OBJECTIVE:To study the improving effects of argatroban on the brain damage of model rats with cerebral hemor-rhage. METHODS:50 SD rats were randomly divided into normal control group(equivalent normal saline),model group(equiva-lent normal saline),argatroban high, medium and low dose groups (0.75,1.5,3 mg/kg). All groups, except for normal control group, were reproduced models with cerebral hemorrhage, and the rats in each group were treated by corresponding drugs after 30 min,ip,once a day. The rats were executed after continuous administration for 3 days. The data was detected, including the con-tents of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),activity of cysteine-containing aspartic protease 3(Caspase-3), expressions of Bcl-2,Bax,matrix metalloproteinase-2(MMP-2)and MMP-9,and the phosphorylations of nuclear factor κB(NF-κB) p65 and IκBα. RESULTS:Compared with the normal control group, the contents of TNF-α and IL-1β,the activity of Cas-pase-3,the expressions of MMP-2,MMP-9 and Bax proteins were increased;the expression of Bcl-2 was decreased;and phos-phorylations of NF- κB p65 and IκBα in model group were increased, with significant differences(P<0.01). Compared with the model group, the contents of TNF-α and IL-1β,the activity of Caspase-3,the expressions of MMP-2,MMP-9 and Bax were de-creased;the expression of Bcl-2 was increased;and phosphorylations of NF-κB p65 and IκBα in argatroban low, medium and high dose groups were decreased, with significant differences (P<0.01 or P<0.05). And it was positively correlated with the dose. CONCLUSIONS:Argatroban has dose-dependent inhibition effects on the inflammatory cytokine secretion and apoptosis of brain tissue of model rats with brain hemorrhage, and the mechanism may be related to NF-κB signaling pathway.

Objective To investigate the organic pollutants in the contaminated water irrigation area soil and to study the lipid peroxidation effect of the pollutants on mouse. Methods The organic pollutants of the polluted irrigation soil and control soil were extracted using the supersonic oscillator. The ingredients of the organic pollutants were measured by the GC-MS spectrum. The 40 sanitary Kunming mice were randomly divided into five groups: the dimethylsulfoxide control group, low dose group of control soil, high dose group of control soil, low dose group of polluted soil, high dose group of polluted soil. The administered doses of the low and high dose groups were 25.5 g dry soil/kg body weight per day by oral for two weeks. The activities of T-SOD,GSH-Px and the content of MDA in serum of mice were determined. Results Phenanthrene,yrene,diphenyl,fluorene,naphthalene,dimethyl-naphthalene,2,3,6-trimethyl-naphthalene,anthracene,dimethyl-anthracene,1,4-dimethyl-anthracene,fluoranthene,4-methyl-pyrene,dibenzothiophen,4-methyl-dibenzofuran,dibenzofuran,butylated-hydroxytoluene,dodecane,neopentyl-cyclohexane,trimethyl-heptane,1,2-dimethyl-cyclohexanimine,butethal were detected in the polluted irrigation soil and their levels were 14.3,2.1,1.3,1.1,0.8,1.3,0.4,2.9,7.2,2.3,9.1,0.2,0.4,1.4,1.7,0.37,0.9,0.11,0.02,0.02 and 0.13 mg/kg respectively in the polluted irrigation soil. Trimethyl-heptane,dimethyl-heptane and 1,2-dimethyl-cyclohexanimine were detected, and their levels were all 0.01 mg/kg in control soil. Compared with the solvent of dimethylsulfoxide group, the activities of T-SOD and GSH-Px of the high dose group of control soil and two dose groups of polluted soil all decreased, the differences were all significant(P0.05). Conclusion The soil has been contaminated with organic pollutants in the investigated area and the soil extract may produce a certain effect on lipid peroxidation in mice.

