1.Effect of adenovirus-mediated bcl-2 transfer on regeneration and repair of peripheral nerves in rats

Objective To explore the effect of adenovirus-mediated bcl-2 transfer on the regeneration and repair of peripheral nerves in rats. Methods The sciatic nerve-transected model was selected and sutured end to end with epineurium. Then, adenovirus/sense-B-lymphoma/leukemia-2 (Ad/s-bcl-2), adenovirus/anti-sense-B-lymphoma/leukemia-2 (Ad/as-bcl-2), adenovirus/?-galactosidase (Ad/LacZ) and normal saline (NS) were injected into the sciatic nerve 0.5 cm away from the sutured point respectively. The rats were perfused with 4% paraformaldehyde at the 48th hour, 7th, 15th and 30th days respectively after operation. The spinal cords L 4-6 were harvested, frozen sectioned and given x-gal staining, bcl-2 in situ hybrid and immunohistochemical staining and GAP-43 immunohistochemical staining. The postoperative functional recovery of the rear limbs of rats was dynamically observed by using sciatic nerve functional index (SFI). Results The GAP-43 expression in the motor neurons of anterior horn of spinal cord in group Ad/s-bcl-2 was higher than that in other groups (P0.05). Conclusions Gene transfer of bcl-2 can promote regeneration and functional recovery of the peripheral nerves following sciatic nerve injury.
2.Effect of pelvic inflammatory disease grades on in vitro fertilization-embryo transfer outcome

Objective To evaluate the effect of different grades of pelvic inflammatory disease (PID)and of salpingectomy on outcomes of in vitro fertilization-embryo transfer(IVF-ET).Methods Two hundred and twenty three cycles of IVF-ET were divided into three groups,including mild group,moderate group and severe group,according to different grades of sequelae of PID finding in exploratory operations before IVF.Patients in each group were divided into two subgroups according to receiving salpingectomy or not.The data of total dose of gonadotrophin(Gn),oocyte number,low response rate,fertilization rate,good embryo number and pregnancy rate were analysed between three groups and the two subgroups of each group, respectively.Results All parameters were related to PID grades except fertilization rate.The total dose of Gn,oocyte number,good embryo number,low response rate and pregnancy rate were(2057?503)IU/L, (16?6),(6.0?4.3),4.2%,63.9% in mild group;(2204?603)IU/L,(12?6),(4.5?3.5), 13.9%,46.8% in moderate group;and(2372?1018)IU/L,(9?6),(3.1?2.9),33.8%,41.2% in severe group.The total dose of Gn and low response rate increased with the aggravation of the grades of PID (P
3.Risk factors for preeclampsia in women with irregular prenatal care

Objective To investigate the risk factors for severe and mild preeclampsia (PE) in women with irregular prenatal care,and to identify practical measures to reduce the occurrence of severe PE.Methods A retrospective study of 222 PE patients with irregular prenatal care,who delivered in Peking University Third Hospital from January 2007 to December 2011,was performed.The risk factors for PE and the status of prenatal care were analyzed.The non-parametric test,Chi-square test,Fisher's exact test,trendy Chi-square test and Logistic regression analysis were used for statistical analysis.Results There were 207 (93.2%) cases of severe PE and 15 (6.8%) cases of mild PE.In 207 severe PE patients,there were 95 cases (45.9%) of early-onset PE (diagnosed before 32 gestational weeks) and 112 cases (54.1%) of late-onset PE.In the 15 mild PE patients,there were two early-onset cases and 13 late-onset cases.The percentage of early-onset cases in severe PE patients was higher than that in mild PE patients [45.9% (95/207) vs 2/15,x2=6.027,P=0.015].After excluding 9 cases without any prenatal care,213 PE patients were analyzed,and it was found that the proportion of severe PE diagnosed in hospitals of grade 3,2 and 1 were significantly different [5/9,94.2% (131/139) vs 96.9% (63/65),x2=8.600,P=0.003].Compared with mild PE patients,the prenatal care interval for PE diagnosis in severe PE patients was longer [M(Q),8.0(4.0) vs 4.8(4.4) weeks,Z=2.695,P=0.007];the frequency of prenatal care after 20 gestational weeks was less [1(1) vs 3(3) times,Z=-4.195,P=0.000];the gestational week of PE diagnosis and referral to grade 3 hospitals were earlier [32.4(5.6) vs 35.4(4.3) weeks,Z=-3.075,P=0.002;33.1(5.3) vs 35.4(3.9) weeks,respectively,Z=-2.608,P=0.009];and the interval between PE diagnosis and referral was longer [0.1 (0.7) vs 0.0(0.0) weeks,respectively,Z=2.904,P=0.004].Multivariate logistic regression showed that the frequency of prenatal care after 20 gestational weeks was an independent risk factor for severe PE (OR=0.115,95%CI:0.046-0.285,P=0.000).Conclusion In women without regular prenatal care,the onset of severe PE is related to low-level prenatal hospital care,lack of prenatal care after 20 gestational weeks and longer prenatal care intervals as well as referral to grade 3 hospitals.
4.Investigation of teachers' cognition and behavior of PBL teaching in nursing college

