±0.14)%,(0.57±0.11)% ; had no significant differences with the others in vitro (P>0.05);while the expression of MHCⅠ,CD80 and CD86 on the surface of 9L/IL-18 were(67.51± 1.40)%,(12.51±1.57)%,(6.95±0.56)%which were higher than 9L/LXSN and 9L cells (P<0.05),the expressions of MHCⅡwere no significant difference(P>0.05) in vivo.Conclusion: IL-18 did not affect the immunogenicity of 9L in vitro,but improve the immunogenicity and tumor antigen presentation in vivo.
