1.MicroRNAs Differential Expression Patterns in the Hypertrophic Left Ventricular Tissue in Spontaneously Hypertensive Rats

Objective To investigate the molecular mechanisms for the development of cardiac hypertrophy in hypertension by scanning the expression profile of microRNA (miRNA) in the left ventricular hypertrophic tissue in spontaneously hypertensive rats(SHR)using miRNA microarray chip technique.Methods 17-week-old male SHR with left ventricle hypertrophy (LVH) (n=10) and 8-week-old male SHR without LVH were employed in this study (n=10).Left ventricular mass index (LVMI),HE staining and measurement of transverse diameter of myocardium (TDM) were used to evaluate LVH.The mRNAs of the two groups were extracted from left ventricle tissue and the differential expression of miRNA was detected by using miRNA microarray chip.Results LVMI and TDM were greater in rats with LVH vs without [LVMI(2.87?0.15)vs(2.17?0.14)mg/g;TDM(15.32?0.61)vs(12.01?0.43)?m,P
2.Design of Integrated Result Query Service System in HIS Based on Secure Data-exchange

Objective To solve the conflict between data security and sharing in hospital so that patients can get their clinical data through result query service system.Methods Advanced secure data-exchange technology was used to realize network isolation.Integrated result query service system adopted the embedded system as hardware and the embedded LINUX operation system as software.Results Through LAN query module,internet query module and the secure data-exchange module,the design of HIS multimedia query system was realized.Conclusion Via the application of HIS integrated query platform,the design of the hospital secure data exchange is proved to be feasible,reliable and practical.
3.The correlation between the hypertrophic ventricular tissue of SHR rats and ventricular arrhythmia occurred

Objective To explore the susceptibility differences of ventricular arrhythmia (VA ) in SHR rats with/without left ventricular hypertrophy(LVH) and the significance .Methods The experiments were performed on isolated hearts of 8‐week(the non‐LVH control group ,n=12) ,16 week‐old SHR(the LVH group ,n=12) and age‐matched Wistar Kyoto rats(Wistar ,the blank control group ,n=12) ,which were perfused in Langendorff mode with oxygenated Krebs‐Henseleit solution followed by a K+‐defi‐cient solution .The epicardial electrocardiogram was continuously monitored during all experiments .HE staining and collagen vol‐ume fraction(CVF)was used to evaluate the condition of LVH .Results Compared with the non‐LVH control group and the blank control group ,the low K+ induced ventricular arrhythmia occurred earlier with increased incidences and duration in the hearts of the LVH Group ,the incidences of ventricular tachycardia(VT) ,transient ventricular fibrillation(TVF) ,sustained ventricular fibrillation (SVF) and CVF were higher in the hearts of the LVH Group(P<0 .05) ,myocardial cell hypertrophy and myocardial cells intersti‐tial increased .Conclusion The ventricular arrhythmia occurred earlier with increased incidences and duration in the LVH tissue of SHR rats ,which implies that LVH is associated with VA .
4.Clinical efficacy analysis and surgical treatment for tibial plateau fracture of Schatzker Ⅴ and Ⅵ

[Objective]To investigate the characteristics,treatment and prognosis of tibial plateau fracture of Schatzker type Ⅴ and Ⅵ and recommend the option of the surgical treatment.[Method]From May 2002 to March 2006,arthroscopically assisted double-plating fixation,locking plate with cannulated screws and external fixation with limited internal fixation were performed in 35 patients with tibial plateau fracture of Schatzker Ⅴ and Ⅵ.[Result]All patients were followed up from 12 to 35 months with an average time of 16.5 months.Bone union was achieved in all cases.According to Rasmussen's functional grading system,the excellent,good,fair and poor cases were 19,10,4 and 2,respectively.[Conclusion]CT 3D reconstruction,evaluation of soft tissues injury,suitable operation timing and approaches,anatomical reduction,bone transplantation,proper fixation and right rehabilitation training are all key factors to improve the therapeutic effects on tibial plateau fracture of Schatzker Ⅴ and Ⅵ.
5.Effect of phacoemulsification on corneal endothelium of cataract with lupus nephritis

To evaluate the influence of phacoemulsification on corneal endothelium of senile cataract with lupus nephritis (LN).
●METHODS:This clinical trial involved 40 cataract patients with lupus nephritis (40 eyes), and 50 cases (50 eyes) without lupus nephritis. All of them underwent phacoemulsification+lOL implantation. The parameters of corneal endothelial cell including central corneal endothelium cell density ( CED ), average area of endothelial cell ( AVE) and coefficient of variation ( CV) were recorded by corneal endothelial microscope pre -operation and at one month after operation. The data were analyzed by SPSS 13. 0 statistical software.
● RESULTS: Both LN group and control group, the morphology of coneal endothelial was statistical significant differences between pre - operation and 1mo postoperation. The CED was lower, AVE and CV were higher ( P < 0. 05, respectively). A significant decrease in CED was seen in the LN group than did in the control group (P < 0. 05). Compared to control group,the post -operative AVE and CV in LN group was significantly increased (P<0. 05).
● CONCLUSlON: The corneal endothelial cell in lupus nephritis patients is more fragile. Safe and reliable operation should be selected for these patients.
6.Magnetic nanoparticles versus a commercial kit for apolipoprotein E gene polymorphism analysis

