OBJECTIVETo elucidate the necessary and research of accelerating basic research of Chinese standard pieces as standard materials.
METHODAccording to over 10 years accumulated experience and be keenly aware of the author, the evaluation method of standardized processing technology and Chinese pieces quality, aimed at consummated the standard material of the quality evaluation of Chinese herbal pieces at the current situation, and inaccordance with the need of improving quality standard system of Chinese herbal pieces, illustrate the necessity of accelerating basic research of Chinese standard pieces as standard materials; from the technical specification for collecting and processing of raw materials, and the technical specification, homogenized sample, packaging, storage and etc., for processing of candidate standard pieces, determine the methods and steps of technical specifications for standard pieces as the standard substance, determine the methods and steps of technical specifications for standard pieces as the standard substance.
RESULT AND CONCLUSIONTo speed up the basic research of standard of Chinese medicine pieces as of standard material is very necessary. The research objective is to specificate the processing technical for a number of standard pieces, to identify technical specifications and to ascertain the guiding principle and technical specification of decoction pieces as standard substance. This research will provide basic scientific data relevant national departments to apply for the accreditation of the standard substance.

3.Characteristics of research achievement and its transfer strategy in hospital system

Objective To propose new policies and strategies that will optimize the transformation of medical research achievements at hospital level.Methods In this paper,we systematically analyzed the characteristics and influencing factors of the transformation of medical research achievements based on the status quo in our hospital.Results In our hospital as an example,major problems in the transformation of medical research achievements identified as low investment,low conversion rate,complex procedures,multi-links existed and more multi disciplinary cooperation needed.Conclusions The hospital can take some measurements to optimize the transformation of medical research achievements,such as more stream lined management of scientific research project,establishing and improve the platform for scientific research transfer,increasing the funding,strengthening cooperation among different disciplinaries,building a professional team to promote the transfer and so on.
4.Vegetative state treated with acupoint injection combined with plum-blossom needle in children: a randomized controlled trial.

OBJECTIVETo observe the clinical efficacy difference on vegetative state in children between acupoint injection combined with plum-blossom needle and western medication based on basic treatment.
METHODSForty-eight children of vegetative state were randomized into an observation group and a control group, 24 cases in each one. On the basis of the treatment of transcranial magnetic stimulation apparatus, balancing treatment apparatus and massage, the acupoint injection and tapping method with plum-blossom needle were adopted in the observation group, in which Xingnaojing injection, mouse nerve growth factor (mNGF) injection, monosialotetrahexosylganglioside sodium injection (MSI), compound Danshen injection were divided in 6 pairs and were injected respectively in Baihui (GV 20), Yongquan (KI 1), Fengfu (GV 16), Yamen (GV 15) and the others, 0.5 mL in each acupoint, once a day for continuous 10 days. Additionally, the tapping method with plum-blossom needle was used on the Governor Vessel and Jiaji (EX-B 2) on the back. In the control group, the intravenous infusion was adopted with citicoline sodium injection, mannitol injection and dexamethasone injection. The treatment was given once a day, 20 days of treatment made one session and totally 3 sessions were required in the two groups. The clinical efficacy, the vegetative state score and the mean curing time were observed after 20 days, 40 days and 60 days of treatment between the two groups.
RESULTSThe effective rates were 58.3% (14/24), 70.8% (17/24) and 79.2% (19/24) in 20 days, 40 days and 60 days of treatment in the observation group and 20.8% (5/24), 45.8% (11/24) and 58.3% (14/24) in the control group respectively. The efficacy in the observation group was superior to those in the control group (P < 0.01, P < 0.05). The vegetative state score was improved apparently after 20 days, 40 days and 60 days of treatment as compared with those before treatment separately (all P < 0.05). It was improved obviously at the each time point after treatment in the observation group as compared with that in the control group (3.34 +/- 2.41 vs 2.64 +/- 11.56, 6.20 +/- 1.46 vs 4.34 +/- 1.64, 11.26 +/- 2.63 vs 8.75 +/- 2.18, all P < 0.05). The mean curing time was (45.67 +/- 16.24) days in the observation group, which was shorter apparently than that of (55.34 +/- 4.57) days in the control group (P < 0.05).
CONCLUSIONBased on basic treatment acupoint injection combined with tapping method of plum-blossom needle achieve the reliable efficacy on vegetative state in children.

6.Study of gonadal hormone drugs in blocking filovirus entry of cells in vitro.

