1.Approach to the relationship between the expression of metallothionein-Ⅲ mRNA and free zinc contents

Objective To investigate the induction mechanism of MT ⅢmRNA in the brain after cerebral ischemia.Methods The forebrain ischemia reperfusion model was established in rats. The changes of the expression of MT Ⅲ mRNA in hippocampus after forebrain ischemia reperfusion were observed by in situ hybridization method. The changes of free Zn 2+ in hippocampus after forebrain ischemia reperfusion was examined using Zn 2+ specific fluorescent probe(TSQ). The Zn 2+ chelator (CaEDTA) was injected into the lateral ventricles for determining influences of Zn 2+ on the MT Ⅲ mRNA expression and the neuronal damage after forebrain ischemia/reperfusion.Results (1) The expression of MT ⅢmRNA in hippocampus increased gradually after cerebral ischemia and reached the peak in 96 hours after reperfusion. Seven days after reperfusion the expression of MT ⅢmRNA reduced to the normal level. (2) Zn 2+ fluorescence in the hilus of dentate gyrus, CA 3 region and the stratum radiatum and stratum oriens of CA 1 decreased slightly at 48 hours after reperfusion. From 72 to 96 hours after reperfusion, the fluorescence returned to normal, but some new fluorescence dots appeared in pyramidal neurons of CA 1 and the hilus of dentate gyrus increased gradually. Seven days after reperfusion, the fluorescence returned to normal. (3) The cell membrane impermeable Zn 2+ chelator could reduce the intracellular concentration of free Zn 2+ and the expression of MT Ⅲ mRNA.Conclusion The expression of MT Ⅲ mRNA can be induced by the increase in the concentration of intracellular free Zn 2+ after forebrain ischemia/reperfusion.
2.Effect of zoledronate on the clinical efficacy and IL-8, IL-10 and osteocalcin levels in perimenopausal with osteoporosis

Objective To investigate the effect of zoledronate on the clinical efficacy and IL-8, IL-10 and osteocalcin levels in perimenopausal women with osteoporosis.Methods A total of 72 cases menopausal syndrome with osteoporosis from January 2014 to March 2015 in Hangzhou Third People's Hospital of Yuhang District were randomly divided into two groups with 36 cases in each group.Patients in the control group were treated with routine oral administration of calcium carbonate D3 and alpha D3, and the observation group was treated with zoledronate acid sodium on this basis for four weeks.Serum inflammatory factor and bone metabolism index were measured before and after treatment , VAS score and bone mineral density were measured, and the clinical effect was compared.Results Compared with before treatment, two groups of serum ALP, BGP levels and VAS score were significantly decreased (P<0.05), IL-8 and TNF-αlevels decreased (P<0.05), IL-10 level increased (P<0.05), lumbar(L1 ~4), bilateral hip and femoral neck BMD increased (P<0.05); compared with the control group, serum ALP, BGP levels and VAS score in the observation group was lower (P<0.05), IL-8, TNF-αlevels were lower (P<0.05), IL-10 level was higher (P<0.05), lumbar(L1 ~4), bilateral hip and femoral neck BMD were higher (P<0.05), the treatment effective rate was higher (P <0.05).Conclusion Zoledronate in the treatment of perimenopausal syndrome with osteoporosis is significant clinical efficacy, it can effectively relieve the inflammatory symptoms and bone and joint pain, inhibit bone reduction.

4.Progress in stem cell transplantation for treatment of retinal ganglion cell damage diseases

Retinal ganglion cells are crucial in the formation of vision. Injury or death of retinal ganglion cells may lead to irreversibly damage of visual function. Glaucoma, diabetic retinopathy, hypertension, and other blind leading diseases can cause the damage or progressively apoptosis of retinal ganglion cells. Currently, there is no specific treatment to restore vision damage caused by those diseases. Scholars at home and abroad focus on stem cells transplantation in order to recover the visual function of patients. Two categories are mainly involved in stem cell transplantation, one is the replacement therapy based on stem cells, the other is to promote the secretion of some factors to protect ganglion cells through stem cell transplantation. In this review, we aim to summarize the potential of stems cell transplantation to treat retinal ganglion cells related diseases, and discuss the differentiation of different types of stem cells to retinal ganglion cells.
5.Study on capillary endothelium injury in the lung with ischemia-reperfusion

AIM: To confirm the effects of capillary endothelium injury in the lung with ischemia reperfusion. METHODS: The rabbits pulmonary models of ischemia reperfusion (I-R)injury were established. Plasma nitric oxide and endothelin levels, arterial oxygen tension and wet/dry weight ratios were determined in different periods in control and I-R groups, and the pulmonary ultrastructure abnormities were analyzed under electromicroscope. RESULTS: The plasma levels of NO and ET-1 in I-R animals increased significantly ,compared with those in sham treated control groups. The level of ET-1 had significantly negative correlation with PaO 2 and positive correlation with the value of the wet/dry weight ratios. The swelling and karyopyknosis of capillary endothelium in the lung with I-R groups were observed in ischemia periods. The injuries of endotheliums and typeⅠandⅡ alveolar cells in 0 5 h of reperfusion were more severe than those in ischemia time, and the injuries began to repair in 2 h of reperfusion time. CONCLUSION: In the I-R group, pulmonary capillary endothelium have been injuried, which may play a prominent role in I-R injury and dysfunction of the lung.
6.Clinical characteristics and outcomes of preoperactive short-acting octreotide treatment in patients with thyrotropin-secreting pituitary adenomas

Objective:To explore the clinical characteristics of patients with pituitary thyrotropin-secreting adenoma and evaluate the effect of preoperative short-acting octreotide treatment on hyperthyroidism.Methods:A retrospective analysis was performed in 40 patients with pituitary thyrotropin adenoma diagnosed in Beijing Tiantan Hospital from January 2008 to January 2018. The general data, laboratory examinations and imaging findings were reviewed and analyzed. The clinical effect of preoperative octreotide on hyperthyroidism was evaluated.Results:The age of onset year of the 40 patients (male: female = 24∶16) was (30.5±5.1) years. Among them, 35 patients (87.5%) were with macroadenoma. The most common symptoms were thyroid hypermetabolism syndrome, followed by headache, dizziness, visual field damage and hypogonadism. The thyroid function of 30 patients (75%) recovered to normal within 3-5 days after the octreotide treatment. The total effective rate of the octreotide was 90.0%. The level of free thyroxine (FT 4) before treatment in patients with more than 10 times of effective cumulative dose was significantly higher than that in patients with less than 10 doses. Conclusions:Thyroid hypermetabolism syndrome and pituitary occupying effect are the most common clinical manifestations of thyrotropin-secreting adenoma. Preoperative octreotide treatment can effectively control hyperthyroidism. The level of FT 4 is a crucial factor affecting the efficacy of octreotide.

AIM : To construct and analyze eukaryotic expression plasmid inserted by Cx50 with V64G mutation through bioinformatics software.METHODS: The full coding domain sequence of Cx50 with V64G mutation was acquired from the blood of patients with cataract and was cloned into pcDNA3.1 /Amp (+).The constructed plasmid was identified with PCR , enzyme digestion and sequencing. The analysis of Cx50 with V64G mutation was performed with bioinformatics software.RESULTS : Cx50 with V64G mutation was successfully amplified and its eukaryotic expression plasmid was constructed. Valine-64 is well conserved in the first extracellular loop of connexin 50 in different species and also in different human α -type gap junctional proteins.CONCLUSION : The successive reconstruction and verification of eukaryotic expression plasmid containing Cx50 with V64G mutation established the foundation for further studying the mechanism of cataract.
