1.Effects of iptaklim hydrochloride on glutamate induced cytotoxicity in cultured pheochromocytoma (PC12) cells

AIM To investigate the protective effects of iptakalim hydrochloride (Ipt) against glutamate induced neurotoxity. METHODS Neurotoxity injury was induced by with glutamic acid in PC12 cells. The cell viability was measured by MTT assay. RESULTS Ipt at the concentrations of 1 ? 1(T-10 - 1 ? 1(T4 mol ? L-1 markedly mitigated glutamate-induced neurotoxicity. The protec- live effect was nullified by glibenclamide, an ATP-sensitive potassium channel antagnist. CONCLUSION Ipt provides neuroprotection on glutamate-induced cytotoxixity in cultured PC12 cells and this effect may be related with ATP-sensitive potassium channels.
2.Effects of sevoflurane preconditioning on myocardial connexin 43 during myocardial ischemia-reperfusion in rats

ObjectiveTo investigate the effects of sevoflurane preconditioning on myocardial connexin 43 (Cx43) during myocardial ischemia-reperfusion (I/R) in rats.MethodsMale adult Wistar rats weighing 220-250 g were anesthetized with intraperitoneal pentobarbital 40 mg/kg and heparin 500 IU/kg.Their hearts were excised and perfused in a Langendorff apparatus with K-H solution saturated with 95% O2-5% CO2 at 37 ℃ for 30 min.Sixteen isolated rat hearts were randomly assigned into 2 groups ( n =8 each):I/R group and sevoflurane preconditioning group (group SP).At 30 min of equilibration,group I/R received perfusion for another 20 min,the hearts were purfused with K-H solution saturated with 2.4% sevoflurane for 15 min followed by 5 min washout with K-H solution in group SP.Then all the hearts underwent 40 min of ischemia followed by 60 min of reperfusion.HR,left ventricular developed pressure (LVDP),+ dp/dtmax and - dp/dtmax were recorded at the end of equilibration,immediately before ischemia and at 30 and 60 min of reperfusion.Myocardial tissues were obtained from apex for microscopic examination and from left ventricle for observation of distribution of Cx43 and determination of Cx43 expression.ResultsCompared with the baseline value at the end of equilibration, + dp/dtmax and - dp/dtmax immediately before ischemia and HR,LVDP,+ dp/dtmax and - dp/dtmax at 30 and 60 min of reperfusion were significantlydecreased in groups I/R and SP (P < 0.01 ).Compared with that immediately before ischemia,HR,LVDP, + dp/dtmax and - dp/dtmax were significantly decreased at 30 and 60 min of reperfusion in groups I/R and SP (P < 0.05 or 0.01).Compared with group I/R,HR,LVDP,+ dp/dtmax and - dp/dtmax were significantly decreased immediately before ischemia,HR,LVDP, + dp/dtmax and - dp/dtmax were significantly increased at 30 and 60 min of reperfusion ( P < 0.01 ),and pathological injury was attenuated in group I/R.Myocardial Cx43 was unevenly distributed in group I/R,while evenly distributed in group SP.There was no significant difference in Cx43 expression between the two groups.ConclusionThe mechanism by which sevoflurane preconditioning protects myocardium against I/R injury may be related to redistribution of Cx43,but not Cx43 expression.
3.Histopathology and biology of epithelial ovarian cancer

Ovarian epithelial cancer is the most common type of ovarian cancers. It can be further classified according to the histopathology and cell differentiation of the tumor, which are key factors determining the biologic behavior and prognosis of ovarian epithelial cancer. Understanding the subtype-specific histopathology and molecular biology of ovarian epithelial cancer may provide clues to novel tumor markers for early detection and new therapeutic intervention strategies.
4.New light on neuronal ATP-sensitive potassium channels

ATP sensitive potassium (K ATP ) channels widely expressed in many tissue and cell types including neurons are heteromultimers of sulfonylurea receptor (SUR) and inward rectifier K + channel (Kir6) subunits associated with 1∶1 stoichiometry as a tetramer (SUR/Kir6) 4. The activity of K ATP channels is modulated by intracellular ATP concentrations. Neuronal K ATP channels are involved in the mediation of glucose sensing and metabolic stress. Under physiological condition, they also possess important pathophysiological functions in acute brain ischemic and chronic neurodegenerative diseases.
5.Effects of MIP-1β and TGF-β antibody on the expansion of CD34+ cells from cord blood in stroma-contact culture system

