2.Therapeutic Effects of Interferon-? and Polyresistin on Measles in Children

10-15 years were 7 cases(6.3%),they were randomly divided into treatment group and contol group.The treatment group with 60 patients were given INF-? and polyresistin,age 3 years,given IFN-? 200 000 U per day;and polyresistin 5 mg per time,3 times every day,the course of therapy were 5 days.The control group was given traditional Chinese medicine at the same time.Results After therapy,in treatment group,53 cases(88.3%) of children′s cooling time were lower than 5 days,47 cases(78.3%) of children′s rash subsided time were lower than 6 days,48 cases(75%) of children′s Catarrhal symptoms disappeared time were lower than 5 days,and the rate of complications′ occurrence were 15 cases(18.3%).In control group,28 cases(53.8%) of children′s cooling time were lower than 5 days,14 cases(26.9%) of children′s rash subsided time were lower than 6 days,28 cases(53.8%) of children′s Catarrhal symptoms disappeared time were lower than 5 days,and the rate of complications′ occurrence were 37 cases(71.5%),there were significant differences between both groups(Pa
4.Establishment of a HL-60 cell line with stable RYBP silencing

Objective To establish stable HL-60 cell line with stable RYBP gene silencing using lentivirus-mediated RNA interference. Methods Five special shRNAs for RYBP gene were cloned to lentivirus vector.Recombinate lentivirus vectors were packed into lentivirus,which were used to infect HL-60 cells, and took empty vector and non-specific shRNA as control groups. Stable infected cells were selected with puromycin in 8 μg/ml concentration.The expression levels of RYBP were analyzed by Western blot,and confirmed the most effective RYBP shRNA.Then the level of mRNA was analyzed by real-time PCR.Results Stable infected cells were selected by puromycin successfully.Comparing to control groups,the expression of RYBP were reduced at different degrees (P < 0.01). And RYBP shRNA2 took the most silencing effect, the RYBP mRNA was decreased by more than 95 % (P < 0.05).Conclusion The shRNA2 targeting RYBP gene can effectively inhibit the expression of RYBP. HL-60 cell line with stable RYBP gene silencing were constructed successfully,which had provided experiment fundament for further studying the function of RYBP.
5.Identification and the mRNA expression of HS1-associated protein X-1 in the peripheral blood mononuclear cells of patients with systemic lupus erythematosus

Objective To study the mRNA expression of HS1-associated protein X-1(Hax-1),an an- ti-apoptosis genc,in the peripheral blood mononuclear cells(PBMC)of patients with systemic lupus erythe- matosus(SLE),and further investigate the roles and significance of Hax-1 in the pathogenesis of SLE.Meth- ods Generation of longer cDNA fragments from serial analysis of gene expression(SAGE)tags for gene identi- fication(GLGI)was applied to identify the gene Hax-1 according to the Long SAGE tag.Then reverse tran- scription-polymerase chain reaction(RT-PCR)technique was used to semiquantitatively analyze mRNA ex- pressions of Hax-1 in PBMC from 34 active SLE patients and 25 healthy subjects.Results Compared with healthy controls,there was significant difference between SLE patients in the active stage and the normal controls(Z=-4.556,P<0.01).The average level of mRNA expression in active SLE group was higher than that in healthy controls.Significant difference was found between the group with mild SLE and either the moderate or the severe one(P<0.01).Conclusion The mRNA expression level of Hax-1 in active SLE group increase markedly,and to some extent,it is related to the activity of SLE.This provides a valuable basis for the further study on the role of apoptosis in SLE.
6.Characterization of biophysical properties of insulin-producing cells labeled with SPIO-PLL in vitro

Objective To evaluate the biophysical properties of insulin-producing cells labeled and unlabeled with superparamagnctic iron oxide nanoparticals and poly-L-lysine(SPIO-PLL)in vitro,and then monitor cellular imaging with 1.5 T MR.Methods BMSCs were isolated from tibia and femur of 6~8 weeks normal Spragne-Dawley rats,purified on the basis of their ability to adhere to the matrix,and expanded through their self-renewal.Two-step strategy was adopted with BMSCs induced into insulin-producing cells, After that,the cells were incubated with SPIO-PLL.Prussian blue stain was employed for identifying intracellular irons.Radioimmunology assay was used to measure the insulin secretion by the labeled and unlabeled cells,and later on underwent MR imaging with T_1WI、T_2WI、T_2*WI sequences,Results lntracytoplasmic nanoparticales were stained with Prussian blue possessing insulin-producing cells labeled with SPIO-PLL.The amount of insulin secreted by the labeled and unlabeled ceils had no statistical significant difference.The signal intensity of labeled cells decreased significantly on T_2*WI,as well as the stronger proportional variations for signal intensity.Conclusion Insulin-producing cells can be labelled effectively with SPIO-PLL and be imaged by 1.5 T MRI.(J Intervent Radiol,2007,16:104-108)

