Objective:To realize Tangshan City’s medical institutions of health and human resources. Methods: Using gray predict model to prognosis Tangshan City’s main medical human resources, and find out the phenomena of lacking of medical administrative staff, disproportional of nursing staff, and shorting of assistant departments staff, such as pharmacy and medical laboratory.Results: In the next few years, we should focus on strengthening the ability of the health technical personnel, except practicing(assistant)doctors.Conclusion:Through increasing the orientation training and recruitment, improving treatment, changing ideas, etc, to achieve the rationalization of health personnel allocation. In order to coping with health care reform, we should complete the allocation and reserve of health human resources.

There are many problems existing in humanities and social science courses in education and teaching of medical colleges and universities.Changing concepts of education,adjusting the curriculum,improving teaching methods and improving the quality of teachers are effective measures in medical colleges to strengthen the humanities and social science courses.We have made a preliminary exploration on teaching practice for humanistic education and professional education.

AIM: To study the application of macular ganglion cell complex (mGCC) and peripapillary retinal nerve fiber layer thickness (pRNFL) measured by optical coherence tomography (OCT) in the early diagnosis of glaucoma.METHODS: Case-control study.Eighty-six subjects, including 30 eyes in normal subjects, 27 eyes in suspected primary open angle glaucoma, 29 eyes in primary open angle glaucoma were enrolled in this study.The thickness of mGCC and pRNFL were measured by OCT.The area under the receiver operating characteristic(AROC) curve at fixed specificities were calculated for each parameter.RESULTS: There were significant differences in mean pRNFL thickness, superior pRNFL thickness and inferior pRNFL thickness between normal group, suspected glaucoma group and early glaucoma group (P=0.001, 0.004, 0.011).The mean mGCC thickness, the thickness of the top mGCC, the thickness of the lower mGCC were statistically significant (P=0.008, 0.002, 0.003);the difference of general loss of volume (GLV) and focal loss of volume (FLV) between the three groups was statistically significant (P=0.002).Compared with the normal group, all the pRNFL and the mGCC parameters were higher in the suspected glaucoma group, and the FLV had the highest AROC (0.801), all the remaining AROC was >0.700 except above Prnlf (0.688).Compared with the normal group and the early glaucoma group, all the pRNFL and the mGCC had higher AROC, average mGCC was hightest(0.804), all parameters AROC were >0.700 except mean pRNFL (0.683).In suspected glaucoma group, 58% patients had abnormal mGCC thickness and 23% had abnormal pRNFL thickness;in early glaucoma group, 98%patients had abnormal mGCC thickness and 90% had abnormal pRNFL thickness;in normal group, 93%patients had abnormal mGCC thickness and 93%had abnormal pRNFL thickness, the correlation between the three groups was statistically significant (x2=12.11, P<0.05).CONCLUSION: OCT measurement of mGCC thickness and pRNFL thickness in early glaucoma have good diagnostic ability;mGCC thickness measurement can be used as an effective method for early diagnosis of glaucoma.

Objective To investigate the role of asctrocytes in the process of chronic degeneration of dopaminergic neurons in intracephalic inflammation rat model induced by intracerebroventricularly injection of lipopolysaccharide.Methods Sixty healthy male SD rats were assigned into lipopolysaccharide group or saline control group randomly.All injections were made intracerebroventricularly on right side of the rats.Ethovison software was used to measure the movement distance of rats within 30 minutes.Specific antibody for glial fibrillary acid protein (GFAP) was used in immunohistochemistry stain to detect the changes of asctrocytes in the substantia nigra of rats.Results Movement distance of lipopolysaccharide-injected rats decreased by 21.2% compared with saline-injected rats at 16 weeks after injection (t=2.54,P<0.05)by 27.0% (t=3.55,P<0.01) at 24 weeks and by 31.4% (t=3.91,P<0.01) at 28 weeks after lipopolysaccharide injection.The asctrocytes were activated obviously in the substantia nigra of lipopolysaccharide-injected group at 2 weeks,while the numbers of GFAP-positively stained cells (228.60 + 22.35) increased significantly compared with saline-injected group ( 165.20 ± 25.97) (t = 4.14,P< 0.05).The activation of asctrocytes was not found in lipopolysaccharide-injected group at 4 weeks and 12 weeks.The asctrocytes were re-activated in the substantia nigra of lipopolysaccharide-injected group at 24 weeks,while the numbers of GFAP-positively stained cells (220.00±21.01 ) increased significantly compared with saline-injected group (169.00± 19.00) (t= 4.03,P<0.05).The activation of asctrocytes was not seen at any time point in saline-injected group.Conclusions Intracephalic inflammation induces chronic degeneration of substantia nigral dopaminergic neurons in rats.The asctrocytes exhibite "acute activation-quiescing-reactivation" state,indicating that they might be involved in the mechanism of dopaminergic neurons degeneration.

OBJECTIVE:To optimize the parameters of the extraction and purification technologies of total flavonoids from Di-ospyros kaki Thunb. leaves,and then to compare the contents of flavonoids in fresh and dried D. kaki Thunb. leaves. METHODS:Taking the yield of total flavonoids as the investigated index,the influences of liquid-solid ratio,the volume fraction of ethanol and ultrasonic extraction time on the extraction effect were discussed by single-factor and orthogonal test. With the purity of total flavo-noids as the investigated index,the purification effects of such three kinds of methods as ethyl acetate extraction method,alcohol deposition method and macroreticular resin purification method combined with petroleum ether degreasing on the extracted solution were compared. The optimal extraction technology was adopted to extract the total flavonoids from fresh and dried D. kaki Thunb. leaves and their contents were compared. RESULTS:The optimal extraction technology was as follows as liquid-solid ratio of 25∶1 (ml/g),volume fraction of ethanol of 70%,ultrasonic extraction time of 30 min,extraction temperature of 30 ℃. The results of the verification tests showed the average content of total flavonoids was 1.75%(RSD=2.00%,n=3). The total flavonoids in the extracted solution purified by the above-mentioned three purification methods had a purity of 24.92%,15.94% and 35.52% respec-tively,in which the macroreticular resin purification method with petroleum ether degreasing combined with AB-8 resin purification was optimal. The content of flavonoids in fresh D. kaki Thunb. leaves(1.75%) was about twice as much as that in dried leaves (0.87%). CONCLUSIONS:The optimal extraction and purification technologies are simple with good effect,and suitable for large-scale production. Fresh D. kaki Thunb. leaves should be used as raw materials for extracting flavonoids.

