1.Regulation of gene expression via control of transcription elongation during immune responses
Chinese Journal of Immunology 2017;33(8):1121-1128
Transcription is a multi-stage process that coordinates several steps during the transcription cycle including pre-initiation,initiation and elongation.Recent advances by genome-wide study suggest that control of transcription elongation is a critical step for precise regulation of gene expression across species from Drosophila to mammals. Here we review the molecular mechanisms of how transcription elongation of RNA polymeraseⅡ(PolⅡ) is modulated by the major pausing factors-NELF,DSIF and the positive elongation factor P-TEFb,which is the key player in pause release. We also discuss the potential implications of regulation of transcription elongation in pathogenesis of cancer, inflammation and virus infection.

Objective To discuss the role of matrix metalloproteinase-8(MMP-8) mRNA in apical granuloma and periapical cyst by detecting the its expressions in the periapical granuloma and periapical cyst. Methods Reverse transcription polymerase chain reaction(RT-PCR) technique was used to measure the expressions of MMP-8 mRNA in 22 cases of periapical granuloma,11 cases of periapical cyst and 10 cases of normal periapical tissues. Results The positive rates and the levels of MMP-8 mRNA expression in apical granulomas and periapical cysts were significantly higher than those in normal tissues(P0.05).Conclusion The expression of MMP-8 may play an important role in the pathogenesy of apical granuloma and periapical cyst, and it may be involved in the process of bone destruction.

The therapeutic effects and side effects were evaluated according to the WHO criteria. Results In all 45 patients, 53.3 % of overall response rate were achieved CR for 9 cases, 15 cases PR, 13 cases SD and 8 cases PD. Main side effects were myelosuppression, nausea, vomit, alopecia, etc. Conclusion MINE regimen is an effective and safe salvage regimen in treating patients with recurrent and refractory intermediate grade NHL. Further clinical study on the application of MINE regimen is recommended.

Journal of Jilin University(Medicine Edition) 2001;27(1):68-70
Objective:To investigate the changes in plasma ET-1 and ANF in IDDM and its microvascular complication. Methods:The plasma levels of ET-1 and ANF were measured by radioimmunoassy in 31 patients with IDDM and 25 normal controls. Results:The plasma levels of ET-1 and ANF significantly increased in IDDM compared with the normal controls (P<0.01);and the levels were also markedly higher in diabetics with microangiopathy complications than in those withou the complications (P<0.01).The more kinds of microangiopathey complications, the higher level of the plasma ET-1 and ANF,which suggests that the elevation of plasma ET-1 and ANF may be concerned with the microangiopathy and ET-1 may reflect the degree of vascular endothelial cell damage. There were no significant correlation between plasma ET-1 or ANF and blood glucose, fructosamine and C-peptide,but the plasma levels of ET-1 and ANF both correlated with the duration of diabetes mellitus. And the plasma level of ET-1 positively correlated with ANF1 (P<0.01).Conclusion:The results of study suggest that ET-1 and ANF are both associated with the development, progress and severity of microvascular complications.

Objective To determine the content of rutin in fruits, stems, and leaves of Sambucus williamsii Hance, and provide an essential reference for the exploitation and utilization of the resources. Methods The content of rutin was analyzed on an Agilent ZORBAX Eclispse XDB C18 column(150 mmí4. 6 mm,5 μm). The mobile phase was methanol-0. 1%phosphoric acid (4555),the column temperature was set at 25 ℃,the flow rate was 0. 8 mL·min-1,and the detection wavelength was set at 360 nm. Results The linear range of rutin was 0. 105-0. 525 μg (r=0. 999 6),the average recovery was 97. 86%,and RSD was 0. 49%. The content of rutin in stems, leaves, and fruits of Sambucus williamsii Hance was 0. 171, 0. 522, and 0. 165 mg·g-1 ,respectively. Conclusion The stems, leaves and fruits of Sambucus williamsii Hance all contain rutin, but at a significant different content level. The leaves contain the highest level of rutin.

Objective To evaluate the structure and function of left ventricle in diabetes mellitus (DM) patients without complications by myocardial velocity gradient (MVG) measured by myocardial velocity profile (MVP). Methods Thirty type 2 DM patients without complications and 30 healthy volunteers as controls were enrolled. The heart structure, systolic function and diastolic function of left ventricle were measured by echocardiography and the left ventricular mass index (LVMI) was calculated. Mitral annular systolic movement (Sm) , early rapid filling phase movement ( Em) and atrial contraction movement(Am) were measured by tissue Doppler image and the left ventricular MVG at diastole and systole in subendocardium and subepicardium (MVGs,MVGd) were measured by MVP. Results The diameter of left atria and left ventricle, thickness of interventricular septum and LVMI were higher in DM group than those of control group ( P <0. 05 or P <0. 01) , MVGs, MVGd, and Em were lower in DM group than those of control group( P <0. 05 or P <0. 01). There were no significant differences on E/A, Em/Am and E/Em between two groups. In addition, there were also no significant differences on Sm,left ventricular ejection fraction and left ventricular fraction shortening between two groups. Conclusions Structure and function of left ventricle have changed in patients of DM without complications. MVG measured by MVP is an accurate and sensitive index to assess left ventricular systolic and diastolic function.

Chinese Journal of Modern Nursing 2020;26(15):1972-1976
Objective:To explore the current situation of emotion labor among emergency nurses and to analyze its influence on job engagement. Methods:From May to August 2018, this study selected 448 emergency nurses from 6 ClassⅢ general hospitals in Beijing as subjects by convenience sampling. All nurses were investigated with the General Information Scale, Emotional Labor Scale (ELS) and Utrecht Work Engagement Scale (UWES) . SPSS 22.0 was used to data entry and analysis. T test, variance analysis, Pearson correlation was used to single factor analysis, and multiple hierarchical regression was used to influencing factor analysis. Results:Among 448 emergency nurses, the total average scores of ELS and UWES were (3.94±0.75) and (3.43±0.69) respectively. There were statistical differences in the scores of UWES among emergency nurses with different genders and posts ( t=-2.048, -4.553; P<0.05) . Pearson correlation analysis showed that surface acting and expression of emotions in ELS had negative correlations with dimensions in UWES among nurses with statistical differences ( r=-0.108--0.330, P<0.05) ; deep acting in ELS had positive correlations with dimensions in UWES with statistical differences ( r=0.220-0.256, P<0.01) . Hierarchical regression analysis showed that deep acting positively predicted job engagement, and surface acting negatively predicted job engagement which explained 17.9% of the variance of job engagement. Conclusions:Emergency nurses of Class Ⅲ general hospitals have middle level of emotion labor and upper middle level of job engagement. Nursing managers should pay more attention to emotion labor of nurses and improve the job engagement by emotion labor so as to improve the nursing service quality.
