2.Study on composition of essential oil in above-ground and root of Bupleurum malconense and root of B. chinense by AMDIS and retention index.

Chemical constituents of the essential oil in above-ground and root of Bupleurum malconense and root of B. chinense were investigated by GC-MS compiled with automated mass spectral deconvolution and identification system (AMDIS) and retention index. The results showd that the components of essential oil in B. malconense have some similarities with the one in B. chinense, and both of them have the higher content of caryophyllene oxide which is an active component of anti-inflammatory and analgesic. These results suggested that as a local substitute, B. malconense has a certain scientific basis of the treatment for cold fever.

Object To compare the contents of alkaloids of callus at different period with tissue culture of fresh bulbs of Fritillaria mellea S. Y. Tang et S. C. Yueh.Methods To study the best condition of induction frequency of callus about some phytohormones, temperature and light, and then to determine the contents of alkaloids of callus in F. mellea. Results The best condition of induction frequency of callus above was NAA 1 mg/L+ 6BA 3 mg/L; temperature at (22?1) ℃ and scattered light; the contents of alkaloids were kept highest in about 20th day ofter the callus appeared.Conclusion The contents of alkaloids of callus are kept higher level than fresh bulbs of F. mellea.

7.Effect of ATP-binding cassette transporter A1 on cholesterol efflux from cells

Objective To investigate the potential change of cellular cholesterol efflux and the functional changes of ABCA1 on macrophages from diabetes animals. Methods High energy diet was given to golden hamster to make animal model of hyperlipidemia. Diabetes was induced by a single intraperitoneal injection of STZ (30 mg/kg). Macrophages were isolated and incubated with apoA-I or 8-br-cAMP in vitro. Cellular cholesterol content was measureal before and after treatment by HPLC. Results Intracellular cholesterol content of diabetic animals was higher than that in high energy diet and normal controls. Expression of ABCA1 mRNA was upregulated in diabetic group after incubation with apoA-I and cAMP. When macrophages were incubated with apoA-I, cholesterol efflux increased with the prolongation of incubation time.The amount of intracellular cholesterol efflux in 3 groups of animals showed the following relationship: diabetic animals exceed high energy diet fed animals, and the latter exceeds normal animals. Conclusion Intracellular lipid efflux depends mainly on the presence of extracellular apoA1 as well as membrane ABCA1 protein. The deposition of cholesterol in the macrophage of diabetic and insulin resistant animals may be related to the characteristic changes of quantity and function of apoA1 in these animals.
8.Effects of intrathecal Ro25-6981 on focal cerebral ischemia-reperfusion injury in rats

ObjectiveTo investigate the effect of intrathecal (IT) Ro25-6981 (a selective NR2B receptor antagonist) on focal cerebral ischemia-reperfusion (I/R) injury in rats.MethodsSixty healthy male SD rats weighing 280-320 g in which IT catheter was successfully implanted without complication were randomly divided into 4 groups ( n = 15 each):group sham operation (group S) ; group focal cerebral I/R(group I/R) ; group I/R + Ro25-6981 (group Ro) and group I/R+ normal saline (group NS).The right middle cerebral artery was occluded for 2 h with a nylon thread with rounded tip which was inserted into right internal carotid artery and advanced cranially until resistance was met in groups I/R,Ro and NS.In group Ro Ro25-6981 100 μg/10 μl was injected IT at 0,2,23 h of reperfusion,while in group NS NS was injected IT instead of Ro25-6981.Neurological dificit was assessed and scored (0 = no deficit,3 = severe deficit) at 24 h of reperfusion.The infarct size was determined by 2,3,5-triphenyl tetrazolium chloride staining.ResultsIT administration of Ro25-6981 significantly reduced cerebral I/R-induced infarct sized and neurological deficit score in group Ro as compared with groups I/R and NS.ConclusionRo25-6981 injected IT can protect the brain against local cerebral I/R injury.
9.Clinical, histopathologic and ultrastructural characteristics of achromic naevus

