1.Observation on the effects of massage on treatment of attention deficit hyperactivity disorder in children

BACKGROUND:Attention deficit hyperactivity disorder(ADHD) is treated clinically mainly with medication,psychotherapy and acupuncture,which is seldom treated with simple massage.During working in Swaziland, the writer found a part of children only accept massage for they did not receive remarkable results by medication and also were scared of acupuncture. OBJECTIVE:To probe manipulation and mechanism of massage on ADHD in children. DESIGN:Cases analysis. SETTING:Sino Japan Friendship Hospital. PARTICIPANTS:All the cases were collected from April 2000 to April 2002 while the writer was working in Swaziland. Thirty three children with ADHD were in accordance with the symptomatic diagnostic standards of American DSM IV.No control was selected. INTERVENTIONS:Simple massage manipulation was adopted in treatment of the 33 ADHD children.The common manipulations included head manipulation,chest abdominal manipulation and back manipulation.According to various clinical manifestations,the manipulations were modified based on differentiation of syndromes. MAIN OUTCOME MEASURES:Therapeutic results on patients with massage manipulations. RESULTS:Of the 33 cases of ADHD children treated with massage, 10 were cured,9 were remarkably effective,9 were improved and 5 had no effect. CONCLUSION:The therapeutic results of massage on ADHD in children are satisfactory and safe without side effects.
2.Clinical Observation on the Treatment of Chronic Nonspecific Ulcerative Colitis by Acupuncture and Moxibustion

The main acupoints ofZhongwan (CV 12), Tianshu (ST 25), Zusanli (ST 36), Shangjuxu (ST 37), Dachangshu (BL 25) and Shenque (CV 8), in combination with the corresponding acupoints based on the syndrome differentiation, were selected to treat 28 cases of chronic nonspecific ulcerative colitis, the result showed clinical cure in 18 cases, clinical effect in 15 cases, failure in 5 cases. The most treatment times were 46, and the least 25.
3.Acupuncture Treatment of Depression by Pattern Identification

In the treatment of 63 cases of depression, by bilateral Baihui (GV 20),Sishencong (Ex-HN 1), Juque (CV 14) and Neiguan (PC 6) as the main acupoints, based upon pattern identification, with bilateral Zhigou (TE 6), Zusanli (ST 36), Yanglingquan (GB 34)and Taichong (LR 3) added for liver qi stagnation and spleen deficiency, with bilateral Hegu (LI 4), Xuehai (SP 10), Sanyinjiao (SP 6) and Taichong (LR 3) added for stagnation of liver blood, with bilateral Shenmen (HT 7), Zusanli (ST 36), Sanyinjiao (SP 6) and Taibai (SP 3)added for deficiency in both the heart and spleen, and with bilateral Sanyinjiao (SP 6), Taibai(SP 3), Taixi (KI 3) and Guanyuan (CV 4), by moxibustion on Guanyuan (CV4) and needling techniques on the rest acupoints, the results showed clinical cure in 21 cases, remarkable effect in 18 cases, improvement in 20 cases and failure in 4 cases, in the treatments from 13times to 45 times, at the average treatments of 26 times.
4.Accelerated solvent extraction of ecdysterone from Achyranthes bidentata

Objective To apply accelerated solvent extraction (ASE) technique to extract Achyranthes bidentata and to explore the application of this technique in quality control of Chinese materia medica. Methods Investigation of single factor was used to optimize the conditions that affected the efficiency for ASE from A. bidentata by RP-HPLC using ecdysterone as a quantitative marker. Results The optimized conditions for ASE of A. bidentata were obtained as follows: methanol as solvent, particle size between 0.3 and 0.45 mm, temperature at 100 ℃, pressure under 10.34 MPa, 6 min duration and once extraction. Conclusion ASE technique can be used to extract A. bidentata quickly and effectively.
5.Multicenter clinical study on patients with inflammatory bowel disease in Kunming city

