1.T cell subsets and the related changes in cytokine in children with recurrent respiratory tract infection

Objectlve To investigate recurrent respiratory tract infection in children with T-cell subsets and related eytokines change.Methods Choose meet the RRTI diagnostic criteria for children and healthy infants,application APAAP double antibody ELISA method and the sandwich method ty detect RRTI other healthy infants and children with the T-lymphocyte subsets and cytokines interferon γ(IFN-γ),interleuldn 10(IL-IO),interleukin 6 (IL-6)levels,and compared,Results Children with recurrent respiratory tract infection of T lymphocyte subsets CD+3,CD+4decreased co;increased serum IL-6(2.5±1.6 ku/L),IL-10(3.1±2.4 ku/L)and IFN-γ(12.6±6.4μg/L)levels were significantly below the healthy group(6.8±4.7 ku/L、7.4±3.5 ku/L、21.5±9.7μg/L)(P<0.01).Conclusion The boay of the RRTI children humoral and cellular immune existed varying degrees of functional defects.
2.Comparative study on diagnosis of fetal dysplasia of spine and spinal cord between B ultrasonography and magnetic resonance imaging

Objective To compare the value of ultrasound and magnetic resonance imaging (MRI) in detecting fetal spine and spinal cord dysplasia Methods Thirty women with complicated pregnancies,age from 22 to 41 years,gestation from 23 - 38 weeks,were studied with MRI within 72 hours after ultrasound studies and revealed 19 cases of fetal spine with irregular,7 cases of vertebral canal widened partly and 4 cases of anomalies of spinal curvature.Autopsy or imaging was conducted when pregnancy was terminated.A close follow-up was given for cases continuing pregnancy within 12 months after delivery.Results (1)Nineteen cases of fetal spine with irregular in ultrasound,the coincidence of antenatal sonographic diagnosis was 42% (8/19),the coincidence of antenatal MRI diagnosis was 89% (17/19).Seven cases of vertebral canal widened partly in ultrasound,the coincidence of antenatal sonographic diagnosis was 0,the coincidence of antenatal MRI diagnosis was 7/7.Four cases of anomalies of spinal curvature in ultrasound,the coincidence of antenatal sonographic diagnosis was 2/4,the coincidence of antenatal MRI diagnosis was 3/4.(2)By MRI study,one cases were conformed their ultrasound diagnosis,eleven cases were completed their ultrasound diagnosis,nine cases were made the same diagnosis as ultrasound and six cases were corrected diagnosis.By MRI and ultrasound study one case of hemivertebra,one case of butterfly vertebra and one case of dorsal dermal sinus were missed diagnosis.Ultrasond had limits and low specificity especially in fetal spinal cord.Conclusions MRI can show the spinal cord and its lesion intuitively and has advantages in displaying fetal anomaly of spine and spinal cord.MRI is a valuable complement to sonography in difficult cases and diagnostic accuracy can be significantly improved.
3.Human immunodeficiency virus vulnerabilities and related health risk behaviors of out-of-school children of the floating population in Beijing

Objective:To explore the vulnerability and related risk behaviors of the out-of-school adolescents floating with their parents from country to city.Methods:From September to October in 2005,260 out-of-school adolescents who were 14 to 20 years old were investigated in our study.Results:The out-of-school adolescents lacked of HIV/AIDS knowledge and life skills,and their psychosocial competence was at a lower level.The adolescents were open-minded to sex,but they were not conscious of using condoms.Attitude towards people with HIV was positively correlated with a well-being family,harmonious relation between their parents,a stable job,knowledge,life skills and psychosocial competence(such as in control of emotions,empathy).Their life skills were positively correlated with their mothers'schooling satisfaction with family income,a stable job,high scores of knowledge,social responsibility,and interpersonal relationship.Competence of HIV/AIDS prevention was positively correlated with schooling,a stable job,knowledge,life skills,and psychosocial competence.Conclusion:Out-of-school adolescents were vulnerable to HIV/AIDS.Individual level intervention combined with structural intervention could effectively reduce their vulnerability.Effective and feasible health education approach to enhance their life skills and psychosocial competence was a key point to health education on HIV/AIDS prevention.
4.Experimental Study of Transctheter Bilaterial Internal Iliac Artery Infusion with HCPT