Objective To study the effects of MIP-1β and TGF-β antibody on CD34+ cells from cord blood in stroma-contact culture system. Methods Immunomagnetic selected CD34+ cells were inoculated onto the pre-established irradiated human stroma layer. MIP-1β, TGF-β antibody (20 μg/ml) and MIP-1β+anti-TGF-β (5 μg/ml) were added on day 0 and day 5. On day 5 and day 10, cell counting, hematopoietic progenitor cells count were made by semi-solid culture and CD34+ cells were assayed by FACS. Results On day 5, no significant difference of total cell number was observed as compared with the control group (P>0.05), but the number of CD34+cells, CFC, and HPP-CFC in TGF-β antibody and MIP-1β+ TGF-β antibody groups was significantly higher than that in the control (P<0.05). On day 10, the number of total cell, CD34+ cells, CFC, and HPP-CFC in TGF-β antibody and MIP-1β+ TGF-β antibody groups was significantly higher than that in the control (P<0.05). No significant difference was observed between groups MIP-1β and control either on day 5 or day 10 (P>0.05). Conclusion MIP-1β has no significant effect on the CD34+ cells as compared with the control while the CD34+ cells can be expanded 1-3 folds with TGF-β antibody (20 μg/ml) or MIP-1β+ TGF-β antibody (5 μg/ml) in 10 days. There are synergic interactions between MIP-1β and TGF-β antibody.
6.Treatment of periodontal disease by double circumferential fibrotomy combined with orthodontic method

Objective:To explore the effects of double circumferential fibrotomy(CF)in the improvement of periodontal attachment of the teeth with periodontal disease.Methods:22 migrated incisors of 4 adults were randomly assigned to one-time CF group (T1 )and double CF group(T2)by center line.Periapical intraoral radiographic examination was performed using paralleling technique at pre-and post-leveling-alignment stage of the teeth.Root length,crown-root ratio and the distance of CEJ-AC were measured.The periodontal in-dicators were also recorded during the orthodontic treatment.Data were statistically analysed by comparative t-test.Results:CEJ-AC and crown root ratio reduction was greater in T2 group than those in T1 group(P <0.01 ),but the periodontal indices did not show sig-nificant difference between the 2 groups(P >0.05).Conclusion:Double circumferential fibrotomy can improve periodontal attachment, reasonable orthodontic force does not lead to remarkable root resorption of the periodontitis inflicted teeth.
7.Survey analysis on distribution of type A behavior characteristics in patients with periodontitis

Objective:To investigate the influence of type A behavior in the occurrence and development of chromic periodontitis and compare the differences among people with different behavior types in the occurrence and development of chronic periodontitis, and to explore the influence of psychological factors in the process of periodontitis.Methods:According the same standard,132 patients with periodontitis were selected as periodontitis group,and 126 patients without periodontitis were used as control group.OralSurveys Basic Methods proposed by WHO in 1987 and ReferenceStandard of Diagnosis and Treatment of Periodontitis made by AAP in 2000 were used to dignose the periodontitis.All subjects finished type A behavior and SCL-90 questionnaire (ZHANG Boyuan 1983,revision in 1985 ), their scores were recorded and the results were analyzed by t test. Results:The proportion of type A behavior in periodontits group (68.94%)was higher than that in control group (27.78%) (P<0.05)compared with type M and type B behavior.The scores of hostility (1.54±0.38),anxiety (1.47± 0.39),depression (1.41 ± 0.37),interpersonal sensitivity (1.23 ± 0.39),compulsive (1.72 ± 0.46),and somatizition (1.47 ± 0.38)were significantly higher than those in control group (1.32 ± 0.30, 1.29 ± 0.24, 1.25±0.23,1.04± 0.17,1.41 ± 0.35,1.25 ± 0.24).The calculus index (CI)of the people with type A behavior was higher than those of the people with other types of behavior (P<0.05).Conclusion:The people with tyep A behawior is easier to get periodontitis than other people.
8.Expressions of matrix metalloproteinase 8 mRNA in apical granuloma and periapical cyst and their significances

Objective To discuss the role of matrix metalloproteinase-8(MMP-8) mRNA in apical granuloma and periapical cyst by detecting the its expressions in the periapical granuloma and periapical cyst.Methods Reverse transcription polymerase chain reaction(RT-PCR) technique was used to measure the expressions of MMP-8 mRNA in 22 cases of periapical granuloma,11 cases of periapical cyst and 10 cases of normal periapical tissues.Results The positive rates and the levels of MMP-8 mRNA expression in apical granulomas and periapical cysts were significantly higher than those in normal tissues(P0.05).Conclusion The expression of MMP-8 may play an important role in the pathogenesy of apical granuloma and periapical cyst,and it may be involved in the process of bone destruction.

AIM To investigate the effects of iptakalim hydrochloride (Ipt) on cerebral neuronal sodium, calcium and potassium channels. METHODS The effects of Ipt on sodium, calcium and potassium channels in cultured hippocampal neurons was observed using whole-cell recording technique. RESULTS Ipt had no significant effect on Na +, Ca 2+ currents, but significantly increased the outward K + currents. This enhancement of Ipt was inhibited by synchronously application of glibenclamide. 10.Advances in study of cervical small cell carcinoma

Small cell carcinoma of cervix (SCCC) is a rare and high malignant neuroendocrine tumor.Because of its strong invasion,metastasis in early stage and easy recurrence,the prognosis is poor.Its diagnosis is based on pathomorphology,and immune-histochemistry biomarkers can improve its diagnosis.Combined therapy is recommended.Postoperative platinum/etoposide (PE),vincristine/adriamycin/cyclophosphamide (VAC) or taxel/carboplatin(TP) chemotherapy can improve the prognosis.
