1.ENDOTHELIAL CELL DERIVED CYTOKINES IN THE LIVER AND LUNGS DURING SEPSIS

To investigate the role of endothelial cell derived cytokines in sepsis induced liver and lung injury. Sepsis was induced in mice by cecal ligation and puncture(CLP). Sham operation group underwent the same manipulation but without CLP. The gene expressions of pro inflammatory cytokines (TNF?, IL 1? and IL 6) in pulmonary and hepatic microvascular endothelial cells were assessed by RT PCR. The results showed that TNF? and IL 1? mRNA levels markedly increased at 3h after CLP in both pulmonary and hepatic microvascular endothelial cells. Though at 12h after CLP both of them declined, the levels were still higher than those in sham group. IL 6 mRNA expression also increased significantly at 3h after CLP in both pulmonary and hepatic microvascular endothelial cells. At 12h after CLP, the expression of IL 6 mRNA in hepatic microvascular endothelial cells remained at a high level, while the level in pulmonary microvascular endothelial cells became higher. It suggested that endothelium is an important source of pro inflammatory cytokines in the liver and lung tissues during sepsis.

Myofascial pain syndrome (MPS) is a common skeletal muscle pain and dysfunction,characterized by the presence of myofascial pain points (MTrPs).At present,the main method of locating MTrPs is palpation which has lower intra-rater reliability,and lacks an objective evaluation approach for therapeutic effects.Therefore,the research on objective evaluation of the characteristics of MPS has been receiving great attention.These studies involve stiffness of trigger points,fascia thickness,surface temperature and other aspects through various methods,such as ultrasonic imaging,magnetic resonance imaging,infrared thermal imaging.In this paper,the research progress and methods of MPS in imaging were surveyed,which can provide the basis for the clinical diagnosis and objective evaluation of therapeutic effects.
3.Effect of erythrocytes in hypercholesterolemia rabbits irradiation by low level 650 nm laser

Objective To study the structure and function changes of erythrocyte in hypercholesterolemia rabbits that treated with irradiation in auricle root by low level 650 nm laser.Methods Sixteen male rabbits were selected to establish the model animal of hypercholesterolemia,successful model rabbits were divided into 2 groups (model and treatment groups),6 rabbits per group,and 6 normal rabbits were selected as control group.650 nm laser of 100 mW were given in auricle root in rabbits of treatment group for 16 weeks (6 d one week,twice a day,30 min one time).Blood from ear vein was collected every 4 weeks to perform routine and biochemical blood examination,and one-drop blood test under microscope was used for blood accumulation observation.Results Total number of red blood cells of treatment group decreased,while model group showed an increasing trend than normal group (P<0.05).Hemoglobin in three groups were lower than other two groups (P>0.05).Hematocrit (HCT) in treatment group were lower than that in normal group,while HCT in model group were higher than that in other two groups and showed a higher aggregation degree also.Less than model group,treatment group showed less aggregation degree but with higher total bilirubin.Conclusions Low level laser of 650 nm showed an effect on total number of erythrocytes,HCT and red blood cells aggregation,high dose of laser might be injury for erythrocytes and lead to total bilirubin increasement.
4.Xenotransplantation of Microencapsulated Rabbit Parathyroid Tissue for Treatment of Hypoparathyroidism In Rats

Objective To investigate the effect of xenotransplantation of microencapsulated rabbit parathyroid tissue in different sites in rats for the treatment of hypoparathyroidism. Methods The parathyroid glands from Wistar rats were removed to make them aparathyroid. Ultimately, sixteen rats were included because their serum calcium values were continuously below 1.6 mmol/L. We also encapsulated the cultured rabbit parathyroid tissue with alginate-BaCl2 microcapsule. According to the transplantation sites, rats were randomly divided into two groups: renal adipose microcapsule group and peritoneal microcapsule group, eight in each group. Encapsulated rabbit parathyroid tissues were then transplanted accordingly to different microcapsule groups. The calcium serum contents were examined on 5,15,25,35,45,55 and 65 d respectively after transplantation and the grafts were observed through electron microscope on the 65 d in particular. Results The calcium contents after transplantation in renal adipose microcapsule group restored to normal and the observation outcomes of grafts showed that they survived well. The calcium contents of posttransplantation in peritoneal group also restored to normal with an exception that it dropped to a level lower than 1.6 mmol/L on the 65 d. Electron microscope also showed that there were necrotic tissues in the center and only a few cells survived on the edge of the grafts. Within peritoneal microcapsule group, the values were significantly lower than others taken at different phases. Conclusion Microencapsulated rabbit parathyroid tissue that was xenotransplanted into rats can survive and function without administration of immunodepressant. There are significant differences of calcium contents at varying phases between two transplantation sites, which demonstrate that renal adipose may be an optimal site for microcapsule xenotransplantation.
5.Cytokine genes expression of hepatic sinusoidal endothelial in mice with sepsis

