1.The impact of the Jiawei Xiaochaihu particle and it's prescription on immune function of ITP mice

Objective:To observe the impact of the Jiawei Xiaochaihu particle and it's prescription on immune organ and immune function of ITP mice.Methods:The ITP mice were divided into five groups at random:(1)Nomal group;(2)ITP model group;(3)Jiawei Xiaochaihu particle group;(4)Xiaochaihu particle group;(5)Danshen and Sanqi particle group.All ITP mice were administrated 7days later by 30ml per kilogram the rat's weight once a day for 14 days.21 days after the model mice were made,peripheral blood T lymphocyte subpopulation was detected with flow cytometry.Antibody against platelets was measured by ELISA.Peripheral blood IL-2 and IL-10 were measured by radio immuno assay(RIA).Thoracic gland and spleen were weighted to compute the immune organ index.Results:Spleens of ITP mice had intumesce of spleens and a rise of the index;Peripheral blood platelet antibody was also increased;A disproportion was found in T lymphocyte subpopulation,CD8 upgrade,CD3 and CD4 descent,CD4/CD8 also descent.As one of the Th1 cytokine,IL-2 expression accentuated while IL-10,the kind of Th2 cytokine,was inhibited.There were significances when compared with the nomal group(P0.05).Conclusion:There are indeed dysfunction in immune organ and its function of ITP mice.Jiawei Xiaochaihu particle and Xiaochaihu particle can exert a regulation,but the Danshen and Sanqi particle not.
2.Effect of different prone time/supine time ratios to pathology of mini-pig with acute lung injury

Objective To compare the effects of different prone and supine position duration within 24-hour period on mini-pig acute lung injury (ALI)models. Methods Fifteen female mini-pigs,mean weight (30.2±3.2) kg, length (76.5±5.9) cm were anesthetized and ventilated. Hydrochloric acid (pH 1.0)was aspirated intratracheally to create ALI model. Mini-pigs were assigned randomly into three groups: p rone 8 h group, prone 8h then supine 16 h; 16 h group, prone 16 h then supine 8 h; 24 h group, prone 24 h continuously. Min-pigs were euthanasia at the end of the experiment to take out the lung, examine the tissue change and calculate the wet to dry weight ratio of lung (W/D). Sample was cut into slices and carried on hematoxylin and eosin (HE) stain which was intended to calculate injury score by semi ration score criteria. Results W/D, and injury score were lower in 16 h group and 24 h group than the 8 h group,whereas no significant differences among the three groups. Conclusions Prone position for 16 hours or longer could decrease oxygenation by decreasing inflammation in acute lung injury(ALI)animal models.
3.Effects of peroxisome proliferator-activated receptor γ agonist on serum adiponectin, urine microalbumin and renal pathology of obese mice

Objective To investigate the effects of peroxisome proliferator-activated receptor γ (PPARγ) agonist on serum adiponectin (ADP),urine microalbumin (mALB) and kidney pathology of obese mice,and discuss the significance of PPARγagonists preventing obesity-related glomerulopathy (ORG).Methods Sixteen male ob/ob mice and 8 male C57BL/6 mice which were 8 weeks old were selected in this study.Ob/ob mice were dividing into 2 groups according to body mass:obesity group (M group) and pioglitazone intervention group (T group) which were fed with high lipid chow.C57BL/6 mice were control group (C group) which were fed with ordinary chow for 12 weeks.Body mass,blood glucose,serum ADP,urine mALB were assayed in each group and compared.The morphological changes of kidney were observe by HE staining,glomerular diameters were measured and compared.Zonula occludens-1 (ZO-1) and Wilms tumor 1 (WT1) were positioned by immunohistochemistry,the level of ZO-1 expression in podocyte and podocyte number which was signed with WT1 in each group were evaluated and compared.And then the correlations between serum ADP,urine mALB,body mass,blood glucose,kidney mass,glomerular diameter,the level of ZO-1 expression,podocyte number were analyzed.Results The serum ADP of obese mice decreased (P < 0.05),and the urine mALB increased (P < 0.05),when compared with that of control group,and there was a negative correlation between ADP and mALB (r =-0.538,P < 0.01).Renal pathology showed glomerular hypertrophy,in part associated with focal segmental glomerulosclerosis (FSGS),the expression of ZO-1 in podocyte and podocyte number were lower than that of control group (P < 0.05).After the intervention with pioglitazone,the urine mALB of obese mice reduced (P < 0.01),and the serum ADP level of them was higher than that of obese mice without intervention (P < 0.01).There was no glomerular hypertrophy and glomerulosclerosis in kidney of obese mice,the expression of ZO-1 in podocyte and podocyte number were higher tban that of obese mice without intervention (P < 0.05).Conclusions The kidneys of obese mice have clinical and pathological changes indicating that obesity can lead to kidney damage.Pioglitazone can make the low serum ADP level of obese mice ameliorated,and make the urine mALB of them reduced.Renal pathological change significantly is alleviated,the expression of ZO-1 in podocyte and podocyte number are increased,indicating pioglitazone can improve renal injury related to obesity.