Objective To retrospectively investigate the characteristics of patients with inflammatory bowel disease (IBD)in last 10 years in Kunming city. Methods Four hundred and thirty consecutive patients with IBD, who had hospitalized in 7 hospitals between January 1998 to March 2007, were investigated. Among them, 379 patinets had ulcerative colitis (UC) and 51 had Crohn's disease (CD). The patients who received coloscopy, histopathological and bariam enema examination accunted for 98.2%, 56.2% and 2.6% in UC group,respectively, and 72.5%,78.4% and 31.4% in CD group,respectively. The gender, age, occupation and the clinical manifestation of the patients, and the results of the colonoscopy and pathological examination were analyzed. Results Most of the UC patients were aged 30 to 39 years and 50 to 59 years with average age of (46.9±15.8) years, whereas CD patients were aged 20 to 29 years with average age of (41.6±17.2)years. The male and urban patients were predominat in both UC and CD groups. Mental workers were common in UC group. The symptoms of diarrhea (302,79.7%), abdominal pain (285,75.2%) and bloody stools (290,76.5%) were major events in UC patients, and those of abdominal pain (44,86.3%), diarrhea (28,54.9%) and weight loss (28,54.9%) were major events in CD patients. The coincidence ratio among colonoseopy, histopathology and bariam enema examinations was 88.4 % ( 329/372 ), 24.4 % ( 52/213 ) and 4/10 in UC patients, respectively, and 86.5%(32/37), 27.5%(11/40) and 75%(12/16) in CD patients, respectively. All of the UC patients were in active stage of disease,including 38.3 % in mild,42.2% in moderate and 19.5% in severe. In CD group, 7.8% was in relief stage and 92.2% in active stage,including 15.7% in mild,43.1% in morderate and 41.2% in severe.Conclusion The understanding of prevalence and clinical characteristics of IBD in last 10 years in Kunming city will be helpful in diagnosis and treatment of the disease.
6.The Role of Hydrogen Sulfide in Acute Liver Injury Induced by Traumatic Stress in Rats

Objective To explore the role of hydrogen sulfide (H 2S ) in acute liver injury induced by crush-ing hind lim bs of rats. Methods The rats w ere random ly divided into the follow ing groups:control, crush-ing, H 2S donor sodium hydrosulfide (NaHS) + crushing, H 2S inhibitor propargylglycine (PAG ) + crush-ing group. The acute liver injury m odel w as established by crushing the hind lim bs of rats w ith standard w eight. R ats w ere sacrificed at 30 m in and 120 m in after the crush. The activities of serum aspartate am inotransferase (AST) and alanine am inotransferase (ALT) w ere m easured by colorim etric m ethod, and the content of H 2S in plasm a and the contents of m alondialdehyde (MDA), protein carbonyl, glutathione (GSH) in the liver and the activity of H 2S generating enzym e (cystathionine γ-lyase, CSE) w ere deter-m ined by chem ical m ethod. The expression of CSEm R N Ain liver w as detected by R T-PCR . Results For crush injury group, the levels of ASTand ALTin serum , MDAand protein carbonyl in liver in-creased. The levels of GSH, CSE, CSEm R N Ain liver and H 2S in serum decreased. The adm inistration of NaHS before lim bs crush could attenuate the changes of liver injury, but the pre-treatm ent w ith PAG could exacerbate the changes. Conclusion The decrease of H 2S production could involve in m ediating the acute liver injury induced by traum atic stress in rats.
7.Effects of HO-1 on Lipopolysaccharide-induced Endoplasmic Reticulum Stress of Rat Hepatocytes

Objective To investigate effects of antioxidant stress protein hem e oxygenase-1 (HO-1) on lipopolysaccharide (LPS)-induced endoplasm ic reticulum stress (ERS) of rat hepatocytes. Methods The BRL cells (rat hepatocyte cell line) were cultured. The hepatocytes were treated with LPS, LPS+HO-1 si RNA , HO-1 siRNA and PB S solution, respectively. The cell viability was m easured by trypan blue ex-clusion test. The apoptosis cells were detected by the fluorescent dye Hoechst 33258. E xpressions of GR P78, C HO P, caspase-12 and HO-1 were detected by Western blotting. Results LPS caused an in-crease of HO-1 protein expression of rat hepatocytes in a dose-dependent and tim e-dependent m anner, a up-regulation of GRP78, CHO P and caspase-12, a decrease in cellviability,and an increase in apopto-sis rate of hepatocytes. Pretreatm ent of HO-1 siRNA inhibited the up-regulation of LPS-induced HO-1, however, aggravated ERS and cellular injury. Conclusion HO-1 inhibites ERS-m ediated cellular injury of rat hepatocytes induced by LPS.
8.Drug interactions on P-glycoprotein transport level and its application.

Potential drug interactions on P-glycoprotein transport level were discussed in this article. As an efflux transporter protein widely distributed in various tissues, P-glycoprotein plays an important role in many drugs interactions, via affecting the metabolic process to alter the concentration of drug in plasma and tissue. Interactions between herb and western drug may occur by inducing or inhibiting P-glycoprotein, resulting in either efficacy enhancement or adverse effects. So it is important for clinical drug use by rational utilization of the interaction between herb and western drug which medicated by P-glycoprotein.

