1.Effect of Moxa Smoke on Amino Acids Neurotransmitters in Brain of Senescence Accelerated Mice

Objective To observe the effect of moxa smoke (Artemisia vulgaris) on amino acids neurotransmitters in the brain of senescence accelerated mouse (SAMP8) and explore the anti-aging effect of moxa smoke. Methods Totally 70 SAMP8 were randomized into one model control group and 6 intervention groups, and 10 SAMR1 mice were used as normal control group. There were low, medium and high concentrations of moxa smoke during intervention. Moxa smoke intervention was performed 15 min/30 min each day for 28 days. High performance liquid chromatographic method was used to determine the levels of Glu, Asp and GABA in the brain of the mice. Results Compared with the normal control group, the levels of Asp and Glu in the model control group were significantly higher, while the GABA was significantly lower. The levels of Asp and Glu in 6 intervention groups were significantly lower than those in the model control group, while GABA was significantly higher than or the same as the model control group. There was no significant difference among different intervention groups in the levels of Asp and Glu, while the 30 min-medium concentration moxa smoke had the most significant effect in increasing the level of GABA. Conclusion Moxa smoke intervention could decrease the high level of Asp and Glu, and increase the low level of GABA in SAMP8. It could achieve the effect of anti-aging through adjusting the abnormal metabolism of amino acids in the brain.
2.Study on mechanism of active ingredients in Sanhuang Xiexin Decoction on acute kidney injury induced by lipopolysaccharide

Objective To research on reversal mechanism of active ingredients of Sanhuang Xiexin Decoction on lipopolysaccharide-induced acute kidney injury.Methods C57BL/6 mice were randomly divided into 6 groups: control group, LPS group, rhein group, berberine group, baicalin group and dexamethasone group.8 mice in each group.Control group were injected intraperitoneally with 0.9%NaCl 0.5 mL, and the remaining groups were injected intraperitoneally with LPS 0.1 mL/g.The mice of rhein group, berberine group, baicalin group and dexamethasone group were respectively injected intraperitoneally rhein, berberine, baicalin ( 0.1 mL/g ) and dexamethasone ( 0.0001 mL/g ) 2 h before LPS injection.One day after LPS treatment, mice were decapitated and blood collected, diagnostic kits and automatic biochemical analyzer were used to detected urea nitrogen and creatinine.TNF-αand IL-6 concentrations in serum were detected by ELISA kit.HE staining and immunohistochemistry were performed in the kidney tissues.expression of apoptosis protein detected by Western blot.ResuIts Compared to control group, the blood urea nitrogen, serum creatnine, TNF-αand IL-6 level were significantly lower ( P<0.05 ) .HE staining found the renal tubular epithelial cell which treated with LPS were swelling, degeneration, loss, narrow lumen and tube type and exudate, renal interstitial edema, inflammatory cells infiltration.And symptoms of the other group were significantly slight.Immunohistochemical results showed that the expression of Bax and Caspase3 in LPS group was obviously increased, while the expression of the others group was decreased, neither did Bcl-2.Western blot results showed that apoptosis proteins Bax, Cleave-caspase3, CytoC expression increased of LPS group, while the other groups were lower than LPS group.But Bcl-2 expression reduced in LPS group and increased in others group.ConcIusion The effective constituent of Sanhuang Xiexin Decoction can reversal of LPS induced acute kidney injury and its mechanism of action is similar to that of dexamethason.
3.Effect of different doses of lidocaine on expression of HMGB1 in small intestine in septic rats

