1.The compliance of schizophrenia patiemts in rehabilitation

Objective To research the compliance in the treatment rehabilitation of outpatients with schizophrenia Method The compliance of 415 cases of schizophrenia were evaluated by face to face interview, and their genaral populations data,antipsychotics use situation,factors affecting compliance were collected by comprehensive interview.The corretaled factors were also analysised.Achi-square statistic was used.Results Among all the enrolled cases,55.9% were fully compliant,23.6% partially compliant and 20.5% non-compliant.Factors affecting compliance were ages,dosage,famillies-environment relationship and personality.Conclusions It is necessary to increase compliance in the treatment rehabilitation of schizophrenia.In this way,the effect can be strengthened and the relapse in rehabilitation can be decreased.
2.Treatment of obsession with electric-acupuncture and clomipramine

BACKGROUND: The combination of electric acupuncture and clomipramine is the behavior attempt in the integration of Chinese and western medicine in the treatment of obsession.OBJECTIVE: To probe into the long-term therapeutic effects of the combination of acupuncture and medication on obsession and its influence on social functional recovery.DESIGN: Controlled study with observation in which the patients were taken as the objects.SETTING: Wards of a municipal mental hygiene center PARTICIPANTS: The research was performed in the Fifth People's Hospital of Zibo. Totally 30 inpatients were selected from January 1997 to October 2003 in the hospital, aged varied from 18 to 57 years, at the average of (33 ± 12) years. Thirty patients were divided randomly into two groups,named as experimental group, 15 cases with 8 males and 7 females; and the control group, 15 cases with 7 males and 8 females.METHODS: In the experimental group, electric acupuncture was applied on neiguan, renzhong and chengjiang, together with clomipramine orally. In the control group, only clomipramine was applied. The therapeutic effects and side effects were evaluated with Y-Bocs, TESS and clinical evaluation standards successively before the treatment, at the 4th, 6th and 8th weekends after treatment and by 6 months following visits. SDSS evaluation was carried on at the 8th week of the treatment.MAIN OUTCOME MEASURES: Comparisons of evaluating results with Y-Bocs, TESS and SDSS.RESULTS: At the 4th, 6th and 8th weekends of the treatment, the score-reducing rates of Y-Bocs in the experimental group were higher than those in the control group, but the difference was not significant ( P > 0.05). The scores of TESSin the control group were remarkably higher than those in the experimental group( P < 0. 01 ) . Factor scores of "profession and work" in SDSS in the experimental group were lower than that in the control group ( P < 0.05 ).CONCLUSION: Electric acupuncture on neiguan, renzhong and chengjiang reduces the side effect of clomipramine in the treatment of obsession and improve the living quality of the patients with obsession.

Objective To investigate the feasibility of tissue-specific gene therapy for bladder cancer using human UroplakinⅡ(UPⅡ) promoter. Methods The recombinant adenoviruses,Ad-hUPⅡ-TNF carrying tumor necrosis factor (TNF-?) under control of the hUPⅡ,were generated.ELISA showed the production and secretion of TNF by bladder cancer cells infected with Ad-hUPⅡ-TNF.The level of TNF in urine was identified with ELISA. Results ELISA showed that production and secretion of TNF by bladder cancer cells infected with Ad-hUPⅡ-TNF was distinctly higher than by non-urothelium cells infected with Ad-hUPⅡ-TNF and MTT showed that proliferation of bladder cancer cells was obviously inhibited.Conditioned medium from bladder cancer cells apparently inhibited cells of L929 proliferation,compared with conditioned medium from non-bladder cells by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay.Intravesical inoculation of Ad-hUPⅡ-TNF also caused decreased tumor growth in orthotopic human bladder cancer model.The sustained high level of TNF in urine could be identified with ELISA. Conclusions TNF driven by hUPⅡ promoter has effective active in the inhibition of bladder cancer growth both in vivo and in vitro.These will undoubtedly yield a new approach of therapy for bladder cancer.
5.Prevention of cell death in LNCaP cells by overexpression of clusterin