This study was designed to discover filovirus entry inhibitors in a drug library of commercial medicines. One thousand and six hundred drugs were screened using the ZEBOV-GP/HIV model, a pseudovirus formed by an HIV-core packed with the Zaire Ebola virus glycoprotein. We identified 12 gonadal hormone drugs with inhibitory activities in ZEBOV-GP/HIV entry at final concentration of 10 μmol x L(-1). Among them, three drugs exhibited strong activities with IC50 < 1 μmol x L(-1), such as toremifene citrate (IC50: 0.19 ± 0.02 μmol x L(-1)), tamoxifen citrate (IC50: 0.32 ± 0.01 μmol x L(-1)) and clomiphene citrate (IC50: 0.53 ± 0.02 μmol x L(-1)); seven drugs had moderate activities with IC50 between 1 and 10 μmol x L(-1), such as estradiol benzoate (IC50: 1.83 ± 5.69 μmol x L(-1)), raloxifene hydrochloride (IC50: 3.48 ± 0.07 μmol x L(-1)), equilin (IC50: 4.00 ± 9.94 μmol x L(-1)), estradiol (IC50: 5.26 ± 9.92 μmol x L(-1)), quinestrol (IC50: 6.36?5.37 gmol-L1), estrone (IC50: 6.87 ± 0.03 μmol-L1) and finasteride (IC50: 9.94 ± 0.45 μmol x L(-1)); two drugs, hexestrol (IC50: 14.20 ± 0.55 μmol x L(-1)) and chlormadinone acetate (IC50: 24.60 ± 0.36 μmol x L(-1)), had weak activities against ZEBOV. Further, toremifene citrate, tamoxifen citrate, clomiphene citrate, raloxifene hydrochloride and quinestrol could block both pseudovirus type Sudan ebola virus (SEBOV-GP/HIV) and Marburg virus (MARV-GP/HIV) entries.

7.Quantification of peramivir in rat plasma by liquid chromatography/tandem mass spectrometry employing precolumn derivatization and its application in pharmacokinetics

Objective:To develop a liquid chromatography/tandem mass spectrometry employing precolumn derivatization method for determination of peramivir in rat plasma.Methods: The sample preparation consisted of a protein precipitation extraction followed by derivatization with hydrochloric acid (10 mol/L) methanol (10∶90, v/v) and determined by liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). The positive ion SRM mode was performed and the precursor-to-the-product ion transitions of m/z 343→284 and 299→152 were used to measure the derivative of peramivir and Ro 64-0802 (I.S.). The chromatographic separation was achieved using a Zorbax RX-C8 (2.1 mm × 150 mm × 5 μm) analytical column with an isocratic mobile phase composed of acetonitrile-water-formic acid (30∶70∶0.1, v/v/v, 0.2 ml/min). Results:The method was linear over a concentration range of 10 to 10 000 ng/ml. The LLOQ was 10 ng/ml. The average inter-day/intra-day precision values ranged from 5.0% to 7.1%, respectively, while the accuracy values ranged from 89.9% to 106.1%.Conclusion: In this method, retention time is greatly improved, the matrix effects are decreased by chemical derivatization. This method has been successfully applied to the preclinical and clinical research of peramivir.
8.The significance of standardized fetal ultrasonographic screening in the first trimester

Objective To evaluate the significance of standardized ultrasound examination of fetal structures in the ifrst trimester (11-13+6 weeks). Methods The ultrasound data of 29 858 fetuses who underwent the ifrst trimester screening were analyzed retrospectively in Beijing Obstetrics and Gynecology Hospital of Capital Medical University from January 2010 to December 2012 and followed up the cases with structural abnormalities and increased nuchal translucency (NT). Results Among 29 858 fetuses, 328 structural defects were detected in 284 fetuses (0.95%) by ultrasound in the first trimester, including fetal hydrops (124 cases), choroid plexus cysts (36 cases), exencephaly (32 cases), gastroschisis (24 cases), exomphalos (22 cases), cardiac defects (19 cases), megacystis (14 cases), spine abnormalities (10 cases), meningoceles/encephaloceles (9 cases), alobar holoprosencephaly (8 cases), hydrocephaly (7 cases), abnormalities of extremities (7 cases), acrania (6 cases), amniotic band syndrome (4 cases), abdominal cyst (3 cases) and conjoined twins (3 cases). Pregnancy was terminated in all cases (115) with exencephaly, gastroschisis, exomphalos, meningoceles/encephaloceles, alobar holoprosencephaly, hydrocephaly,abnormalities of extremities, acrania, amniotic band syndrome and conjoined twins, and the defects were verified by the gross appearance of fetuses. Two cases of megacystis were turned out to be normal during follow-up while 1 case was proved to be aneuploid and 1 other case progressed to multiple abnormalities during the second trimester. Most choroid plexus cysts, except 1 case of aneuploid, were normal during the second and the third trimester follow-ups by ultrasound. Thirteen cases of fetal hydrops were found to be aneuploids and 4 hydrops cases were proved to be complicated with other structural defects during the second and the third trimester. NT was increased in 422 cases (1.41%), among which 122 cases (28.91%) were complicated with structural defects and/or fetal hydrops. Ninty-nine cases with increased NT underwent chromosomal examination and 38 cases were found to be aneuploides. During the follow-up of 298 cases with increased NT who continued pregnancy, 21 structural abnormalities were found, including cardiac defects (14 cases), neural abnormalities (4 cases) and diaphragmatic hernia (3 cases). Conclusion The standardized fetal ultrasound in the first trimester is a effective tool for the screening of aneupolides and major structural abnormalities.
9.Research on correlation between autophagy related gene frequency distribution characteristics and pulmonary tuberculosis