Anaplastic lymphoma kinase (ALK) rearrangement is one of the most potent carcinogenic genes in non-small cell lung cancer (NSCLC).The first-generation ALK inhibitor such as crizotinib is superior to chemotherapy for NSCLC patients with ALK rearrangement. At the same time, more and more studies have reported ALK inhibitors in brain metastases of NSCLC patients with intracranial efficiency. However, despite the initial clinical data of first-generation ALK inhibitors in the treatment of ALK-positive NSCLC with brain metastases, different degrees of recurrence of tumors after acquired resistance have posed new challenges for follow-up treatment of cancer patients.A new generation of ALK inhibitors, such as alectinib;ceritinib,AP26113 and PF-06463922 have emerged to solve this problem.

Object To establish a method for chemical pattern recognition on the quality assessment of Radix Astragali. Methods The contents of astragaloside in 18 samples of Astragalus Linn. in different species and origins were determinated by dual-wavelength TLCS method. The developing solvent was CHCl 3-MeOH-H 2O (65∶30∶10,), the UV detection was set at ? s=390 nm; ? R=590 nm. Astragaloside was regarded as the quality assurance of medicinal Radix Astragalus. Based on the TLCS method, the chemical data were obtained. Hierarchical clustering analyses were applied to the chemical pattern recognition. Results The content of astragaloside in Astragalus mongholicus (Bge.) Hsiao and A. membranaceus (Fisch.) Bge. was relatively higher than that in the other samples. This is consistent with the Pharmacopoeia of the People's Republic of China in which the two sorts of Astragalus Linn. were regarded as goods. Conclusion This method is a practicable in the quality assessment of Radix Astragali.

Objective To explore the change of proportion of peripheral blood dendritic cells (DCs) in patients with stroke. Methods 56 patients (30 cases of cerebral infarction and 26 cases of cerebral hemorrhage) in Beijing Bo'ai hospital from June to September, 2014 and 14 healthy controls were investigated. The severity of stroke was assessed with the National Institutes of Health Stroke Scale (NIHSS). Flow cy-tometry analysis was employed to detect the proportion of DCs subtypes in the peripheral blood. Results No obvious difference was found in DCs between the stroke patients and the controls. Compared to the control group, the percentages of peripheral blood myeloid dendritic cells (mDCs) decreased in the cerebral hemorrhage and the cerebral infarction subgroups (P<0.001). The percentages of plasmacytoid den-dritic cells (pDCs) reduced significantly in the cerebral hemorrhage and the cerebral infarction subgroups (P<0.05). The stroke patients were divided into NIHSS≤7 subgroup and NIHSS>7 subgroup. The percentages of pDCs in the cerebral hemorrhage and the cerebral infarction patients were significantly lower in the NIHSS>7 subgroup than in the NIHSS≤7 subgroup (P<0.05). While there was no statistical differ-ence between NIHSS≤7 subgroups and NIHSS>7 subgroups in the percentages of mDCs in the cerebral hemorrhage and cerebral infarction patients. Conclusion The proportion of DCs subtypes in the peripheral blood in stroke patients changed significantly, indicating inflamma-tion responds play a role in stroke.

To investigate the clinical features, genetic diagnosis, and treatment of a patient with Prader-Willi syndrome(PWS). For a case with clinically suspected PWS, methylation specific PCR(MSPCR)amplification was applied to CpG islands of SNRPN(exon α)gene locus in the 15q11-q13. Furthermore, the diagnosis was comfirmed by the method of bisulfite sequencing PCR(BSPCR). Metabolic status before and after the operation of sleeve gastrectomy were compared. Absence of amplification of paternal allele on chromosome 15q11-q13 was detected in the case by MSPCR, different from the normal control. Results of BSPCR further proved a full methylation of CpG islands in the SNRPN gene locus. Four months after sleeve gastrectomy, systemic metabolic status and ventricular function were improved. MSPCR and BSPCR were both consistent with genetic diagnosis of PWS. Weight loss surgery is expected to be a major therapy of this disease.