Objective: To evaluate the effect of clusterin with and without leader sequence on overexpression preventing apoptosis in human prostate LNCaP cells. Methods:The plasmid pIRES2 EGFP was used to generate the clusterin expression constructs with full length or without the leader sequence (designated as pIRES2 EGFP/cluac, pIRES2 EGFP/clubc, respectively). Western blot analysis was employed to compare clusterin expression levels in the lysis and supernatant fluid of clusterin transfected LNCaP cells in vitro . The distribution of different functional domains of clusterin in cells was detected with Immunocytochemical staining. The clusterin's protective role of Na 2SeO 3 induced apoptosis in LNCaP cells was examined by flow cytometry (FCM) and fluorescence microscope. Results: Clusterin expression was detected in the lysis and supernatant fluid of pIRES2 EGFP/cluac transfected LNCaP cells, while clusterin was found only in lysis liquid of pIRES2 EGFP/clubc transfected LNCaP cells, but not found in their supernatant fluid. The distribution of cluserin in the plasm of pIRES2 EGFP/cluac transfected cells was aggregative, and on the other hand, clusterin distributed dispersedly in pIRES2 EGFP/clubc transfected cells. Its anti apoptotic property in LNCaP cells was proved by FCM and fluorescence microscope.Conclusion: It is apparent that clusterin plays an important role in preventing apoptosis in prostate cancer, and the presence of the leader sequence is necessary for clusterin's anti apoptotic function.
6.Ginsenosides combined with dexamethasone in preventing and treating postembolization syndrome following transcatheter arterial chemoembolization: a randomized, controlled and double-blinded prospective trial

OBJECTIVE: To observe the effect of ginsenosides (GS) and low dose glucocorticoid in preventing and treating the postembolization syndrome following transcatheter arterial chemoembolization (TACE). METHODS: Eighty patients with primary liver carcinoma were randomly divided into 4 double-blinded groups, with 20 patients in each group. Patients in groups A, B, C, D were treated with placebo, dexamethasone (Dex), GS, Dex and GS, respectively. The changes of clinical symptoms and laboratory tests after TACE were observed. RESULTS: Dex combined with GS markedly decreased the occurrence ratio and lasting time of the symptoms such as nausea, vomiting, fever and pain, and protected the function of liver as compared with the placebo (P<0.05). Single use of Dex or GS improved some symptoms as compared with the placebo, but it was not as good as the combination of Dex and GS. CONCLUSION: Dex combined with GS can effectively prevent and treat the postembolization syndrome following TACE.
7.Regulation of expression of pituitary tumor transforming gene 1 (PTTG1) by androgen in prostate cancer

Objective:To identify the androgen-responsive genes in prostate and screen the molecular targets for further studying human prostate cancer. Methods:The potential androgen-responsive gene pituitary tumor transforming gene 1 (PTTG1) was selected which had been previously screened by cDNA microarray in rat prostate and its mRNA level was detected by Northern blot in the castrated rat prostate with and without replacement of Mibolerone. Immunohistochemistry was performed to determine the expression and location of PTTG1 in human prostate tissues. Then human androgen-dependent prostate cancer cells LNCaP were used as a model to study the regulation of PTTG1 by Mibolerone. Results: PTTG1 mRNA was hardly detectable in the prostate of 7-day castrated rats, while it was up-regulated dramatically in the prostate of 7-day castrated rats treated with Mibolerone for 2 days. It was showed that high expression of PTTG1 was localized to the epithelial cells of human prostate cancer but not to the stromal cells with Immunohistochemistry. Northern blot analysis indicated that LNCaP cells treated with 0.1 nmol/L Mibolerone for 2 days led to the high PTTG1 mRNA expression. The basic expression of PTTG1 in human androgen-independent prostate cancer cell lines PC3 or DU145 was even higher than that in the human androgen-dependent prostate cancer cells LNCaP treated with Mibolerone. Conclusion: Androgen can up-regulate the PTTG1 expression in castrated rat prostate and human prostate cancer cell LNCaP. It suggests that PTTG1 is potential to play an important role in human prostate cancer progression.
