1.Assaying of Fluoxetine in Microdialysate in Rat's Brain by HPLC

OBJECTIVE: To establish a HPLC method for the assaying of fluoxetine in microdialysate in rat's brain by HPLC. METHODS: Fluoxetine, which was extracted with N-hexane, was separated on ODS chromatographic column after derivatization by NBD-COCl,acetonitrile-water (50∶50) was the mobile phase with a flow rate of 1.2ml/min, column temperature of 35℃,fluorescence detection wave length ?ex at 478nm and ?em at 537nm. RESULTS: The linear range for fluoxetine in microdialysate in rats' brain was 10~100 nmol/L(r=0.9 996), and the quantitation limit was 10 nmol/L. CONCLUSIONS: The method is specific, sensitive and accurate,it is suitable for the assaying of fluoxetine in biological specimen.
2.Clinical observation of acupoint injection in the treatment of nonneoplastic epithelial disorders of vulva

Objective To observe the curative effect of acupoint injection of compound Danshen injection in the treatment of nonneoplastic epithelial disorders of vulva.Methods 96 patients with vaginal epithelial non tumor like lesions were enrolled in this study,including 38 cases of squamous epithelial hyperplasia,lichen sclerosus in 42 cases,lichen sclerosus associated with squamous cell hyperplasia in 16 cases.Take Zhōngjí,Guīlái,Huìyīn, Sānyīnjīao,Zhàohaǐ,and Tàichōng six points,each acupoint injected compound Danshen injection 2mL,injected twice a week,10 times for a course of treatment,after 2 weeks began next course of treatment,a total of three courses.The clinical curative effect and incidence of adverse reaction of three types of nonneoplastic epithelial disorders of vulva were compared.Results The effective rates (effective rate =clinical control +effective +valid)of three types of epi-thelial disorders of vulva tumor like lesions were squamous epithelial hyperplasia 92.11%,lichen sclerosus 90.48%, hardening of lichen planus associated with squamous cell hyperplasia 87.50%,there was no significant difference among the three types of lesions(χ2 =4.102,P >0.05).The total effective rate of three types of nonneqlastie epithe-lial disorders of tumor was 90.62% by acupoint injection,and there were no serious adverse reactions in all patients during therapy.A patient of lichen sclerosus appeared the symptoms of nausea,sweating during treatment due to nerv-ous,and after psychological guidance the symptoms disappeared,re -treatment did not appear.Conclusion The acu-point injection of compound Danshen injection in the treatment of vulvar intraepithelial non -tumor like lesions has better clinical effect,and has no serious adverse reactions.

5.High mobility group box 1 protein and intracerebral hemorrhage

High mobility group box l protein (HMGB1) is a highly conserved non-histone DNA binding protein,which is widely distributed in eukaryotic cells.HMGB1 is an important proinflammatory cytokine,involving in various pathological and physiological processes,such as inflammation,neurogenesis and angiogenesis.In the early stage of intracerebral hemorrhage,HMGB1 can induce secondary brain injury,such as inflammatory reaction,neuronal necrosis,and brain edema.However,in the late stage of intracerebral hemorrhage,HMGB1 can promote angiogenesis and neurogenesis,and thus improving neurological function.HMGB1 will become a novel therapeutic target for intracerebral hemorrhage in the future.

Objective:Investigate the awareness of their own rights and the safeguarding of the rights in practice,raise the patients' sense of safeguarding of the rights and help them defend the legal rights.Methods:A survey was taken among patients by applying the questionnaire.Results:There are 5 items about the patients' knowledge about their own rights and 3 items about defending their rights in practice,in which more than 85 percent of 111 patients chose the answer "always or almost".From this we can see that the awareness and safeguarding of their rights are not good enough.Conclusion: The government is appealed to break up the existence of the monopoly mold in medical treatment and to take measures to protect the rights of patients.The medical personnel should renew their concepts,pay more attention to publicizing and defending patients' rights,and help patients realize their rights.
9.Capability of internal transcribed spacer (ITS) region sequencing analysis to identify clinical isolates of filamentous fungi

Objective To evaluate the capability of internal transcribed spacer (ITS) region sequencing analysis to identify clinical isolates of filamentous fungi.Methods A total of 267 filamentous fungi isolates collected from clinical specimens were analyzed by ITS region sequencing analysis.Alignment of acquired sequences with known sequences in GenBank and MycoBank was conducted to identify the species of those isolates.Results ITS sequences of the 267 isolates were amplified successfully.Among these isolates, 53.9% (144/267) were identified to species level and 44.2% (118/267) to genus level.Only five isolates were failed to be identified at genus level as they shared >95% homology in ITS sequence with multiple genera.Conclusion ITS region sequencing analysis is preferred for identification of clinical isolates of filamentous fungi at genus level for its high universality and great capability.When species-level identification is required, some informative DNA markers besides ITS region should be included accordingly.
10.Prognostic significance of lymphatic density and mircovessel vessel density in breast invasive ductal carcinoma

Objective To investigate tumor lymphatic and mircovascular densities as prognostic markers in 69 cases of invasive breast cancer treated with partial or total mastectomy and lymph node dissection.Methods 69 cases of untreated primary unilateral invasive ductal breast carcinomas were selected.All cases were immunostained with D2-40 and CD31.Positively stained microvessels were counted in densely vascular/lymphatic foci (hot spots).The relationship between lymphatic vessel density (LVD),microvessel density(MVD) and prognosis was analyzed.Results The mean ± SD peritumoral lymphatic vessel density (P-LVD) was significantly higher than intratumoral LVD(I-LVD) (P < 0.01).There was a positive correlation of D2-40 LVD(peritumoral) counts with lymph node metastasis (P =0.003) and clinical stage (P =0.026),and CD31 microvessel density was found significantly associated with clinical stage(P =0.038).No significant association was found between above variants with I-LVD (P > 0.05).Univariate analysis showed that survival time was impaired by higher MVD and higher peritumoral LVD(P =0.007,P =0.008,P =0.014,P =0.024,log-rank test),but not I-LVD.Multivariate survival analysis showed that MVD,peritumoral LVD,TNM stage and lymph node metastasis were independent prognostic factors for overall survival.Conclusions Peritumoral LVD and MVD were significantly correlated with survival status of patients with breast cancer.This is the first attempt to predict prognosis of breast cancer patients by quantifying the peritumoral LVD and MVD.