Objective To understand teachers' cognition and behavior of PBL teaching in nursing college.Methods Eighty teachers were investigated by questionnaire at eight nursing colleges in Hunan province.The percentage was used to do statistical description.Results Totally 75 copies of valid questionnaires were returned,the effective recovery rate was 93.75%.Among all,96% of teachers thought that teacher was the guide;85.3% teachers thought that students were knowledge's seekers;93.3% teachers heard of PBL teaching;81.3% teaches thought that PBL teaching played a positive role ;74.7% teaches understood PBL teaching by communication or reading literature; 72% teachers were willing to try PBL teaching;46.8% teachers emphasized on multidisciplinary knowledge fusion in teaching; 88.3% teachers encouraged student discussion in class.The main factors effecting PBL teaching were case and problem design,teaching result evaluation,lack of expert guidance and teacher instruction.Conclusion Teachers have certain understanding of PBL.Organizers should formulate related policies for teachers and students in order to create good education environment.
5.mRNA expression and promoter methylation of paternally expressed gene 1 and paternally expressed gene 3 in human placenta of abnormal birth weight fetuses

Objective To study the mRNA expression of two imprinted genes,paternally expressed gene 1(PEG1) and paternally expressed gene 3(PEG3),and their promoter methylation in human placenta of abnormal birth weight(BW)neonates. Methods Placentas were obtained after full term delivery,without any maternal complications during pregnancy,and were divided into 3 groups according to BW:high BW group(n=22,BW≥4000 g),normal BW group(n=14,BW>2500 g and<4000 g)and low BW group(n=24,BW≤2500 g).The mRNA expression of PEG1 and PEG3 were determined by real-time quantitative polymerase chain reaction.Promoter methylation was measured by the bisulfite genomic sequencing method.Results among different groups were compared. Results (1)The mRNA expression of PEGl and PEG3 were 11.66±9.01 and 16.45±10.13 in the high BW group,0.84±0.49 and 0.85±0.67 in the low BW group and 1.10±0.77 and 1.11±0.60 in the normal BW group,respectively.Both genes were significantly up-regulated in the high BW group compared to the normal BW group(P<0.05),but no significant difference was found between the normal and low BW group (P>0.05). (2) The levels of PEG1 promoter methylation were (49. 7± 2. 3) %, (50. 2 ± 2. 1 )% and (50. 3 ± 1.9)% in the high, low and normal BW group (P>0.05), while the levels of PEG3 promoter methylation in the high BW was significantly lower than in the normal BW group [(13.1± 2. 7) % vs (16.2 ±1.8)%, P<0. 05], but no difference was shown between the low BW group (16.7± 3.5)% and the normal BW group (P>0.05). (3)Negative correlation was detected between the expression of PEG:3 and DNA methylation level within the objective fragment of promoter region (r= -0. 963, P<0. 01). Conclusions The up-regulation of PEG1 and PEG3 may be associated with high BW. The reduction of methylation pattern in the promoter region of PEG3 might be involved in the up-regulation of PEG3 and contribute to the mechanism of high BW fetus in turn.
6.Preventive effect of early bowel intervention on the gastrointestinal dysfunction in patients with invasive mechanical ventilation

Objective To explore the preventive effect of early bowel intervention on the gastrointestinal dysfunction in patients with invasive mechanical ventilation.Methods Sixty patients with invasive mechanical ventilation were equally randomly divided into the observation group and the control group.patlents in the control group were treated by conventional nursing care,while Patients in the observation group were given the early bowel intervention including abdominal massage and stimulation of rectum.The two groups were compared in terms of gastrointestinal dysfunction.Result The rate of gastrointestinal dysfunction in the observation group was significantly lower than that of the control group(P<0.05).Conclusion The early bowel intervention may effectively promote intestinal peristalsis,protect the intestinal digestion, absorption of nutrients and mucosal barrier function and prevent gastrointestinal dysfunction in patients with invasive mechanical ventilation during enteral nutrition.
7.Effect of TRF1,TRF2 and telomeric stability on As_2O_3 induced MGC803 cell apoptosis

