1.Expression of serum AQP4-Ab and the significance in patients with optic neuritis

AIM:To investigate the serum antibody of aquaporin 4 ( AQP4 - Ab ) in positive expression rate and the significance in patients with neuritis.
●METHODS: A total of 98 cases ( 128 eyes ) of patients with optic neuritis were studied to detect the patient′s serum AQP4-Ab positive rate of antinuclear antibodies ( ANAs) from Jan. 2012 to Dec. 2015 in ophthalmology center of our hospital. According to the expression of AQP4 - Ab group, the best corrected visual acuity between the two groups, peripapillary nerve fiber layer thickness (pRNFL), the volume of the macula, macular RNFL ( mRNFL ) , macular core layer volume ( mlNL ) measurement were compared.
●RESULTS:Ninety-eight patients (128 eyes) with optic neuritis cases diagnosed through examination revealed AQP4-Ab positive in 22 patients ( 22%) , negative in 76 patients ( 78%) , ANAs positive in 21 patients ( 21%) , negative 77 patients ( 79%) . Optic neuritis patients with serum AQP4 - Ab positive rate and ANAs positive significant correlation ( r = 0. 707, P < 0. 05 ). After examination revealed AQP4-Ab patients and negative-positive patients with best corrected visual acuity difference was not statistically significance (P>0. 05). After inspection found pRNFL, macular volume measured value AQP4 - Ab positive patients were significantly less than the negative patients, the differences were statistically significant (P<0. 05). AQP4-Ab positive patients and negative patients the mRNFL, mlNL measured values were not significantly different (P>0. 05).
●CONCLUSION:AQP4-Ab and ANAs expression in optic neuritis patients is a significant correlation. AQP4-Ab positive patients with optic neuritis pRNFL thinning of macular volume are decreased compared with negative patients.
2.Mutation analysis of NR5A1 gene in children with idiopathic hypogonadotropic hypogonadism

Objective To study the NR5A1 gene mutation in patients with idiopathic hypogonadotropic hypogonadism(IHH), and to find the new mutation point.Methods Sixty-one IHH patients and 100 normal control subjects were collected and genomic DNA was extracted from blood samples.These patients were with normal karyotype and no abnormality was discovered in magnetic resonance imaging (MRI) scan of the pituitary.Other endocrine diseases were also excluded.The 2-7 exons and splice-sites of NR5A1 gene were amplified with polymerase chain reaction.DNA of the coding sequence and splice-sites of NR5A 1 were sequenced by double deoxidizing terminal end sequencing method in 61 IHH and 100 normal control subjects.The results of sequencing were compared with their corresponding sequence data.61 IHH kindreds were investigated and the clinical data of these patients were collected.Finally, the phenotype and genotype positive cases were analyzed.Results Six patients carried NR5A1 gene mutational sites in 61 cases of IHH.Analysis of sequencing results from 100 age and ethnicity matched control subjects did not show any of these novel changes.Conclusions One mutation in NR5A1 gene may affect protein structure and function, which should be considered in male IHH patients with normal karyotype and without insufficiency of adrenal function.
3.Pharmacokinetic and distribution of arctiin in rats

OBJECTIVE To study the pharmacokinetic and distribution of arctiin in rats. METHODS Each rat was given a single dose at random by oral administration. The arctiin in serum and organs were determined by use of RP-HPLC. All pharmacokinetic parameters were calculated with a 3P87 program. RESULTS After oral administration of arctiin at the dose of 300mg·kg-1, Arctiin plasma C-T curve conform to open two-compartment model. The Pharmacokinetic parameters were as follow: A=(37.374 5±8.964 7)μg·mL-1;B=(6.210 6±1.489 3)μg·mL-1;α=(0.004 3±0.000 9)min-1;β=(0.000 4±0.000 2)min-1;Kα=(0.420 2±0.167 5)min -1;t1/2α=(115.192 6±14.382 4)min ;t1/2β=(1 485.578 1±161.173 3)min;K10 =(0.001 0±0.000 4)min -1;K21=(0.001 4±0.000 6)min -1 ;K12=(0.002 3±0.001 3)min -1 ;Cmax=(41.786 3±7.521 7)μg·mL-1 ;Tmax=(9.891 9±4.341 4)min;AUC=(22 503.272 7±4 120.182 8)μg·min·mL-1. Liver had the highest concentration of arctiin after oral administration. CONCLUSION RP-HPLC method is rapid, sensitive and specific for the research of arctiin pharmacokinetic and its distribution in rats. Arctiin is distributed and eliminated quickly in rats.
4.Action of sulfonylurea on mRNA levels of components of ATP-sensitive potassium channel(SUR1,SUR2 and Kir6.2)in rat brain

