1.Molecular predictive biomarkers in non-small-cell lung cancer

With the development of phamacogenomic and molecular biology, it has been revealed that the curative effect of non-small-cell lung cancer (NSCLC) is closely related to tumor molecular markers. Predictive biomarkers in NSCLC patients, including ERCC1, BRCA1, hMLH1, hMSH2, RRM1, β-tubulin, TS,EGFR and K-ras, can predict individual patient's response to chemotherapy or targeted therapy, making individualized therapy be possible.
2.Efficacy of three liver-protecting drugs commonly used in treatment of drug-induced liver injury: a comparative analysis

ObjectiveTo compare the efficacy of three liver-protecting drugs commonly used in the treatment of drug-induced liver injury. MethodsThe clinical data of 90 patients with drug-induced liver injury who were treated in Yingcheng People′s Hospital from November 2012 to April 2015 were analyzed retrospectively, and according to the drugs administered, they were divided into groups A, B, and C, with 30 patients in each group. The patients in groups A, B, and C were treated with reduced glutathione, polyene phosphatidylcholine injection, and tiopronin injection, respectively. The efficacy, drug cost, and incidence of adverse events were compared between the three groups. An analysis of variance was used for comparison of continuous data between multiple groups, and the LSD t-test was used for comparison of continuous data between any two groups; the chi-square test was used for comparison of categorical data between multiple groups. ResultsGroup A had a significantly higher overall response rate than groups B and C (90.0% vs 73.3%/76.7%, χ2=2.78 and 3.75, both P<0.05). Group B had a significantly higher drug cost than groups A and C (316.12±4.05 RMB vs 235.13±2.90 and 135.21±7.62 RMB, both P<0.01). The incidence of adverse events showed no significant differences between the three groups (P>0.05). ConclusionIn the clinical treatment of drug-induced liver injury, reduced glutathione has better efficacy compared with polyene phosphatidylcholine and tiopronin, as well as a reasonable cost. Therefore, this drug is preferred in the treatment of this disease.
3.A twin study of behavioral problems and relations to genetic and environmental factors in adolescents

Objective:To explore the relations of adolescents'behavioral problems to genetic and environmental factors.Methods:66 twin pairs aged 12 to 18 years in Chongqing were included in the study.The Youth Self-Report (YSR) was used to evaluate behavioral problems.The blood sample was collected to identify zygosity of twins.Genetic effects were evaluated by comparing the phenotypic correlationamong monozygotic twins(rMZ) and the phenotypic correlation among dizygotic twins(rDZ).Structural equation modeling (SEM) was performed to evaluate the relations of adolescents behavioral problems to the additive genetic factors (A),shared environment factors (C) and individual specific environmental factors (E).Results:MZ correlations of Social problems and Delinquency were less than DZ correlations(rMZ < rDZ).It prompted that data was not suitable for genetic analysis.MZ correlations(rMZ) of any others (including social withdrawal,somatic complaints,anxiety/depression,thinking problem,attention problems,self-destructive/identity problem)were less than 2DZ correlations (2rDZ).It prompted to fit the ACE model.The optimal model was AE model.Among the contribution of the additive genetic factors (A) and individual specific environmental factors (E)to the total variance,the genetic factors ranged from 0.52 to 0.57 and individual specific environmental factors ranged from 0.43 to 0.48 in anxiety/depression and self-destructive/identity problem.For social withdrawal,somatic complaints,thinking problem and attention problems,the genetic factors ranged from 0.17 to 0.45and individual specific environmental factors ranged from 0.55 to 0.83.Conclusion:There is no obvious relationship between genetic effects and adolescent social problems or delinquency.Other behaviors problems (social withdrawal,somatic complaints,anxiety/depression,thinking problem,attention problem,self-destructive/identity problem)are correlated with additive genetic factors and individual specific environmental factors.Additive genetic factors have high-correlation with anxiety/depression and self-destructive/identity problem,and individual specific environmental factors have high-correlation with other factors.
4.Chemotherapy effect and prognosis analysis of elderly patients with advanced non-small-cell lung cancer