Objective To investigate the effect of different doses of lidocaine on expression of the high mobility group box 1 protein (HMGB1) in small intestine in septic rats.Methods Fifty adult male Wistar rats weighing 200-250 g were randomly divided into 5 groups ( n =10 each):sham operation group (group S),sepsis group( group Sep ) and different doses of lidocaine group (group L1~3 ).Group S were not applied cecal ligation and puncture (CLP).Sepsis intestinal damage model was performed in group Sep by CLP.Group L1~3 were given intraperitoneally lidocaine in a dose ofS,10 and 20 mg/kg at immediately,1 and 2 h after CLP,respectively.Isometric normal saline was given intraperitoneally in group S and group Sep.The small intestine tissues were taken at 24 and 48 h after CLP.The small intestine morphology was observed with optical microscope.The expression of the HMGB1 mRNA were examined by PCR and the activity of diamine oxidase (DAO) were determined by ELISA.Results Compared with group S,the expression of HMGB1 mRNA was increased and the activity of DAO decreased in group Sep and groups L1~3 at 24 and 48 h after CLP ( P < 0.05 ).Compared with group Sep,the expression of HMGB1 mRNA was decreased and the activity of DAO increased in a dose-dependent manner in groups L1~3 ( P <0.05),The injury of pathology of small intestine was slighter in groups L1~3 than in group Sep.Conclusion Lidocaine can reduce samll intestine injury through inhibiting HMGB1 expression in septic rats by a dose-dependent manner.
4.A Survey on Epidemic Situation and Associated Risk Factors of Hypertension Among the Empty-Nest elderly in Shanggang commnnity of Shanghai

Objective To study the epidemic situation and associated risk factors of hypertension among the empty-nest elderly in Shangha of community. Methods Using pure random sampling method, a face-to-face questionnaire survey and the blood pressure measuring of the empty-nest elderly aged 60-year-old or above in the two communities of Shanggang were performed by trained investigators. Results There are 634 empty-nest elderlies in the two communities. The prevalence rate of hypertension was 51.7% in the subjects. The associated risk factors of hypertension among the empty-nest elderly include smoking, drinking, excess salt diet, lacking of sports and family history of hypertension. There was significant difference between the groups of hypertension and non-hypertension in the distribution of keeping healthy behaviors. The rate of awareness, management and control were 62. 8% ,49. 2%, and 28.7% respectively in the hypertension subjects. Conclusion The prevalence rate of hypertension among the empty-nest elderly is high, Public health education programs, multiple prevention and intervention measures of hypertension should be strengthened among the empty-nest elderly.
5.Effects of different doses of lidocaine on acute liver injury in septic rats

Objective To investigate the effects of different doses of lidocaine on acute liver injury in septic rats.Methods Fifty male Wistar rats,weighing 200-250 g,aged 8-10 weeks,were randomly divided into 5 groups(n =10 each):sham operation group(group S),sepsis group(group CLP),and different doses of lidocaine groups(groups L1-3).Sepsis was induced by cecal ligation and puncture(CLP)in anesthetized rats.At 0,1 and 2 h after CLP,lidocaine 5,10 and 20 mg/kg(in normal saline 0.5 ml)were injected intraperitoneally in groups L1-3 respectively,while normal saline 0.5 ml was given in groups S and CLP.At 24 h after CLP,blood samples were taken for determination of the plasma alanine aminotran sferase(ALT)concenlralion.The rats were then sacrificed,and the liver was removed for microscopic examination and determination of the hepatic high-mobility group box 1 protein(HMGBI)mRNA expression.Results Compared with group S,the plasma ALT concentration was significandy increased and hepatic HMGBI mRNA expression was up-regulated in groups CLP and L1-3(P < 0.05).Compared with group CLP,HMGBI mRNA expression was down-regulated in groups 14-3,while the plasma ALT concentration was decreased in groups L2 and L3(P < 0.05),The plasma ALT concentration was significantly decreased and HMGBI mRNA expression was down-regulated in groups L2 and L3 com pared with group L1,and in group L3 compared with group L2(P < 0.05).The microscopic examination showed that the pathologic changes were attenuated in groups L1-3,and the changes were least severe in group L3.Concluslon Lidocaine can reduce acute liver injury in septic rats,this effect is dose-related,and inhibition of hepatic HMGBI mRNA expression is involved in the mechanism.
6.Expression of tissue inhibitor of metalloproteinase-4 in human cutaneous malignant melanoma tissue and its relationship with melanoma initiation, invasion and metastasis

