Objective:To study the influence of laboratory mental stress on plasma levels of angiotensin II (Ang II), aldosterone(ALD)and atrial natriuretic peptide (ANP) in individuals with different salt sensitivity Method:48 healthy male undergraduates were divided into salt sensitive group (SS group, n=14) and non-salt-sensitive group (NSS group, n=34) Their blood pressure, plasma levels of Ang II, ALD and ANP were measured before and after mental arithemtic test Result:Blood pressure, plasma levels of Ang II and ALD increased significantly in both groups, with SS group having greater increase Plasma level of ANP had no change after the test Conclusion:Laboratory mental stress increases the plasma levels of Ang II and ALD in male students Salt-sensitivity can magnify this effect

5.Study on Protective Effect of Mongolian Medicine Lomatogonium rotatum Water Extract against Acute Liv-er Injury in Mice Induced by D-GlaN and CCl4

OBJECTIVE:To study the protective effects of Mongolian medicine Lomatogonium rotatum water extract against acute liver injury in mice caused by D-GlaN and CCl4. METHODS:60 mice were randomly divided into normal group(normal sa-line),D-GlaN model group(normal saline),positive control group(Kuihua hugan tablet,0.56 g/kg)and L. rotatum water extract high-dose,medium-dose and low-dose groups [3,1.5,0.75 g(crude drug)/kg] 10 rats in each group. They were given relevant medicine intragastrically once a day for consecutive 23 days. 30 min after last administration,those group were given D-GlaN ip to induce acute liver injury model except normal group. The activity of AST,ALT,ALP,and acetylcholinesterase(CHE)content in serum were detected,and liver index was calculated. Liver histopathology was observed and scored. Other 60 mice were selected, and then grouped,given medicine and detected in lab index with same method with above group,except for modeling method(ig, 0.1% CCl4). RESULTS:Compared with normal group,necrosis or degeneration of liver cells were obvious in 2 model groups, pathological score,the activity of AST,ALT and ALP increased while CHE content decreased;liver index of D-GlaN model group increased while that of CCl4 model group decreased (P<0.05). Compared with D-GlaN model group,the activity of AST,ALT and ALP and liver index decreased in L. rotatum water extract groups,while CHE content increased(P<0.05);pathological mor-phology had been improved,and pathological score decreased. Compared with CCl4 model group,ALT activity decreased in L. rota-tum water extract low-dose group,while CHE content and liver index increased;CHE content of L. rotatum water extract medi-um-dose group decreased,while liver index increased;the activity of AST,ALT and ALP decreased in L. rotatum water extract high-dose group,while CHE content increased(P<0.05). Pathological morphology of L. rotatum water extract groups had been im-proved to certain extent,and pathological score decreased. CONCLUSIONS:L. rotatum water extract could protect acute liver inju-ry in mice induced by D-GlaN and CCl4.

7.Effect of Compatibility of Radix Astragali and Radix Angelicae Sinensis in Different Proportion on Hemorheology and Contents of IL-4, IFN-γ in Blood of Qi Deficiency and Blood Stasis Rat Model

Objective To observe the effects of the compatibility of Radix Astragali and Radix Angelicae Sinensis in different proportion on hemodynamic and balance of Th1/Th2 cells of Qi deficiency and blood stasis rat model. Methods Qi deficiency and blood stasis rats were caused by restricted diet, forced swimming and norepinephrine subcutaneous injecting, and treated by compatibility of Radix Astragali and Radix Angelicae Sinensis in different proportion of 5∶1, 3∶1, 1∶1 and 1∶2 for 21 days. The indexes of hemorheology were detected with hemorheological analyser, and the level of IFN-γand IL-4 in serum were tested by ELISA. Results Compared with model group, the compatibility of Radix Astragali and Radix Angelicae Sinensis in proportion of 1∶1, 3∶1 and 5∶1 groups reduced the low shear blood viscosity. The spleen index of model group decreased, and compatibility of Radix Astragali and Radix Angelicae Sinensis in 5∶1 proportion group increased spleen index. The level of IL-4 increased and IFN-γ decreased in the serum of model group, the compatibility of Radix Astragali and Radix Angelicae Sinensis in proportion of 3∶1 group increased the level of IFN-γ. Astragalus angelica 5∶1 group decreased the level of IL-4 and increased the level of IFN-γ. Conclusion The compatibility of Radix Astragali and Radix Angelicae Sinensis can improve hemorheology, adjust the balance of Th1/Th2 cells of Qi deficiency and blood stasis rats. The effects were better when Radix Astragali’s dosage greater than that of Radix Angelicae Sinensis, and the group of 5∶1 proportion was the best.

