Objective To study the function of let-7 and miR-24 in UVB-induced apoptosis.Methods After NIH3T3 cells were irradiated with UVB.Hoechest33342/PI staining was used to study the cell apoptosis and RT-PCR was used to uralyzc the expression level of let-7 and miR-24.In addition,potential target genes of these miRNAs in PicTar were classified into different function categories through GOstat software.Results Compared to the control,the NIH3T3 cells exposure to UVB appeared typical apoptotic and necrotic ceils by fluorescence microscope.The exprossion level of let-7 and miR-24 in NIH3T3 cells after UVB irradiation was higher than that of the control.Among the target genes,casp3,bc1212,map3kl and cdk5 were also involved in UVB-induced apoptosis mechanism.Conclusion Let-7 and miR-24 play a role in UVB-induced apoptosis.

Objective To study the electrophysiologic characteristics of atrial-ventricular reentrant tachycardia(AVRT)characterized by paroxysmal occurrence that slow atrioventricular accessory pathway participatesin.Methods Twenty-one cases were chosen from patients receiring radiofrequency ablation therapy in Peking University People's Hospital from July in 1999 to January of 2005.The patients with slow atrioventricular accessory pathways diagnosed correctly were divided into two groups with paroxysmal tachycardia and permanent tachycardia in terms of the occurrent frequence of AVRT.The electrophysiologic features of AVRT of two groups were contrastively analyzed.Results Compared with the group of permanent AVRT,it was found that antidromic refractory period of slow atrioventricular accessory pathways was longer[(359?46)ms vs (318?31)ms,P

Objective: Effects of surfactant replacement were evaluated on graft pulmonary function in rats. Method: Forty adult Wistar rats were anesthetized with an intraperitoneal injection of pentobarbital sodium(30 mg?kg~(-1)), and intubated through tracheolomy. All rats were ventilated with a ventilator. Surfactant of 50 mg was injected into lungs through tracheal. Cold Ringer's solution(0℃-4℃)was infused into the lungs through pulmonary artery, and then they were kept in cold solution. Result: The static lung compliance (SLC)in the both surfactant-replaced S(no airway pressurc)and SP(0.98kPa airway pressure)groups did not markedly decrease after cold storage. However,SLC in the no surfactant-replaced NP (no airway pressure)and P (0.98kPa airway pressure) groups both significantly decreased after cold storage. Objective To study the expression level of miR-21 in UVB irradiated HaCaT cells and A431 cells. Methods Real-time qPCR was used to examine the expression level of miR-21 in HaCaT cells and A431 cells after 50 J/m2UVB radiation. The possible target genes were predicted by PicTar and performed function categories with Gostat analysis. Results Compared with the HaCaT cells, miR-21 the expression level in A431 cells increased over 4 times. At 2h and 4h after UVB irradiation, the expression level of miR-21 in HaCaT cells were up regulated, and it lowered 2 times at 8 h compared with the control.There was no further change in the expression level of miR-21 after 12 h. While miR-21 expression levels in A431 cells were not changed signifcantly. The results of target prediction and Gostat analysis suggested that PIK3R1, BCL2 and E2F3 were involved in the cell differentiation and cell process. Conclusion miR-21 possibly involved in the pathogenesis of epidermal squamous cell carcinoma and the mechanism of UVB-induced injury.

Objective To study the effects of the active components of eight kin ds of Chinese herbs on cytochrome P450 enzymes (CYP) 1A1,2E1,3A4 and 3A5 mRNA expression. Methods The mRNA expression levels of four CYP enzymes were determin ed by real-time quantitative reverse-transcriptase polymerase chain reaction. Results Baicalin,baicalein and artemisinin induced CYP1A1 expression at differe nt concentrations. Compared with baicalin and baicalein,the effect of artemisin in was weaker. The expression of CYP3A4 gene was significantly obvious after ind uced by sodium aescinate,baicalein and artemisinin. Conclusion HepG2 cells shou ld be an appropriate in-vitro system for investigating potential human CYP indu cing agents. CYP1A1 and CYP3A4 expression could be significantly induced by baic alin,baicalein,artemisinin and sodium aescinate,which would supply the eviden ce for the interaction of herbal medicine and western medicine based on cytochro me P450 and toxicology.

