Objective: To detect the novel apoptosis-related protein PDCD5 expression in granulosa cells of polysystic ovary syndrome(PCOS) and normal ovary, and explore the pathogenesis of PCOS. Methods:The granulosa cells were collected from 30 cases of PCOS and normal ovary in IVF-ET. Expression of PDCD5 was detected by flow cytometry; immunofluorescence and immunohistochemistry. Cell apoptosis was detected by Propidium Iodide (PI) staining. Results: The number of hypodiploidy cells associated with apoptosis in granulosa cells of PCOS was greater than that of the normal control. Objective: To study the expression and localization of apoptosis-related protein TFAR19 in TF-1 cells undergoing apoptosis. Methods: Using monoclonal antibody against TFAR19, the expression level and cell localization of TFAR19 were examined by fluorescence microscope, confocal laser scan microscope(CLSM) and flow cytometry. Simultaneously, we also analyzed the relationship of TFAR19 protein with phosphatidylserine (PS) externalization and cell nuclear DNA fragmentation. Results: The level of TFAR19 proteins expressed in TF-1 cells treated with GM-CSF withdrawal was significantly increased compared with normal TF-1 cells, then translocated rapidly from cytoplasm to the nucleus of cells. Appearance of TFAR19 in the nucleus of apoptotic cells preceded the detection of PS externalization and DNA fragmentation. Conclusion: Nuclear translocation of TFAR19 protein is one of the earliest events of cell apoptotic process. These data provided a new clue to further approach to the biological function of TFAR19 and study of cell apoptosis.
3.Apoptosis-associated Gene PDCD5 Expression in Esophageal Cancer in Xinjiang Kazakh and Han Nationality

Objective: To study the expression of PDCD5 mRNA and its significance in esophageal cancer in Xinjiang Kazakh and Han nationality. Methods: RT-PCR was used to detect the mRNA of PDCD5 in 40 cases of esophageal cancer (18 cases of Kazakh, 22 cases of Han). Results: The positive rate of PDCD5 mRNA expression in 40 samples of esopha-geal cancer tissue, adjacent tissue, and normal tissue was 80.0% (32/40), 80.0% (32/40) and 87.5% (35/40), respectively, with no significant difference (P>0.05). The Ods of cancer tissues, adjacent tissues, and normal tissues were 0.7644± 0.1444, 0.9341 ±0.1631 and 1.8703±0.4767, respectively. The expression of PDCD5 was significantly increased in cancer tissues compared with that in normal tissues (P<0.05). The expression level of PDCD5 mRNA was not significantly correlat-ed with the degree of differentiation and lymph node metastasis. Conclusion: No significant difference was found in PDCD5 mRNA expression in esophageal cancer between Xinjiang kazakh and Han nationality (P>0.05). The expression of PDCD5 is not correlated with the degree of differentiation, depth of invasion or lymph node metastasis. Detection of PDCD5 mRNA expression in esophageal cancer tissues may provide valuable information for patient prognosis.

Traditional 2D cell culture technology is widely used in cell culture field, but it lacks the formation of 3D matrix scaffold and lacks specific cytokines in vivo.Cells in 3D cell culture system are similar to cells in vivo for gene expression and cell activity. The article focuses on the application of three-dimensional cell culture technology in cancer research, including the construction of tumor microenvironment,tumor biological behavior, tumor angiogenesis, drug resistance and so on and provides a reference for cancer research workers.
5.The role of TF-1 cell apoptosis-related gene 19 in systemic lupus erythematosus

AIM: To clarify the role of new apoptosis-related gene, TF-1 cell apoptosis-related gene 19(TFAR19), in the pathogenesis of systemic lupus erythematosis (SLE) and the relationship between TFAR19 and SLE. METHODS: DNA Ladder detection, Western blotting, immunological fluorescence method, ELISA and so on were used to test if ultraviolet B(UVB) could induce HaCaT cell apoptosis and TFAR19 expression. RESULTS: HaCaT cell apoptosis could be detected after 24 hours of 30 mj/cm 2 UVB irradiation. Also, we found that in active SLE patients, the TFAR19 antibody was increased, but not significant compared to the normal control. CONCLUSION: TFAR 19 is involved in the process of UVB induced ketatinocyte line HaCaT apoptosis and SLE pathogenesis.
6.A Novel Mutation p.L461P in KRT5 Causing Localized Epidermolysis Bullosa Simplex

Background:
Epidermolysis bullosa (EB) is a rare genetic disease with widely different clinical manifestations, but the relationship between genotype and phenotype is not fully understood. In the present study, we recruited a Chinese family in which two members had been diagnosed with localized EB simplex (EBS), with clinical manifestation, including blisters and erosions on the soles of the feet since infancy.
Objective:
To identify and confirm the genetic variation in a Chinese family diagnosed as localized EBS.
Methods:
Our study included two patients, other healthy members of the family, and 100 normal controls. Genomic DNA samples were isolated from each participant, and then polymerase chain reaction (PCR) direct sequencing was performed.
Results:
The results of PCR direct sequencing revealed a novel heterozygous missense mutation in codon 461 of exon 7 of KRT5 (c.1382T>C), which led to an amino acid change (p.L461P) in the patients with EBS but was absent in unaffected family members and 100 unrelated control samples.
Conclusion
The present study broadens the mutational spectrum of EBS, and this knowledge could be harnessed for prenatal screening, gene diagnosis, and gene therapy for lo-calized EBS.
7.Non-invasive prenatal diagnosis of Duchenne muscular dystrophy