The effects of streptozotocin-induced diabetes or long-term glyburide administration on mRNA levels of components of ATP-sensitive potassium channel(SUR1,SUR2,Kir6.2)in rat brain were observed. Streptozotocin-induced diabetes itself did not affect the mRNA levels of SUR1,SUR2,and Kir6.2 in the brain, and glyburide-treatment increased the Kir6.2 mRNA level in brain by 23% in non-diabetic rats than those in normal control but did not change SUR1 and SUR2 levels.The effects of glyburide on SUR1,SUR2 and Kit6.2 mRNA levels did not manifest in brain of diabetic rats.
5.Jugular foramen schwannomas: a review of 17 cases.

OBJECTIVETo analysis the clinical presentation, radiological findings, surgical techniques and outcomes of jugular foramen (JF) schwannomas.
METHODSWe reviewed our 10-year experience in the surgical treatment of 17 patients suffered from JF schwannomas in Hua Shan Hospital, Shanghai. A total of 8 males and 9 females with a mean age of 42 years underwent surgical procedures. A relative long period of 53-month symptomatic history was shown before surgery. The main clinical presentation are vertigo and hearing difficulty in 10 cases, atrophy of unilateral muscles of tongue in 9 cases, involvement of lower cranial nerve in 8 cases. The classification of tumors was type A (at cerebellopontine angle with minimal enlargement of the JF) in five cases, type B (JF with intracranial extension) in 3 cases, type C (extracranial tumors with JF extension) in 2 cases and type D (dumbbell-shaped with both intra-and extracranial components) in 7 cases.
RESULTSFar lateral approaches were used in 10 cases, retrosigmoid suboccipital approaches were used in 5 cases. Submandibular approaches were selected in other 2 cases. Gross total removal was achieved in 12 cases, and subtotal removal in 5 cases. Follow-up revealed marked improvement from preoperative symptoms in 9 cases and no additional deficits in 3 cases. 5 cases suffered from additional neurological deficits. There were two cases of temporary hoarseness and gradually improved within follow-up. Two patients suffered from swallowing problems as a new deficit. One patient had facial palsy.
CONCLUSIONSJF schwannomas can be surgically treated with relative good outcomes. Surgical approaches should be tailored according to the tumor extension.

6.Expression of Intercellular Adhesion Molecule-1 in Rat Heart with Ischemia/Reperfusion Injury and Its Association with Ischemia/Reperfusion Injury

To study the expression of intercellular adhesion molecule-1 in rat heart with ischemia/reperfusion injury and its association with ischemia/reperfusion injury. Methods:Fourty-two male Sprague-Dawley rats were subjected to coronary artery ligation as ischemia/reperfusion heart model and were randomly divided into seven groups of six rats each. The ischemia time was 30 min.The rat hearts were excised at 0,30,60,360,540,720 min after reperfusion. The ischemic and reperfused myocardium was examined immunohistochemically to localize ICAM-1. Malondialdehyde(MDA) and nitric oxide (NO) contents in the myocardium were also determined by thiobarbituric acid and Griss method,respectively. Results: ①At the time of 540 min after reperfusion,expression of ICAM-l in rat hearts was upragulated. ②The MDA values were increased during reperfusion (P<0. 05) and reached the peak 1 hour after reperfusion. ③The NO values get lower and lower during reperfusion. Conclusion :Expession of ICAM-1 in rat hearts subjected to ischemia followed by reperfusion was increased in the late-stage of reperfusion,which might be associated with delayed reperfusion injury.
7.Effect of arctiin on hemorheology of experimental rats with blood stasis ayndrone

