Hippocampus is an important structure of neural memory circuit , which plays an important role in learning and memory integration in human and animal .Vision is the most important channel for people to accept information , while the vision system is the most common organ contacting information from outside .The retina as an important part of the visual system , plays a key role in human cognition of the objective world .The study found that many of pathological changes of the body will make the patients cognitive function decline , but molecular mechanism remains unclear .With the research on cognitive dysfunction was penetrated , the link be-tween how VEGF , PKC expression in hippocampus and retinal tissue and cognitive dysfunction was reviewed in this article .

Cognitive impairment in diabetes ( CID) is one of the complications of diabetes .The features of mild or moderate cognitive disorder and the decread abilities in memory and studying are the main symptoms of CID .But it′s mechanism is still un-known .NF-κB is the original signal activator , it can activate other signal pathway of cell dysfunction .With the influence of hyperglyce-mia and oxidative stress, the expression of NF-κB is enhanced, that leads to the changes of the expression of NOS , MMP-9, TNF-α, then, these changes lead to the apoptosis of hippocampus neuron cell .This review focuses on NF-κB in order to provide evidences in studying CID by reviewing the relationship between NF-κB and it′s relative factors of NOS, MMP-9, TNF-αand CID.
3.Effect of elemene on radiosensitivity of A549 cells and its possible molecular mechanism

Objective To investigate the effect of elemene on the radiosensitivity of A549 cells and its possible molecular mechanism.Methods The effect of radiosensitivity was detected by colony forming assay.The protein expressions of DNA-PKcs,Bcl-2 and P53 were detected with Western blot.The correlation between the protein expression of DNA-PKcs and Bcl-2,DNA-PKcs and P53 was analyzed.Results Elemene had radiosensitizing effect on A549 cells,with the SERDo and SERDq 1.54 ± 0.20 and 1.43±0.15,respectively for 10 μg/ml elemene,and 1.63 ±0.32 and 1.75 ±0.19,respectively for 20 μg/ml elemene.Compared with irradiation group,the expression of DNA-PKcs was reduced significantly in 10,20 μg/ml elemene combined with radiation group ( t =7.52,8.33,P < 0.05 ),so was for Bcl-2(t =10.74,11.33,P <0.05).The expression of P53 protein increased significantly (t =-9.25,7.66,P <0.05).There was a remarkable negative correlation between the expression of DNA-PKcs and P53(r =-0.569,P <0.05),and a remarkable positive correlation between DNA-PKcs and Bcl-2 (r =0.755,P <0.05 ).Conclusions Elemene has radiosensitizing effect on A549 cells,which might be related to down-regulation of DNA-PKcs gene expression,up-regulation of P53 and down-regulation of Bcl-2.
4.Screening radiosensitizing-related genes mediated by element in lung adenocarcinoma A549 cells by using gene chip

Objective To screen radiosensitizing-related genes mediated by elemene in lung adenocarcinoma A549 cells by using gene chip. Methods MTT test was used to calculate the IC50 of elemene. ①The effect of radiosensitivity was detected by colony forming assay. A549 cells were divided into 2 groups: radiation group and radiation + elemene group. Oligonucleotide chip was used to screen the gene expression changes of A549 cells from these 2 groups. The up-regulated gene Egr-1 and the down-regulated gene CyclinDl were selected to undergo RT-PCR so as to confirm the reliability of the result. Results MTT test showed the elemene inhibited the proliferation of the A549 cells dose-dependently. The IC50 value of elemene on the A549 cells was 120 mg/L. ②10 mg/L elemene had radiosensiting effect on A549 cells.The values of SERDO and SERDq obtained from the survival curve were (1.54±0. 20) and (1.43±0. 15 )respectively. Gene chip screened 122 differentially-expressed genes, including 89 up-regulated genes and33 down-regulated genes. ③These altered genes could be related to cell structure, substance metabolism,cell proliferation, cell differentiation, signal transduction, material transport, DNA repair, apoptosis,immune response and so forth. The RT-PCR results of Egr-1 and Cyclin D1 were consistent with the genenchip analysis.Conclusions The mechanism of elemene enhancing the radiosensitivity of lung adenoearcinoma A549 cells is the result of participation and collaboration of multiple genes. Further study of the newly-discovered differentially-expressed gene helps find out new radiosensitizational targets of elemene.
5.Effect of β-elemene Combined with Radiotherapy on the Expression of KU70 mRNA and Apoptosis of Lung Adenocarcinoma Cell Line A549

