1.Study on the Effect of Anti Osteopontin Antibody in Treating Rats’Collagen Induced Rheumatoid Arthritis
Journal of China Medical University 2015;(5):405-410
Objective To investigate the anti?OPN antibody′s protective effect towards the synovial membrane of rats with rheumatoid arthritis,in?cluding the mechanism of such protective action. Methods From a total of 40 SD rats,6 were randomly selected as the normal control group while the remaining 34 were used to establish the collagen induced arthritis model. A total of 24 rats with arthritis index higher than 2 were chosen(exclud?ing the rest 10 rats),and randomly divided into model group and groups being treated with low, medium and high dose of anti?OPN anti?body,re?spectively. Weight, arthritis index and the volume of the left hind leg toe of the rats were recorded. Then on day 35,the rats were killed and their joints were taken out to make HE colored sections for observation of the pathological changes. The synovial membrane of each rat was taken out and WB method was used to test the protein expression of OPN and TNF?αin such membrane. ELISA method was used to test the change of the content of OPN and TNF?αin each rat′s blood. Results The rats in treatment group gained weight steadily from day 16 and their swollen joints were gradu?ally relieved upon medication. Comparing with the control group, the differences in weight, toe volume and arthritis index of the rats in model group were statistically significant(P<0.05). Comparing with the model group, the differences in weight, toe volume and arthritis index of the rats in the treatment groups were also statistically significant(P<0.05). Pathologic change of synovial membrane:for rats in treatment groups, synovial cell hy?perplasia was visibly reduced with a little inflammatory cell infiltration and smooth articular cartilage surfaces with no visible damage. Expression of OPN and TNF?αin synovial membrane and blood serum of rats:Judging from the OPN measured in synovial membrane and blood serum,we could see that the expression of OPN in rats of the model group increased obviously if compared with the control group. Comparing with the model group, the expression of OPN in the treatment groups was reduced(P<0.05). Judging from the TNF?αmeasured in synovial membrane and blood serum, we could see that the expression of TNF?αin rats of the model group increased obviously if compared with the control group, thus such difference was statistically significant between different groups(P<0.05). Conclusion The anti?OPN anti?body could significantly reduce the articular symp?toms of the rats with collagen induced arthritis, restrain the synovial cell hyperplasia and alleviate the inflammatory reaction. In the process of protect?ing the joints of the rats with collagen induced arthritis, anti?OPN anti?body restrained only the expression of OPN in the synovial membrane and blood serum, leaving the expression of TNF?αunaffected.

Objective To investigate the three methods for sun protection factor determination in vivo in Chinese under the same condition, and to evaluate the feasibility and reliability of the three methods. Methods There were 45 healthy volunteers in Guangzhou area were involved in the study. All of them were randomly divided into three groups. Among them 15 volunteers were in the current method group, 5 volunteers in the simplified method group, and 25 volunteers were in the single-exposure method group. Two standard sunscreens of SPF 4 and SPF 15 and one sunscreen labeled as SPF 20 were tested by all the three methods in the study. Two experienced observers read results, respectively, under the same conditions. Results The volunteers in the current method group received the largest dose of UV light. The simple method was next to it and the single-exposure reduced 4-5 folds of UV dose received. The results of the two observers in the current method group and the single-exposure group were very close. But the test results of P3 were not completely coincident between the two observers in the simplified method group. SPFs value of the three sunscreens were 4. 12, 16.09, and 22.60 obtained by the observer 1 and 4. 18, 15.92, and 21.97 obtained by the observer 2, respectively. In the simplified method group, SPFs values of <4, >15, >20 and <4, <15, >20 were obtained by the two observers. In the single-exposure method group, SPFs values of the three sunscreens were 4.04, 15.34, 21.33 by the observer 1 and 3. 96, 15.82, 21.33 by the observer 2, respectively. Conclusion The single-exposure method is convenient and easily to operate and may replace the current method some day. The simple method is not valuable in determination of SPF labeled on the packages of sunscreens, but may be useful in supervision of the market sunscreen products.

Chinese Journal of Applied Physiology 2015;31(3):220-224
OBJECTIVETo determine the interaction between miR-21 and DNA methylation in different breast cancer cells.
METHODSFluorescence tagged miR-21 inhibitor and its negative control (NC) were transient transfected into MCF-7 and MDA-MB-231 cell, the transfection efficiency was observed using fluorescence microscopy, and the miR-21 expression level and genome DNA methylation status before and after transfection were assessed by real-time PCR and bisulfite-qMSP respectively. To investigate the regulation effect of DNA methylation on miR-21, cells were treated with 5-AZA (2.5 µmol/L) for 72 h, with dimethyl sulfoxide (DMSO) treatment as its negative control (NC), and the expression level of phosphatase and tensin homolog deleted on chromosome ten (PTEN) and AKT(also known as Protein Kinase B), two downstream genes of miR-21 were detected by Western blot.
RESULTSThe expression of miR-21 in MCF-7 cell was significantly knocked down (P < 0.01) by miR-21 inhibitor, with the genome DNA methylation level (P < 0.05) and all the three Dnmts: Dnmt1, Dnmt3a, and Dnmt3b unregulated. In contrast, the miR-21 expression in MDA-MB-231 cell was elevated ( P < 0.01) by miR-21 inhibitor, meanwhile, down- regulated of genome DNA methylation (P < 0.05) and Dnmt3b expression, upregulation of Dnmt3a were also observed. In addition, treated with 5-AZA resulted in significant increases of miR-21 expression in both MCF-7 and MDA-MB-231 cells (P < 0.01), with the protein level of PTEN increased in MCF-7 cell, which was further involved in the downregulation of AKT.
CONCLUSIONThe regulation effects of DNA methylation by transient transfection of miR-21 in MCF-7 and MDA-MB-231 cells are almost opposite, whilst the expression of miR-21 in two cell lines were all upregulated by decreased DNA methylation level and our results may provide some experimental evidences for the future development of rational therapy for different breast cancer.
