1.Effect of quateranary ammonium salt derivative (F2) of haloperidol on calcium in vascular smooth muscle cells

AIM To observe the effect of quateranary ammonium salt derivative (F2) of haloperidol on calcium level in vascular smooth muscle cells (VSMCs) isolated from thoracic aortas of rat. METHODS VSMCs were loaded with Fluo-3-AM, a calcium sensitive fluorescent dye, and [Ca2+]i was determined by the use of laser scanning confocal microscope (LCSM). RESULTS In Ca2+ (1.26 mmol*L-1) bath solution, intracellular calcium fluorescent intensity (FI) in VSMCs was increased when application with 30 mmol*L-1 KCl, and then was inhibited after addition with F2. The FI of the ASMCs with different concentrations of F2 (0.1,1,10,100 μmmol*L-1)within three minutes were 64%±9%(n=16, P<0.05),16%±5%(n=20, P<0.01),0.6%±0.8%(n=20, P<0.01),0.1%±0.3%(n=15, P<0.01), respectively. CONCLUSION F2 could decrease the intracellular Ca2+ level in the VSMCs by blocking the voltage-dependent calcium channel.

3.Application of continuing quality improvement on reducing incidence of PICC-related phlebitis in breast cancer patients undergoing chemotherapy

Objective To investigate the effect of the continuing quality improvement on PICC-related phlebitis in breast cancer patients undergoing chemotherapy. Methods Eighty breast cancer patients undergoing PICC for chemotherapy in our hospital from September to December 2014 were selected as control group, receiving routine nursing care. Another eighty patients undergoing PICC for chemotherapy from January to March in 2015 were selected as experiment group and the quality control cycle was used, the measures including investigating the present situation aiming to theme, goal setting and analyzing, working out and using countermeasures and effect comfirming. Result After applying continuing quality improvement in the experiment group the incidence rate of phlebitis in the continuing quality improvement group was significantly lower than those before continuing quality improvement and in the control group (P<0.05). Conclusion The implementation of continuing quality improvement activities can reduce the incidence of pzce-related phlebitis in patients with breast cancer during chemotherapy. It can improve the safety of PICC catheter and therefore, it is worthy of popularization.
4.Study on the regulation of ERαon NK1R-Tr in breast cancer cells

Objective To analyze the regulation of estrogen receptor α (ERα) on truncated neurokinin-1 receptor (NK1R-Tr), and the influence of this regulation on cell proliferation in estrogen receptor-positive breast cancer cell lines. Methods The chromatin immune coprecipitation (CHIP) was used to observe the transcriptional regulation function of ERαon NK1R-Tr in breast cancer cells. Luciferase reporter gene assay was used to verify whether ERα played a positive regulatory role in the expression of NK1R-Tr. Western blot assay and real-time-PCR were used to detect the expression of ERα and NK1R-Tr in breast cancer cells, MCF-7 and T47D, as well as the expression of NK1R-Tr protein and mRNA level. NK1R-Tr levels were also detected after using estradiol (E2, ERα agonist) and small interfering RNA (knock out ERα). CCK-8 and clone formation experimen were used to detect the proliferation ability of breast cancer cells after knocking out NK1R-Tr with small interfering RNAs. Results CHIP test and Luciferase reporter gene assay proved that ERα can positively regulate the expression of NK1R-Tr via the ERα sequences in the upstream of the NK1R-Tr gene promoter. The expression of NK1R-Tr at both protein level and mRNA level dropped in the estrogen receptor-positive breast cancer cell line MCF-7 upon knocking out ERα. After knocking out NK1R-Tr, the proliferation ability of estrogen receptor-positive breast cancer cells was lower than that of the control group. Conclusion The ERα positively regulates the expression of NK1R-Tr, resulting in the increased cell proliferation in estrogen positive breast cancer cells.
5.Determination of Vitexin in Cajanus cajan(L.) Millsp. by HPLC

Objective To develop a quantitative method to determine vitexin in Cajanus cajan (L.) Millsp. Methods The chromatographic conditions were as follows: column C18(250 mm? 4.6 mm, 5 ? m) , mobile phase being MeOH ∶ 1 % HAC (25 ∶ 75) for 20 mins and then being MeOH ∶ 1 % HAC (30 ∶ 70) after 20 mins, flow rate at 1.0 mL/min, and wavelength at 339 nm. Results The good linearity of this method was in the range of 0.104 4~ 0.522 ? g (r=0.999 4), and the average recovery of vitexin was 100.11 % ( RSD =0.63 % ).Conclusion The method is simple and sensitive with good stability, it can be suitable for the quality control of vitexin in Cajanus cajan (L.) Millsp..
6.Anti-oxidative damage effect in Schisandrin B in mice of Alzheimer's disease and its mechanism

