Objective To observe the relationship of the Janus kinase-signal transducers and activator of transcription pathway and neurogenesis in the lateral ventricle subventricular zone(SVZ) and choroid plexus of neonatal rats with periventricular leukomalacia(PVL).Methods PVL models were established by right common carotid artery ligation followed by 4 h 60 mL/L oxygen exposure in 2-day-rat;the neonatal rats performed a sham operation,without hypoxia-ischemia were used as control group.The rats were sacrificed at 0 h,3 h,6 h,12 h,1 d,3 d and 7 d after hypoxic-ischemic(HI),and brain tissues were collected,immunohistochemistry was used to detect the expression of P-STAT3 and Nestin in the subventricular zone and choroid plexus.Results Very low expressions of P-STAT3 and Nestin were observed in lateral ventricle SVZ and choroid plexus in control group.The expression levels of P-STAT3 and Nestin increased significantly after HI,peaked at 1 d and 3 d separately,and remained at a higher level at 7 d after HI,demonstrating significant differences at each time point compared with those in control group(Pa
2.Quantitative Proteomic Analysis of Bromotetrandrine and Tetrandrine in K562 Cell Line Using 18O-labeling Method

ObjectiveTo compare quantitative proteomic analysis of bromotetrandrine (W198) which was a Class Ⅰ new antitumor drug in China and tetrandrine (Tet) in K562 cell line using 18O-labeling method.MethodsTo illustrate its mechanism,a shotgun quantitative proteomic strategy employing 2D LC-MS-MS and trypsin catalyzed 18O-labeling quantification was carried out in this study.Compared to normal chronic leukemia cell line K562 and K562 induced by Tet,the proteomic changes of K562 induced by W198 were investigated.In order to validate the quantitation by the 18O-labeling,the analysis was done on an equivalent sample composed of the same amount of labeled and unlabeled proteins from normally cultured cells to act as a reference to the comparative sample.ResultsA threshold of ± 2-fold change for deciding whether a protein concentration was changed was settled for the following experiments.Comparing the 105 identified soluble proteins' expression levels of the apoptosis starting up K562 cells after W198 induction with the normally cultured cells,16 proteins were found with significantly altered expression levels after W198 treatment.Eight proteins were up-expressed including HMGB2,peroxiredoxin-2,and eIF4A-I,etc.Eight proteins were down-expressed including TCP-1,GRP94,GST-π,and SFGHs,etc.Compared to K562 induced by Tet,eight proteins of K562 were found with significantly altered expression levels after W198 treatment.Five proteins were up-expressed including HSP 90-β and 40S ribosomal protein S15a,etc.Three proteins were down-expressed including phosphoglycerate kinase 1,isoform 5 of interleukin enhancer-binding factor 3,etc.ConclusionThe 18O-labeling MS-MS-based method is ideal as a discovery tool,but it is not suitable for validation using a large number of samples.Other more effective methods,such as Western blotting should be used for further validation of candidate cancer proteins discovered from 18O-labeling samples.In total,105 soluble proteins were discovered,and 16 proteins were found with significantly altered expression levels after W198 treatment.These repressed or activated proteins are the potential drug targets of W198,which may provide novel targets for future development ofbiomarkers for cancer therapy.

Mini-clinical evaluation exercise(Mini-CEX) is a new tool used to evaluate clinical skill of internal residents.Through necessity,operation and practice of Mini-CEX applied in internal residents' clinical teaching,the paper makes it known that Mini-CEX is consistent with six requisitions of resident competence raised by Accreditation Council of Graduate Medical Education (ACGME).It increases medical and directorial quality,and is combined with doctor training abroad.Mini-CEX should be spread in training and examination of internal residents.
4.Studs on effect of nebulised budesonide in treatment of neonatal laryneal edema caused by mechanical ventilation