Objective To investigate the correlation between autophagy related gene(ATG) locus polymorphism and the pulmonary tuberculosis(PTB) susceptibility according to the frequency distribution characteristics of autophagy related gene in the patients with PTB.Methods Eighteen single nucleotide polymorphism (SNP) loci of autophagy related genes in 202 patients with PTB as the case group and 222 healthy controls were genotyped by SequenomMassArray mass spectrometry array technology.The correlation between the each locus genotype and the PTB susceptibility was statistically analyzed.Results In the PTB patients group and healthy control group,after correcting the factors of sex and age,the binary Logistic regression analysis found that the frequency distribution of genotype and allele had statistical difference between rs5973822 and rs807185 sites in ATG4A gene (P<0.05).The stratified analysis by body mass index (BMI) found that this difference was more significant in the high BMI population,moreover the distribution frequency in the patient group was lower than that in the control group,and the other 16 SNP loci had no statistical difference.Conclusion ATG4A gene rs5973822 and rs807185 loci polymorphism may be negatively correlated with PTB susceptibility,moreover which is more significant in the high BMI group.
10.The CT and MRI advantage manifestation of primary central nervous system lymphoma of immunocompetent patients and immunodeficiency patients

Objective To analyze the CT and MRI features of primary central nervous system lymphoma ( PCNSL) and improve the accuracy of MRI diagnosis.Methods MRI features of 13 cases with PCNSL confirmed by pathology were retrospectively analyzed.All the cases were accepted MRI plain scanning and enhanced scanning. Tumor location, tumor size, signal intensity, enhancement characteristics, age distribution, cystic changes and the presence of calcification and intratumoral hemorrhage were assessed.Results Single lesion was found in 5 cases,and multiple lesions were found in 8 cases.Supra-tentorial lesions were found in 11 cases,the lesions involved both supra and infra tentorial were found in two cases.18 lesions located in deep-brain tissues,such as the thalamus,basal gan-glia,corpus callosum,midbrain and pons.7 lesions located at the surface of the brain,such as the temporal lobe,fron-tal lobe,occipital lobe and parietal lobe.The PCNSLs were found varied shapes in MRI,17 PCNSLs manifested irregu-lar shape,5 PCNSLs manifested round or ovoid shape,3 manifested butterfly shape.The average size of these lesions was 4.8cm.The PCNSLs manifested uniform or low T1 WI signal,uniform or slight high T2 WI signal,slight high or high signal on FLAIR,and slight high signal on DWI.11 cases manifested uniform signal and 2 cases manifested non-uniform signal.Different degrees of edema and mass effect were found combined with the lesions.The CT scan showed 6 cases were slighted high density,4 cases were equal density,3 cases were showed low density.Cystic chan-ges and calcifications were not found.In all cases,significant homogeneous enhancement was shown in 3 cases,moder-ate heterogeneous enhancement was shown in 1 case and mild enhancement was shown in 1 case.In multifocal dis-ease,significant homogeneous enhancement was shown in 14 cases,moderate heterogeneous enhancement was shown in 6 cases and mild enhancement was not shown.Conclusion The CT and MRI features of PCNSL of immunocompe-tent patients are always uniformity, whether T1 WIor T2 WI, while of immunodeficiency patients are complicated, all these features are helpful to make correct diagnosis.
