Angiogenesis plays an important role in solid tumors and hematologic malignancies. Angiopoietins act as essential regulators in this process,complementary with another mediator-VEGF. In the development of hematologic malignancies, the expression of Angs/Tie-2 system is identified abnormal, especially for Ang-2, whose expression and prognostic impact have gained significant attention recently.
3.Clinical application of distally based sural neurovascular flap with anastomosing small saphenous vein

Objective To explore the clinical application of sural neurovascular flap with anastomosing small saphenous vein to be transferred distally for repairing soft tissue defect of the leg.Methods Seven patients with soft tissue defects in the distal one third of anterior tibia,the foot dorsum,the heel or the ankle were treated with the distally based sural neurovascular flap with anastomosing small saphenous vein.The size of the flap ranged 8 cm?6 cm ~ 18 cm?10 cm.Results The flaps survived in all the 7 patients,with excellent color and texture.Their appearance and function were satisfactory during 6~40 months of follow-up. Conclusions Use of distally based sural neurovascular flap with anastomosing small saphenous vein is reliable.The performance is simple.This flap has a rich blood supply without the sacrifice of major arteries.Venous blood reflux was unobstructed.It can be used for the reconstruction of soft tissue defects in the distal one third of anterior tibia,the foot dorsum,the heel or the ankle.
4.Clinical analysis of trans laparoscope hysterectomy

Objective To assess the clinical analysis of trans laparoscope hysterectomy(TLH).Methods Retrospectively analyzed the TLH in 62 patients Which were compared with the trans alxtominal hysterectomy(TAH)in 64 patients.Results Blood loss during operation,analgesic use after operation,time to first passing flatus,hospital stay in TVH group were significantly lower than those in TAH group(P<0.05),and mean operating time has no significant difference in TVH group and TAH group(P>0.05).Conclusion TVH has been shown to be an effective modus operandi of mini-invasive and safe.
5.Isolation and biological characteristics of rat umbilical cord mesenchymal stem cells

BACKGROUND:There are many studies concerning rat bone marrow mesenchymal stem cells for immune tolerance following transplantation and tissue repair.However,there are no reports on umbilical cord mesenchymal stem cells(UCMSCs).OBJECTIVE:To establish a method of separating mesenchymal stem cells(MSCs)from rat umbilical cord,and to study its biological characteristics.METHODS:MSCs were separated from rat umbilical cord with enzyme method and tissue mass method,and then incubated in DMEM-LG medium.Cell morphology was observed under an inverted microscope.Growth curves of cells were drawn using cell counting.Cell cycle and surface antigen were detected with flow cytometry.Adipogenic differentiation and osteogenic differentiation were tested by immunohistochemistry.RESULTS AND CONCLUSION:Both of the two methods could obtain plenty of MSCs from rat umbilical cord.Primary culture showed that the efficiency of enzyme method was higher than tissue mass method.Passage time of the former was about 10 days and the latter was 14 days.The passage time of latter except primary culture was the same.Immunophenotype analysis showed that MSCs from rat umbilical cord expressed adhesion molecule and stromal cell markers,CD90 and CD106,but did not express hematopoietic cell markers,CD34 and CD45.In vitro induction test verified that rat UCMSCs have the potentials of adipogenic and osteogenic differentiation.
6.Acute T-B cell biphenotypic leukemia:report of four cases with literature review

Objective To strengh the awareness of acute T-B cell biphenotypic leukemia.Methods Four new cases of acute T-B cell biphenotypic leukemia were reported and the related literature were reviewed.Results Fourteen patients with acute T-B cell biphenotypic leukemia.including 4 diagnosed at our hospital and other 10 cases reported in literature,were retrospectively analyzed.Similar clinical fleatures as the typical acute lymphocytic leukemia(ALL) were presented.Ten of 14 cases were male and were within 17 to 46 years old.The disease were refactory to the conventional ALL chemotherapy regimens and deteriorated progressively.Six patients died within 12 months after their diagnosed with the median survival time of ten months. Conclusion Acute T-B cell biphenotypic leukemia is one of the most rare type of leukemia.A better understanding of the clinical and hematological features of this type of leukemia and new therapeutic strategies are needed.
7.The role of BCR/ABL isoforms in the presentations and outcome of Philadelphia-positive acute lymphoblastic leukemia in adult patients

Objective To investigate the difference of clinical characteristics and outcomes between different isoforms of BCR/ABL in adults with Philadelphia-positive acute lymphoblastic leukemia (ALL).Methods The data of 106 adults with Ph+ALL diagnosed in our hospital from January 1, 1996 to December 31, 2007 were reviewed. The difference of clinical characteristics between different subgroups of BCR/ABL was compared and their relation with outcomes was studied. Results The median age of the 106 patients was 34 years and the median white blood cell count at baseline was 28. 5 × 109/L. Comparative analysis demonstrated that patients in p210 group had an older age, higher blood platelet count (BPC) and more frequent occurrence of splenomegaly. Referring to the outcomes, the complete remission (CR) rate of the two groups were 92. 2% and 93.9%, respectively. The median overall survival (OS) and relapse free survival (RFS) in p190 group were 13 months and 10 months, the 1,3-year estimated OS were (54. 7±6. 7)% and (5.5±5.2)%, and the 1,3-year estimated RFS were (40. 2±6. 8)% and (7. 8±6. 7)%,while in p210 group, the median OS and RFS were 15 months and 10 months, respectively, the 1,3-year estimated OS were (65.8±8. 9)% and (14. 5±7.4)%, and the 1,3-year estimated RFS were (48. 3±9. 4)% and (12. 9±7. 7)%. All of the above data had no statistic significance between the two groups.Conclusion Majority of the adults with Ph+ALL is p190 positive and patients with p210 have older age, higher BPC and more frequent occurrence of splenomegaly, while there is no significant difference between p190 group and p210 group in CR rate, RFS and OS.
8.Establishment of rat skin organ culture model and initiation of hair follicular morphogenesis with its correlative dynamic expression of Wnt10 b/β-catenin