BACKGROUND:Relative to blood samples, mouth swab samples are more beneficial for apolipoprotein E gene polymorphism analysis among large cohorts. However, agreement has not yet been reached about how to extract genomic DNA form mouth swab samples.
OBJECTIVE:To develop an appropriate method to extract genomic DNA form mouth swab samples, which are suitable for apolipoprotein E gene polymorphism analysis.
METHODS:Fifty mouth swab samples from patients with sporadic Alzheimer’s disease were col ected. Magnetic nanoparticles and PicoDNA trace nucleic acid extraction kit were used to extract genomic DNA form mouth swab samples. And the purity and concentration of the genomic DNA extracted by the two methods were analyzed. Then PCR amplifications and DNA electrophoresis were performed to confirm whether the genomic DNA was able to amplify desired DNA fragments. DNA sequencing was applied to analyze apolipoprotein E gene polymorphisms.
RESULTS AND CONCLUSION:Genomic DNA extracted by the two methods was of high purity. The concentration of genomic DNA extracted by magnetic nanoparticles was higher than by PicoDNA trace nucleic acid extraction kit, and the difference had statistical significance (P<0.05). Al the genomic DNA were able to performed PCR amplifications to obtain desired PCR products, but results of DNA electrophoresis showed that DNA fragments were more clear by nanoparticles method. The results of DNA sequencing were the same by the two methods. The distribution ofε2,ε3,ε4 genotypes of apolipoprotein E gene was 6%, 71%, 23%, respectively. Magnetic nanoparticles were better than PicoDNA trace nucleic acid extraction kit for extracting genomic DNA form mouth swab samples for apolipoprotein E gene polymorphism analysis.
8.The effects of telmisartan treatment on the abdominal fat deposit in metabolic syndrome combined with essential hypertension

Objective To observe the effects of telmisartan treatment on the abdominal fat deposit in metabolic syndrome (MS) combined with essential hypertension (EH).Methods Sixty patients of MS combined with EH were divided into two groups according to random number table method,with 30 cases in each group.The patients in experimental group were given telmisartan orally 40 mg once a day,and in control group were given valsartan 80 mg once a day,until average ambulatory blood pressure monitoring blood pressure was < 140/90 mmHg (1 mmHg =0.133 kPa) in 24 weeks.The visceral fat area (VFA) and subdermal fat area (SFA) was measured by multislice CT,at the same time the body mass index (BMI),blood pressure,blood glucose,homeostasis model assessment of insulin resistance (HOMA-IR) and glyeosylated hemoglobin (HbA1c) of the subjects was measured.Results The blood pressure,BMI,blood glucose,HbA1c,and blood lipids between two groups before treatment had no statistical significance (P >0.05).VFA in experimental group after treatment was lower than that before treatment [(127.8 ± 16.6) cm2 vs.(150.5 ± 15.4) cm2] (P < 0.05).But there was no statistical significance of VAF in control group before and after treatment (P >0.05).The SFA in experimental group and control group had no statistical significance before and after the treatment (P > 0.05).The HOMA-IR in experimental group after treatment was reduced compared with that before treatment (1.9 ± 0.3 vs.4.2 ± 0.9),and adiponectin was increased [(5.77 ±0.71) mg/L vs.(3.16 ±0.72) mg/L] (P <0.05).Conclusion Compared with valsartan,telmisartan can significantly reduce the visceral fat deposition in MS combined with EH patients and improve insulin resistance.
9.ALT abnormality among workers from food industry and related people from public places and influencing factors

Objective To explore the influencing factors of ALT abnormality among workers from food industry and related people from public places in Shenzhen and influencing factors.Methods 2,411 workers from food industry and people from public places who had physical examinations in our department from May to October in 2013 were involved in the investigation.ALT abnormality rate and the influencing factors were analyzed.Results The incidence of ALT abnormalities among workers from food industry and related people from public places in Shenzhen was 9.37%.The ALT abnormalities were correlated with censue register,gender,age,marital status,work duration in Shenzhen,education level and monthly income respectively(all P<0.05). Conclusion The ALT abnormality has a higher rate among people from cities,of male gender and old age,with longer work duration in Shenzhen and relatively high level of education and higher monthly income.Therefore,for the population,the health education should be strengthened and the healthy lifestyles should be advocated to effectively reduce the ALT abnormality rate.
10.Drug resistance of Helicobacter pylori in nodular gastritis in children

Objective To analyze the drug resistance of Helicobacter pylori (H.pylori) in nodular gastritis in children and to further explore the rational use of antibiotics for drug-resistant H.pylori strains. Methods A total of 473 children with upper gastrointestinal symptoms undergoing gastroscopy from January 2013 to June 2014 in our hospital were enrolled. Two pieces of gastric antral mucosa in children with nodular gastritis were collected for H.pylori rapid urease test and H.pylori culture. The resistance of H.pylori to amoxicillin, clarithromycin, metronidazole, moxilfoxacin and levolfoxacin was detected by agar dilution method and E-test. Results In 473 cases of nodular gastritis, 258 cases were H.pylori culture-positive. The resistance rate of H. pylori isolates to amoxicillin, clarithromycin, metronidazole, moxilfoxacin, levolfoxacin was 6.2%, 34.9%, 49.2%, 8.9%and 5.0%, respectively. Dual resistance to clarithromycin and metronidazole was 23.3%. Moreover, 405 cases had a family history of H.pylori infection. Conclusions Nodular gastritis is a special sign of H.pylori infection in children. H.pylori infection has obvious familial aggregation. The low resistance rate of H.pylori to amoxicillin in children with nodular gastritis indicates that amoxicillin can be used as the main drug for eradication of H.pylori. Meanwhile, clarithromycin should be applied according to the drug sensitive test due to high resistance rate of H.pylori to clarithromycin.
