1.Application and evaluation of questionnaire based on professional knowledge method with problem - based learning in ophthalmology short -term teaching

AIM: To evaluate the application and effect of questionnaire based on professional knowledge method combined with problem - based learning ( PBL ) in ophthalmology short-term teaching.
METHODS: Ninety non ophthalmic students of Beijing Hospital of Traditional Chinese Medicine from January to August 2015 were randomly divided into observation group (45 students) and control group (45 students). Observation group: using questionnaire based on professional knowledge method combined with PBL teaching method; the control group: using traditional teaching method.
RESULTS: The survey results showed that: non ophthalmic students were unfamiliar with eye diseases knowledge; there was no significant difference in the clinical operation examination between the 2 groups (P>0. 05 ). There were significant differences in the eye disease knowledge, eye disease knowledge related to their own major and the total score between the 2 groups (P< 0. 05), especially students in the observation group had higher scores of eye disease knowledge related to their own major. There was no significant difference in the rate of improvement of learning pressure between the 2 groups (P> 0. 05), the differences of improvement rate on other problems between 2 groups was statistically significant ( P < 0. 05 ), and the improvement rate was higher in the observation group.
CONCLUSION: Questionnaire based on professional knowledge combined with PBL teaching method can effectively improve the learning effect in ophthalmology teaching.
2.Retrospective analysis of association between human papilloma virus infection and antisperm antibodies in male infertile patients in Xinjiang area

Objective To explore the association between human papilloma virus(HPV)infection and antisperm antibodies(As-Ab)in male infertile patients in Xinjiang area.Methods A total of 127 cases of infertile male patients were selected as experimental group,other 130 cases of normal male were enrolled in the control group.HPV sperm infection was detected by using reverse dot blot hybridization kit.The levels of AsAbs in semen specimen were measured by using the enzyme-linked immunosorbent assay (ELISA).Results The infection rate of male infertile patients(67.7%)was higher than that of normal male(1 6.2%),with signifi-cant differences(P <0.05).The positive rate of AsAb of male infertile patients with HPV infection(38.4%)was higher than that of male infertile patients without HPV infection(1 7.1%),with significant differences(P <0.05).Compared with male infertile pa-tients without HPV infection,the percentages of sperm motility and grade (a+b)sperm motility were decreased in male infertile pa-tients with HPV infection,wihile the sperm malformation rate was increased in male infertile patients with HPV infection,with sig-nificant differences(P <0.05).Conclusion The HPV infection rate of male infertile patients is relatively high in this area.HPV sperm infection could be associated with AsAb that may further reduce male fertility.

4.Comparison of pathogenicity between Ureaplasma urealyticum serotype 1 and 3 in genital tract of female BALB/c mice

Objective To investigate the pathogenicity of the same concentration of Ureaplasma urealyticum serotype1 (Uu1) and 3 (Uu3),alone or in combination,in genital tract of female BALB/c mice.Methods A total of 144 estradiol-pretreated adult female BALB/c mice were randomly and equally divided into 3 groups to be inoculated intravaginally with the same concentration (106 copies/g) of Uu1 and Uu3 alone or in combination.Twelve estradiol-pretreated female BALB/c mice were inoculated with sterile culture solution and served as the control group.At 1,3,7,14,21,35 days after the inoculation,8 mice in each Uu-inoculated group and 2 mice in the control group were sacrificed.Tissue specimens were obtained from the cervix,endometria and fallopian tube and subjected to hematoxylin-eosin staining followed by light microscopy.Scanning electron microscopy was used to observe the tissue specimens collected at 14 days from cervical mucosa of mice infected by Uu.Chi-square test was performed for statistical evaluation.Results The total infection rate of Uu within 3-35 days after the inoculation was 35.0% (14/40),47.5% (19/40) and 62.5% (25/40) in the Uu1 group,Uu3 group,and combination group respectively (x2 =6.07,P < 0.05).None of these mice was infected by Uu in the control group.Conclusions At the same inoculation concentration,the combination of Uu1 and Uu3 shows the strongest pathogenicity in genital tract of BALB/c mice,followed sequentially by Uu3 and Uu1 alone.
5.The causes of short stature in Turner syndrome

