Objective To investigate whether the splenic metastasis of B16 melanoma cells in C57BL/6 mice occur through the blood. Methods Cultured B16 melanoma cells were collected, adjusted into 1?10~7/ml and inoculated into the left armpit of C57BL/6 mice,200 ?l each. After 15 days, the mice were sac- rificed and metastasis was examined in various tissues and organs by HE stain.1.5 % Evan Blue(EB),2 ml/kg, was administered to normal C57BL/6 mice through the tail vein, and the region stained blue was compared with that of metastasis. Results All of 12 mice inoculated were burdened with melanoma.Splenic metastasis occurred in 9 of them, among which 3 were accompanied by lung metastasis, and all of 9 mice showed differ- ent degree of local lymph node metastasis. The dorsal 1/4 part of the positive spleen was stained black by the metastatic tumor cells.Under the microscope, distribution of the metastatic tumor cells in the spleen was scat- tered, their growth inhibited, and maturation accelerated. The region stained blue by EB also located at the dorsal 1/4 part of the spleen, which was almost the same as the positive part of metastasis. Conclusions Metastasis of B16 melanoma cells in C57BL/6 miee occurs earlier in the spleen than in other organs, and this metastasis is achieved most possibly through the blood.

Corresponding author: SONG Guang yao Human umbilical venous endothelial cells (HUVEC) were incubated with free fatty acids (FFAs) of various degrees of saturation. C18:1 and C18:2 inhibited the synthesis of nitric oxide (NO) and endothelin (ET) in HUVEC, and ET synthesis was inhibited much more than NO synthesis, and C18:0 increased ET synthesis in HUVEC, suggesting that the effects of FFAs on HUVEC were related to their degree of saturation.

OBJECTIVE:To systematically review the effectiveness and safety of methotrexate(MTX)in the treatment of rheuma-toid arthritis, and provide evidence-based reference for clinical treatment. METHODS:Retrieved from Medline,EMBase,Cochrane Library,PubMed,CJFD,CBM,VIP and Wanfang Database, randomized controlled trials(RCT)about MTX in the treatment of rheu-matoid arthritis were collected,Meta-analysis was performed after data extraction and quality evaluation. RESULTS & CONCLU-SIONS:33 effectiveness evaluation were included, involving 8 253 patients, and 53 safety evaluation were included, involving 4 803 patients. Results of analysis show, the efficacy of MTX was not higher than leflunomide in the treatment of rheumatoid arthritis,superi-or to cyclosporine A,and similar to sulfasalazine. In addition,MTX shows similar efficacy with etanercept(7.5-20 mg per week),goli-mumab,adalimumab or rituximab, but less effective than trastuzumab. The incidence of adverse reactions of MTX is high, but mainly mild or moderate, and the most common adverse reactions are gastrointestinal symptoms. Oral administration of MTX is more secure than intramuscular injection and subcutaneous injection.

AIMTo detect the expression of a novel protein, hemangiopoietin (HAPO), in the human fetal liver at the protein level.
METHODSMonoclonal antibodies (moAbs) against HAPO were produced by traditional hybridoma technique. Their affinities were calculated from the results of non-competitive ELISA and the antibodies were purified by protein G affinity chromatography. Expression of HAPO in the liver was detected by SDS-PAGE and Western blot.
RESULTSFive strains of moAbs were screened out in total, among which three were IgG1 and the other two were IgM. Their light trains were all belonged to kappa. The relative affinities of the three IgG1 form moAbs were 3.06 x 10(9) mol/L, 6.07 x 10(8) mol/L and 1.71 x 10(10) mol/L respectively. After purification, the purity of the moAbs could reach more than 99%. HAPO expression was detected at the protein level in the human fetal liver, and the apparent molecular weight of the nature HAPO was very close to but a little higher than our recombinant one.
CONCLUSIONHAPO was expressed in the human fetal liver at the protein level.

To evaluate changes in quality- of- life of adults with concomitant exotropia before and after surgery.
●METHODS:A retrospective cohort method was used in this research. Sixty - five patients with concomitant exotropia ( ranged from 18 - 30 years) were enrolled. Quality of life was studied with 2 different questionnaires [the Adult Strabismus - 20 ( AS - 20) and the MOS 36 -item Short - Form health survey ( SF - 36 )], which patients completed preoperatively and at 3mo postoperatively.
●RESULTS: With the AS - 20, 3mo after surgery, the mean psychosocial and visual function scores of AS- 20 improved significantly (P<0. 01). Similarly, with the SF-36, the mean score in 7 of 8 areas improved significantly ( P < 0. 01 ), including physiological function, role limitations due to physiological health, general health, vitality, social function, role limitations due to emotional problems, mental health and the overall mean score. However, with respect to bodily pain, no significant improvement was found after surgery(P>0. 05).
● CONCLUSlON: Surgical treatment of concomitant exotropia in adults gives a highly significant improvement in quality - of - life scores. We should pay more attention to the impact of strabismus on quality of life clinically to improve the outcome of the surgery.

