OBJECTIVE:To establish the quality standard of Bikemin capsule to control the quality METHODS:TLC method was used for the identification of Ephedra sinica and the detection of limitation of aconitine in Bikemin capsule Ephedrine and pseudoephedrine in the capsule were determined by HPLC on C18 column with the detection wavelength at 215nm RESULTS:The standard curves of ephedrine and pseudoephedrine were linear in the ranges of 0 02~0 10mg/ml (r=0 9 993) and 0 01~0 05mg/ml(r=0 9 996)respectively The average recoveries were (101 4?1 78)% for ephedrine and (98 08?1 81)% for pseudoephedrine CONCLUSION:The identification method is specific and the determination method is sensitive The quality standard is suitable for control of quality of Bikemin capsule
3.Determination of ampicillin and probenecid concentration in human plasma by high performance liquid chromatography

Objective To develop a high performance liquid chromatography(HPLC) method to determine the concentration of ampicillin and probenecid in human plasma.Methods A Symmetry ShieldTM RP18 column(5 ?m,150 mm?3.9 mm)was used.The mobile phases were 0.068 mol/L KH_2PO_4(pH 3.0)∶acetonitre(92∶8,V/V)for ampicillin,H_2O∶acetonitrile∶1 mol/L KH_2PO_4∶1 mol/L acetic acid(59.5∶39.8∶0.62∶0.062,V/V,0.15% triethylamine added to reach pH 2.76) for probenecid.The detective wavelength were at 254 nm for probenecid and 210 nm for ampicillin.Results The calibration curves obtained were linear in the range of 0.625-20 ?g/ml (r=0.999 9) of probenecid and 1.25-40 ?g/ml of ampicillin.Conclusion This analysis method is simple,sensitive and rapid,suitable for studying pharmacokinetics of ampicillin and probenecid.

5.An important role for type Ⅲ interferon(IFN-lambda) in anti-HIV activity

Objective To examine whether IFN-λ has the ability to inhibit HIV-1 infection of blood monocyte-derived macrophages and its mechanism(s). Methods Macrophages were pretreated with IFN-λ/ IFN-λ2 for 24 h before infected by HIV-1 R5 strains (Bal, Jago, and JRFL). And then the culture supernatants were detected HIV-1 reverse transcription (RT) activity and p24 protein expression by HIV-1 BT assay and ELISA. The expressions of IFN-λ receptor, CD4, CCRS, CXCR4 were evaluated by real-time PCR. Results Both IFN-λ1 and IFN-λ2, when added to macrophage cultures, inhibited HIV-1 infection and replication. This IFN-λ-mediated anti-HIV-I activity is broad, as IFN-λ could inhibit infection by both laboratory-adapted and clinical strains of HIV-1. Investigations of mechanism(s) responsible for the IFN-λ action showed that although IFN-λ had little effect on HIV-1 entry receptor CD4 and co-receptor CCR5 and CXCR4 expression, IFN-λ inhibited HIV-I infection of macrophages through connecting with IFN-λ recep-tor. Conclusion IFN-λ could inhibit HIV-I replication in macrophages. These findings indicate that IFN-λ may have a therapeutic value in the treatment of HIV-1 infection.
6.The expression of NOD2 and TLR9 in gastric carcinoma and precancerous lesions

Objective To investigate the expression of NOD2 and TLR9 in gastric carcinoma and precancerous lesions and their clinical significance. Methods The expression of NOD2 and TLR9 was measured by immunohistochemical staining (S-P method) in 84 patients with atrophic gastritis, 48 eases with gastric ulcer, 80 cases with gastric adenocarcinoma and 40 eases with superficial gastritis. Results NOD2 and TLR9 expression was up-regnlated in superficial gastritis, atrophic gastritis, gastric ulcer and gastric adenocarcinoma. The positive expression rate of NOD2 was 35% ,21% ,33% ,40% respectively, and the positive expression rate of TLR9 were 15%, 12% ,21% ,22% respectively. The expression of NOD2 and TLR9 in Hp complicated atrophic gastritis, gastric carcinoma was significantly higher than that of lip not complicated disease entities ( P < 0. 05 ). Conclusions The expression of NOD2 and TLR9 may be valuable index for predicting the development of gastric mucosal damage from superficial to atrophic gastritis, gastric ulcer and gastric carcinoma.
7.Study on Formulation of Xiqingguo Buccal Tablet