Purpose:To evaluate the different concentration of HCPT in rabbit blood and bladder tissue between intraarterial and intravenous penfusion groups.To indentify the probability of using HCPT as a new type of anti-tumor agent delivered by the intra-arterial pesfusion method for the treating of bladder cancer.Materials and methods:Group A:40 white female rabbits were divided into 2 groups(intraarterial and intravenous)and each was subdivided according to the time intervalsof bladder tissue sampling,of 5,15,30,60,120 minites after the drug delivering.Group B:16 white female rabbits equally divieded into 2 groups(intraarterial and intravenous)for dynamic blood sampling by the intervals of 5,15,30,60,120 minitus after the drug delivering.The intra-arterial method using an artifial bilaterial super-select internal iliac artery perfunsion model defined previouysly.The intravenous method was to deliver drugs via earmarginal vena of rabbit.1mg/kg as the concentration of the drug in bladder tissure was measured by reversed-phase HPLC.Results:The concentration of the drug in bladder tissue was significantly different between the intra-arterial and intionenous groups.Drug in the bladder tissue delivered by intra-artial method was 2.17,3.41,1.51,1.52,2.33 times as much as by intra-venous method.While drugs in blood by intra-venous method was 1.92,2.03,1.77, 1.92,1.40 times as much as by intra-arterial method.Conclusion:HCPT is suitable for trans- arterial perfusion for the treating of bladder cancer.
5.Effect of vascular endothelial growth factor on vascular regeneration of reconstructed posterior cruciate ligament allograft transplantation in rabbits

BACKGROUND: Currently,the research of posterior cruciate ligament (PCL) construction mainly focuses on the surgical technique or graft selection. However,studies on vascular regeneration after construction are few,especially those on allograft construction. OBJECTIVE: To observe the effect of vascular endothelial growth factor (VEGF) on vascular regeneration of reconstructed PCL with femur-anterior cruciate ligament (ACL)-tibia allograft in rabbits. DESIGN,TIME AND SETTING: The factorial design experiment was performed at the Orthopedics Laboratory of the Third Affiliated Hospital of Weifang Medical College from March 2006 to September 2007. MATERIALS: Sixty-eight adult female Japanese white rabbits,weighing (3.3?0.1) kg,were adopted. Twenty-three of them were used to excide the femur-ACL-tibia complexes to establish the animal model of PCL reconstruction with allograft. METHODS: Forty-five rabbits were randomly divided into 3 groups,15 rabbits in each group. In the control group,no additional treatments were applied. In the phosphate-buffered saline (PBS) group,0.2 mL of PBS was injected into the knee joint. In the VEGF group,30 ?g VEGF mixed with 0.2 mL PBS was injected into the knee joint. MAIN OUTCOME MEASURES: The immunological rejection of allograft was observed after reconstruction. Five rabbits were randomly selected in each group at 3,6,12 weeks,respectively,for immunohistochemistry staining; and the microvessel density of the medio-one-third ligament part of allograft was evaluated with the Chalkley scoring method. RESULTS: Forty-five rabbits were involved in the result analysis. There were no hydrops articuli or aneretic cartilage when every rabbit’s knee joint was opened. The gross morphology of the grafts was similar to normal posterior cruciate ligament. Immunological rejection,such as degeneration,necrosis,dilapsus or defluxion was not appeared. Plenty of good vascular tissue in the arthrosis could be seen in the VEGF group,but could not be found in the control group or in the PBS group. This study is to investigate the role of endogenous CSE/H2S in regulating apoptosis of HepG2 cells. MTT and Trypan blue assay were performed to determine the effect of CSE inhibitor PAG and CSE siRNA on proliferation of HepG2. Production of H2S from HepG2 cells was assessed spectrophotometrically using N, N-dimethyl-p-phenylenediamine-dihydrochloride. Cells apoptosis was detected by means of double staining of Hoechst 33342 and PI with Array Scan V(TI)HCS600 High-Contents. Dihydroethidine (DHE) and 2', 7'-dichlorodihydrofluorescein diacetate (DCFH-DA) assay was used to determine intracellular superoxide anion and ROS level. Reduced glutathione (GSH) was determined by OxiSelect Total Glutathione Assay Kit. Recombinant plasmid pcDNA 3.1/myc-His(-)-CSE was constructed and transfected into 293T cells to rescue the ROS and GSH level to further investigate the effect of CSE/H2S on ROS and GSH. Western blotting was performed to test the effect of CSE siRNA on expression of activated caspase 3 and p-AKT and Nrf2 protein. The results showed that PAG and CSE siRNA could significantly decrease the production of H2S in HepG2 cells and inhibit the proliferation of HepG2 cells at a dose-dependent and time-dependent manner, respectively. PAG and CSE siRNA could promote the cell apoptosis of HepG2 cells. Moreover, PAG and CSE siRNA induced increased ROS generation and depletion of the critical antioxidant GSH and recombinant plasmid pcDNA 3.1/myc-His(-)-CSE rescued the level of ROS and GSH. Meanwhile, CSE siRNA increased the expression of activated caspase 3, but CSE siRNA did not affect the expression of p-AKT and Nrf2. These results suggested that the CSE/H2S pathway was involved in suppression of HepG2 cell growth and promoted apoptosis of HepG2 cells in an oxidative stress-dependent manner.
7.Effect of Dietary Medium-and Long-chain Triacylglycerols on Body Fat Accumulation in Subjects with Different Levels of Hypertriglyceridemia