Objective To investigate the Lymphocyte-specific protein-tyrosine kinase(Lck)gene ex- pression in the renal tubule epithelial cells(TECs)of lupus nephritis,and the effect of interlenkin-2(IL-2) stimulation on its expression.Methods Proximal TECs derived from 6 weeks old spontaneous systemic lupus erythematosus(SLE)BXSB mice were exposed to IL-2(100 U/ml),the expression of Lck mRNA and protein was examined by reverse transcription-polymerase chain reaction(RT-PCR)and immunoblotting respectively. Objective:To explore the effect of hydrotherapy combined with breathing training on lung function and mobility of patients with thoracolumbar spinal cord injury.Methods:A total of 80 patients with thoracolumbar spinal cord injury admitted to Beijing Rehabilitation Hospital Affiliated to Capital Medical University from April 2018 to March 2020 were selected as observation objects.A prospective cohort study was conducted and randomly divided into observation group and control group with 40 cases in each group.The control group was given routine rehabilitation therapy combined with respiratory training.On the basis of the control group, the observation group was treated with water therapy.The indexes of lung function, motor function, lower limb muscle tension, function evaluation and activities of daily living were compared between the two groups.Results:After intervention, the observation group′s forced vital capacity (FVC) was (3.86±0.82) L, the forced expiratory volume in the first second (FEV1) was (3.76±0.68) L, the maximum ventilation (MVV) was (102.34±10.38) L/min, the maximum suction pressure (MIP) is (50.36±4.62) cmH 2O; the control group FVC was (3.41±0.76) L, and FEV1 was ( 3.35±0.63) L, MVV was (90.67±11.68) L/min, MIP was (44.38±4.85) cmH 2O, the difference between the two groups is statistically significant ( t=2.546, 2.797, 4.723, 5.646, respectively, all P<0.05). After the intervention, the motor function score of the american spinal injury association (ASIA) of the observation group was (58.62±7.56) points, and the modified ashworth scale (MAS) score was (2.74±0.89) points; The ASIA motor function score of the control group was (42.24±6.40) points, and the MAS score was (3.36±0.94) points.The difference between the two groups was statistically significant ( t=10.459 and -3.029, respectively, all P<0.05). After intervention, the observation group′s spinal cord independence measure (SCIM III) score was (75.33±10.72) points, and the modified barthel index (MBI) was (66.64±6.34) points; the SCIM III score of the control group was (68.34±9.55) points, and the MBI score was (57.52±6.77) points, the difference between the two groups was statistically significant ( t=3.079 and 6.219, respectively, all P<0.05). Conclusion:Hydrotherapy combined with breathing training can significantly improve lung function and respiratory muscle strength in patients with thoracolumbar spinal cord injury, and improve motor function and ability of daily living.
10.Effect of hyperbaric oxygen treatment on the expression of FasL and caspase-3 in renal tissue after renal ischemia reperfusion injury

Objective To observe the effects of hyperbaric oxygen (HBO) on the expression of FasL mRNA and caspase-3 protein in renal tissue after renal ischemia-reperfusion injury (IRI) in order to elucidate the underlying mechanisms.Methods Rats were randomly divided into thrcc groups: sham group(n=8),IRI group(n=8) and IRI+HBO group(n=8).The IRI group and the IRI+HBO group recieved 45 minutes hibateral renal ischima and the IRI +HBO group received additional HBO therapy at the 1st,24th and 48th hour after ischemia.The kidneys were removed at the end of HBO therapy.Malondialdehyde (MDA) level and superoxide dismutase (SOD) activity were measured to determine the extent of oxidative stress.The expression of FasL mRNA and caspase-3 protein was detected by quantitative real-time PCR and immunohistochemical staining in renal tissue respectively.Results Compared with the sham group,MDA level increased markedly and SOD activity decreased markedly after ischemia.After HBO treatment,MDA level decreased and SOD activity increased significantly (P <0.05).In IRI group,the expression of FasL mRNA and caspase-3 protein were higher than those in the sham group (P<0.01),which were reduced significantly by HBO treatment (P<0.01).Conclusion The expression of FasL mRNA and caspase-3 protein increases along with the lasting of reperfusion and HBO exhibites protection against cell apoptosis through improving the antioxidant-oxidant balance and reducing IRI in acute stage of IRI.
