1.Human stomach cancer SGC-7901 cell apoptosis induced by n-butanol extract of Lagotis Brevitaba Maxim

AIM: To study inhibitive effect of Lagotis Brevituba Maxim extracted on human stomach cancer SGC-7901 cells proliferation and/or induce apoptosis. METHODS: Human stomach cancer SGC-7901 cells were treated with Lagotis Brevituba Maxim extracted from n-butanol of 0.34~21.60 mg/mL for 24 h~72 h.Cell proliferation was evaluated by MTT assay.Morphological changes of apoptosis were examined by fluorescence microscope and electron microscope.DNA fragmentation was visualized by agarose gel electrophoresis.The amount of apoptosis cells was measured by flow cytometry. RESULTS: Growth of SGC-7901 cells was obviously inhibited by Lagotis Brevituba Maxim extract with value from 21.60 mg/mL to 0.34 mg/mL.After incubation of SGC-7901 cells with 2.7 mg/mL Lagotis Brevituba Maxim extract for 48 h, morphological changes of apoptosis were observed.DNA ladder was identified by agarose gel electrophoresis of DNA ladder. CONCLUSION: The n-butanol extract of Lagotis Brevituba Maxim can inhibit SGC-7901 cell growth and induce apoptosis.
2.Association of vascular cognitive impairment with acute cerebral infarction in different parts of the brain

Objective To explore the correlation between acute infarctions in different parts of the brain and the risk of vascular cognitive impairment (VCI). Methods 212 patients with acute cerebral infarction were tested using Mini Mental State Examination(MMSE)and Montreal cognitive assessment(MoCA),and were divid-ed into normal cognition group,VCI group,vascular dementia group,and mixed dementia group. We analyzed the gender,age,past medical history,personal history,MMSE and MoCA scores,and imaging data. Results Of the 212 patients,61(28.77%)had normal cognition level,74(34.91%)had VCI,56(26.42%)had vascular demen-tia,and 21(9.91%)had mixed dementia. Multiple regression analysis showed that frontal lobe infarct increased the risk of VCI(OR,41.72)and vascular dementia(OR,48.49);cerebellar infarction also increased the risk of vascular dementia(OR,4.70)and mixed dementia(OR,12.38);and temporal lobe infarction increased the risk of mixed dementia significantly(OR,56.98). Conclusions Approximately 71.3%of the patients with acute cere-bral infarction develop vascular cognitive impairment. The infarcts occurring in the frontal lobe ,temporal lobe and cerebellum increase the risk of VCI significantly ,which should be given interventional therapies.

Underwater exercise and hydrotherapy have rapidly developed in recent years. As an noninvasive and convenient tool, surface electromyography (sEMG) has been applied in underwater research. In favour of experiment procedure, underwater sEMG needs localized waterproof seal or whole-body waterproof suit, and are mainly detected by telemetry system. The muscle activity pattern of underwater exercise is different from on land exercise, the sMEG amplitude is correlated with motion velocity. Compound underwater motion analysis includes sEMG, video and foot pressure will be more effective assessment tool for underwater exercise and hydrotherapy research.
4.Inhibitory effects of siRNA targeting protein kinase CK2?on the invasion of laryngeal carcinoma cells

OBJECTIVE To construct siRNA eukaryotic expression vector targeting protein kinase CK2?and to investigate its inhibitory effect on invasion of the HEp-2 cell line in human laryngeal carcinoma. METHODS siRNA expression vector psiRNA-hH1neo-CK2 targeting protein kinase CK2?was constructed by gene recombination,and then was transfected into the HEp-2 cells by lipofectamine methods. Protein kinase CK2?mRNA and protein of the transfected cells were detected by reverse transcription polymerase chain reaction(RT-PCR) and Western blot,respectively. The invasion of the transfected cells was measured by Boyden chamber.SP method was used to examine the expressions of MMP2 and TIMP2 protein of the transfected HEp-2 cells. RESULTS Protein kinase CK2?siRNA expression vector was successfully constructed by gene recombination. Objective To discuss the protective effects and mechanisms of dexamethasone and Shenfu sepa-rate and joint administration of flap after ischemia-reperfusion injury. Methods 40-month-old fairly healthy rats were randomly divided into 4 group as A, B, C, D, and to product the abdominal island flap, then blocking the flow of blood of the pedicle artery respectively before 30 minutes when injecting with normal saline (1 ml/kg), dexametha-sone (1 ml/kg), Senate (10 ml/kg), joint injection with dexamethasone (1 ml/kg) and Shenfu (10 ml/kg). 4 groups of animals' blood samples were collected from the pedicle vein before the time of I hour when blocking vascu-lar pedicle and reperfusion after the time of 1 h, 6 h, 12 h, 24 h, respectively. Then the plasma concentration of TNF-α was measured. Results The concentration of TNF-α in the treatment group was significantly lower than the blank group(P <0.01). And the group D and the group B、C has significant differences either(P <0.01). Conclu-sion Using Dexamethasone, Shenfu injection in early can reduce the concentration of TNFα in repeffusion injury of flap and has a protective effect on the flap, but make better effect in combined.
6.Expression of vascular endothelial growth factor and cyclooxygenase-2 in laryngeal squamous cell carcinoma and its significance.