Objective: To study the effect of β-elemene combined with radiotherapy on the expression of KU70 mRNA and apoptosis of lung adenocarcinoma cell line A549. Methods: A549 cells were divided into the control group (c), irridation group (IR), β-elemene group (0.1×IC50 and 0.2×IC50 I.e. 10 μg/mL and 20pg/mL β-elemene) and β-elemene combined with irridation group (0.1×IC50 + IR and 0.2×IC50 + IR). IC50 was obtained through MTT method and cell survival rate was analyzed by colony formation test. Cell cycle and apoptosis were detected with flow cytometry. The expression level of KU70 mRNA was detected by RT-PCR. Results: MTT assay showed that IC50 value of A549 cells was 120 μg/mL. The number of cell clones and survival rate of β-elemene groups were declined significantly. The radiosensitivity of A549 cells can be enhanced by β-elemene. The flow cytometry confirmed that the ratio of G_2/M and apoptosis were significantly in-creased under the effect of β-elemene, statistically different from those in the control group (P <0.05). The expression level of KU70 mRNA in β-elemene with radiotherapy group was declined significantly, statistically different from those in the con-trol group (P<0.05). Cendusion: The rediosensitization effect of β-elemene on A549 cells is associated with induction of cell apoptosis and down-regulation of KU70 mRNA expression.
6.Safety and stability of posterior chamber phakic intraocular lens for correction of extreme myopia: A follow-up of 11 cases

BACKGROUND: The phakic intraocular lens implantation has advantages of reversibility and accommodative capability, which is a promising refractive operation. Posterior chamber phakic intraocular lens implantation is a developing method for the correction of extreme myopia. OBJECTIVE: To evaluate the efficacy, safety and stability of posterior chamber phakic intraocular lens implantation for the correction of extreme myopia. METHODS: This study included 18 eyes of 11 patients whose received posterior chamber phakic intraocular lens implantation at Shierming Eye Hospital from March 2006 to January 2009, including 4 males (7 eyes) and 7 females (11 eyes). All patients were followed-up 3-6 months, mean 5.3 months. The refraction, intraocular pressure, contrast sensitivity, glare sensitivity, endothelial cell morphometry, as well as lens opacities were observed. RESULTS AND CONCLUSION: After operation, naked visions of the 18 eyes were better than or equal to preoperative corrective vision, and the astigmatism was relieved obviously. The naked contrast sensitivity and glare sensitivity was better than that before the operation. The anterior chamber depth, intraocular pressure and corneal endothelium counting before and after the operation had no significant difference. No serious complication was detected during the follow-up. The vision decent at night and dazzling sensation occurred in 2 cases. Shot-term observation shows that implanting intraocular lens in posterior chamber to correct high myopia is safe, stable and predictable. This operation is a promising method for correcting high myopia.
7.Construction, expression and activity evaluation of recombinant NuBCP-9/ Tumstatin(74-98) fusion polypeptide

