2.Effects of anti-VEGF antibody on angiogenesis and proliferation and apoptosis of human osteosarcoma cell line OS-732

Objective To investigate the effects of anti-VEGF antibody on angiogenesis and on the proliferation and apoptosis of the osteosarcoma OS-732 cells and the vascular endothelial cells. Methods The effects of polyclonal antibody against VEGF on OS-732 induced angiogenesis were observed by a stero-microscope and light microscope in a tumor model of the chick embryo chorioallantoic membrane (CAM). Furtheremore, the proliferation and apoptosis of tumor cells and vascular endothelial cells were examined by TdT-mediated dUTP nick and labelling (TUNEL) and immunohistochemical staining by using monoclonal antibody for proliferating cell nuclear antigen (PCNA). Results The number of both the newly formed vessels in the tumor and the tumor population were significantly decreased in the anti-VEGF antibody group as compared with that of the PBS control group. The apoptotic index for tumor cell was higher in the anti-VEGF antibody treated group than that in the negative control group, but the proliferation index was not significantly different between them. At the same time, increased apoptotic cells and decreased proliferation in vascular endothelial cells were also noted. Conclusion VEGF antibody can inhibit the neovascularization of OS-732, probabaly by the mechanism of inhibi-ting the proliferation and promoting the apoptosis of vascular endothelial cells, further, might be contributive to the apoptosis of tumor cells and result in suppression of tumor growth.
3.Expression of VEGF and bFGF in xenografts of human osteosarcoma OS-732 on chick embryo chorioallantoic membrane and its significance

Purpose:To study the characterics of angiogenesis induced by osteosarcoma OS-732 cell line. Methods:With a tumor model of the chick embryo chorioallantoic membrane(CAM),the angiogenesis induced by OS-732 cell line was observed by a stereo-microscope and transmission electron microscope, and the expression of angiogenic factors vascular endothelial growth factor(VEGF) and basic fibroblast growth factor(bFGF) in xenografts of OS-732 cell line on CAM was detected by immunohistochemical technique. Results:This cell line was strongly angiogenic, which probably is associated with production of many factors.The new capillaries converging upon the tumor can be observed by a stereo-microscope. Furthermore,under transmission electron microscope, it also can be noted that the new blood vessel consisted of a monolayer of vascular endothelial cells with enlarged fissures,and the basal membrane was not intact. The expression of VEGF and bFGF was positive in xenografts on CAM, and VEGF was expressed at high levels constantly. Conclusions:The new blood vessels induced by tumors have pathologic and physiopathologic characteristics and many angiogenic factors are alm involved in the angiogenic process of OS-732. Furthermore, the application of angiogenic factors as a target in antiangiogenic therapy of osteosarcoma may be useful to improve the prognosis of patients with this disease.
4.Therapeutic Effect of Kummerowia Striata on IgA Nephropathy Rats

Objective To examine the therapeutic effect of Kummerowia striata (KS) on IgA nephropathy (IgAN) rats.Methods Sixty SD rats were equally randomized into 6 groups after adaptive feeding for 3 days:blank control group,model control group,tripterygium glycosides (TG) group (in the dose of 0.0165 g/kg),and high-,middle-and low-dose KS groups (in the dose of 16.0,8.0,4.0 g/kg respectively).IgAN rat model was established with bovine serum albumin (BSA),lipopolysaccharide (LPS) and carbon tetrachloride.After modeling,rats except those in blank control group received gastric gavage of corresponding drugs for 10 continuous weeks.Levels of 24-hour proteinuria and hematuria in each group were measured,serum levels of total protein (TP),serum albumin (ALB),aspartate aminotransferase (AST),and alanine aminotransferase (ALT) were tested.The pathological changes of renal tissue were observed under light microscope by HE staining.Results Water decoction of KS could significantly decrease the proteinuria and hematuria levels,and reduce renal pathological changes of IgAN model rats.However,water decoction had no effect on indicators of liver function such as TP,ALB,AST,and ALT in IgAN rats.Conclusion KS has certain therapeutic effect on IgAN model rats.
5.The cytotoxity of paclitaxel-nanoparticles with dual ligands combined with ultrasound irradiation on drug-resistant breast cancer cells

Objective To screening optimized ultrasound (US)condition for MCF-7 cells and MCF-7/Tax cells,and to investigate the effect of dual-ligands nanoparticles (NPs ) combined with US on overcoming multidrug resistance (MDR).Methods To obtain optimized US condition,cell viability and cellular uptake of NPs treated with MCF-7 cells and MCF-7/Tax cells were detected under different ultrasonic parameters including ultrasound intensity,exposure duration and microbubble concentration. MTT assay was performed to observe the effect of NPs combined with US on cells.Results The optimal US conditions for MCF-7 cells was irradiation intensity 0.6 W/cm2 ,irradiation time 80 s,and the MBs concentration with volume ratio of microbubbles to medium was 1∶1 0;and for MCF-7/Tax cells,irradiation intensity 0.6 W/cm2 ,irradiation time 80 s,and the MBs concentration with volume ratio of microbubbles to medium was 1∶20.Compared with individual NPs,NPs assisted by US exhibited greater killing ability for MCF-7 (IC50 1 .43 ng/ml)and MCF-7/Tax cells (IC50<800 ng/ml).Conclusions US effect could enhance drug sensitivity of the dual-functionized NPs for MCF-7/Tax cells,indicating that US irradiation had potential assisted modality to reverse tumor MDR.
6.Auricular Composite Tissue Flap for Repairing Nasal Ala Defect