OBJECTIVE To study influence of arctiin from seeds of Arctium lappa on hemorheology of experimental rats with the blood stasis syndrone. METHODS The blood hemorheology parameters, Fib, aPTT and PT of experimental rats with the blood stasis syndrone were evaluated using semi-automatic biochemical analysis. RESULTS Arctiin obviously decreased their high shear, middle shear, low shear, the blood viscosity, red blood cell aggregation index, red blood cell rigidity index and reductive viscosity. It also significantly prolonged the time of aPTT and PT and lowed the Fib concentration. CONCLUSION Arctiin apparently ameliorated the blood rheology abnormality and enhanced anti-coagulation effect on experimental rats with the blood stasis.
8.Auricular reconstruction for concha-type microtia.

OBJECTIVETo investigate the method of auricular reconstruction for concha-type microtia.
METHODSTwo-staged auricular reconstruction was applied in 13 cases (14 ears) with concha-type microtia. The cartilage auricular framework was fabricated and implanted in the first stage, followed by ear elevation and cranio-auricle angle formation at the second stage.
RESULTSThe patients were followed up for 2 months to 2 years with satisfactory aesthetic result. The reconstructed ears had a good appearance and position, and were symmetric to the healthy ears.
CONCLUSIONSThe two-staged auricular reconstruction with autologous cartilage framework is ideal for concha-type microtia.

9.Dyuamical studies on metabolic chemistry of lignans from seeds of Arctium lappa.

OBJECTIVETo study the metabolic chemistry and pharmaco-dynamics characters of ligan from seeds of Arctium lappa.
METHODHPLC method was used in the study. The analysis was carried out on C18 column. The mobile phase was CH3CN-0.05% H3PO4 (36:64) with flow-rate at 0.6 mL x min(-1) and wave-length of 210 nm. The column temperature was kept at 25 degrees C.
RESULTThe results indicated that the ligan was detected in plasma and the main organs 5 min after po. The main metabolic production in plasma was arctigenin. In addition, arctigenin and an unknown product were found in metabolic production in the organs.
CONCLUSIONThe method was stable,simple and reproducible. It can be used to determine the metabolic product of the ligan. The metabolic chemistry of ligan in plasma was obviously different from that in the main organs.

10.Application Fourier transform near infrared spectrometer in rapid estimation of soluble solids content of intact citrus fruits.

Nondestructive method of measuring soluble solids content (SSC) of citrus fruits was developed using Fourier transform near infrared reflectance (FT-NIR) measurements collected through optics fiber. The models describing the relationship between SSC and the NIR spectra of citrus fruits were developed and evaluated. Different spectra correction algorithms (standard normal variate (SNV), multiplicative signal correction (MSC)) were used in this study. The relationship between laboratory SSC and FT-NIR spectra of citrus fruits was analyzed via principle component regression (PCR) and partial least squares (PLS) regression method. Models based on the different spectral ranges were compared in this research. The first derivative and second derivative were applied to all spectra to reduce the effects of sample size, light scattering, instrument noise, etc. Different baseline correction methods were applied to improve the spectral data quality. Among them the second derivative method after baseline correction produced best noise removing capability and yielded optimal calibration models. A total of 170 NIR spectra were acquired; 135 NIR spectra were used to develop the calibration model; the remaining spectra were used to validate the model. The developed PLS model describing the relationship between SSC and NIR reflectance spectra could predict SSC of 35 samples with correlation coefficient of 0.995 and RMSEP of 0.79 degrees Brix.
