1.Clinical efficacies of Jiangzhi Decoction for lipid metabolism disorder patients with traditional Chinese medicine syndrome of phlegm and turbidity retardation

A total of 101 lipid metabolism disorder patients with traditional Chinese medicine (TCM) syndrome of phlegm and turbidity retardation were collected and randomly divided into group A:Jiangzhi Decoction (JZF) alone (n =30),group B:atorvastatin (ATV) alone (n =39) and group C:JZF plus ATV (n =32).Lipid level,liver and kidney function,total effective rate and TCM scores were observed after a course of 2 weeks.Total effective rate:Group C was significantly superior to group B (90.62% vs.76.92%,u =1.97,P < 0.05) ; TCM score:Group C had a statistically superior advantage over groups A andB (C vs.A,90.62% vs.80.00%,u=1.98,P<0.05; Cvs.B,90.62% vs.51.28%,u=4.11,P <0.01); TC and Improvements of total cholesterol (TC) and low-density lipoprotein-cholesterol (LDL-C):Group C demonstrated marked decreases in both TC and LDL (CTC & CLDL-C vs.group A,P < 0.01 ; CTC & CLDL-C vs.group B,P < 0.01),no abnormal liver or kidney function occurred during the follow-up period.Thus JZF and ATV may have synergic lipid-lowering effects.And single uses of JZF and ATV have no marked differences in short term efficacy.
2.Study on clinical efficacy and endothelial function protection of Tianma Gouteng Granule in interfering primary hypertension patients with hyperactivity of liver-yang

Objective To investigate clinical anti-hypertension effect,clinical traditional Chinese medicine (TCM) syndrome improvement and endothelium function such as cardio-ankle vascular index (CAVI),flow-mediated dilatation (FMD) and endothelial micro-particle (EMP) protection of Tianma Gouteng Granule in interfering primary hypertension patients with hyperactivity of liver-yang.Methods Sixty six primary hypertension patients with hyperactivity of liver-yang were randomly divided into the control group (standardized treatment,n =35) and the treatment group (Tianma Gouteng Granule + standardized treatment,n =31) according to the random number method.In four weeks' follow-up treatment,the changes in clinical efficiency,TCM syndrome improvement,CAVI,FMD and EMP were observed.Results (1)Total clinical efficiency ratio was 71.43% in control vs.70.97% in treatment without statistical significance (P >0.05).(2) TCM syndrome improvement was better in treatment group than that in control group (93.55% vs 74.29%,x2 =4.39,P < 0.05),especially in improving symptoms such as dissiness,headache,dysphoria in chest,palms and soles,insomia and tinnitus (P < 0.05).(3) In the aspect of endothelium protection,treatment group can greatly improve CAVI compared with control (L-CAVI ∶ t =2.11,P <0.05;R-CAVI∶ t =2.15,P <0.05) ; the levels of FMD(t =-4.65,P <0.01) and EMP (t =4.35,P < 0.01)were significantly improved after treatment (P < 0.05) while no statistical meaning was found between two group.Conclusions Tianma Gouteng Granule can improve TCM syndrome,decrease CAVI level.FMD and EMP were significantly decreased in treatment group.Multi-factor regression analysis showed that EMP level was related to pulse pressure.
3.Inhibitory effect of cajanonic acid A on lipogenesis and lipolysis in 3 T3-L1 adipocytes

Aim To investigate the effects of cajanonic acid A (CAA) on lipid metabolism in murine 3T3-L1 adipocytes. Methods 3T3-L1 cells induced to differ-entiated into mature adipocytes were treated with CAA in different dosages for 48 h, then total lipids as well as triglyceride, free fatty acid and glycerol were meas-ured. The expression levels of genes related to lipid metabolism were quantitatively analyzed by real-time fluorescent quantitative polymearase chain reaction ( RTFQ-PCR) . Results Total lipids and triglyceride in 3T3-L1 adipocytes were markedly reduced by CAA. The release of free fatty acid and glycerol was lower than that of control. This coincided with decreased mRNA levels of the key enzymes involved in de novo lipogenesis ( acetyl CoA carboxylase and fatty acid syn-thase) , fatty acid uptake ( lipoprotein lipase) , and li-polysis ( hormone sensitive lipase and adipose triglycer-ide lipase ) . While the expression of fatty acid oxida-tive genes including acyl CoA oxidase and carnitine palmitoyl transferase1 was increased after CAA treat-ment. Conclusion CAA may inhibit lipogenesis and lipolysis,reduce circulating free fatty acid and improve the lipid metabolism in adipocytes by regulating gene expressions.
4.Analysis of the Essential Oil from Cassia Mimosoides

Objective To identify the volatile chemical components of cassia mimosoides. Method The volatile components were distilled with steam distillation,and then their compounds were confirmed by GC-MS. Results Fifteen peaks were detected by GC-MS,and 14 components of them were identified. The identified compounds occupied 98.97% of the volatile oil. The main component is 6,10,14-trimethyl-2-pentadecanone (43.86%),then is phytol (36.54%).Conclusion 6,10,14-trimethyl-2-pentadecanone can be used for the identification of this plant.