9.Effect of inhibition of GSK-3βactivity on sevoflurane postconditioning-induced cardioprotection in diabetic rats

Objective To investigate the effect of inhibition of glycogen synthase kinase?3 beta ( GSK?3β) activity on sevoflurane postconditioning?induced cardioprotection in diabetic rats. Methods Healthy adult male Sprague?Dawley rats, weighing 250-300 g, in which diabetes mellitus was induced by intraperitoneal 1% streptozotocin 60 mg∕kg combined with high?fat and high?sucrose diet and confirmed by blood glucose level >16. 7 mmol∕L. Forty rats with diabetes mellitus were divided into 5 groups ( n=8 each) using a random number table: sham operation group ( S group ) , ischemia?reperfusion ( I∕R ) group, sevoflurane postconditioning group ( SP group) , GSK?3β inhibitor SB216763 group ( SB group) , and sevoflurane postconditioning plus SB216763 group ( SS group ) . Myocardial ischemia was induced by 30 min occlusion of the left anterior descending branch of the coronary artery followed by 120 min reperfu?
sion. The rats inhaled sevoflurane with the end?tidal concentration of 2.5% for 5 min starting from 1 min be?fore reperfusion in group SP. SB216763 0.2 mg∕kg was injected via the caudal vein at 1 min before reperfu?sion in group SB. In group SS, the rats inhaled sevoflurane with the end?tidal concentration of 2.5% for 5 min starting from 1 min before reperfusion, and SB216763 0.2 mg∕kg was injected via the caudal vein at 1 min before reperfusion. At 120 min of reperfusion, blood samples were collected from the carotid artery for determination of serum creatine kinase?MB (CK?MB) activity and cardiac troponin I (cTnI) concentra?tions. Myocardial specimens were collected at 120 min of reperfusion for microscopic examination of the pathological changes and for determination of myocardial infarct size ( by 2,3,5?triphenyltetrazolium chlo?ride staining) and phosphorylated GSK?3β (p?GSK?3β) expression (by Western blot). Results Com?pared with group S, the myocardial infarct size and serum CK?MB activity and cTnI concentration were sig?nificantly increased, and the expression of p?GSK?3βwas significantly down?regulated in I∕R, SP, SB and SS groups (P<0.05). Compared with group I∕R, the myocardial infarct size and serum CK?MB activity and cTnI concentration were significantly decreased, and the expression of p?GSK?3β was significantly up?regulated in SB and SS groups (P<0.05), and no significant change was found in the parameters men?tioned above in group SP ( P>0.05) . Compared with group SB, the myocardial infarct size and serum CK?MB activity and cTnI concentration were significantly decreased, and the expression of p?GSK?3β was sig?nificantly up?regulated in group SS (P<0.05). The pathological changes of myocardium were significantly attenuated in SB and SS groups as compared with group I∕R and group SP . Conclusion Inhibition of GSK?3β activity can improve sevoflurane postconditioning?induced cardioprotection in diabetic rats.
10.Effect of Shaoyao Gancao Tang on function and expression of P-glycoprotein in Caco-2 cells.

OBJECTIVETo investigate the effect of Shao Yao-Gan Cao-Tang on function and expression of P-glycoprotein (P-gp) in Caco-2 cells.
METHOD3H-digoxin (Dig), a substrate of P-glycoprotein, was used as a probe to measure the P-gp-mediated drug efflux transport, which indicated the function of P-gp in Caco-2 cells, while Verapamil (Ver) was used as a positive P-gp inhibitor. P-gp expression in Caco-2 cells was tested by immunohistochemistry staining. Inhibition effect of SGT on P-gp-mediated drug efflux transport and P-gp expression were investigated.
RESULTDig was shown a positive absorption mode in Caco-2 cell monolayer, characterized as the ratio of apparent permeabilities (Papp) from basolateral side to apical side Papp (BL-->AP) and from apical side to basolateral side Papp (AP-->BL) of Dig was 27.07. Addition of Ver into Dig transport media significantly inhibited P-gp activity which was indicated by increasing the Papp (AP-->BL) of Dig by 3.82 times, whereas Ver had no significant effect on Papp (BL-->AP). SGT (at the concentrations of 1/25 IC5, 1/5 IC5, IC,) could promote Papp (AP-->BL) of Dig by 159.83%, 217.95% ,160.26%. Papp (AP-->BL) of Dig was mildly increased by 59.16%, 50.73% by SGT at 1/25 IC5, 1/5 IC, respectively. Immunohistochemistry staining showed that SGT inhibited the expression of P-gp of Caco-2 cells.
CONCLUSIONSGT showed the potential inhibition to the function and expression of P-gp, leading to the increase absorption of P-gp's substrates.