Objective:To investigate the preventive and protective effects of Schisandrin B in the mice of Alzheimer's disease (AD),and to clarify its mechanism.Methods:Fifty Balb/c mice were randomly divided into blank group,model group,pasitive control group,low dose of Schisandrin B group(0.1 g·kg-1)and high dose of Schisandrin B group(0.5 g·kg-1);there were 10 mice in each group.Step-through test was conducted after last administration to detect the latencies and number of errors of the mice in various groups,and the brain tissue was taken.Congo red staining was to detect the morphology changes of cells and neuronal amyloidosis in brain tissue of the mice.The levels of ROS in brain tissue of the mice were tested by Flow Cytometry.The contents of MDA,the levels of LDH,and the activities of CAT,GSH-Px and SOD in brain tissue of the mice were tested by biochemical method.Western blotting method was used to detect the expression levels of signaling pathway proteins Nrf2 and Keap1 in brain tissue of the mice.Results:Compared with model group,the latencies of the mice in low and high dose of Schisandrin B groups were increased (P<0.01) and the number of errors in step-through tests was decreased (P<0.05 or P<0.01).The Congo red staining results showed that compared with model group,the neuronal amyloidosis in brain tissue of the mice in Schisandrin B groups was decreased significantly.Compared with model group,the levels of ROS,LDH and the contents of MDA in brain tissue of the mice in low and high doses of Schisandrin B groups were decreased (P<0.05 or P<0.01),and the activities of CAT,SOD and GSH-Px were increased (P<0.01).Compared with low dose of Schisandrin B group,the content of MDA and the activities of SOD and GSH-Px in brain tissue of the mice in high dose of Schisandrin B group were increased (P<0.001).Compared with model group,the expression level of Nrf2 protein in brain tissue of the mice in low dose of Schisandrin B group was increased (P<0.01),while the expression level of Nrf2 protein in brain tissue of the mice in high dose of Schisandrin B group was decreased (P<0.01);the expression levels of Keap1 protein in brain tissue of the mice in low and high doses of Schisandrin B groups was decreased (P<0.01).Conclusion:Schisandrin B could decrease the level of peroxidation in brain tissue of the mice and reduce the oxidative damage and improve the memory function of the AD mice.The mechanism is related to the activation of Nrf2 signaling pathway which improve the activity of antioxidant enzymes.

"Postpartum Body pain"refers to such symptoms as body pain,numbness,heaviness,arthralgia and even paralysis in a woman during the postpartum period.Besides the above symptoms,the patient may also have other symptoms like sleep disorder,emotional hindrance,somatization symptoms such as fatigue,sleepy,and chest distress,etc.This disease may be considered as somatization symptoms due to postpartum depression,but actually it is a psychogenic disorder.Clinical observation showed there were no clinical changes of objective signs such as red,swelling and fever;Laboratory examinations showed there was no ahnormal manifestation.This article primarily discussed"postpagtum body pain"from medicopsychological angle,hoping to help people get a right recognition of this disease.
8.Quality standard of Fuke Yangkun Pill

AIM:To improve the quality standard of Fuke Yangkun Pill. METHODS: Fructus Viticis was(identified) by micrology,Radix Angelica sinensis,Rhizoma Chuanxiong,Rhizoma Cyperi,Radix Aucklandiae,Rhizoma Corydalis,Radix Rehmanniae,Radix Rehmanniae preparata,Radix Paeoniae alba,Radix et Rhizoma Glycyrrhizae,Fructus Amomi were identified by TLC,baicalin content was determined by HPLC. RESULTS: The TLC spots were fairly clear and distinguishable,the linear range of baicalin was within 3.28-131.2 ?g/mL.The average recovery was 100.4% with RSD 1.0%(n=6). CONCLUSION: The method is simple,and specific,and can be used for quality control of Fuke Yangkun Pill.

Surgical therapy and nursing of a rare case of semi-conjoined deformed twins was summarized. To relieve life threaten, correct deformity and retain function, surgical therapy was performed. Afterwards, patient received nursing strategies including preventing elevation of abdominal pressure, incision care of hemipelvectomy, pelvic fixation, prevention of pressure sores, multi-canal nursing, nutrition support and infection control. Survival of patient was prolonged through a comprehensive therapy and nursing, and the patient was successfully discharged.