Objective To evaluate the kinetics of peripheral blood cells in DTC patients before and after 131I treatment.Methods A total of 64 patients were divided into 2 groups with different therapeutic doses:high-dose group (3.70-5.55 GBq,n =24) and low-dose group (1.11 GBq,n =40).The WBC,neutrophils (NEUT),lymphocytes (LY),RBC and PLT were counted before operation,before 131I treatment,and on 3 d and 7 d after 131I treatment.One-way analysis of variance and two-sample t test were used to analyze the data.Results The counts of WBC and NEUT in both groups along with the LY in high-dose group varied significantly before,and on 3 d and 7 d after 131I treatment(WBC:high-dose group,(6.30±1.04),(8.86±2.07),(6.59±1.64) × 109/L;low-dose group,(6.65±1.48),(10.17±3.04),(7.17± 1.57) ×109/L; NEUT:high-dose group,(3.75±0.88),(6.42± 1.91),(4.53± 1.54) × 109/L; low-dose group,(3.88±0.90),(7.12±2.77),(4.40±1.17) × 109/L;LY:(2.11±0.67),(2.06±0.74),(1.59±0.49) × 109/L;F values:3.88 to 30.20,all P＜0.05).The counts of WBC and NEUT in both groups were significantly higher on 3 d after 131I treatment than that before treatment (all P＜0.05).The counts of WBC and NEUT in both groups along with the LY in high-dose group decreased significantly on 7 d compared to that on 3 d after 131I treatment (all P＜0.05).The counts of LY in high-dose group also significantly decreased on 7 d after 131I treatment than before treatment(P＜0.05).The counts of RBC before 131I treatment and LY on 7 d after 131I treatment were significantly different between the 2 groups(t=2.36,-4.30,both P＜0.05).Compared with the counts before operation,LY,RBC and PLT were significantly higher (t values:from-4.92 to-2.45,all P＜0.05) during hypothyroid state induced by thyroxine withdrawal before 131I treatment.Conclusions Short-term kinetics of WBC and NEUT present as an increase first followed by a decrease after 131I treatment; while LY of high-dose group presents as a gradually decrease.Hypothyroid state induced by levo-thyroxine withdrawal leads to increased counts of LY,RBC and PLT before 131I treatment.

Objective:To determine the effect of apatinib combined with Xiaoyan decoction for the treatment of non-squamous non-small cell lung cancer. Methods:Thirty-eight patients with non-squamous non-small cell lung cancer were randomly categorized into apatinib group (group A, 18 cases) and apatinib combined with Xiaoyan decoction group (group B, 20 cases). All patients did not under-go surgical treatment, radiotherapy, or chemotherapy during the study. Results:The median progression free survival (mPFS) of ad-vanced non-squamous non-small cell lung cancer patients reached up to 3 months. The mPFS, objective response rate, and disease control rate of the apatinib combined with Xiaoyan decoction group showed no significant difference and statistical significance (P>0.05). The apatinib combined with Xiaoyan decoction group was superior to the apatinib group with regard to alleviating clinical symp-toms and adverse reactions (P<0.05). Conclusion:Xiaoyan decoction combined with apatinib can improve the clinical symptoms of pa-tients and reduce the incidence of adverse reactions in the treatment of advanced non-squamous non-small cell lung cancer.

Objective To study the role of psychological factors in the onset of psoriasis,to investigate the role of monoamine substances in the course of psychological factors-associated psoriasis,and to reveal the psychoneurologic pathogenesis of psoriasis.Methods The psychological factors were assessed with life events scale and questionnaire of A character.The serum levels of norepinephrine(NE),dopamine(DA)and5-hydroxyindoleacetic acid(5-HIAA)were detected with high-performance liquid chromatography(HPLC).Results It was shown that there was an overall tendency of weak A character and moderate depression and anxiety in psoriasis patients.The concentrations of NE,DA and5-HIAA were higher in psoriatic patients than those in normal controls(P

Objective To explore the relationship between embryo damage and anxiety,depression and life events during pregnancy. Methods A case control study was carried out. 192 women diagnosed with embryo damage were selected as the case group,and 192 normal pregnant women or normal parturition within 7 days were selected as the control group from March to June,2014.All cases were from two 3-A-grade general hospitals of Jining. Self-rating anxiety scale ( SAS ) , center for epidemiological survey, depression scale( CES-D) and life events scale for pregnant women( LESPW) were used to evaluate the anxiety,depres-sion and life events of the two groups,so as to analyze the association between these factors and embryo dam-age. Results Multiple non-conditional logistic regression analysis showed that anxiety ( β=0. 409, OR=1.050,95%CI =1.003-1.009) ,depression( β=0.085, OR=1.088,95%CI =1.035-1.145) ,life events dur-ing pregnancy( β=0.003, OR=1.003,95%CI =1.001-1.005) were the risk factors of embryo damage,while education ( β=-0.749, OR=0.452,95%CI=0.298-0.685) ,pre pregnancy health status ( β=-1.366, OR=0.255,95%CI =0.098-0.655) and taking the initiative to acquire knowledge of eugenics ( β=-0.884, OR=0.413,95%CI =0.203-0.840) were the protective factors of embryo damage. Conclusion Anxiety,depres-sion and life events during pregnancy are related to the occurrence of embryo damage. High degree of educa-tion,pre pregnancy in good health and taking the initiative to acquire eugenics knowledge can reduce the oc-currence of embryo damage.