Objective To establish a rat skin organ culture model to study the initiation of hair follicle morphogenesis, and the dynamic expression of Wnt10b/β-catenin in the developing hair follicle. Methods The dorsal skins of SD rat at embryos 14-17 (E14-E17) were cultured on a gelatin sponge-supported tissue culture system at the air/liquid interface of DMEM with 10 % fetal bovine serum (FBS) for 3-6 days, some of which removed from El5 were cultured in DMEM with FBS of different concentrations. HE staining and fluoroimmunoassay were adopted. Results The model we established allowed skin tissues isolated from E14-E15 to develop in a manner that was histologically and temporarily similar to the process in vivo. However, the developing hair follicle ceased to continue when their morphogenetic process reached the forth stage, and the concentration of FBS did not show any significant effect on the development of hair follicle. Expression of Wnt10b/β-catenin was induced in culture, as it also occurred in vivo,but grew weak till it disappeared in culture for 6 days, which was accompanied by development halt of hair follicle. Conclusions A rat skin organ culture model is established in which the morphogenesis of hair follicle takes place in a manner similar to in vivo, and Wnt10b/β-catenin probably has a close connection with the early morphogenesis of hair follicle.
9.Application of diffusion weighted magnetic resonance imaging in assessing the efficacy of chemoradiotherapy for uterine cervical carcinoma

Objective To explore the feasibility of diffusion-weighted imaging (DWI) in assessing the efficacy of chemoradiotherapy for cervical cancer.Methods Data of magnetic resonance imaging (MRI) and DWI were analyzed in 32 patients with uterine cervical cancer received conventional prior to chemoradiotherapy and after 1 and 3 months of therapy.13 cases of normal controls also had been examed by MRI and DWL DWI with b values of (0,300) s/mm2 and b values of (0,600) s/mm2 were performed in all patients.Pretreatment post-treatmentADC values were compared between the health group and patients group.Results When the b =300 s/mm2,normal cervical average ADC value was (1.72±0.31)×10-3 mm2/s,cervical cancer was (1.10±0.24)×10-3 mm2/s before treatment and was (1.61±0.23)×10-3 mm2/s after treatment.When the b =600 s/mm2,normal cervical average ADC value was (1.46±0.25)×10-3 mm2/s cervical cancer was (0.89±0.21)×10-3 mm2/s before treatment and was (1.54±0.18)×10-3 mm2/s after treatment.When b =300 s/mm2,ADC value was higher than when b =600 s/mm2.ADC values of cervical cancer was significantly lower than that of the normal cervix group,ADC values of cervical lesions after chemoradiotherapy was significantly higher than that before chemoradiotherapy (P < 0.05).In the same group with different b values,ADC value was not significant (P > 0.05).Conclusion Joint observation of DWI and ADC values could be more objective and accurate in the analysis of the disease and would help to evaluate the efficacy of chemoradiotherapy.
10.Immunomodulatory effects of xenogenous umbilical cord mesenchymal stem cells on rat heart transplantation

BACKGROUND: Bone marrow mesenchymal stem cells (BMSCs) can prolong the survival time of mice and baboons' alloskin graft and degrade acute and chronic graft-versus-host disease (GVHD) incidence after hematopoietic stem cell transplantation. But, at present there is no report that rat umbilical cord mesenchymal stem cells (UC-MSCs) reduced rejection response following heart transplantation. OBJECTIVE: To study the immunomodulatory effects of rat UC-MSCs on a model of allogeneic heart transplantation. METHODS: A total of 20 DA rats served as donors, and 20 Lewis rats as recipients. They were equally and randomly assigned to 2 groups: drug intervention and control groups (n=10). Using double cannulation, the left pulmonary artery and innominate artery of rat donors and external jugular vein and common carotid artery of rat recipients received end-to-end anastomosis under a microscope to establish heterotopic cardiac transplantation. One Wistar pregnant rat was selected to harvest UC-MSCs by collagenase digestion method. Following model establishment, rats in the cell transplantation group received UC-MSCs via caudal vein. Rats in the control group received sodium chloride. Survival time of the transplanted heart was determined. The transplanted heart received histopathology score using the acute rejection diagnosis criteria. Lymphocyte infiltration of transplanted heart was observed using hematoxylin-eosin staining. RESULTS AND CONCLUSION: Compared with the control group, the survival time of the transplanted heart was significantly longer in the cell transplantation group (P = 0.001), and the pathological score of acute rejection was significantly reduced (P = 0.000 4). There were lots of lymphocyte and monocyte infiltration in the myocardium in the control group. Little lymphocyte infiltration was detected in the myocardium in the cell transplantation group, with the presence of mild edema of myocardial interstitial substance. Results verified that rat UC-MSCs can induce immune tolerance of heart transplantation, soften immunological rejection and prolong xenograft survival.