Objectives To investigate the causes of short stature in Turner syndrome (TS). Methods 86 patients were di-agnosed with TS by karyotypes from 2004 to 2013. According to the deletion types of the X chromosome short arm, growth hor-mone (GH), insulin-like growth factor-1 (IGF-1) and thyroid function, the TS patients were divided into different groups and com-parison was made among groups. Results Ht SDS in three groups with different extent of the deletion of the X chromosome short arm were (-4.39±1.08), (-3.26±1.25) and (-2.84±0.15) (P<0.05). The proportion of growth hormone deifciency (GHD) in the three groups were 62.5%, 38.9%and 0%(P<0.05). Ht SDS in groups with different degree of IGF-1 level were (-4.37±1.10), (-3.82±1.07) and (-3.25±0.91) (P<0.05). There was no signiifcant difference of Ht SDS between hypothyroidism patients with and without GHD (P>0.05). Conclusions The deletion of X chromosome short arm may cause the short stature in TS. The GH-IGF-1 axis in TS is impaired, but GHD is not related to short stature in TS.
6.Effects of hippocampal neurogenesis in spatial learning and memory in rats

Objective To investigate the effects of hippocampal neurogenesis in spatial learning and memory,especially in spatial long-term memory.Methods Totally 24 wistar rats were randomly divided into two groups:normal control group (NC group,n =12) and irradiation group (IRR group,n =12).We use low-dose irradiation at subgranular zone to inhibit adult hippocampal neurogenesis and new neurons were investigated by 5-Bromo-2-deoxyUridine/neuron specific nuclear protein double-labeling.Four weeks after irradiation,Morris water maze,including navigation test and space exploration assay,was used to detect spatial leaming and memory.Then,in the day 8 and day 15 after the navigation test,space exploration assay was repeated to detect spatial long-term memory.The expression levels of postsynaptic density protein 95 and synaptophysin were evaluated using western blot and real-time quantitative PCR.Results Hippocampal neurogenesis was inhibited by low dose irradiation(2.80 ± 0.44 vs 23.50 ± 1.12,t =20.21,P < 0.01).After the inhibition,the escape latency did not change,while the time spent in target quadrant was significantly declined in the day 2((14.76 ±.2.04) s vs (20.47 ± 1.29) s),day 8((11.95 ± 1.34) s vs (19.52±1.43) s)and day 15((11.79 ±1.35) s vs (21.58 ±1.07) s) after the navigation test (t=2.45,P<0.05; t =3.76,P<0.01; t =5.39,P<0.01).The postsynaptic density protein 95 and synaptophysin expressions were significantly decreased in IRR group.As to the RNA levels,there was also a significant difference between the two groups.Conclusion Hippocampal neurogenesis in the dentate gyrus plays an important role for the formation of spatial long-term memory.
7.Ghrelin participates in the two-way regulation of the motility of the small intestinal smooth muscle in rats

Objective To investigate the effect and mechanism of Ghrelin on the contraction and relaxation of rat small intestinal smooth muscle. Methods The effect of different concentrations of Ghrelin (0;20;40;80 μg/kg) on the small intestinal transit in vagotomized rats in vivo and Ghrelin (0.01; 0.1;0.5 ; 1.0 μmol/L) on the contraction and relaxation of rat small intestinal smooth muscle strips in vitro was observed,the locations of Ghrelin receptors (GHS-R1 a) in small intestinal muscle layers were detected by immunofluorescency. Results Ghrelin dose-dependently increases small intestinal transit (( 25.4 ± 1.0)%,(33.7 ± 1.9)%,(39.3 ±2.4)%,(44.7 ±2.1)%),enhances the contraction ((67.0 ±2.4)%,(149.5 ±3.3)%,(187.1 ±4.7)%,(213.5 ±3.4)%) and relaxation ((35.3 ± 1.1)%,(62.9 ± 3.8 ) %,( 79.6 ± 2.7 ) %,( 94.6 ± 2.2 ) % ) of smooth muscle strips mediated by carbachol.Ghrelin receptors were mainly located on membrane of the nerve cells in the muscle layers,while no receptors exist on membrane of smooth muscle cells. Conclusions Ghrelin enhances the effect of contraction and relaxation of rat small intestinal smooth muscle mediated by cholinergic neurotransmitters activating nerve cells in the enteric plexus.
8.Experimental study on differentiation of different passages of human bone marrow mesenchymal stem cells into neurons in vitro