Objective To evaluate the curative effect and toxicities of platinum-based double regimens for patients aged ≥ 65 with advanced non-small-cell lung cancer (NSCLC) and identify the prognosis factors. Methods 70 patients aged ≥65 with staged ⅢA-Ⅳ NSCLC, who received platinum-based double regimens as first line treatment, were emrolled.Response rates and toxicities were evaluated.Progression free survival (PFS) and overall survival (OS) were estimated using the Kaplan-Meier method. Cox regression analysis was used to identify the potential prognosis factors.Results The median chemotherapy cycles was 3.The overall response rate was 41.5 % (22/53), and there was no difference between patients aged <70 and ≥70 (χ2 =1.945, P =0.378).The median PFS and OS were 6.0 months and 12.5 months.The chemotherapyrelated hematologic toxicities were common.Multivariate analysis revealed that performance status, numbers of metastasis, chemotherapy cycles were significant independent predictive factors for OS. Conclusion In elderly advanced NSCLC, platinum-based doublets show inspiring efficacy, but with more adverse events, and could not be all well tolerated. It should be personalized. Patients with poor performance status and multiple organs metastasis are hard to benefit from combined chemotherapy.Three to six cycles of chemotherapy is the optimal duration for patients who could be well tolerated.
5.The A-204C Polymorphism in CYP7A1 Gene Affects Its Promoter Activity

cholesterol 7α-hydroxylase gene ( CYP7A 1 ) plays a key role in the catabolism of cholesterol into bile acids. To investigate whether the A-204C polymorphism in CYP7A1 gene affects the gene expression,using luciferase as the reporter gene, four recombinants were constructed by inserting forward or reverse sequence with A or C allele at the polymorphism site into the promoter-less vector pGL3-basic. The constructs were then transfected into four cell lines and the luciferase activity of each expression vector was examined by dual luciferase reporter gene assay system. The results showed that activities of the forward sequence of both genotypes were higher than that of reverse sequence. Promoter activity of the recombinants with A allele was about one third lower than that with C allele. According to the analysis with TRANSFAC database, there may exist a Zic3 binding site when there is the C allele at -204. Our study indicates that the A-204 C polymorphism in CYP7A1 promoter region decreases its promoter activity and thus represses the gene expression, possibly due to the lack of a potential Zic3 binding site.
6.Relationship between hepatitis B virus infection and B-cell non-Hodgkin lymphoma

Objective To evaluate the association between B-cell non-Hodgkin lymphoma (NHL) and hepatitis B virus (HBV).Methods The positive rates of HBV markers in 284 patients of B-cell NHL who were admitted to our department between January 2003 and December 2009 were investigated.The positive rates of HBV markers in colorectal cancer patients were used as controls.Results The HBsAg-positive rates of patients aged 18~ 39 and stage m/Ⅳ patients were 42.6 % (26/61) and 37.0 % (50/135),which was higher than other groups.The x2 value and P value were 7.573 and 6.874,0.023 and 0.009,respectively.Compared with the control group, the B-cell NHL had significantly higher prevalence of positive HBsAg and positive HBeAg (29.6 % vs 14.5 %,6.7 % vs 0.8 % ).The Wald values were 25.174 and 20.496,respectively.Both of the P value were <0.001 and lower prevalence of positive anti-HBs (45.4 % vs 58.0 %,Wald =11.062,P =0.001).The coexpression of HBsAg, HBeAg and anti-HBc was higher in the B-NHL group than in the control group (6.0 % vs 0.8 %,x2 =31.619,P <0.001).Similarly,the coexpression of HBsAg,anti-HBe,and anti-HBc was higher in the B-NHL group (16.2 % vs 11.5 %,x2 =4.542,P =0.033).Significantly higher rate of positive anti-HBc and negative anti-HBs was observed in the B-NHL group (37.0 % vs 24.5 %,Wald =17.708,P < 0.001),whereas the same group showed a lower rate of negative anti-HBs compared with the control group (20.8 % vs 27.8 %, Wald =5.646, P =0.017).Conclusion This finding of a positive association between HBV infection and B-NHL suggests that HBV may play an etiologic role in the induction of B-NHL.
7.A twin study on influential factors of adolescent depressive symptoms

Objective To explore the influence of genetic and environmental factors on adolescents depressive symptoms. Methods The depressive mood were investigated in 107 twin pairs from Chongqing (aged from 11 to 18 years old) using Beck Depression Inventory (BDI). The Parenting Styles and Dimensions Questionaire (PSDQ), Family Stresses Questionnaire (FSQ), Family Adaptability and Cohesion Scale 2nd Edition Chinese Version (FACES II-CV) and Stressful Life Event (SLE) were applied to assess their environment factors. The blood samples were collected to identify zygosity of twins. Structural equation model was performed to evaluate the effects of the additive genetic factors (A), shared envi?ronment factors (C) and individual specific environmental factors (E) on the adolescents depressive mood. Results The scores of depressive mood were negatively correlated with father’s education level (r=-0.15, P=0.03), mother’s educa?tion level (r=-0.17, P=0.01), scores of the authoritative-parenting-style (r=-0.18, P<0.01), scores of the family adaptabil? ity (r=-0.27, P<0.01) and family cohesion (r=-0.20, P<0.01). There were significantly positively correlation between de?pressive mood and family stresses scores (r=0.14, P=0.04). The scores of adolescents depressive mood were not signifi?cantly correlated with stress life events (P>0.05). The effects of A and E on adolescents depressive symptoms were 0.37 (95%CI:0.14~0.57) and 0.63 (95%CI:0.43~0.86), respectively. Conclusion The authoritative-parenting-style and fami?ly adaptability are negatively correlated with depressive mood. Family stresses are positively correlated with depressive mood. Adolescents depressive mood is influenced by genetic factors and, to a greater extent, specific environmental factors.
8.Relationship between non-Hodgkin' s lymphoma and hepatitis B virus infection