Objective To confirm the immunomodulatory effect of ursolic acid on normal and immunosuppressive mice caused by cyclophosphamide.Methods We assayed the function of T lymphocyte transformation with MTT test,examined the function of B lymphocyte with hemolysis spectrophotography,determined the function of non-specific immunity with phagocytosis of macrophages,and measured the content changes of Th_(1) cytokine IL-2 and Th_(2) cytokine IL-6 in serum with ELISA.Results Ursolic acid had a suppressant effect on the function of humoral immunity and enhanced cell-mediated immune response at the low dosage but presented opposite effect at the high dosage in normal mice.Ursolic acid significantly improved immune function on immunosuppressive mice caused by cyclophosphamide(P

Objective To study the relationship among the angiotensinogen (AGT)gene T174M,M235T polymorphisma,blood glucose level and atherosclerotic cerebral infarction.Methods The polymerase chain reaction-restriction fragment length polymor-phism (PCR-RFLP)method was adopted to detecte the gene polymorphisms of AGT gene 174,235 sites and the fully automatic bi-ochemical analyzer was used to detect the biochemical indexes of GLU,etc.in 396 patients with atherosclerotic cerebral infarction and 360 normal controls.Results The GLU level in the patients of the ACI group carrying genotype TT and TM at AGT gene T174M site was higher than that in the normal control group with statistical differences(P <0.05),the glucose level had no statisti-cal difference between the different 2 kinds of genotype (P >0.05);the glucose level in the patients carrying genotypes MM,MT and TT at M235T site in the ACI group was higher than that in the normal group,and the difference was statistically significant(P<0.05).The blood glucose level between the two groups carrying 2 different kinds of genotype showed no statistically significant difference (P >0.05 ).Conclusion No correlation is found among AGT gene T174M,M235T polymorphism,blood glucose level and atherosclerotic cerebral infarction;hyperglycemia is one of the risk factors of atherosclerotic cerebral infarction occurrence.
8.Influence of growth hormone receptor gene knockout in pancreaticβcells in STZ-induced type 1 diabetes in mice

Objective To investigate the influence of tissue-specific growth hormone receptor (GHR)deficiency in type 1 diabetes in the mice at the gene level using pancreaticβcells combined with streptozotocin (STZ)-induced type 1 diabetes model.Methods The experiment was divided into four groups:knockout mice group (LLc knockout group), using the homozygotes (LLc:LL+Cre) producted by pancreaticβ cell-specific expressed recombinant enzyme mice (RIP-Cre)and Cre-LoxP system modified GHR mice (Floxed,LL);LL control group, containing Floxed GHR allele homozygous mice (LL);LLc STZ group and LL STZ group (STZ was used for inducing type 1 diabetes model mice). The mice with feeding glucose≥25 mmol · L-1 were considered to be successful models.The Glucose Tolerance Test (GTT),pancreas tissue HE staining and immunohistochemistry were performed in the mice.Results The blood glucose of the mice in LL STZ group and LLc STZ group and LLc STZ group were increased after inj ection of STZ and the models achieved the diagnostic criteria for diabetes 1 6 d later.The results of GTT showed that compared with LLc control group and LLc knockout group, the blood glucose levels of the mice in LL STZ and LLc STZ groups were increased (P<0.05).There was no significant change of morphology and structure of islets between LL control group and LLc knockout group detected by HE staining. The immunohistochemistry results showed that the insulin level of the mice in LL STZ group was significantly reduced compared with LL control group;the insulin level of the mice in LLc STZ group was reduced compared with LLc control group.Conclusion Pancreaticβcell GHR gene knockout has no effect on the blood glucose and the function ofβcells in the mice with STZ-induced type 1 diabetes.
9.Effect of stroke-associated pneumonia on the prognosis of the elderly hospitalized for acute stroke in the intensive care unit

Objective To investigate the risk factors for death within 30 days of stroke associated pneumonia(SAP) in elderly patients of the intensive care unit (ICU).Methods Clinical data of 116 patients with SAP who were admitted to ICU were reviewed.The predicting factors of death within 30 days were analyzed through variable analysis method.Results The hospitalization periods of the SAP group and control group were (27±8) d and (12±5) d,the difference was statistically significant(t =1 1.30,P =0.002).The most common pathogenic bacteria in SAP were Klebsiella pneumoniae,Bauman Acinetobacter,Escherichia coli,and gram negative bacteria.There was significant difference between the two groups in term of mortality rate(37.9%(44/ 116) of SAP group,22.0% (22/100) of control group,x2 =6.423,P =0.011).Logistic regression analysis showed that basic diseases(OR =2.778,95% CI:1.205-6.401),high CRUB-65 score (OR =1.978,95% CI:0.871-11.098),lower Glasgow coma score(GCS) (OR=3.601,95%CI:0.244-9.477),bucking(OR=3.020,95%CI:1.305-10.603),mechanical ventilation(OR=2.654,95%CI:1.176-5.990),shock(OR=2.636,95% CI:1.164-5.969) and high plasma CRP level(OR=2.333,95%CI:1.046-5.206) were risk factors for SAP (all P<0.05).In Cox regression analysis,a GCS score of < 9 was an independent risk factor for 30 d mortality of patientswithSAP(HR=7.23,95%Cl:2.24-20.11,P=0.001).Conclusion SAP is one of the serious complications of the elderly hospitalized for the acute stroke in the ICU,which may affect the prognosis of the patients.