Objective To discuss the clinicd effect of using nebulised budesonide to treat neonatal laryneal edema caused by mechanical ventilation.Methods 43 newborns with mechanical ventilation were included in this study and divided into two groups:The Observational group(23 newborns)were treated with nebulised budesonide right before excubation once and three times after that,and the control group(20 newborns)were treated with dexamethasone once right before excubation and nebulised adrenine after that for three times.Comparisons were made between two groups about dismissing time of wheeze or hoarseness,tachypnea,PaO2 and PaCO2.Results Observational group:dismissing time of wheeze or hoarseness was(10.26±4.01)hours,dismissing time of tachypnea was(1.63±0.55)days,which was shorter than that of control group[(14.20±5.58)hours/(2.05±0.58)days],P<0.05).PaO2 and PaCO2 of two groups had no statistical difference(P>0.05).Conclusion Nebulised budesonide was effective in treatment neonatal laryneal edema caused by mechanical ventilation,using.
8.Curative Effect Observation of Ginkgo Biloba Extract on Back Chapped Skin Model in Rats

Objective:To determine the curative effect of Ginkgo Biloba extract on the back chapped skin in SD rats. Methods:Totally 60 SD rats were randomly divided into 6 groups with 10 rats in each. One group was the blank control group, while the other five groups with the same back chapped skin were respectively treated with different therapy regimen, namely one group was the model group without any treatment, one group was urea group treated with the commercially available urea, and the last three groups were re-spectively treated with Ginkgo Biloba extract at the dose of 5%, 1% and 0. 2%. The pathological tissue was observed after HE stai-ning, and the content of hydroxyproline ( Hyp) in skin was determined by alkaline hydrolysis. Results:The skin lesion and the score of pathological section were studied and the content of Hyp in the damaged skin was determined. The comprehensive score for the groups was 0, 13. 2, 7. 4, 11. 0, 5. 8 and 6. 6, respectively, and the content of Hyp was (4. 57 ± 0. 07), (2. 41 ± 0. 27), (3. 29 ± 0.57), (2.55 ±0.31), (3.24 ±0.26) and (3.37 ±0.24) μg·mg-1,respectively. There was significant difference in the curative effect between the Ginkgo Biloba extract groups at high dose (5%) and medium dose (1%) and the model group (P<0. 05), which showed no obvious difference when compared with the urea group (P>0. 05). Conclusion:The extract of Ginkgo Biloba has a certain effect on the back chapped skin in SD rats, which may contribute to the external preparations of Ginkgo Biloba extract.
9. Study on HPTLC chromatographic fingerprint analysis of Ilex pubescens and its related species

OBJECTIVE: To develop the high-performance thin-layer chromatographic (HPTLC) fingerprints of triterpenoid from Ilex pubescens and compare the fingerprints similarity with its related species, such as Ilex rotunda and Ilex asprella. METHODS: The separation was performed on pre-coated HPTLC GF254 silica-gel plate stationary phase with trichloromethane-ethyl acetate-methanol-water-formic acid(3:22:5:2:0.2) as solvent system and 10% sulfuric acid alcoholic solution as color development reagent. The common pattern of HPTLC fingerprints were obtained through Chromafinger2005 solution software, and authentication and quality assessment were analyzed by similarity and Principle Commonent Analysis. RESULTS: The common pattern of the pileus of Ilex pubescens consists of 13 characteristic peaks. There are notable differences among the fingerprints of different parts of Ilex pubescen, and those between Ilex pubescens and its related species. CONCLUSION: The different parts of Ilex pubescens can be distinguished by comparison of the HPTLC chromatographs. The other relatives can be easily differentiated from Ilex pubescens too. 10.Role of graph-text-combined health education in dieting health education for patients with hematologic diseases

Objective To explore the role of graph-text-combined health education in dieting health education for patients with hematologic diseases. Methods About 56 patients with hematologic diseases hospitalized in February 2015 were assigned as the control group and another 57 patients with the same diseases during March to April 2015 as the experiment group. In the control group, routine dieting health education was done and in the latter the graph-text-combined health education was used for the dieting health education. The two groups were compared in view of recognition on dieting knowledge, dieting compliance and patient's satisfaction level. Result The recognition on dieting knowledge, dieting compliance and patient's satisfaction level were all higher those of that the control group (P<0.05). Conclusions The graph-text-combined health education can improve the recognition on dieting knowledge, dieting compliance and patient's satisfaction level when it is used in the health education for the patients with hematologic diseases. Therefore, it is worth clinically popularizing.