Objective To evaluate the association between non-Hodgkin's lymphoma (NHL) and hepatitis B virus (HBV).Methods The positive rate of hepatitis B virus surface antigen (HBsAg) in 393 patients of NHL who were admitted. The colorectal cancer patients and healthy persons were also enrolled as control. Results The positive rate of HBsAg in NHL patients (26.5 %) was significantly higher than that in colorectal cancer patients (14.5 %) and healthy persons (8.8 %),respectively (x2=55.713,P<0.001).The characteristics of HBV-infected patients with NHL were similar to those who were HBV-uninfected in terms of sex,stage and B symptoms,whereas the HBV-infected patients were younger than the HBV-uninfected patients (the median age was 47ys vs 52ys,t =-1.911,P=0.021).In the HBsAg-positive NHL group,B-cell subtype was much more common than T-cell subtype (80.8 % vs 15.4 %,P=0.043).Regarding colorectal cancer patients and healthy persons as control groups,the positive rate of HBsAg was significantly higher in B-cell NHL patients than that in the control groups, respectively ( 29.6 % vs15.5 % vs 8.8 %,Wald value were 25.174 and 55.139,respectively,P<0.001).The positive rate of HBsAg of HBV between T-cell NHL (16.7 %) and control groups were not significantly different. Conclusions An association is present between HBV infection and NHL,especially in B-cell subtype.
9.Purification and stability studies of prokaryotic PDCD5 protein

Objective:To set up an effective and simple purification method to obtain highly purified prokaryotic protein of PDCD5 and study its stability. Methods:Recombinant PDCD5 protein expressed in E. coli was accumulated as an inclusion body. After washing, the inclusion body was denatured, renatured, digested with thrombine and then purified by two steps of chromatography. The purity of the products was analyzed by capillary electrophoresis and the stability was identified by SDS-PAGE. Results:Capillary electrophoresis showed that the purity of protein was 100%, and molecular weight was 15 800 with pI 5.9. Further bioactivity assay indicated that the purified PDCD5 could enhance the apoptosis of HL 60 cells withdrawing cytokine, which was in a dose dependent manner. Stability analysis showed that the PDCD5 protein was sensitive to temperature and easy to degrade at 4 ℃ and 25 ℃. However, it was relatively stable at -20 ℃ or lyophilized. Conclusion:Highly purified and stable recombinant PDCD5 protein was obtained, which lays a foundation for the functional study and application investigation of PDCD5 .
10.Phase Ⅱ trial of second-line chemotherapy with docetaxel and capecitabine in advanced esophageal squamous cell carcinoma

Objective To evaluate the effects of antitumor,toxicity and survival of second-line chemotherapy with docetaxel and capecitabine in patients with advanced esophageal squamous cell carcinoma.Methods Thirty eligible patients with measurable lesions received 1-hour intravenous treatment of docetaxel (60 mg/m2 on day 1) plus oral capecitabine (825 mg/m2 twice daily on days 1-14) every 3 weeks for up to 6 cycles.Results Patients received a median of two cycles of treatment (range 2-6).The median follow-up interview was 15.4 months (3.0-31.5 months).Intent-to-treat efficacy analysis demonstrated an overall response rate of 23.3 % (0 complete and 7 partial) and stability of 43.3 % (13 cases).The median time to progression was 3.0 months (95 % CI 1.929-4.071).The median survival was 8.3 months (95 % CI6.848-9.752).Severe adverse events (grade 3/4) reported were neutropenia (10 cases),anaemia (5 cases),thrombocytopenia (3 cases),hand-foot syndrome (4 cases),and fatigue (3 cases).Conclusion Docetaxel plus capecitabine have a manageable adverse event profile and promising activity in advance esophageal squamous cell carcinoma as a second-line treatment.