In order to study the expressions of vascular endothelial growth factor (VEGF) and cyclooxygenase-2 (COX-2) in human laryngeal squamous cell carcinoma (LSCC) and its significance, the expression of VEGF mRNA and COX-2 mRNA in 62 cases of LSCC and 54 adjacent noncancerous laryngeal tissues and 9 normal human laryngeal mucous tissues was detected by using techniques of semi-quantitative RT-PCR. It was found that the expression level of VEGF and COX-2 mRNA was significantly increased in LSCC as compared with that in the normal human laryngeal mucous tissues (both P < 0.01), and the expression level of VEGF and COX-2 mRNA were significantly increased in stage Ill + IV tissues of LSCC as compared with the stage I + II tissues of LSCC (P < 0.01). There was a high positive correlation between VEGF and COX-2 expression in LSCC (r = 0.756, P < 0.01). These data raise the possibility that VEGF and COX-2 may play key roles in the growth, invasion and metastasis of LSCC.

7.Effect of DRB on the biological characteristics of human laryngeal carcinoma Hep-2 cell line.

In order to study the effect of 5, 6-Dichloro-1-beta-D-ribofuranosyl-benzimidazole (DRB) on the biological characteristics of human laryngeal carcinoma Hep-2 cell line in vitro, Hep-2 cells cultured in vitro were treated with different concentrations of DRB. Changes in cell proliferation, apoptotic rate and invasiveness were detected by MTT assay, flow cytometry (FCM) and matrigel in vitro invasion assay, respectively. It was found that DRB inhibited the proliferation of Hep-2 cells in a dose-and time-dependent manner. After being treated with 0, 10, 20, 40, 80 microm mol/L DRB for 24 h, the apoptotic rate in Hep-2 cells was (0.68+/-0.19)%, (1.95+/-0.12)%, (8.51+/-0.26)%, (11.26+/-0.17)% and (14.99+/-0.32)%, respectively. The matrigel in vitro invasion assay revealed that DRB began to inhibit the invasion of Hep-2 cells at the concentration of 5 microm mol/L, and with the increase of DRB concentration, the inhibitory effect was enhanced. It was suggested that DRB could influence the essential biological characteristics of Hep-2 cells, inhibit Hep-2 cells proliferation, reduce invasive ability and induce apoptosis of Hep-2 cells.
8.Techniques and improvements of establishing orthotopic liver transplantation rat models

BACKGROUND:At present, liver transplantation is the only way to cure end-stage liver disease, but the complications after transplantation is stil an important factor of affecting the long-term survival of patients who received orthotopic liver transplantation, therefore it is necessary to establish a stable animal transplantation model. OBJECTIVE:To establish rat models of orthotopic liver transplantation. METHODS:After inhalation anesthesia with ether, 204 SD rats were perfused with 2-4℃ Ringer’s solution through the abdominal aorta. In order to reduce warm ischemia of the liver, the liver was not turned over before perfusion. The suprahepatic inferior vena cava was cut off along the phrenic ring after perfusion. No further trimming was needed when dressing, so as not to damage the vena cava. The donor liver was removed and preserved in 4℃Ringer’s liquid. The receptor liver was cut off and alogeneic orthotopic liver transplantation was performed using modified two-cuff method. After transplantation, rats could automaticaly turn over and drink water. Surviving more than 3 days is regarded as a successful transplantation. RESULTS AND CONCLUSION:102 liver transplantations were performed in 204 rats, with 86 rats surviving more than 3 days. The success rate of transplantation was 84%. The results demonstrate that rat models of orthotropic liver transplantation can be constructed successfuly through improving techniques.
9.A new caffeate compound from Nardostachys chinensis.

A new caffeate compound, (E)-erythro-syringylglyceryl caffeate (1), was isolated from the roots and rhizomes of Nardostachys chinensis Batal., together with nine known phenolic compounds, including (+)-licarin A (2), naringenin 4', 7-dimethyl ether (3), pinoresinol-4-O-β-D-glucoside (4), caraphenol A (5), Z-miyabenol C (6), protocatechuic acid (7), caffeic acid (8), gallic acid (9) and vanillic acid (10). Their chemical structures were elucidated on the basis of spectroscopic data and physicochemical properties. Furthermore, this is the first report of compounds 2, 5 and 6 from Nardostachys genus.
10.Inhibitory effects of siRNA expression plasmid specific to protein kinase CK2alpha on human laryngeal carcinoma xenograft in nude mice.

OBJECTIVE:
To study the effect on the expression of protein kinase CK2alpha and growth of human laryngeal carcinoma xenograft in nude mice by applying small interfering RNA (siRNA) specific to protein kinase CK2alpha.
METHOD:
Human laryngeal carcinoma Hep-2 cells were implanted under the skin of nude mice. After the tumors grew to a definite size, the tumors were injected with siRNA expression plasmid specific to protein kinase CK2alpha. The weight and volume of subcutaneous tumors were measured. The expression level of protein kinase CK2alpha mRNA and protein of tumors were measured with reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemical technique, respectively.
RESULT:
Protein kinase CK2alpha mRNA and protein expressions were significantly decreased in tumors transfected with siRNA expression plasmid specific to protein kinase CK2alpha (P<0.05). The tumor grew slowly after transfected with siRNA expression plasmid specific to protein kinase CK2alpha (P<0.01).
CONCLUSION
The siRNA expression plasmid specific to protein kinase CK2alpha may suppress the growth and the protein kinase CK2alpha expression of subcutaneous tumors. RNA interfering technology may be a new strategy for the treatment laryngeal cancer.