Objective To detect the expression of tissue inhibitor of metalloproteinase-4 (TIMP-4) in cutaneous malignant melanoma (CMM) tissue and to assess its relationship with melanoma proliferation,invasion and metastasis.Methods Western blot was conducted to measure the protein expression of TIMP-4 in five fresh lesional and paratumoral tissue specimens of CMM and three fresh tissue specimens of nevi.Immunohistochemistry was carried out to quantify the expression of TIMP-4,Ki-67,matrix metalloproteinase-2 (MMP-2),vascular endothelial growth factor (VEGF) and CD63 in paraffin-embedded tissue samples from 43 cases of CMM and 51 cases of nevi.The degree of malignancy of melanoma was evaluated in these lesions.Results Western blot analysis showed that the expression of TIMP-4 was significantly higher in 4 of 5 CMM tissue specimens than in corresponding paratumoral tissue specimens and nevus tissue specimens.Immunohistochemistry revealed that the expression rate of TIMP-4 was 86.04% (37/43) in melanoma tissue,compared to 19.6% (10/51) in nevus tissue (x2 =31.55,P < 0.05).The expression of TIMP-4 increased sequentially from in situ melanoma to invasive and metastatic melanoma (rs =0.309,P < 0.05).As far as CMM was concerned,the TIMP-4 expression was uncorrelated with any of the known prognostic variables including clinical stage,Clark level,Breslow depth,presence of ulcer,and Ki-67 expression (all P > 0.05),but positively correlated with the expressions of VEGF (rs =0.345,P < 0.05) and CD63 (rs =0.555,P < 0.01).The median expression level of TIMP-4 was significantly higher in MMP-2-positive than in MMP-2-negative melanoma tissue samples (3 vs.0,P < 0.01).Conclusions TIMP-4 protein is highly expressed in CMM tissue,which may be closely associated with the initiation and progression of CMM,especially with the metastasis of and angiogenesis in CMM.
7.Effect of catalpol on RAW264.7 macrophage polarizationmediated by AGEs-stimulated mouse mesangial cells

Aim To investigate the effect that catalpol intervenes macrophage polarization mediated by mouse mesangial cells(MMCs) stimulated by advanced glycation end products(AGEs).Methods RAW264.7 macrophages and MMCs were co-cultured in vitro and divided into model group(100 mg·L-1 AGEs), control group(100 mg·L-1 BSA), catalpol(0.1, 1.0, 10.0 μmol·L-1) group, and aminoguanidine(1.0 μmol·L-1) group which was set as positive control.After being incubated with catalpol for 1 h, MMCs were stimulated by AGEs for 23 h.The proliferation-inhibition rate of MMCs was measured by MTT assay.MCP-1 in supernatant liquid of MMCs was detected by ELISA method.The expression of iNOS, CD16/32, TNF-α, COX-2, CD206 and Arg-1 was detected by Western blot.Simultaneously, the percentage of iNOS and CD206 was also measured by flow cytometry.Results AGEs could increase the level of MCP-1 secreted by MMCs.The expression of iNOS, TNF-α, CD16/32 and COX-2 protein of macrophage was up-regulated after MMCs stimulated by AGEs, while the expression of CD206 and Arg-1 was down-regulated.After being intervened by catalpol, these effects could be reversed.All the changes were concentration-related.Conclusions Catalpol can inhibit macrophages M1-type polarization process and promote M2-type polarization, which may be mediated through MCP-1 secreted by MMCs after AGEs stimulation.Catalpol can ameliorate inflammation and relieve diabetic kidney injury.
8.The relationship between total IgE and atopic dermatitis in maternal serum and neonatal umbilical cord blood and allergen testing

Objective To explore the correlation of total IgE and childhood atopic dermatitis (AD) in maternal serum and newborn cord blood, as well as its clinical significance of allergen testing. Methods Thirty-five cases diagnosed as AD (AD group) were selected, and other 35 children who were not diagnosed as AD (control group) were randomly selected from a birth cohort established in 2009—2011. The total IgE levels were detected by ELISA in maternal serum and newborn cord blood. The serum specific IgE antibody level was detected by quantitative immunoblotting method. Results The serum total IgE level was significantly higher in mother and newborn cord blood in AD group than that in control group (χ2=16.568 and 14.933, P<0.01). Compared to control group, there was a significantly higher positive rate of mother serum allergen includ?ing dust mites, house dust, ragweed pollen, song kind of pollen, poplar, surname and elm pollen, mould, shrimp, marine fish, in AD group (P<0.05). There was a significantly higher positive rate of artemisia pollen and fungi IgE in newborn cord blood in AD group (P<0.05). Conclusion The increased total IgE in maternal serum may play a predictive effect on infants suf?fering from AD. There is no obvious consistency in allergic state between mothers and infants.
9.Association between Norovirus epidemics and school toilets design and management in Chengdu City