Aim: To construct a prokaryotic expression vector carrying NuBCP-9-tumstatin(74-98) (abbreviated as NT) gene and to obtain the fusion peptide with antitumor activity. Methods: Nucleotide sequences of antitumor peptides, NuBCP-9 and Tumstatin( 74-98), were connected via a linker(G_4S)_3 based on biased codons of E. coli the fused NT gene was reconstructed using SOE PCR, and inserted into pET32a(+) vector, and transformed in E. coli BL21(DE3). After expression, the novel fusion peptide was purified through nickel-affinity chromatogra-phy, Factor Xa digestion and ultrafiltration. Biological activity of the fusion peptide on ECV304 and A549 cells was evaluated by MTT assay. Results: A prokaryotic expression system with NT gene was successfully constructed. The soluble fusion peptide was accounted for approximately 25% when induced by 0. 5 mmol/L IPTG at 30 ℃ for 4 h. The purified fusion peptide could inhibit cell growth of ECV304 and A549 with inhibition rates of 60. 8% and 65. 2% at 20 μmol/L, respectively. Conclusion: A novel fusion peptide with antitumor activity was cloned, expressed and purified.
8.Effect of follow-ups on medical compliance behaviors and quality of life of patients with gastroesophageal reflux disease

ObjectiveTo investigate the effect of follow-ups on medical compliance behaviors and quality of life of patients with gastroesophageal reflux disease (GERD).Methods One hundred GERD patients who were hospitalized in our department from June 2012 to June 2013 were divided into intervention group (n=50) and control group (n=50) according to the random number table. Both groups received the same drug treatment and conventional nursing during hospitalization.The control group received periodically outpatient follow-ups,while the intervention group received professional follow-ups by trained nurses after discharge from hospital. The compliance behaviors and quality of life in these patients were assessed and statistically analyzed after 6 months nursing intervention or outpatient follow-up.Results With the implement of follow-up in patients with GERD,the intervention group was better than the control group in compliance behaviors,including the decrement of smoking and drinking,strong tea,coffee,having midnight snack and spicy food (P<0.05),but there was no difference in the behavior of staying-up between the two groups.Concerning the quality of life,the intervention group was significantly better than the control group in the 6 items excluding the physical and physiological functions (P<0.05).Conclusion Professional follow-ups can improve compliance behavior and quality of life of GERD patients.
9.Predictive value of 18F-FDG PET-CT for lymph node metastasis of peripheral lungadenocarcinoma

Objective Classification of non-small cell lung lymph (NSCLC) node (N) is one of the key factors influencing treatment, however, the cilinical noninvasive and invasive approaches to N classification have their limitations.This study aimed to investigate the risk factors of lymph node metastasis of peripheral lungadenocarcinoma by using CT and PET / CT scans.Methods Retrospective analysis had been done on a total of 248 patients who underwent surgical resection from February 2010 to November 2015 in our hospital.All of them underwent chest CT and 80 patients underwent PET/CT examination.Univariate analysis was applied in the relation of lymph node metastasis to gender, age, smoking situation, CEA, SUV, cancer size, pathological variants, and the degree of differentiation.Multivariable logistic regression analysiss were performed in the prediction of risk factors for lymph node metastasis.ResultsSeventy-four patients (29.8%) had regional lymph node metastases.Univariate analysis showed that lymph node metastasis was related to the serum CEA level, degree of differentiation, SUVmax, tumor size, lobulation/spiculation, pleural retraction, mediastinal or hilar lymphadenopathy (P<0.05).In the multivariable analysis of risk factors, including serum CEA, SUVmax and CT features, for predicting lymph node metastasis, the most important and significantly independent risk factors identified were SUVmax, CEA level, mediastinal or hilar lymphadenopathy, cavitation/bubble-likelucency and pleural retraction (P<0.05).Conclusion The lymph node metastasis is associated with SUVmax of primary tumor, serum CEA level, mediastinal or hilar lymphadenopathy, cavitation/bubble-likelucency and pleural retraction.The combination of radiographic features and serum CEA can help to predict more accurately the risk of lymph node metastasis in patients with peripheral lung adenocarcinoma.
10.Research Progress of the Relationship between Cognitive Impairment in Diabetes and NMDA Receptor and Subunits NR2

Diabetic cognitive impairment is one of the chronic complications of diabetes,and the pathogenesis has not yet been fully clarified. In recent years, more and more studies of CID showed that the changes of NMDA receptor and subunits NR2A and NR2B might be important during the period of the diabetes,and they are associated with the mechanism of cognitive impairment. This article makes a summary on these researches so as to lay the foundation for a further study.
