1.Effect of sevoflurane anesthesia on DNA methyltransferase mRNA expression in neonatal rat amygdala

Objective To investigate the effect of sevoflurane anesthesia on DNA methyltransferase (DNMT) mRNA expression in neonatal rat amygdala. Methods Forty-two 8-day-old SD rats were randomly divided into 2 groups ( n = 21 each): control group and experimental group. 5% sevoflurane was inhaled for 1 min, and then the inhaled concentration of sevoflurane was decreased to 3 % and maintained for 4 h. The rats were sacrificed at the end of sevoflurane inhalation and 24 h after the end of sevoflurane inhalation, and amygdala was removed for determination of DNMT, mRNA, DNMT3, mRNA and DNMT3b mRNA expression by RT-PCR. Blood samples were taken at the end of sevoflurane inhalation for blood gas analysis. Results Compared with control group, the DNMT, mRNA and DNMT3, mRNA expression was down-regulated (P < 0.05). There were no significant differences in DNMT3b mRNA expression and parameters of blood gas analysis between the two groups (P > 0.05). Conclusion Sevoflurane anesthesia can down-regulate DNMT, mRNA and DNMT,, mRNA expression in neonatal rat amygdala, which may result in functional deficits during the development of central nervous system.
2.The relationship between peritoneal lavage cytology in gastric carcinoma and lymph node metastasis

Objective To explore the relationship between peritoneal lavage cytology in gastric carcinoma and lymph node metastasis.Methods From June 2012 to May 2014,intraoperative peritoneal lavage for cancer cells was carried out in 226 gastric cancer patients undergoing gastrectomy.During the process 63 patients were irrigated with 500 ml of normal saline,while the others were using 1 000 ml.D2+ lymph nodes dissection was performed to evaluate the relationship between peritoneal lavage cytology in gastric carcinoma and lymph nodes metastasis.Results The positive rates of exfoliative carcinoma cells were 11.1% in the 500 ml group and 9.2% in the 1 000 ml group (x2 =0.188,P=0.695).In both groups,rate of exforliative cancer cells were positively related to lymph node metastasis,the numbers of positive lymph nodes,the station of lymph nodes metastasis,the rate of the lymph nodes metastasis and that of the second station (all P < 0.05),but not to the numbers of the lymph node cleaned (P > 0.05).Conclusions Lymph nodes metastasis was an important risk factor that contribute to exfoliated carcinoma cells in patients with gastric carcinoma,and especially in those with lymph nodes metastasis at the second station.
3.Prognostic value of CEA and CA19-9 expression in peripheral blood of patients with advanced gastric cancer

Objective The aims of this study were to investigate the prognostic value of CEA and CA19-9 expression in advanced gastric cancer and the prognostic factors of advanced gastric cancer.Methods A total of 255 patients with advanced gastric cancer who underwent radical R0 radical surgery were enrolled in the Department of Gastrointestinal Surgery,Harbin Medical University Cancer Hospital from January 2010 to December 2010,and were divided into group A(CEA<2.19ng/mL and CA19-9<10.78U/mL),group B(CEA≥2.19ng/mL or CA19-9≥10.78U/mL)and group C(CEA≥2.19ng/mL and CA19-9≥10.78U/mL).These groups were analyzed the differences in gastric cancer patients with clinical pathology and survival,and prognostic factors of patients with advanced gastric cancer factors.Results There were statistically significant differences in clinical N staging,clinical TNM staging and pathological TNM staging in three groups of patients with advanced gastric cancer(P<0.05).There was no significant difference in the median survival time in three groups of patients with advanced gastric cancer(P>0.05).The 1-year,3-year and 5-year survival rates of group A were higher than those of group B and group C(P>0.05).The factors affecting the prognosis of gastric cancer included the degree of tumor differentiation,pathological T staging,pathological TNM staging,the total number of lymph nodes,the number of positive lymph nodes and lymph node metastasis.Conclusion Combined detection of CEA and CA19-9 expression indicates a certain reference value for prognosis of gastric cancer.

5.The impact of activin A neutralizing antibody on mRNA expression of cytokines in murine liver tissue

Objective To verify the effect of activin A neutralizing antibody (AANA) on inhibition of hepatic fibrosis and mRNA expression of various cytokines. Methods Mouse hepatic fibrosis model was induced by CCl_4. Male Kun-ming mice were divided into 5 groups: normal control group,olive oil control group, CCl_4 model group, AANA group and antibody control group. The mRNA expressions of activin A, transfer growth factor (TGF)-?_1, platelet derived growth factor (PDGF) and tissue inhibitors of metalloproteinas (TIMP)-1 in liver tissue were determined by semiquantitative reverse transcription-polymerase chain reaction(RT-PCR) analysis. 6.Effects of epidural capsaicin on pain threshold, neurological function and spinal neurons in rabbits