5.Construction of bioartificial renal tubule assist device and determination of transport to creatinine,amino acids and sodium in vitro

Objective To investigate the method of constructing bioartificial renal tubule assist device(RAD) and its transport function to creatinine,glucose,sodium and asparagine,theonine,serine,glutamic acid in vitro.Methods The cultured LLC PK1 cells were seeded into the inner space of the hemofilter(FH66)and the hemofilter was incubated for two weeks.Then the filtration in vitro was simulated to detect the function of the RAD.The transport rates of sodium,creatinine,glucose and four kinds of amino acids were measured.Meanwhile,the effects of oubaine and phlorizin on the transport rates of sodium,glucose and amino acids were determined and compared with no LLC PK1 device.Results The creatinine transport rate of RAD was significantly decreased as compared with control group(P

Objective To explore the relationship between serum creatinine ( Scr ) , blood urea nitrogen ( BUN) and lung image changes in uremic patients .Methods According to the chest CT ,X-ray and related findings , 256 patients with uremia were divided into 84 cases of uremia ( uremic lung group ) and non-uremic lung 172 cases ( non-uremic lung group ) .The chest CT and X-ray examination ,Scr and BUN levels were observed .Results Uremic lung group had five types of image findings,including alveolar edema (67.86%),pulmonary interstitial edema (16.67%),pulmonary congestion syndrome(8.33%),pleural effusion(4.76%) and pulmonary interstitial fibrosis (2.38%).The Scr[(1 145.6 ±236.4)μmol/L]and BUN [(38.9 ±6.1)mmol/L]levels in uremic lung group were significantly higher than those in the non-uremic lung group[(905.3 ±138.9)μmol/L,(23.8 ±5.6)mmol/L](t=8.128,4.782,all P<0.05).Conclusion The image of uremic lung is mainly pulmonary edema ,and radiological and pathological changes are closely related with Scr and BUN levels .
7.Protective effect of diphenyleneiodonium, a NADPH oxidase inhibitor, on hyperpermeability of endothelial cells exposed to AOPP-HSA in vitro

[ ABSTRACT ] AIM: To investigate the effect of advanced oxidation protein product-human serum albumin ( AOPP-HSA) at different concentrations on the permeability of human umbilical vein endothelial cell ( HUVEC) monolayer and the protective effect of NADPH oxidase inhibitor diphenyleneiodonium ( DPI ) against AOPP-HSA exposure. METHODS: Cultured HUVECs were exposed to 200 mg/L HSA (control) or AOPP-HSA (50, 100 and 200 mg/L).The permeability of the endothelial monolayer was assessed by measuring CMFDA-labeled THP-1 cells across the endothelial cells.The cultured HUVECs were treated with HSA (200 mg/L), AOPP-HSA (200 mg/L), or AOPP-HSA (200 mg/L)+DPI (100 μmol/L), and the activation of NADPH oxidase, endothelial monolayer permeability and cytoskeleton rear-rangement were evaluated.RESULTS: AOPP-HSA increased the permeability of the endothelial cell monolayer, and AOPP-HSA at 200 mg/L significantly increased the phosphorylation level of NADPH oxidase in the cells.Treatment with 100 μmol/L DPI obviously attenuated AOPP-HSA-induced NADPH oxidase activation, the increase in the permeability of the cell monolayer and the cytoskeleton rearrangement.CONCLUSION: AOPP-HSA increases the hyperpermeability of HUVEC monolayer via the phosphorylation of NADPH oxidase, and the NADPH oxidase inhibitor DPI reverses such effects.

9.Expression of monocyte chemotactic protein-1 and its receptor in sudden coronary death.

OBJECTIVE:
To investigate the expression of monocyte chemotactic protein-1 (MCP-1) and its receptor CC chemokine receptor-2 (CCR-2) in coronary atherosclerosis plaques between sidden coronary death (SCD) and non-SCD. Methods The expression levels of MCP-1 and CCR-2 in SCD group, coronary atherosclerosis group (non-SCD), control group (normal coronary artery) were detected by immunohistochemistry.
RESULTS:
Positive rates of MCP-1 among the three groups were 78%, 47%, and 0%, respectively, with significant expressing differences between each two groups (P<0.05). Positive rates of CCR-2 among three groups were 72%, 47%, and 0%, respectively, with significant expressing differences between the SCD group and coronary atherosclerosis group as well as between the SCD group and control group (P<0.05), but with no significant expressing difference between coronary atherosclerosis group and control group (P>0.05).
CONCLUSION
Overexpression of MCP-1 and CCR-2 in coronary atherosclerotic plaques is closely correlated with SCD.

10.Life style and periodontitis

Objective To evaluate the correlation between periodontitis and life style. Methods By using randomized cluster sampling method,2990 health check-up receivers were enrolled in this cross sectional survey from September 2007 to September 2008. The participants were requested to fill in a questionnaire to assess community periodontal index and synthesis grades. Multiple linear regression was used for data analysis. Results Gender (P = 0. 000) , age (P = 0. 000 ) , educational level ( P = 0. 001) , smoking (P = 0.011 ) and duration of teeth brush ( P = 0.026 ) were positively correlated with periodontitis. However, periodontitis showed no relationship with alcohol consumption, food intake, daily activity,frequency of teeth brush, periodontal bleeding. Conclusion Periodontitis is a condition with multiple risk factors. Improving life style may help to prevent general population from periodontitis.