Objective To investigate the role of cytokines genes expression of liver sinusoidal endothelial cells in liver damage by sepsis.Methods Septic mice models were established with cecal ligation and perforation (CLP), while sham operation group received the same procedure exclusive CLP. The genes expression of TNF?, IL 1? and IL 6 in liver sinusoidal endothelial cells were assessed by RT PCR. Results A significant increase of TNF?, IL 1? genes expression was observed at 3h, and a slight decline at 12h after operation, but still significantly higher than that in the sham group; while IL 6 gene expression showed signficantly higher at 3h and remained at the high level at 12h. Conclusions Liver sinusoidal endothelial cell is an important source of cytokine production in mice with sepsis.
6.Study of ironizing radiation sensitivity decrease of stem cell induced by 635 nm low level laser

Objective To study the radioresistance increased by low level laser irradiation (LLLI) in stem cells.Methods Umbilical cord mesenchymal stem cells (UC-MSCs) were divided into control group and experimental groups (LLLI only,γ-ray irradiation only and LLLI combined with γ-ray irradiation groups).635 nm (10 mW/cm2,12 J/cm2) laser irradiation was applied using for 3 days (twice a day),and cells were exposed in 2 Gy dose γ-ray irradiation on the third day.DNA injury was detected by alkaline single cell gel electrophoresis,reactive oxygen species (ROS),superoxide dismutase (SOD) and catalase (CAT) activity were valued using detection kits.Results DNA injury in LLLI combined with γ-ray irradiation group was lower than that in γ-ray irradiation exposed group,activity of oxidation stress kinases was enhanced,and level of ROS was inhibited.Conclusions Ironizing radioresistance of UC-MSCs can be increased after treatment of 635 nm LLLI,which will induce the radiation sensitivity of transplanting cells.
7.Design of a dual wavelength multi-channel laser acupuncture instrument

Objective To design a laser acupuncture instrument with dual wavelength and multi-channel,under the guidance of traditional Chinese medicine acupuncture theory and according to low-level laser induced effect in bio-tissues and compatibility characteristics of acupuncture points.Methods Using SCM and semiconductor laser,hardware circuitry was designed to achieve the basic functions of the instrument,and the corresponding PC software was developed.Results The laser acupuncture treatment for several acupuncture points could be carried out at the same time with the same or different laser parameters (wavelength,power density,continuous/pulse),and each channel could output 635 nm and 808 nm laser,with continuous adjustable power.Conclusions The dual wavelength multi-channel laser acupuncture instrument can meet the needs of compatibility during clinical acupuncture treatment and is easy to operate.
8.Relationship between HLA antibodies strength and C1q binding ability

Objective To investigate the relationship between HLA antibodies strength and complement-binding ability in sensitized renal patients waiting for renal transplantation.Method Serum samples of 31 sensitized renal patients waiting for renal transplantation were retrospectively analyzed by single-antigen bead array (SAB) to identify HLA antibodies and in parallel by C1q-SAB to determine the complement binding of HLA antibodies.Result C1q-positive HLA antibodies had significantly higher MFI than C1q-negative HLA antibody (for Class Ⅰ,11052 ± 3291 vs.4506 ± 2960,P<0.05;for Class Ⅱ,13347 ± 4076 vs.4448 ± 3602,P<0.05).The mean fluorescence intensities (MFI) of IgG-SAB were correlated with the MFI of C1q-SAB for the same antibodies (Spearman correlation; Class Ⅰ,r =0.665,P < 0.01 ; Class Ⅱ,r =0.761,P < 0.01).Receiver operating characteristics (ROC) curve analysis showed that the MFIs of HLA antibodies by IgG-SAB could predict their C1q-binding abilities [area under the curve (AUC)Class Ⅰ =0.917; AUCclass Ⅱ =0.927).Using MFI cut-off value of 8238 and 6754 for HLA Class Ⅰ and Class Ⅱ antibodies,respectively,the sensitivity and specificity for C1q binding were 82.4% and 87.4% for Class Ⅰ antibodies,and 90.9% and 82% for Class Ⅱ antibodies,respectively.Conclusion The MFI of HLA antibodies by IgG-SAB can predict the C1q binding capability at a certain extent before transplantation.
9.ANALYSIS OF CT IMAGE AND EFFECT OF ANTI-CYSTICERCUS THERAPY FOR 300 PATIENTS WITH CEREBRAL CYSTICERCOSIS

Objective To understand the effect of anti-cysticercus therapy for patients with cerebral cysticercosis and the changes of cysticercus on CT image after treatment. Methods The patients with cerebral cysticercosis were classified by the presentation of their brain CT image before treatment, then the effect of anti-cysticercus therapy on them after treatment was analyzed and the presentations of their brain CT images between before and after treatment were compared. Results There were different changes on CT image of cysticercus in brain tissues after anti-cysticercus therapy for different types of patients with cerebral cysticercosis. Type Ⅰ: the focus was absorbed completely after treatment in the majority of patients and calcificated in the minority. Almost all the patients were cured clinically after anticysticercus therapy. Type Ⅱ: the focus was absorbed completely in the minority, and one to two or more calcification dots were observed in the majority of patients. Anti-cysticercus therapy was effective. Type Ⅲ and Ⅳ: the absorption of focus was not very good and the effect of anti-cysticercus therapy was lower relatively. Conclusion The changes of CT image such as absorption, calcification, has important significance in forecasting prognosis and instructing clinical usage.