Background Corneal chemical burn is one of blinding eye diseases.Previous therapies for corneal chemical burn is limited to certain extent. However, transplantation of adipose-derived mesenchymal stem cells (ADSCs) for corneal diseases is drawing more and more attention. Objective This study was to observe the effect of rabbit ADSCs transplantation for ocular alkali burns and explore its mechanism. Methods Rabbit corneal stromal cells (CSCs) were isolated and cultured by suspended matrix method, and rabbit ADSCs were obtained and digested from inguinal fat tissue with enzyme digestion method (0.25% trypsin) and identified by flow cytometry.CSCs cocultured with ADSCs,and CSCs-induced ADSCs were identified by double-label of with immunofluorescence and reverse transcription PCR (RT-PCR).Then induced or uninduced ADSCs were inoculated on amniotic membrane to prepared ADSCs-amnion patch.Corneal alkali burn models were established in the right eyes of 60 New Zealand rabbits by placing a filter paper with 1% NaOH solution at the central cornea for 50 seconds. The models were randomized into the induced ADSCs+ amnion implanted group, the uninduced ADSCs + amnion implanting group, amnion implanted group and model group.Corneal opacification and neovascular area were examined and corneal inflammation was graded by slit lamp microscope 1 week,2 weeks and 1 month after surgery. The contents of CD45,interferon-γ (IFN-γ) and interleukin-10 (IL-10) in corneal homogenate as well as vascular endothelial growth factor (VEGF) in aqueous humor were detected by ELISA assay. The use and care of experiment animals followed the Statement of ARVO.Results ADSCs showed the positive responses for CD105,CD29,CD44 with the positive rate 90.23 %,88.56% and 98.88%,respectively.CSCs was positively reactive for vimentin. The double-label staining was positive after coculture of CSCs with ADSCs.Hematoxylin-eosin stain exhibited that ADSCs grew well on the amnion.Corneal porcelain opacity and a lot of new blood vessels were seen in the model group, and corneal was clear in the induced ADSCs+ amnion implanted group 1 month after surgery. The inflammatory scores were 1.65 ±0.18,2.05 ± 0.17,2.68±0.25,2.90 ±0.18,and the areas of neovasculization were (10.59 ± 1.78),(22.58 ± 1.63),(37.98 ± 1.90),(45.37±1.65)mm2 respectively in the induced ADSCs+ amnion implanted group, uninduced ADSCs+ amnion implanted group, amnion implanted group and the model group. The inflammatory scores of 1 week,2 weeks,1 month after operation among the four groups had statistically significant differences (F =280.826,330.172,465.707,all at P =0.000),and the areas of neovasculization of 1 week,2 weeks,1 month after operation among the four groups had statistically significant differences (F=60.020,670.811,1 510.231,all at P =0.000),the inflammatory scores in the induced ADSCs + amnion implanted group were remarkably lower than those of the other groups, the areas of neovasculization in the induced ADSCs+ amnion inplanted group were smaller than those of the other groups (all at P<0.01).In 1 month after surgery, the contents of CD45,IL-10,IFN-γ in cornea and VEGF in aqueous humor were statistically different among the groups(F =916.545,1 739.358,462.134,129.126,all at P =0.000).Compared with the uninduced ADSCs+ amnion implanted group, amnion implanted group and the model group,CD45 and IFN-γ contents were declined, and IL-10 content was elavated in the induced ADSCs+ amnion implanted group (all at P< 0.01).In addition,VEGF contents in aqueous humor were significantly lower than those in the other groups (all at P<0.01).Conclusions Rabbit CSCs-induced ADSCs amnion patch transplantation is effective for the reconstruction of ocular surface after alkali damage probably by differentiation of ADSCs into epithelial-like cell after CSCs induced. Moreover, amnion can alleviate immuno-inflammatory response and suppress neovascularization.

The SRY gene from buffalo (Bubalus bubalis) genome was amplified by the polymerase chain reaction ( PCR) with primers based on the sequence of Hostein SRY gene. The amplified fragment was 2005 bp include 5UTR ( 1 - 504bp) and 3'UTR(1196 - 2005bp). And the amplified fragment was cloned and sequenced. Sequence analysis showed that the coding region of SRY gene (505 - 1195bp) from buffalo was highly homologous with those of other bovine counterpart genes (96% homology) , especially in the HMG box region (99%homology). It was found that there were only signal on male buffalo genome on Southern blot, which indicate SRY gene are highly conservative on evolves.