Objective To investigate the effect of medium- and long-chain triacylglycerols on body fat accumulation in subjects with different levels of hypertriglyceridemia. Methods 112 subjects with hypertriglyceridemia were randomly divided into two groups: one group was demanded to adopt long-chain triacylglycerol (LCT) oil as their daily use and the other one to medium- and long-chain triacylglycerol (MLCT) oil for consecutive 8 weeks. Anthropometric measurements of body weight, body mass index (BMI), waist circumference (WC), hip circumference(HC), WHR (ratio of WC/HC), triceps skinfold thickness (TSF), subscapular skinfold thickness (SSF), body fat weight, body fat percentage, total fat area, visceral and subcutaneous fat area in abdominal and concentration of blood triglycerides (TG) were measured at the initial and final time of the study. In both groups, the cutpoint of TG 2.3 mmol/L was used to distinguish the different levels of hypertriglyceridemia. Results Compared with LCT group, body weight, BMI, WC, body fat weight, total fat area, visceral and subcutaneous fat area in abdominal in those of TG≤2.3 mmol/L in MLCT group were significantly decreased (P 2.3 mmol/L between the two groups. Furthermore, the concentrations of TG in hypertriglyceridemia subjects of different levels in MLCT group were both significantly lower than those in LCT group after 8 weeks (P 2.3mmol/L.
8.VDD Pacemaker in an Unipolar Lead for Clinical Practice

Objective To inquire into the forward clinical results of ventricular pacing, dual-chamber sensing, atrial-triggered, and ventricular-inhibited (VDD) Pacemaker in an unipolar lead.Methods 16 patients with the pacemaker were studied from 1993 to 2002, including 12 male patients and 4 female patients at 49 to 75 years old (average age 64 4 7 8 years old). They were in normal sinus rhythmia with complete or high degree atrial-ventricular block. Lead electrode was inserted through subclavian venous access, and the pacemaker was implanted in the same side as the electrode.Results All patients were followed up from 1 to 112 months (average 68 8 6 3 months) after pacemaker implantation. Their heart functions were improved, symptoms disappeared, life quality remarkably improved. Atrial sensing of VDD was good. One patient was died from coronary heart disease with heart failure by following up for 42 months, but the rest were healthy. Conclusions VDD pacemaker implantation was simple, easy to perform if its indication was appropriate. It could alleviate patients symptom and its therapy was effective. We should think highly of using VDD pacemaker, especially at poor region.
9.Determination of Thiomersal in Hepatitis B Vaccine by Flow Injection-Hydride Generation Atomic Absorption Spectrometry

OBJECTIVE: To establish a method for the determination of the content of thiomersal in hepatitis B vaccine. METHODS: The content of thiomersal in hepatitis B vaccine was determined by flow injection-hydride generation atomic absorption spectrometry. The samples were treated by reflux digestion method with 4% sulphuric acid as medium,0.5% potassium borohydride as reducing agent,and 0.5% sulphuric acid as supporting liquid. RESULTS: The linear range of thiomersal was 5.0~100 ?g?L-1,with an average recovery rate of 100.6%(RSD=0.89%) and the lowest detection limit of 1.00 ?g?L-1. CONCLUSION: The method is simple,rapid and sensitive,and suitable for the determination of the content of thiomersal in hepatitis B vaccine.
10.Supercritical CO_2 extraction of Rhizoma Chuanxiong volatile oil

AIM: Supercritical CO_2 extraction of Rhizoma Chuanxiong volatile oil was studied under conditions of extraction temperature between 33-48 ℃,pressure between 10-25 MPa and CO_2 flow rate between 2-4 L?min -1 .Influence of extraction conditions on solubility and mass transfer rate were analyzed. METHODS: Based on plug flow in fixed bed,experimental extraction curves were evaluated using a model put forward by Stastova.And the effects of extraction conditions on mass transfer coefficients were also analyzed. RESULTS: Coefficient of mass transfer was slowly increased as the extraction temperature elevated,and was in accord with flow and in contrast with pressure. CONCLUSION:The method has a practical use value in the extraction of Rhizoma Chuanxiong volatile oil.