Objective To evaluate the effects of different doses of epidural capsaicin on pain threshold, neurological function and spinal neurons.Methods Thirty-six male New Zealand white rabbits weighing 2.5-3.0 kg were randomly divided into 4 groups (n = 9 each): control group and 3 capsaicin groups (Cap 1, 2, 3). The animals were anesthetized with 10% chloral hydrate 50-75 mg?kg-1 i.v. . An intrathecal catheter was inserted at L6,7 interspace and correct placement was confirmed by outflow of CSF. 1 ml of 0.1 % ( Cap1) , 0.25 % ( Cap2 ) or 0.5% (Cap3 ) capsaicin was injected intrathecally 24 h after IT catheter was placed. Threshold to noxious thermal stimuli was measured and gait of the hind limbs were assessed using Johnson score (5 = normal, 0 = completely paralyzed), before and 1, 3, 7, 10, 15, 20, 25, 30 days after IT injection. On the 2nd day after IT injection 3 animals were killed in each group and the lumbar segment of spinal cord (L6,7) was immediately removed for light and electron microscopic examination. Although chemotherapy is an effective way for advanced gastric cancer ,it does not significant-ly improve the prognosis of patients .Recently ,further researches on the mechanism of gastric cancer have led to the identification of ′driver gene′and the development of new agents that target these genes .By inhibiting the hu-man epidermal growth factor receptor 2(Her-2)for advanced gastric cancer ,it provides a new treatment for pa-tients.Trastuzumab is a kind of Her -2 monoclonal antibody ,it is the first kind of targeted therapy drug for the treatment of advanced and metastatic gastric cancer .A combination of trastuzumab with cisplatin and fluorouracil based chemotherapy can benefit patients overall survival .Therefore,in this paper,we make a review and summary of a set of recent studies related to the gastric targeted therapy .
8.The Expression of CTGF in Condyle Cartilage under Asymmetric Masticatory Force

Objective To investigate the effect of the asymmetric of masticatory force on the remodeling of bilateral mandibular condylar cartilage of SD rats. Methods The animal models were established respectively by excising the unilateral temporalis, and injecting botulinum toxin A into the unilateral masseter. Dividing three batch processing 3 weeks, 6 weeks, and 9 weeks after the animal modeling, six rats were killed in each group. The expression of CTGF in condylar cartilage was observed by immunohistochemical staining. Results CTGF expressed in proliferative layer,chondroblast layer and hypertrophic layer of the condylar cartilage. The expression of CTGF in the experimental groups was increased than control groups (<0.05) . There was no significant difference between the left and right side in the experimental groups ( >0.05), the expression of CTGF after 6 weeks of operation was stronger than the group after 3 weeks of operation, the expression of CTGF after 9 weeks of operation was the strongest than all of other groups ( <0.05) . Conclusions The asymmetric of masticatory force can up-regulate the expression of CTGF mRNA in rat mandibular condylar chondrocytes. CTGF induces the effect of stress-mediated on the mandibular condylar cartilage remodeling.
9.Investigation of correlation between systemic lupus erythematosus and human cytomegalovirus infection

Objective To study the relationship between systemic lupus erythematosus (SLE) and human cytomegalovirus (HCMV) infection.Methods HCMV was isolated from the peripheral blood leucocytes (PBL) of 63 patients with SLE.Indirect immunofluorescence (IIF) was performed to investigate HCMV pp65 antigen and polymerase chain reaction (PCH) was performed to investigate the expression of HCMV UL54 DNA in cell cultures.The clinical and Iaboratory parameters were also assessed.Results The rate of HCMV infection in SLF patients was higher than that in the healthy controls and SLE patients.It was higher in the active phase than in the inactive phase.The total amount of urine protein in 24 houm collection,ESR,SLEDA1,preyalence of arthritis and some autoantibodies were considerably higher in patients with positive HCMV infection than those with negative HCMV infection.Conclusion The HCMV infection rate in SLE patients is higher than healthy controls.HC:MV infection may contribute to the disease flare in some SLE pahents.
10.The relationship between the single nucleotide polymorphism of aminoimidazole carboxamide ribonucleotide transformylase gene and the treatment of methotrexate in rheumatoid arthritis

Objective To investigate the relationship between the single nucleotide polymorphism of aminoimidazole carbexamide ribonucleotide transformylase gene and the efficacy and toxicity of methotrexate treatment in rheumatoid arthritis. Methods Total of 359 patients with RA were divided into mono-therapy with MTX group, combination therapy with other DMARDs group and other DMARDs combination with no MTX treatment group. The clinical and laboratory measurements were evaluated before therapy and 12, 24 weeks after therapy. Efficacy (evaluated by ACR20) and side effects of the drugs were also assessed. Real-time fluorescent quantitative PCR was conducted to test ATIC 347C/G mutation in RA patients and 340 healthy controls. Results There was no statistical significant difference in 347 CC, CG, GG between RA patients and healthy controls. In the MTX mono-therapy group (n=107), 72% (n=77) there was no statistical significant difference in 347CC, CG, GG between patients with good response and patients without efficacy. 32.7%(n=35) of these patients experienced adverse drug reactions. The ATIC G allele carriers (22.4%) experienced a greater frequency of side effects than the CC carriers (OR=2.672, 95%CI, 1.27~5.59, P<0.05). In MTX combined with other DMARDs group (n=128) and other DMARDs combination without MTX group (n=90), the polymorphism in the ATIC gene was not associated with good clinical response and adverse events (P>0.05). Conclusion There is no statistical significant difference between RA and healthy controls in the ATIC347 gene. Polymorphism in the ATIC gene is not associated with clinical response to MTX treatment, but the ATIC347 G allele is associated with MTX toxicity. It maybe used to predict the adverse drug reactions of patients who take MTX.
