1.Effect of thyroid hormone on the expression of deiodinase and myelin basic protein in brain tissue of preterm rabbit with germinal matrix-intraventricular hemorrhage

ObjectivesTo investigate the effect of thyroid hormone on the expression of deiodinase and myelin basic protein in brain tissue of neonatal rats with germinal matrix-intraventricular hemorrhage (GM-IVH), and its nerve protective effect.Methods Sixty premature rabbits of gestational age 29 days were randomly divided into control, model and intervention groups. The prema-ture rabbits in model and intervention groups were given intraperitoneal injection of 50% glycerol to establish GM-IVH animal model and the premature rabbits in control group were given the same dose of saline. The GM-IVH was screened by cranial ultrasound. The premature rabbits with GM-IVH in intervention group were given intraperitoneal injection of triiodothyronine and the other two groups were given the same dose of saline. At 3 days, 7 days and 14 days after birth, the premature rabbits were killed and the brain tissue were quickly separated. The expression of type 2 iodothyronine deiodinase (D2) and type 3 iodothyronine deiodinase (D3) of periventricular zone tissue were detected by western blot method. The expression of myelin basic protein (MBP) were detected by immunohistochem-ical method.ResultsThe expression of D2 in brain tissue increased gradually in three groups. At the same time point, the expression of D2 was the lowest in the model group, the highest in the control group, and the middle in the intervention group. The differences of D2 among three groups were statistically signiifcant (allP<0.001). The expression of D3 increased gradually in the control and model groups, while it was the highest in the intervention group at day 7. At the same time point, the expression of D3 in the control group was the lowest, it was higher in intervention group than that in the model group at 3 days, then it was higher in the model group than that in the intervention group. The differences of D3 among three groups were statistically signiifcant (allP<0.001). The expression of MBP increased gradually in the control and intervention groups, and it decreased gradually in the model group. At the same time point, the MBP was the lowest in the model group, it was the highest in the control group, and it was the middle in the intervention group. The differences of MBP among three groups were statistically signiifcant (P<0.001).ConclusionsThe expressions of D2 and MBP in the brain tissues of premature rabbit with the GM-IVH increased while the expression of D3 decreased in some extent after thyroxine intervention.

Heart rate variability (HRV) is a non-invasive index for quantitative evaluation of cardiac autonomic nervous function, its analysis methods mainly include linear time domain analysis, frequency domain analysis, and nonlinear analysis. This article review the relationship between stroke lesion type, lesion location, degree of neurological deficit, duration and HRV, as well as the possible mechanism and intervention measures of stroke caused HRV.
4.Nursing of elderly patients with osteoporotic intertrochanteric fracture of femur undergoing artificial femoral head replacement

Objective To summarize the nursing methods and effect of artificial femoral head replacement in treatment of osteoporotic interttochanteric fracture in elderly patients. Methods 55 patients with esteoporotic intertrochanteric fracture were selected in our hospital from February 2007 to February 2011.All patients were cared by targeted preoperative and postoperative nursing,helping them conducting functional training in time,giving discharge counseling and follow-up.The hip functional recovery of patients was evaluated by using the Harris evaluation standard,and the excellent rate of surgery was also evaluated. Results After the targeted care,55 patients went through the perioperative period safely,the average length of stay was ( 14.0 ± 3.1 )days,a total of 5 cases (9.09%)patiants suffered varying degrees of complications which were improved after symptomatic treatment.Patients were followed up for 3 to 36 months,only 2 patients died of cerebral vascular accident,all patients gained good recovery of hip function.Harris score was (84.45 ± 9.38),the excellent rate of surgery reached 78.18%. Conclusions Atificial femoral head replacement proves significant effect in treatment of osteoporotic intertrochanteric fractures in elderly patients,but with the accompanying surgical compliuations,special care and timely functional exercise methods should be paid attention to.
5.Model of Salvia miltiorrhiza extraction by alcohol based on ANFIS

AIM:The model of Salvia miltiorrhiza extracted by alcohol based on adaptive network-based fuzzy inference system(ANFIS)was studied in this paper.METHODS:The ANFIS model was set up by a set of experimental data which were gained on a pilot extraction condition.Then the tanshinone ⅡA content of the alcohol extract from Salvia miltiorrhiza in processing conditions could be predicted by this ANFIS model.The Radix Salvia miltiorrhiza was extracted by alcohol thermal reflux;then initially created a fuzzy inference system model based on network and trained by mixing learning arithmetic.RESULTS:The relative errors between the predicted values and the experimental values were in a allowable range,there were only 4 points greater than 10% of totally 90 calibration points.CONCLUSION:The analytical and experimental results show that the method can effectively forecast the content of the effective ingredient according to preset extraction conditions.
6.The Novel Procedures to Increase the Expression Efficiency of Recombinant Human IL-6 in Eukaryotic Cells

In order to improve the expression efficiency of recombinant IL~6 in eukaryotic cells, we constructed a new vector pcDIIL-6 to express IL-6 in eukaryotic cells on the basis of an eukaryotic vector pcDNA3IL-6 by introducin g the first intron of human?-globin gene into the downstream of CMV promoter in the pcDNASIL - 6. The IL-6 level expressed by pcDIIL - 6 is 7. 5 times higher than that of pcDNA3IL - 6. On the other hand , we studied the influence of pAdVAntage vector co - transfection on the expression level of IL-6 in eukaryotic cells and found the level of IL-6 is 3. 6 times higher than that of pcDNA3IL - 6. The results we got are beneficial to express cytokines with high efficiency in eukaryotic cells.