OBJECTIVE:To study the formulation of Xiqingguo buccal tablet,optimize the proportion and quantity of main materials. METHODS:Using appearance,hardness,dissolution and taste as investigation indexes,orthogonal design was adopted to optimize the proportion and quantity of main thinner(lactose,mannitol),wetting agent(ethanol),lubricant(magnesium stea-rate),flavoring agent(aspartame),and critical relative humidity was detected. RESULTS:By wet granulation,the optimal formu-lation were as follows as the ratio of lactose and mannitol was 1:3,ethanol volume fraction was 60%,the dosage of menthol, magnesium stearate,aspartame and orange essence was 0.4%,0.9%,2.0%,0.4%;it was proven that the total score of 3 batches of samples were 2.67,2.67,2.70 (RSD=0.65%,n=3),respectively. The critical relative humidity of granule was 60%. CON-CLUSIONS:The Xiqingguo buccal tablet prepared by optimal prescription meets the requirements.
8.Correlation of hemoglobin level with cytokines neurohormonal activity and their effects on ventricular remodeling in patients with congestive heart failure

Objective To study the relationships of hemoglobin(Hb)level with the levels of neurohormones,and cytokines,and the effect of them on ventricular remodeling in patients with congestive heart failure(CHF). Methods Hb level,serum angiotensinⅡ(Ang Ⅱ ),tumor necrosis factor-α(TNF-α),nitric oxide(NO),soluble intereellular adhesion molecule-1(sICAM-1)and B-type natriuretic peptide(BNP)were measured in 121 CHF patients.The left ventrieular ejection fraction (LVEF)from echocardiography,left ventricular mass index(LVMI),and mean wall stress(MWS)were calculated. Results The levels of Ang Ⅱ,TNF-α,NO,siCAM-1,BNP[(144.5±64.1)ng/L,(92.3±6.4)ng/L,(65.2±4.2)μmol/L,(253.6±26.0)μg/L,(1294.0±223.0)ng/L]and LVMI,MWS in the anemia group of CHF patients were higher than those in the non-anemia group[(76.7±48.5)ng/L,(55.6±10.2)ng/L,(42.1±11.9)μmol/L,(237.18±33.26)μg/L,(437.0±115.0)ng/L,all P<0.01].With the increase of anemia severity,the levels of AngⅡ,TNF-α,NO,siCAM-1,BNP and LVMI,MWS were significantly increased.There were negative correlations between Hb level and the 1evels of AngⅡ,TNF-α,NO,siCAM-1,BNP,LVMI,MWS(r=-0.8173,-0.8509,-0.6001,-0.6692,-0.6283,-0.8604,-0.8733,all P<0.01),and negative correlations between LVMI,MWS and Hb levels and LVEF(P<0.01). Conclusions Neurohormones and cytokines play roles in ventrieular remodeling and anemia in CHF aggravates the severity of ventricular remodeling.
9.Optimization Research on TCM Compound Prescription in Insomnia Treatment

This study was aimed to screen out a safe and effective traditional Chinese medicine (TCM) compound prescription in insomnia treatment, through the method of uniform design and comprehensive scoring method. It provided candidate drugs to further research and development of TCM new drug. U7(76) uniform design was used to divide groups. The test was given on sleep time prolonging effect of mice injected with pentobarbital sodium. The subliminal dosage of pentobarbital hypnosis was tested on mice injected with pentobarbital sodium. The acute toxicity (LD50) was tested on mice. Comprehensive scoring method was applied to evaluate the effect to determine the optimum dose. The results showed the optimized prescription was composed of Radix Sophorae Flavescentis and Schisandra chinensis. The optimum ratio was 1:3. It was concluded that the uniform design was applied in the experimental design with 3 factors and 6 levels. Comprehensive scoring method was used to evaluate the combined effect. The optimized prescription that put benefit and risk into consideration was composed of Radix Sophorae Flavescentis and Schisandra chinensis. The best ratio was 1:3, which can be further developed.
10.Effects of recombined neuregulin on healthy Macaca mulatta hearts.

To explore the effects of recombined Neuregulin on the heart of healthy Macaca mulatta, 10 healthy adult Macaca mulatta were randomly divided into two groups and were injected with the same doses of recombined Neuregulin and normal saline, respectively. At the same time, related indices were detected by 2-dimensional echocardiography and M-mode echocardiography. All indices were compared between the two groups and among different phases. Recombined Neuregulin had effects on LVEDD (left ventricular end-diastolic diameter), LVEDV (left ventricular end-diastolic volume), LVESV (left ventricular end-systolic volume) and SV (Stroke volume), and the effects changed with time. However, no significant changes were seen on EF (Ejection fraction) and FS (Fractional shortening). In conclusion, recombined Neuregulin has effects on the left ventricular volume of healthy Macaca mulatta, but no significant effect on cardiac contractility.