Aim To study the effect of As_2O_3(arsenic trioxide) on the expressions of telomeric repeat binding factor1,2 and telomeric stability of human gastric cancer MGC803 cells, and explore the mechanism of cell apoptosis. Method MGC803 cell growth inhibition was measured with MTT assay. Apoptosis was analyzed with flow cytometry. Influence on chromosome distal end was analyzed with chromosome end-end fusion analysis. Expressions of TRF1 and TRF2 were determined with Western blot analysis. Results MTT assay showed that As_2O_3 clearly inhibited the growth of MGC803 cells, depending on time and dosage. The apoptosis rates were significantly higher than those of the control group in a concentration and time-dependent manner. These changes were not found in the control group. After disposal with 5 ?mol?L -1 As_2O_3, chromosome fusion rate was obviously increased in 48 h. After 48 h of disposal with 5 ?mol?L -1 As_2O_3, the TRF1 of MGC803 cells was up-regulated while TRF2 was down-regulated. Conclusion As_2O_3 induced chromosome fusion and MGC803 cells apoptosis through up-regulating expressin of TRF1 and down-regulating expressin of TRF2.
8.Pathogenic Bacteria in Urinary Tract Infection after Cesarean Section:Their Distribution and Drug Resistance

OBJECTIVE To understand the urinary tract infections after cesarean section,the distribution of pathogenic strains,and drug resistance analysis in order to provide clinical treatment. METHODS The routine urine cultures of the 2652 case were identified and drug resistance was analyzed by K-B method. RESULTS From 458 isolated bacteria,Gram-negative bacilli were main,pathogen accounting for 80.13%,Escherichia coli and Klebsiella,accounted for 71.62% and 4.8% respectively;Gram-positive bacteria accounted for 14.41%,and fungi accounted for 3.71%. CONCLUSIONS G-bacteria in the urinary tract infection are in the absolute superiority;rational use of antibiotics to effectively control the urinary tract infections and avoid drug-resistant strains produceing is especially critical.
9.Clinical feature of 9 patients harboring mtDNA G11778 A mutation with Leber hereditary optic neuropathy

AIM: To evaluate the clinical feature of 9 patients harboring mitochondrial DNA ( mtDNA ) G11778A mutation with Leber hereditary optic neuropathy ( LHON) .
●METHODS: Nine LHON patients were enrolled and followed- up between 2012 to 2015 in Shenzhen Eye Hospital, clinical data were collected and analyzed.
●RESULTS: Six cases had maternal inheritance history ( 67%) . Three were sporadic cases. The patients aged from 9 to 43 years old, with average age of (22. 00±9. 42) years. Simultaneous onset with both eyes was in 5 cases (56%). Successively onset was in other 4 cases (44%). The ratio between male and female was 2:1. In the last follow-up, the visual acuity was finger counted in 2 eyes (11%), 0. 01-0. 1 in 12 eyes (67%), 0. 12-0. 4 in 2 eyes (11%),≥0. 4 in 2 eyes (11%). All patients had pale disc and clear boundary. ln the Humphrey visual field examination, 10 eyes had typically cecocentral or paracentral scotoma, 8 eyes had diffuse visual field defect.
●CONCLUSION: ln the 9 LHON patients with mtDNA G11778A mutation, simultaneous onset cases were more than successively onset cases within 1y cases. In most cases, LHON patients kept stable visual acuity. Rare cases had a raise in visual acuity within 1y. Majority patients had typically cecocentral or paracentral visual field scotoma. In the last stage of LHON, visual field present diffused defect. The secondary affected eye was similar performed defect as the former one.
10.Pattern of motoneuron death following transection of the sciatic nerve in adult rats

Objective To investigate the pattern of motoneuron death induced by transection of the sciatic nerve in adult rats. Methods Sciatic nerve transected model was selected. The rats were sacrificed at 48 h, 7, 15, and 30 d after operation, respectively. The L 4-L 6 segments of the spinal cord were harvested after transcardiac perfusion with 4% paraformaldehyde, and were stained by TUNEL method and examined electron microscopically. Results A typical pattern of morphological changes of motoneuron apoptosis in the anterior horn of spinal cord was observed by both TUNEL staining and electron microscopy. Conclusion Motoneuron apoptosis in the spinal cord anterior horn induced by peripheral nerve transection demonstrates that neuron apoptosis may be the main pathological course of rat spinal cord neuron death following peripheral nerve injury in adult rats.