8.Effect of astragalus parenteral solution on cytokines and angiotensin Ⅱ in patients with congestive heart failure

BACKGROUND: Brain ischemia-reperfusion (IR) injury is closely connected with the activity of Kinesin. Previous research believes that reduced activity of Kinesin, a mierotubule based motor protein, is an early mark for nerve cell death induced by brain ischemia. Erigeron breviscapus can prevent brain IR-induced proteinase C activation, reduce calcium overload, and reduce ischemic infarctional volume, thus attenuating brain IR injury. However, it still remains less reported at present whether the neuroprotective role of erigeron breviscapus is related to Kinesin activity.OBJECTIVE: To observe the effect of erigeron breviscapus on the activity of Kinesin, a microtubule based motor protein, in hippocampal pyramidal cells during brain IR.DESIGN: Randomized controlled study.SETTING: Anesthesia Department, Affiliated Hospital of Jining Medical College; Anesthesiology Key Laboratory, Affiliated Hospital of Xuzhou Medical College.MATERIALS: This experiment was carried out in the Anesthesiology Key Laboratory, the Affiliated Hospital of Xuzhou Medical College, between February and August 1999. Totally 35 male gerbils were included.METHODS: Gerbils were randomized into sham-operation group (n=5), ischemia-reperfusion control group (n=15) and erigeron breviscapus group (n=15), the latter two of which were further divided into three subgroups according to reperfusion time, namely reperfusion group Ⅰ (reperfusion of 6 hours), reperfusion group Ⅱ (reperfusion of 48 hours) and reperfusion group Ⅲ (reperfusion of 96 hours) with 5 in each subgroup. Gerbils in IR group and erigeron breviscapus group were subjected to IR model preparation before experiment by brain arterial occlusion for 10 minutes, while gerbils in sham-operation group had only bilateral common carotid artery isolated without occlusion. Gerbils in erigeron breviscapus group were pretreated 15 minutes before ischemic inducement with intraperitoneal injection of breviscapine (its effective component is erigeron breviscapus) at a dosage of 45 mg/kg, which was replaced with the same volume of isometric normal saline in sham-operation group and IR group. IHC staining was used to detect hippocampus microtubule based motor protein-Kinesin activity with the assistance of computer imaging analysis technology. MAIN OUTCOME MEASURES: Activity and changes of Kinesin of animals in each group.RESULTS: Totally 35 animals were enrolled in this experiment and all entered the result analysis with no one lost during the experiment. In hippocampal CA1 region, Kinesin activity in IR group was found to descend to 58%, 38% and 12% respectively of that in sham-operation group at IR 6 hours, 48 and 96 hours (P < 0.01). In erigeron breviscapus group at IR 6 hours, 48 hours and 96 hours it was 81%, 61% and 21% of that in shamoperation group, and was obviously higher than that in IR control group (P < 0.05). However, the changes of Kinesin activity were not obviously different in hippocampal CA2, CA3 and CA4 regions.CONCLUSION: Erigeron breviscapus can exert brain-protecting function by reducing hippocampal CA1 Kinesin activity during brain IR injury.

10.Clinicopathological characteristics in hereditary nonpolyposis colorectal carcinoma-associated endometrial carcinoma

Objective To study clinicopathological characteristics of hereditary nonpolyposis colorectal carcinoma(HNPCC)-associated endometrial carcinoma(EC).Methods Totally 421 EC patients admitted to General Hospital of Tianjin Medical University from 1981 to 2006 were divided into three groups:group A:sporadic EC;group B:familial aggregation of EC;group C:HNPCC-associated EC.Results HNPCC-associated EC accounted for 6.4%(27/421).Mean age at time of diagnosis was 49.7years in group C,earlier than 56.3 years in group A(P=0.004)and 55.2 years in group B(P=0.035).There were 33.3%(9/27)patients with multiple primary carcinomas in group C.It was higher than 14.3%(9/63)in group B and 5.1%(17/331)in group A respectively(P=0.038,P<0.01).There was no difference among the three groups in histological type or menopausal status(P>0.05).The numbers of patients with low grade EC in group C(70.4%,19/27)and group B(61.3%,38/62)were more than that in Group A(P=0.013,P=0.023).Prognosis for group C was better than that in group A(P=0.021),and 5-and 10-year survival rate in groups A,B and C was 80%,70%;88%,85%;and 96%,96%respectively.Conclusions Mean age at time of diagnosis is earlier in HNPCC-associated EC than that in sporadic EC;patients with HNPCC-associated EC are more frequently complicated with multiple primary carcinomas and of low grade;prognosis is better.