Objective This paper was designed to investigate the feasibility of non-invasive prenetal diagnosis of Duchenne muscular dystrophy(DMD). Methods The nucleated red blood cells(NRBC) were separated with percoll using a discontinuous density gradient method. The cells were smeared on microscope slides using a cyto-centrifuge and then stained by Wright-Giemsa. NRBCs were detected and individually retrieved into glass capillary pipettes using a micromanipulator under microscopic observation. The whole genome of a single cell was amplified by improved primer extension preamplification(PEP). The procedures for making prenatal diagnosis of DMD and determining the origin of NRBCs proceeded at the same time using sex determination and linkage analysis of several STR loci of dystrophin. Genotypes were analyzed by amplifying the 9 STR fragments using fluorescence-PCR technique and NRBCs origin was further determined. Results A case of DMD in male fetus was diagnosed. Conclusion With the use of the method reported, the non-invasive prenatal diagnosis of DMD is possible.
8.Study on the content of health education in discharged patients with acute myocardial infarction based on transcultural nursing

Objective To construct the content of health education based on transcultural nursing in discharged patients who were acute myocardial infarction (AMI) and had different cultural backgrounds,and to instruct the nurses to choose and formulate the individual health education program and prescription according to patients' cultural background.Methods Twenty experts who worked in tertiary A hospitals for 15 years,had secondary senior positions,were bachelor degree or above in nursing,cardiovascular and social science were chosen and received the expert consultation of Delphi method through the self-designed framework consulting table with three-level core content of the health education.The items on the content of health education at every level were quantitatively analyzed.Results The Cr was 0.865,and the Cs was 0.840,and the Ca was 0.890.The kandall coefficients of concordace at every level were respectively 0.425,0.443,0.408,and chi-square test showed that the difference was statistically significant (P < 0.01).The number of items on the core content of health education through two rounds of expert consultation was determined including 11 items Grade Ⅰ,42items Grade Ⅱ and 126 items Grade Ⅲ.Conclusions The expert opinion of items on the content of health education in discharged patients with AMI at every level is intensive,and the reliabihty of the result is high,and which provides the scientific basis for carrying out effective health education in discharged patients who are AMI and have different cultural backgrounds.
9.Comparison of surveillance efficacy between black box and light trap methods

ObjectiveTo evaluate the field mosquito surveillance efficacy between the black box method and the light trap method. MethodsEight light traps and eight black boxes were placed in each of the eight field sites in Baoshan District of Shanghai, with a minimum distance of 10 meters between each traps. Surveillance was conducted twice a month from May to September 2023. ResultsThe total number of mosquitoes (273) and female mosquitoes (228) captured by the black box method were 0.795 and 0.774 times higher than those caught by the light trap method, respectively. There was no statistically significant difference (P>0.05) between the mosquito density (t=1.723 5, P=0.100 2) and the female mosquito density (t=1.805 9, P=0.101 1), and there was a correlation (P<0.001) between the mosquito density (r=0.942 7) and the female mosquito density (r=0.896 5). There were no significant differences in the composition ratio of mosquito species(χ2=0.100 1, P>0.05) and female mosquitoes(χ2=0.394 4, P>0.05) captured by black box and light trap methods. In four different habitats, the composition ratios of mosquito species captured by the black box and light trap methods were: hospitals (38.10% and 38.77%) > rural housing areas (36.26% and 37.61%) > parks (14.65% and 12.54%)> urban housing areas (10.99% and 11.08%), and the composition ratios of female mosquitoes captured by the black box and light trap methods were: rural housing areas (40.35% and 42.52%) > hospitals (37.72% and 36.05%) > parks (13.16% and 13.27%) > urban housing areas (8.77% and 8.16%). In different habitats, there was no significant difference in the composition ratio of mosquitoes captured by two monitoring methods(χ2=0.5987, P>0.05), and there was no significant difference in the composition ratio of female mosquitoes(χ2=0.2993, P>0.05). ConclusionWith widen application and fewer requirements for equipment, operation skills, and use cost the black box method can be interchanged with the light trap method for monitoring mosquito density, especially in disaster areas or remote areas with difficult in having access to electricity.
10.Experience of hospital-to-home transition in stroke patients:a qualitative meta-synthesis

Objective To systematically evaluate the real-life experience of stroke patients in the hospital-home transition period,and to provide a reference for better clinical development of transitional nursing practice.Methods We searched PubMed,Web of Science,Embase,CINAHL,Cochrane library,CNKI,Wanfang Data,VIP database,and China Biomedical Literature Database for qualitative studies on the real experience of stroke patients in the hospital-to-home transition period from the establishment of the database to October 2023.The quality of the literature was evaluated using the Joanna Brigg Institute(JBI)Australian Centre for Evidence-Based Health Care Quality Assessment Criteria for Qualitative Research(2016),and the results were integrated using meta-integration methods.Results A total of 14 studies were included,and 48 research results were extracted and 9 categories were summarized.The final synthesis included 3 integrated results:the discharge preparation period was in contradiction;after being discharged from the hospital,life changed dramatically and there was a variety of transition barriers;active response to illness and the experience of self-growth.Conclusion The hospital-home transition period for stroke patients is a critical period for patients'rehabilitation,and medical staff should strengthen the implementation of the transitional discharge plan,strengthen the connection between hospitals and families and communities,pay attention to the psychological experience of patients,so as to help stroke patients achieve a smooth transition from hospital to home.
