BACKGROUND: Electrostatic layer-by-layer (LbL) self-assembly is a simple, effective and novel approach to carrying macromolecules into microcapsule carriers.OBJECTIVE: To prepare chitosan (CS) microcapsules to control the releasing of heparin (Hep) by LbL.METHODS: CS microspheres were fabricated by a sodium sulfate-based precipitation process and then used as positively charged templates for polyelectrolyte multilayer coatings to carry heparin by the LbL self-assembly technique. The polycation was CS and the polyanion was Hep. The microcapsule was formed via coating {CS/Hep}3 on CS hydrogel template. The microcapsules composed of {CS/ Hep}3 on CS hydrogel templates were confirmed by fluorescence inverted microscopy, confocal laser scanning microscopy and laser particle size analysis. The assembly of polyelectrolyte layers on CS hydrogel microspheres was monitored by Zeta Potential Analyzer.RESULTS AND CONCLUSION: The resulting microcapsules were about 1 μm in average diameter, and allowed spontaneous loading of heparin through electrostatic interaction, with the encapsulation efficiency and carrying capacity of 83.8% and 3.05%, respectively.

AIM: Coding microsphere suspension chip is a novel protein chip technology, which is considered as the developing direction of medical tests. This study investigated the feasibility of coding microsphere suspension chip to detect coronary heart disease (CHD)-associated protein factors. METHODS: The serum samples of 30 patients with CHD were collected including 19 males and 11 females, with average age of (63?10) years in the Department of Cardiology, Zhujiang Hospital of Southern Medical University from August to September 2007. Patients did not have hepatic or renal dysfunction, cerebrovascular accident, acute infection or any anticoagulation medication. The regional ethics committee approved the study protocol and the informed consents were obtained from all subjects. The integrated detection method was established by coding microsphere suspension chip to detect CHD-associated protein factors including high sensitivity C reactive protein (hs-CRP), lipoprotein a (Lpa) and adiponectin (ADPN). Meanwhile, these three factors from 30 CHD cases were detected by the enzyme-linked immunosorbent assay (ELISA). The detection results by both methods were compared. RESULTS: The serum samples of 30 CHD cases were all included in final analysis. The detection results by coding microsphere suspension chip integrated detection method were hs-CRP (2.54?1.97) mg/L, Lpa (102.41?76.74) mg/L and ADPN (15.43? 9.87) mg/L. The detection results by ELISA were hs-CRP (3.93?2.52) mg/L, Lpa (208.63?81.23) mg/L, and ADPN (16.63? 8.21) mg/L. The detection results by coding microsphere suspension chip integrated detection method were correlated significantly to that of ELISA (hs-CRP: r=0.979, P

[Objective] To observe the counteractive action of cyclovirobuxinum D (Cvb-D) for myocardial ischemia and to explore its possible mechanism. [Methods] Sixty SD rats were randomized into blank control group, Cvb-D groups in the dosage of 2.2, 1.1 and 0.55 mg? kg-1 ? d-1 respectively, isosorbide dinitrate group (25 mg? kg-1 ?d-1). The blank control group was treated with distilled water and the other groups were treated with the corresponding drugs by gavage according to the experimental design for 30 days. One hour after the last medication, pituitrin (1.5 U/kg) was injected into the caudal vein of the rats except the blank control to induce coronary artery spasticity myocardial ischemia. After that, electrocardiogram (ECG) was recorded, plasma superoxide dismutase (SOD) activity, malonyldialdehyde (MDA), lactate dehydrogenase (LDH) and phosphocreatine kinase ( CPK) levels were detected. [ Results ] The incidence of ischemic changes in ECG was decreased, plasma SOD activity increased and MDA, LDH and CPK levels lowered in Cvb-D groups as compared with those in the model group (P