Objective To discuss the effects of repairing nasal ala defects by free transplantation of autogenous auricle composite tissue flap. Methods 50 cases with nasal ala defects were repaired by free auricular composite tissue flap transplantation from Janu 2003 to May 2013. The defects size was 0.5cm x 0.7cm~1.3cm x 1.5cm. According to the size of the defects, full thick wedge-shaped auricle composite tissue flap were cut off, then inserted into the nasal ala defects area and fixed stablely, the donor sites were sutured directly avoiding ear cartilage. Salvianolate and hyperbaric oxygen were used in postoperative treatment for 5-7 days. Results 48 cases achieved good results, 2 cases had partial necrosis of composite tissue flap after operation, also achieved good results after reoperation. Over 3 months~5 years follow-up, all cases had satisfactory results. The volume of auricle composite tissue flap reduced less than 10%. Surgical incisions had a linear scar, good color matching, nostril symmetry. All patients were satisfied with the overall appearance. Conclusions Autogenous auricle composite tissue flap free transplantation for repairing middle and small size of nasal ala defects can recover ala formation and structure very well, nasal appearance can be improved greatly with nostril symmetry. There is no hypertrophic scar in donor sites. This method is simple and easy,and is also a good method for repairing nasal ala defects.

In present paper, research achievements of post-harvest handling concerning decorticating, boiling, sul-fur-fumigation, drying and sterilizing as well as quality control of Radix Paeoniae Alba were reviewed. Major prob-lems in the post-harvest handling and quality evaluation of Radix Paeoniae Alba were discussed. Strategies for im-provement in post-harvest handling and quality control were proposed, so that the safety and quality consistency can be assured in the medicinal application of Radix Paeoniae Alba.
8.Clinical observation of different narcotic drugs compatibility in elderly patients with laparoscopic tumor resection

Objective To investigate clinical observation of different narcotic drugs compatibility in elderly patients with laparoscopic resection of tumor .Methods From June 2015 to June 2017 ,86 cases of elderly lapa-roscopic tumor resection were selected from our hospital .They were divided into the observation group and the control group according to the random number table ,each with 43 cases .The control group received propofol and dexmedetomidine anesthesia ,and the observation group received sevoflurane and dexmedetomidine anes-thesia maintenance .Results The observation group anesthesia total satisfaction rate (97 .67%) higher than that of the control group (79 .07%) ,the difference was statistically significant (P<0 .05) ;The two group af-ter 3 h and 24 h OAAS score increased after operation ,the difference was statistically significant (P<0 .05) ;The patients in the observation group 3 h and 24 h OAAS were higher than those of the control group ,the difference was statistically significant (P<0 .05) ;The observation group extubation time ,called the open time and the recovery time of spontaneous breathing faster than the control group ,the difference was statistically significant (P<0 .05) ;Two group T1 time HR MAP compared with T0 time reduction ,the difference was sta-tistically significant (P<0 .05) ;The control group at T2 of HR and T0 of MAP were constantly reduced ,the difference was statistically significant (P<0 .05) ,while the observation group at T2 HR and MAP did not change significantly compared with T0 time ,the difference was not statistically significant (P>0 .05) ;Two group T0 ,T1 and T2 HR ,MAP and SpO2time was not statistically significant (P>0 .05) ;The observation group T1 and T2 HR ,MAP times higher than that of the control group ,the difference was statistically signifi-cant(P<0 .05) .Conclusion Sevoflurane combined with dexmedetomidine combined anesthesia in elderly pa- tients with laparoscopic resection surgery is obviously good effect ,little influence on sedation ,patients with HR ,MAP and SpO2.

Mitochondrial DNA (mt-DNA) is an important carrier of extranuclear genetic information. Recent research results show that mt-DNA is closely related to the occurrence and metastasis of various malignant tumors, and can be used for early diagnosis and targeted therapy of cancer. Therefore, further research on the mechanism of mt-DNA in digestive system malignant tumors has important clinical significance for screening and identifying tumor molecular markers for anti-tumor drug targets, cancer diagnosis and prognosis analysis. This article reviews the research progress on the potential relationship, clinical application and therapeutic targets of mt-DNA and digestive system malignancies.

Objective To discuss clinical curative effects of using Forehead skin expansion combine with auricular cartilage for repairing nose alar full-thickness defects.Methods From August 2010 to August 2010,36 patients with nose alar full-thickness defects in the second affiliated hospital of kunmin medical university,The defect range exceed 1.5 cm× 1.2 cm.50-80 ml expander was implanted in forehead and injected saline water to expand in order to acquire extra skin.We Turn around the skin of defect as the lining of nose,harvest ipsilateral auricular cartilage for nose ala framework,Expanded forehead pedicle flap was transferred to cover framework.The donor area was sutured directly.The pedicle of flap was cut and trimmed after 2 months.Results Follow-up time of 3-18 months after the operation,All flaps are survive,nose alar defects are repaired successfully.Some cases were performed second surgery,postoperative,nose alar color,thickness,nostril size and shape the same with the contralateral side.Donor site healed with linear scar.Conclusions This method could be easy to obtain excess skin,for repairing large sides nose alar full-thickness defect.Frontal scar is not obvious,It is a practical.