Objective:
To analyze the relationship between the toilet design and sanitary condition of primary and secondary schools in Chengdu and the epidemic situation of Norovirus infection, so as to provide scientific references for school toilet design and Norovirus prevention and control.
Methods:
A total of 78 cases of norovirus epidemics from 2019 to 2020 were included in this study. These epidemics happened in 78 schools and the causes were indicated as human-to-human transmission. The Chi square and Mann-Whitney U test were used to compare categorical and continuous data respectively. Unconditional binary Logistic regression was used to examine the multivariate associations.
Results:
Most epidemics happened from October in 2019 to March in 2020(79.49%, 62 cases), in primary schools (71.79%, 56 cases) and in the center area (52.56%, 41 cases). The median of the case number in each epidemic was 10. Overall, 56 schools (71.79%) were equipped with toilets which were flushed independently, and 22 schools (28.21%) were equipped with toilets which were flushed uniformly. There were 27 schools (34.62%) that did not have enough water taps. Logistic regression analysis found that those with toilets flushed uniformly had a stronger epidemic, compared to the schools with toilets flushed independently( OR=5.53, 95%CI=1.63-18.76, P <0.05).
Conclusion
In order to prevent intestinal infectious diseases , it is suggested that schools should design or reconstruct independent flushing toilets with adequate faucets.
10.Related risk factors of posterior circulation cerebral infarction and its prognostic factors

Objective To probe into the risk factors associated with posterior circulation cerebral infarction,and to evaluate the factors affecting the prognosis of posterior circulation cerebral infarction.Methods One thousand and five hundred thirty-six patients with cerebral infarction,admitted to our hospital from June 2010 to June2012,were chosen in our study; 218 patients were excluded according to the exclusion criteria; 410 patients were eliminated for incomplete information (time of onset,admission systolic blood pressure,diastolic blood pressure,glycated hemoglobin,fasting glucose,2 h postprandial glucose,previous use ofhypoglycemic agents,previous use of insulin,coronary heart disease,atrial fibrillation [AF],smoking,previous infarction or transient ischemic history of seizures,cerebral family history,triglycerides,total cholesterol,high density lipoprotein,low density lipoprotein,apolipoprotein A1,fibrinogen [FIB],and infarct type).The 908 eligible patients were divided into posterior circulation cerebral infarction group (n=431) and anterior circulation cerebral infarction group (n=477) according to their MRI features.Then the 431 patients with posterior circulation cerebral infarction were divided into possible good prognosis group (n=140) and possible poor prognosis group (n=291) according to their Barthel indexes on admission.All data were analyzed with Logistic regression analysis.Results Occurrence of posterior circulation cerebral infarction were mainly related to HbA1c≥ 8.5% (OR=1.919,P=0.004,95%CI:1.230-2.993) and history of using insulin (OR=2.108,P=0.089,95%CI:0.892-4.982).And that of anterior circulation cerebral infarction was mainly related to AF (OR=0.655,P=0.028,95%CI:0.449-0.955).The possible prognostic factors of posterior circulation cerebral infarction were HbA1c≥ 8.5% (OR=2.502,P=0.013,95%CI:1.217-5.147),FIB (OR=2.318,P=0.001,95%CI:1.381-3.891),time of onset (OR=2.120,P=0.011,95%CI:1.191-3.772),and age (OR=1.022,P=0.015,95%CI:1.004-1.039).Conclusion In the prevention of stroke,HbAlc can be one of the clinical biomarkers in judging stroke risk and intervention effect.