Objective:To explore the effect of tectorigenin on myocardial fibrosis(MF) in the rats and clarify the related mechanism,and to provide reference for its clinical application. Methods:Sixty Wistar rats were randomly divided into normal control group,model group,positive drug (capropril) control group, and low,middle,high doses of tectorigenin groups(n=10).Except normal control group, the rats in other groups were used to construct MF models by subcutaneous injection of 5 mg·kg -1·d -1 isoproterenol (Iso) for 7 d.The rats in tectorigenin groups and captopril group were intragastricly administrated with different doses of tectorigenin (25,50,100 mg·kg-1·d-1)and captopril(10 mg·kg -1·d -1) from the second day after modeling for consecutive 28 d.Bl-420E+ biological function experiment system was used to detect the heart function;Heart mass index (HMI) and left ventricular mass index (LVMI) were measured after experiment.UV detection was used to measure the levels of malondialdehyde (MDA) and the activities of superoxide dismutase (SOD) in myocardial tissue.Microplate reader was used to measure the activities of lactic dehydrogenase(LDH) and creatine kinase(CK) and the levels of nitric oxide (NO)in serum.ELISA were used to detect the levels of collagen typeⅠ(ColⅠ) and collagen type Ⅲ (Col Ⅲ) in myocardium tissue of the rats.The pathological changes of myocardium tissue of the rats in various groups were observed by HE staining.Results:Compared with normal control group,the HR of rats in model groups was increased,and the left ventricular systolic pressure (LVSP) was decreased(P<0.01);the HMI and LVMI were increased(P<0.05),the levels of MDA in left ventricular myocardial tissue was increased(P<0.01),and the activity of SOD was decreased,the levels of serum ColⅠ,Col Ⅲ and the activities of LDH , CK were also increased(P<0.01);the level of NO in serum was decreased(P<0.01).Compared with model groups, the HR were decreased,LVSP were increased, and HMI and LVMI of the rats in different doses of tectorigenin groups were decreased in a dose-dependent manner;the levels of MDA were reduced;the activities of SOD were increased in myocardium tissue,and the CK activities and the ColⅠ and ColⅢ levels were decreased(P<0.05 or P<0.01);the LDH activities in middle and high doses of tectorgenin groups were decreased(P<0.01);and the levels of NO in serum in different doses of tectorigenin groups were significantly increased(P<0.05 or P<0.01) .Conclusion:Tectorigenin could inhibit the MF induced by Iso in the rats, and its mechanism may be related to antioxidation,scavenging free radical and inhibition of collagen synthesis.

Objective To investigate the long-term effect of inflammation on behavior,microglias and dopaminergic (DA) neurons in the substantia nigra of intracephalic inflammation rat models induced by intracerebroventricular injection of low-dose(10μg) lipopolysaccharide (LPS).To analyze the relationship between activation of microglias and DA neurons degeneration in order to explore the mechanism of inflammation in the progressive process of Parkinson' s disease (PD).Methods 50 healthy male SD rats were randomly assigned into saline-injected control group and 10μg LPS-injected group.All injections were made intracerebroventricularly on right side of rats with saline or LPS.Moving speed was measured at different time points.At 24 weeks and 40 weeks after saline or LPS injection,specific antibodies of OX-42 and OX-6 were used separately to detect the changes of microglia in the substantia nigra of rat.The changes in morphology and numbers of substantia nigra DA neurons were observed by tyrosine hydroxylase(TH) immunohistochemical staining.The expression and distribution of the degenerated neurons in substantia nigra were detected by using Fluoro-Jade B(FJB).Results ①Analysis of moving speed sho wed that the moving speed of 10μg LPS-injected group rats and saline-injected group rats was similar from 4 weeks to 36 weeks after injection.At 40 weeks post injection,moving speed of 10μg LPS-injected group rats decreased by 24.6％ compared with that of saline-injected group rats (P＞ 0.05 ).②At 24 weeks and 40 weeks after injection,there were many activated OX-42 positive microglias in the substantia nigra of 10μg LPS-injected group rats,but there was almost no significant activated OX-42 positive microglia in saline-injected group.OX-6 positive microglias were not found in the substantia nigra of both of two groups.③At 24 weeks and 40 weeks post injection,the number of TH-positive neurons in the substantia nigra of 10μg LPS-injected group rats decreasedby 24.2％ ( t=4.803,P＜0.01) and 27.6％ ( t=3.212,P＜0.01) respectively compared with those of salineinjected group.④ There was no FJB positive neurons in the substantia nigra of the two group rats.Conclusion Intraventricular injection of low-dose LPS ( l0μg) in rats may induce long-term activation of microglias and chronic degeneration of DA neurons in the subs tantia nigra of rats although the necrosis are not occurs to DA neurons till 40 weeks post LPS injection.Intraventricular injection of low-dose LPS in rats could be ideal model to study the mechanism of chronic degeneration of DA neurons in PD.