BACKGROUND: Bone marrow mesenchymal stem cell (MSC) is a kind of stem cell with potential of self-repair and multi-differentiation. It may differentiate into neuron, adipose cell and osteoblasts.OBJECTIVE: To observe the transforming efficacy of human bone marrow mesencymal stem cells (hMSCs) into neurons in vitro in different generations so as to provide reliable experimental data for the clinical application of MSCs.DESIGN: Single sample was designed.SETTING: Department of Neurology, Sino-Japan Friendship Hospital,Jilin UniversityPARTICIPANTS: Marrow tissue was collected from 9 cases of spinal fusion in Department of Orthopedics of First Hospital affiliated to Jilin University. Of 9 cases, all of them were in known of the experiment.METHODS: The experiment was performed in Sino-Japan Hospital affiliated to Jilin University from September 2002 to February 2003. The primary and generative culture of hMSCs was given. Experimental and the control groups were divided.-mercaptoethanol was taken as inducer. hMSCs of the 2rd, 4th, 6th and 8th generations were selected for induction in vitro for 6 h. Cytochemistry staining and immunohistochemistry staining were used to assay the expressions of neuronal and astrocytic marked proteins.MAIN OUTCOME MEASURES: ① Growth curve analysis on genera tive culture of hMSCs. ② Nissel staining. ③ Immunohistochemical staining.RESULTS: ① Common characters of generative culture: The latent phase of generative culture was 12-24 hours, exponential phase was 7-10 days and 11-13 days later, cell culture entered the platform phase. ② After induction of the 2nd, 4th and 6th generated hMSCs, deep blue granular Nissl body presented in cytoplasm. In 6 hours on the 8th induction, there was no obvious deep blue Nissl structure presented in cytoplasm.③ Except GFAP, NSE and NF-M were expressed in hMSCs of different generations after induction for 6 hours. There was no significant difference in positive rates of the 2nd, 4th and 6th generations (P > 0.05), but the significant difference presented in comparison between the 8th generation and the 2nd, 4thand 6th generations (P < 0.05). CONCLUSION:-mercaptoethanol can induce hMSCs differentiating into neuronal cells in vitro. The positive rates of the 2nd, 4th and 6th generationsare higher remarkably than the 8th generation.
9.Mutation analysis of steroid acute regulatory protein gene in a patient affected with congenital lipoid adrenal hyperplasia

Objective To analyze the clinical characteristic s of an infant with congenital lipoid adrenal hyperplasia (CLAH),and to sequence the acute regulatory protein (steroid acute regulatory,StAR) gene of the infant patient and her pedigree.Methods Physical examination,laboratory tests,and imaging examination of the 11-month-old patient with CLAH were collected.DNA was extracted from blood samples of the patient and her parents.The 7 exons of StAR gene were amplified by PCR and then sequenced.Results Dark skin,girl vulva,and one each 1.0 cm×1.0 cm palpable lump in inguinal area bilaterally were observed.The adrenocorticotropic hormone (ACTH) was 253 pg/ml,cortisol was 27.6 nmol/L at 8 am,17-hydroxyprogesterone was 3 nmol/L.Uhrasound showed that sign of testicular ultrasonography existed in bilateral inguinal regions.Karyotype analysis showed 46,XY.Sequencing of PCR amplified fragments showed that there were two heterozygous mutations c.229C > T,p (Gln77X) and C.659A>G,p (His220Arg) of StAR gene in this patient.By rectifying the disturbance of electrolyte,and treating with hydrocortisone and 9α fludrocortisone,etc,the patient has been stable so far.Conclusion The patient presents typical clinical manifestations.Two heterozygous mutations including c.229C >T,p (Gln77X) from maternal and C.659A>G,p (His220Arg) from paternal of StAR gene were detected.Wherein the c.659A>G,p.(His220Arg) as a novel point mutation of StAR gene,has not been reported so far.

Atrial fibrillation is a common cardiac arrhythmia encountered closely related to structural remodeling such as atrial fibrosis. Galectin-as a biomarker of fibrosis; therefore, it may be involved in atrial remodeling association of Gal-3 with atrial fibrillation.