9.Cyr61 expression influences cancer cell proliferation and apoptosis via PI3K pathway in human ovarian carcinoma cells

Objective To investigate the relationship between cysteine-rich protein 61 ( Cyr61 ) and phosphatidylinositol 3-kinase ( PI3K ) signal pathway on cell proliferation and apoptotic in human ovarian carcinoma cells.Methods Recombinant human Cyr61 (rhCyr61) was pretreated with ovarian carcinoma cells.The expression of Cyr61 protein was detected by confocal spectral microscopy.Then treated the ovarian carcinoma cells with PI3K transduction inhibitors (LY294002) for 24 hours.Cell apoptosis was detected by flow cytometry (FCM).Cell viability was determined by methyl thiazolyl tetrazolium (MTT) method.The mRNA expressions of Cyr61,the protein levels of protein kinase B ( PKB),phospho-PKB and Cyr61 were assaved by real time-PCR and western blot analysis,respectively.Results The Cyr61 and phospho-PKB protein expression in two ovarian carcinoma cells (OV2008 and OVCAR-3 ) were increased in rhCyr61pretreated group.The decreasing of cell apoptosis [ ( 1.4 ±0.9)%,(2.1 ± 1.0)% ] and increasing of cell proliferation [ ( 124.0 ± 1.8)%,( 133.0 ±2.2)% ] was detected in the same time,compared with negative control group,there were significant difference ( P < 0.05 ).After exposed to LY294002 for 24 hours,the apoptosis rate of OV2008 and OVCAR-3 in pretreated with rhCyr61 group exposed to LY294002 were (21.1 ± 1.6)% and (26.4 ± 1.5 )%,respectively.Cells viability [ (59.0 ± 2.3 )%,(51.0 ± 2.0)% ]was also significantly decreased in OV2008 and OVCAR-3 pretreated with rhCyr61 cells.Meanwhile,the mRNA expressions of Cyr61 (3.2 ± 0.8,6.2 ± 1.1 ) and the protein levels of phospho-PKB and Cyr61 were greatly decreased.Compared with negative control group,there were significant difference in OV2008 and OVCAR-3 cells (all P < 0.0l ).Conclusions The activation of PI3K intracellular signaling pathways may lead to up-regulation of Cyr61 expression.Block PI3K signal pathway could significantly inhibit the expression of Cyr61,and may promote the apoptotic effects and inhibit the cell growth of ovarian carcinoma cells.
10.Effect of different fraction interval on tumor control in C57BL mice implanted with Lewis lung cancer

Objective To study the effect of different fraction interval with same total radiation dose on tumor growth delay and survival in C57BL mice implanted with lewis lung cancer,and to determine whether prolonged fraction interval will decrease the tumor response to radiation. Methods Forty-eight mice were implanted with lewis lung cancer in the back legs.When the diameter of transplanted tumor reached 0.8 to 1 cm,the mice were randomized into 6 groups:normal control group,single fraction of 18 Gy group,18 Gy in 2 fractions of 9 Gy at 30 min interval group,18 Gy in 7 fractions of 2.57 Gy at 5 min inter val group,18 Gy in 2 fractions of 9 Gy at 60 min interval group and 18 Gy in 7 fractions of 2.57 Gy at 10 rain interval group.The maximal and minimal diameters of the tumor were measured and record every other day to study the tumor growth tendency,the tumor growth delay and the mice survival time. Results The tumor growth delay of groups at prolonged fraction interval was shorter than the group with single fraction of 18 Gy (P < 0.05).The tumor growth delay of groups at fraction interval of 30 rain was longer than that of groups at interval of 60 rain (P < 0.05).There was no significant difference of the tumor growth delay be tween the groups with same delivery time (P >0.05).The mice survival time of the groups with prolonged fraction interval was shortened when omparing to the group with single fraction of 18 Gy.While the difference was not significant between the groups at fraction interval of 30 min and 60 min. Conclusions The pro longed fraction interval but same total radiation dose shortens the tumor growth delay and survival time in the mice implanted with Lewis lung cancer.The longer fraction interval impairs the tumor control more signifi candy.However the difference of the effect on mice survival time is not significant between the groups at fraction interval of 30 min and 60 min.