Objective To study the clinical, histopathologic and ultrastructural characteristics of achromic naevus (AN). Methods Clinical data, including sex, age, age of onset, pattern of lesions, involved sites, shape and number of lesions and associated systemic diseases, were collected from 85 patients with AN. Skin melanin index was detected in 34 lesions of 19 patients with AN, 30 lesions of 12 patients with vitiligo and 64 contralateral normal skin islands of the 31 patients. Reflectance confocal microscopy (RCM) was performed to analyze the lesion, normal skin and junctional area between lesional and normal skin of 62 patients with AN. Tissue samples were obtained from lesions and perilesional normal skin of 17 patients with AN and subjected to pathological examination as well as ultrastructural study with transmission electron microscopy; also, skin biopsy specimens were immunostained for tyrosinase, HMB45, tyrosinase-related protein-1 (TRP-1), TRP-2 and CD117. Results Of the 85 patients with AN, 23 (27.1%) developed lesions at birth, and 21 (24.7%) after 3 years of age; 72 (84.7%) had irregularly shaped lesions, 54 (63.5%) had only a single lesion. The mean melanin index and relative melanin index of AN lesions were 186.56 ± 52.86 and 80 ± 11, respectively, significantly lower than those in normal skin islands (223.88 ± 63.19 and 100, both P < 0.01), but higher than those in depigmented lesions from 12 patients with vitiligo (128.57 ± 64.31 and 60 ± 20, both P < 0.01). RCM revealed a decline in the number of melanocytes and brightness of melanin caps, even distribution of melanin in lesions, as well as obscure demarcation between lesions and normal skin from patients with AN. Fontana-Masson stain showed that the melanin content was lower in lesions than in perilesional skin (1810.12 ± 327.96 vs 2064.24 ± 260.41) from patients with AN. Microscopic examination demonstrated a decrease in melanocyte and melanosome number, presence of immature melanocytes at stage Ⅱ and Ⅲ in cytoplasm and dendrites of melanocytes and keratinocytes, aggregated melanosomes in affected keratinocytes in lesions of AN. In 17 patients with AN, the relative expression levels of tyrosinase and TRP-1 were 1827.35 ± 307.09 and 6102.54 ± 1642.64, respectively, in normal skin specimens, significantly higher than those in lesional skin (1477.35 ± 224.05, 5322.33 ± 1565.26, both P< 0.01); no statistical difference was observed in the expression levels of HMB45, TRP-2 or CD117 between lesional and normal skin. Conclusions AN is an early-onset, nonfamilial aggregated, stable leukoderma with irregular margins, and in lesions of AN, the number of both melanocytes and melanosomes is decreased with the presence of immature melanosomes. The measurement of relative melanin index and reflectance confocal microscopy may offer a non-invasive approach to the diagnosis of AN.
10.Application of body strait jacket design in mental patients

Objective To explore the application effect of self-developed strait jacket in mental patients.Methods A total of 200 mental patients meeting indications for protective restraint were selected from a tertiary first-class psychiatric hospital using convenience sampling,and were restrained using self-developed strait jackets,application effects and numbers of adverse events occurring during restraint were observed;patients' satisfaction with restraint tools was investigated as well.Results Cases of adverse events occurring during protective restraint were:3 patients developed dysaemia (1.50%),3 had self-injury (1.50%),4 were injured by others (2.00%),5 developed skin scratching (2.50%),7 threw off restraint by themselves (3.50%),13 presented violent and aggressive behaviors (6.50%),and no patient developed swelling of limbs.According to satisfaction survey,71.00% of the patients demonstrated acceptance of this restraint tool,76.50% felt comfortable,83.50% held that this strait jacket could meet the needs of free activities,76.00% felt safe,and satisfaction to body strait jacket came to 83.00%.Conclusion The manufacture of body strait jacket is simple and it is comfortable to wear,patient's compliance is high,and restraint effect is satisfactory;this body strait jacket reduces the incidence of adverse events,and improve patient's satisfaction to restraint tool.
